We examined an aqueous extract of Hibiscus sabdariffa calyces extracts (HSE) by close-arterial injection on micturition thresholds (MTs) and on uterine contractions (rate and amplitude). Five doses of HSE were examined (1, 5, 10, 50, and 100 mg/kg) in 3 groups of rats: controls, after bladder inflammation, and after bilateral hypogastric neurectomy. In some rats, uterine contractions were induced by injection of oxytocin (OT) and the effect of HSE was compared with that of nifedipine. HSE increased MTs in a dose-dependent manner in all groups. Neither atropine (0.1 mg/kg) nor propranolol (0.4 mg/kg) had significant effects on cystometric parameters. They also did not affect the responses obtained by HSE on cystometric parameters. As with bladder response, HSE inhibited both the rate and amplitude of uterine contractions in all groups in a dose-dependent manner. The uterine response to HSE was not affected by administration of either atropine or propranolol. A slight, but significant, reduction of contraction amplitude by HSE in the OT precontracted uteri was only noted at a dose of 500 mg/kg. Nifedipine was more potent than HSE in reducing uterine contraction amplitude. The present work documents inhibition by HSE of the rat bladder and uterine contractility in a dose-dependent manner via a mechanism unrelated to local or remote autonomic receptors or calcium channels. However, further investigation is needed to establish the exact mechanism of action.  相似文献

Circular dichroism (CD) is an important spectroscopic technique for monitoring chirality and biological macromolecule conformation. However, during a CD measurement, absorbance, light scattering/turbidity, and fluorescence can also be detected. The simultaneous measurement of these different spectral features for a single sample is the basis of a multi-mode optical spectrometer. This allows time-efficient gathering of complementary information and provides a scheme to ensure that CD measurements are reliable. Aspects of circular polarization differential light scattering, pH, and temperature variation of a protein (antibody) solution are described. A procedure to help ensure that CD measurements are reliable is described.  相似文献

A new series of benzoxazole derivatives were designed and synthesised to have the main essential pharmacophoric features of VEGFR-2 inhibitors. Cytotoxic activities were evaluated for all derivatives against two human cancer cell lines, MCF-7 and HepG2. Also, the effect of the most cytotoxic derivatives on VEGFR-2 protein concentration was assessed by ELISA. Compounds 14o, 14l, and 14b showed the highest activities with VEGFR-2 protein concentrations of 586.3, 636.2, and 705.7 pg/ml, respectively. Additionally, the anti-angiogenic property of compound 14b against human umbilical vascular endothelial cell (HUVEC) was performed using a wound healing migration assay. Compound 14b reduced proliferation and migratory potential of HUVEC cells. Furthermore, compound 14b was subjected to further biological investigations including cell cycle and apoptosis analyses. Compound 14b arrested the HepG2 cell growth at the Pre-G1 phase and induced apoptosis by 16.52%, compared to 0.67% in the control (HepG2) cells. The effect of apoptosis was buttressed by a 4.8-fold increase in caspase-3 level compared to the control cells. Besides, different in silico docking studies were also performed to get better insights into the possible binding mode of the target compounds with VEGFR-2 active sites.  相似文献

Building from the literature on racialization of Muslims, we argue that there are two unique dimensions to anti-Muslim attitudes: Christian nationalism and nativism. Christian nationalism subscribes to the idea of Christianity as being central to American identity, and nativism provides insight into the monopolies regulating citizenship. We then test this framework’s hypotheses on data drawn from the General Social Survey in 2014 to see if these two dimensions predict support for civil rights infringements of Muslim-Americans compared to other outgroups, including atheists, communists, and racists. The results indicate both Christian nationalism and nativism have significant and negative effects on Muslim civil liberties. We also find some differences between the effects of Christian nationalism and nativism on Muslim civil liberties compared to the other outgroups. We interpret these results as an indication that nativism works as an ordering principle to reconstitute who counts as American and who does not.  相似文献

In this study, the effects of sludge retention time (SRT) on NH(4)-N oxidation and NO(x)-N accumulation in the nitritation reactors were studied. The gradually decrease of SRT also caused long reaction time to achieve 99% NH(4)-N removal. Although the target NH(4)-N removal was achieved in a short reaction time at 40 days of SRT, decreasing of SRT from 40 to 30, 25, 20 days, increase the reaction time from 168 to 240 and 265 h, respectively. The inlet NH(4)-N was almost oxidized and the concentration of NO(2)-N accumulated to a high level of 177 mg/l, while NO(2)-N/(NO(3)-N+NO(2)-N) ratio was about 0.9 at SRT of 40 days. However, the concentration of NO(3)-N increased slightly and NO(2)-N/(NO(x)-N) ratio dropped to 0.8 when the SRT was lower than 40 days. During the operation in a cycle, free ammonia concentration in the SBR was decreased from 2.8 to 0.7 mg/l which is below the lowest concentration causing inhibition of nitrite oxidizing bacteria (NOB). It was assumed that combined dissolved oxygen limitation and NH(3)-N inhibition on NOB caused NO(2)-N accumulation under the experimental conditions.  相似文献

The tomato leafminer, Tuta absoluta, is one of the most destructive pests of tomato worldwide. Management of the pest is mainly based on chemical insecticides. Reliance on insecticides is difficult to sustain because of unintended long‐term adverse effects on the environment and human health. Consequently, there is a need to develop pest management strategies that ensure the production of high‐quality products, while at the same time ensuring environmental sustainability and maximum consumer protection. We evaluated the efficacy the biopesticides: Azadirachtin, Bacillus thuringiensis, Steinernema feltiae and Beauveria bassiana individually and in combination against T. absoluta under laboratory and greenhouse conditions. When second instar larvae were exposed to tomato leaf discs treated with Azadirachtin (3 g / L), B. thuringiensis (0.5 g/L) or B. bassiana (1.5 g/L), 70%–86%, 55%–65%, and 45.5%–58.5% mortality was observed, respectively. Steinernema feltiae (1,000 IJs/ml) was the least effective biopesticide, with 26%–42% mortality. In the greenhouse trials on tomato, pest infestation (mines/10 leaves/plant) and fruits damaged were significantly lower on plants treated with Azadirachtin Bthuringiensis or Azadirachtin  +  Bbassiana compared to plants treated with Azadirachtin, B. thuringiensi, B. bassiana or S. feltiae alone. Azadirachtin Bthuringiensis and Azadirachtin  +  Bbassiana resulted in 90% and 81% reduction in fruits damaged in the summer experiments, respectively, and 96% and 91% in winter. The most severe pest infestation was observed on plants treated with S. feltia. The results indicate that the biopesticides, except S. feltia, can contribute to T. absoluta control in greenhouse tomato crops. In particular, the combined use of Azadirachtin with B. thuringiensis or B. bassiana provided the highest level of control of the pest. The potential for including these biopesticides in an overall sustainable integrated pest management programme for T. absoluta is discussed.  相似文献

The build‐up of phosphorus (P) in soil is a major factor limiting the operating life of a wastewater land treatment system. In this study, effects of long‐term wastewater application on changes in chemical properties, P profiles, and P adsorption capacity were evaluated in soils of the Muskegon wastewater land treatment plant that has been treating wastewater for > 30 years. Results indicate that the major soil properties have been changed. In the 15 cm topsoil, the pH increased from ~ 5–6 in 1973 to ~ 7.4–7.8 in 2003; the soil's total organic carbon (TOC) increased by 10–71 %; and the level of exchangeable Ca in 2003 is 8–9 times higher than that in 1973. The amount of Ca/Mg absorbed in the soil affects the P adsorption capability of the soil; Ca‐ and Mg‐bound P accounts for > 70 % of the total P adsorbed in the soil. The net P accumulated in the Rubicon soil increased from ~ 700 in 1993 to ~ 1345 kg/ha soil in 2001, but the plant available P varied between ~ 100–500 kg/ha soil during the same period, indicating a large amount of the applied P has become the fixed P that is unavailable to plants. P sorption in the soil consists of a fast adsorption and a slow transformation process. The soil's maximum P sorption capacity (Pmax) (based on 1‐day isotherm tests) has been increased by ~ 2–4 times since 1973; the actual Pmax of the Muskegon soils could be much higher than the 1‐day Pmax. Therefore, the life expectancy of the Muskegon system has been extended significantly with the application of wastewater.  相似文献
