Scirpus × mariqueter Tang & F.T. Wang (CYPERACEAE) is a pioneer plant of the tidal zone of estuaries of East Asia. Amplified fragment length polymorphisms (AFLP) were used to infer the relationship between S. × mariqueter and its putative progenitors, Scirpus planiculmis F. Schmidt and Scirpus triqueter L. The genetic distance between S. × mariqueter and S. triqueter was much higher than that between S. × mariqueter and S. planiculmis, and samples of S. × mariqueter and those of S. planiculmis did not form a monophyletic group, while the S. triqueter group formed a distinct monophyletic group. However, we also detected an apparent clustering of individuals of S. × mariqueter and S. planiculmis separately. Our results disprove the hybrid origin of S. × mariqueter and rather suggest that it is more closely related to S. planiculmis.  相似文献

Diploid wild-type cells of Saccharomyces and cells homozygous for one of the single genes rs1, rs2 of the two alleles of the rs3 gene (rad 2 gene) were tested for their response to ionizing irradiation in the presence and absence of oxygen. Both immediate and delayed plating (agar-holding) techniques were used.An identical oxygen enhancement ratio (OER) of 2.5 was observed in rs1rs1, rs2rs2 cells and the wild type. In rs3?1rs3?1 and rs3?2rs3?2 cells the OER was reduced. Their sensitivity to ionizing irradiation in oxic conditions was the same as that of the wild type.rs2rs2 cells showed an increase in relative sensitivity by a factor of 4 in both conditions of irradiation. The relative sensitivity of rs1rs1 cells was increased by a factor of 2.5 in hypoxic conditions and the sensitivity in hypoxia was identical to that of wild type cells in oxic conditions. In contrast to haploid rs1 cells the oxygen effect was fully expressed in diploid rs1rs1 cells.The results on diploid strains are discussed in terms of three independent genetically determined repair systems in Saccharomyces. The expression of these systems is influenced by ploidy.  相似文献

An unreported graft copolymer of N,N-dimethylacrylamide (DMA) with chitosan has been synthesized under nitrogen atmosphere using peroxymonosulphate/mandelic acid redox pair. The effect of reaction conditions on grafting parameters i.e. grafting ratio, efficiency, conversion, add on and homopolymer has been studied. Experimental results show that maximum grafting has been obtained at 1.0 g dm−3 concentration of chitosan, 30 × 10−2 mol dm−3 concentration of N,N-dimethylacrylamide and 7.0 × 10−3 mol dm−3 concentration of hydrogen ion. It has also been observed that grafting ratio, add on, conversion and efficiency increase upto 3.2 × 10−3 mol dm−3 of mandelic acid, 12.0 × 10−3 mol dm−3 of potassium peroxymonosulphate, 150 min of time and 40 °C of temperature. Grafted polymer has been characterized by FTIR spectroscopy and thermogravimetric analysis. Water swelling capacity of chitosan-g-N,N-dimethylacrylamide has been determined. It has been observed that the graft copolymer is thermally more stable than parent backbone.  相似文献

A collection of 88 Escherichia coli and 22 Klebsiella pneumoniae strains previously studied for the carriage of bla CTX-M-15 were screened for the presence of bla ampC using multiplex PCR, and analysed for antibiotic resistance and co-carriage of bla ampC and bla CTX-M-15. Cefoxitin resistance was noticed in total of 41.8% isolates. Concomitant resistance to third and fourth generation cephalosporins, aztreonam, gentamicin, trimethoprim and ciprofloxacin was noted. A total of 20% (22/110) isolates (14.5% E. coli and 5.5% K. pneumoniae) carried plasmidic bla ampC of the CIT-family. Sequencing of the representative isolates revealed CMY-6 AmpC-beta-lactamase. Simultaneous occurrence of bla ampC and bla CTX-M was noticed in 77.3% (17/22) isolates. Plasmid analysis of the selected isolates showed the presence of a single plasmid of ca. ~23 kb. Curing and transconjugation experiments provided evidence of carriage of bla CMY and bla CTX-M on same plasmid. RAPD typing revealed the presence of bla CMY and bla CTX-M in a wider range of strains of E. coli and K. pneumoniae.  相似文献

The zooxanthellate octocoral Sinularia flexibilis is a producer of potential pharmaceutically important metabolites such as antimicrobial and cytotoxic substances. Controlled rearing of the coral, as an alternative for commercial exploitation of these compounds, requires the study of species-specific growth requirements. In this study, phototrophic vs. heterotrophic daily energy demands of S. flexibilis was investigated through light and Artemia feeding trials in the laboratory. Rate of photosynthetic oxygen by zooxanthellae in light (≈200 μmol quanta m−2 s−1) was measured for the coral colonies with and without feeding on Artemia nauplii. Respiratory oxygen was measured in the dark, again with and without Artemia nauplii. Photosynthesis–irradiance curve at light intensities of 0, 50, 100, 200, and 400 μmol quanta m−2 s−1 showed an increase in photosynthetic oxygen production up to a light intensity between 100 and 200 μmol quanta m−2 s−1. The photosynthesis to respiration ratio (P/R > 1) confirmed phototrophy of S. flexibilis. Both fed and non-fed colonies in the light showed high carbon contribution by zooxanthellae to animal (host) respiration values of 111–127%. Carbon energy equivalents allocated to the coral growth averaged 6–12% of total photosynthesis energy (mg C g 1 buoyant weight day 1) and about 0.02% of the total daily radiant energy. “Light utilization efficiency (ε)” estimated an average ε value of 75% 12 h 1 for coral practical energetics. This study shows that besides a fundamental role of phototrophy vs. heterotrophy in daily energy budget of S. flexibilis, an efficient fraction of irradiance is converted to useable energy.  相似文献

Aenictus is the most species-rich genus of army ants in the subfamily Dorylinae and one of the most species rich ant genera in China and the world. In this paper, a new species of the Aenictus ceylonicus group, Aenictus hoelldobleri sp. n., is described and illustrated based on the worker caste. The new species occurs in the subtropical forests of south-east China and is morphologically most similar to Aenictus henanensis Li & Wang, 2005 and Aenictus wudangshanensis Wang, 2006. Aenictus hoelldobleri sp. n. can be distinguished from both species by the shape of the subpetiolar process. The new species also resembles Aenictus lifuiae Terayama 1984 and Aenictus thailandianus Terayama & Kubota, 1993 but clearly differs in various features of the cuticular sculpture. A key to the Chinese species of the Aenictus ceylonicus group based on the worker caste is provided, which may help to reassess and clarify the taxonomic status of the abundant Chinese records of the true Aenictus ceylonicus (Mayr, 1866), a species which almost certainly does not occur in China. Several new locality records are given, among them the first record of Aenictus watanasiti Jaitrong & Yamane, 2013 from China.  相似文献

The regulation of NO3 assimilation by xylem flux of NO3 was studied in illuminated excised leaves of soybean (Glycine max L. Merr. cv Kingsoy). The supply of exogenous NO3 at various concentrations via the transpiration stream indicated that the xylem flux of NO3 was generally rate-limiting for NO3 reduction. However, NO3 assimilation rate was maintained within narrow limits as compared with the variations of the xylem flux of NO3. This was due to considerable remobilization and assimilation of previously stored endogenous NO3 at low exogenous NO3 delivery, and limitation of NO3 reduction at high xylem flux of NO3, leading to a significant accumulation of exogenous NO3. The supply of 15NO3 to the leaves via the xylem confirmed the labile nature of the NO3 storage pool, since its half-time for exchange was close to 10 hours under steady state conditions. When the xylem flux of 15NO3 increased, the proportion of the available NO3 which was reduced decreased similarly from nearly 100% to less than 50% for both endogenous 14NO3 and exogenous 15NO3. This supports the hypothesis that the assimilatory system does not distinguish between endogenous and exogenous NO3 and that the limitation of NO3 reduction affected equally the utilization of NO3 from both sources. It is proposed that, in the soybean leaf, the NO3 storage pool is particularly involved in the short-term control of NO3 reduction. The dynamics of this pool results in a buffering of NO3 reduction against the variations of the exogenous NO3 delivery.  相似文献

Self-incompatibility in the Solanaceae is mediated by S-RNase alleles expressed in the style, which confer specificity for pollen recognition. Nicotiana alata has been successfully used as an experimental model to elucidate cellular and molecular aspects of S-RNase-based self-incompatibility in Solanaceae. However, S-RNase alleles of this species have not been surveyed from natural populations and consequently the S-haplotype diversity is poorly known. Here the molecular and functional characterization of seven S-RNase candidate sequences, identified from a natural population of N. alata, are reported. Six of these candidates, S 5 , S 27 , S 70 , S 75 , S 107 , and S 210 , showed plant-specific amplification in the natural population and style-specific expression, which increased gradually during bud maturation, consistent with the reported S-RNase expression. In contrast, the S 63 ribonuclease was present in all plants examined and was ubiquitously expressed in different organs and bud developmental stages. Genetic segregation analysis demonstrated that S 27 , S 70 , S 75 , S 107 , and S 210 alleles were fully functional novel S-RNases, while S 5 and S 63 resulted to be non-S-RNases, although with a clearly distinct pattern of expression. These results reveal the importance of performing functional analysis in studies of S-RNase allelic diversity. Comparative phylogenetic analysis of six species of Solanaceae showed that N. alata S-RNases were included in eight transgeneric S-lineages. Phylogenetic pattern obtained from the inclusion of the novel S-RNase alleles confirms that N. alata represents a broad sample of the allelic variation at the S-locus of the Solanaceae.  相似文献

The hydraulic conductivity of the lateral walls of early metaxylem vessels (Lpx in m · s–1 · MPa–1) was measured in young, excised roots of maize using a root pressure probe. Values for this parameter were determined by comparing the root hydraulic conductivities before and after steam-ringing a short zone on each root. Killing of living tissue virtually canceled its hydraulic resistance. There were no suberin lamellae present in the endodermis of the roots used. The value of Lpx ranged between 3 · 10–7 and 35 · 10–7 m · s–1 · MPa–1 and was larger than the hydraulic conductivity of the untreated root (Lpr = 0.7 · 10–7 to 4.0 · 10–7 m · s–1 · MPa–1) by factor of 3 to 13. Assuming that all flow through the vessel walls was through the pit membranes, which occupied 14% of the total wall area, an upper limit of the hydraulic conductivity of this structure could be given(Lppm=21 · 10–7 to 250 · 10–7 m · s–1 · MPa–1). The specific hydraulic conductivity (Lpcw) of the wall material of the pit membranes (again an upper limit) ranged from 0.3 · 10–12 to 3.8 · 10–12 m2 · s–1 · MPa–1 and was lower than estimates given in the literature for plant cell walls. From the data, we conclude that the majority of the radial resistance to water movement in the root is contributed by living tissue. Marine invasions are a worldwide problem that involves changes in communities and the acclimation of organisms to them. The invasive Chlorophyte Caulerpa racemosa var. cylindracea is widespread in the Mediterranean and colonises large areas from 0 to 70 m in depth. The omnivorous fish Spondyliosoma cantharus presents a high frequency of occurrence of C. racemosa in the stomach contents at invaded areas (76.3%) while no presence of C. racemosa was detected in control areas. The isotopic composition of muscle differed significantly between invaded and non-invaded sites for δ13C (− 16.67‰ ± 0.09 and − 17.67‰ ± 0.08, respectively), δ15N (10.22‰ ± 0.22 and 9.32‰ ± 0.18, respectively) and the C:N ratio (2.01 ± 0.0002 and 1.96 ± 0.009, respectively). Despite the high frequency of occurrence of C. racemosa in the stomach contents of S. cantharus and its important contribution to the δ13C source (20.7% ± 16.2), the contribution of C. racemosa to the δ15N in S. cantharus food sources was very low (6.6% ± 5.8). Other invertebrate prey such as decapods and polychaetes were more important contributors to the δ15N source at both invaded and non-invaded sites. Activation of enzymatic pathways (catalase, superoxide dismutase, glutathione-s-tranferase, 7-ethoxy resorufin O-de-ethylase) but not a significant increase in lipid peroxidation MDA (0.49 ± 0.01 nmol/mg prot at non-invaded and 0.53 ± 0.01 nmol/mg prot at invaded sites) was observed in S. cantharus individuals living in C. racemosa-invaded sites compared with control specimens. The low δ15N contribution values of C. racemosa by S. cantharus together with the toxicity demonstrated by the activation of the antioxidant defences and the important contribution of invertebrate prey to the δ15N could mean that the ingestion of C. racemosa by S. cantharus might be unintentional during the predation of invertebrate preys living underneath the entanglement of the C. racemosa fronds and stolons mats.  相似文献

Recent cave biodiversity surveys carried out in Du’an County and its adjacent areas of northwestern Guangxi, China, have revealed some exciting scientific findings. In a very limited area seven new species of the cavernicolous trechine genus Dongodytes Deuve, 1993 were found and are described: Dongodytes (s. str.) elongatus sp. n., Dongodytes (s. str.) troglodytes sp. n., Dongodytes (s. str.) lani sp. n., Dongodytes (Dongodytodes) brevipenis sp. n., Dongodytes (Dongodytodes) jinzhuensis sp. n., Dongodytes (Dongodytodes) inexpectatus sp. n. and Dongodytes (Dongodytodes) yaophilus sp. n. Diagnoses and notes on the genus, subgenera, and two known species in Du’an Karst, Dongodytes (s. str.) baxian Tian, 2011 and Dongodytes (Dongodytodes) deharvengi Tian, 2011, are also given. A key to subgenera and all species of Dongodytes is provided. To date, Dongodytes becomes one of the richest in species genera of subterranean carabid trechines in China with 12 species which are arranged into two subgenera. Dongodytes (s. str.) Deuve is composed of seven species, four of which from Du’an County, each of other three from Bama, Fengshan and Tian’e Counties, respectively. All species of the subgenus Dongodytodes Tian, 2011 are recorded from Du’an Karst. By having 10 species (nine Dongodytes and one Libotrechus Uéno, 1998), Du’an Karst holds the richest specific diversity of cavernicolous Trechinae in China. Dongodytes species are distributed in a very limited area of the river Hongshui He drainages in northwestern Guangxi, and the river acts as a natural barrier of Dongodytes dispersal at only a specific level. However, all members of Dongodytodes are recorded from the eastern or northern bank of Hongshui He.  相似文献

Determination of the complete amino acid sequence of the rubredoxin isolated from the sulfate reducing bacterium Desulfovibriogigas showed that the molecule consists of a single polypeptide chain of 52 residues. The sequence of the first 42 residues was determined using an automatic Protein Sequencer. Peptides derived from tryptic hydrolysis and from specific cleavage at tryptophan residue were used to construct the total sequence. Compared with the sequence of Desulfovibriovulgaris rubredoxin, 37 positions are identical, and with the sequences of Clostridiumpasteurianum, Peptostreptococcuselsdenii, Micrococcusaerogenes and D.vulgaris rubredoxins, 20 matching residues occur. A crystallographic study of the D.gigas rubredoxin is in progress.  相似文献

A method for calculating the rate constant (KA1A2) for the oxidation of the primary electron acceptor (A1) by the secondary one (A2) in the photosynthetic electron transport chain of purple bacteria is proposed.The method is based on the analysis of the dark recovery kinetics of reaction centre bacteriochlorophyll (P) following its oxidation by a short single laser pulse at a high oxidation-reduction potential of the medium. It is shown that in Ectothiorhodospira shaposhnikovii there is little difference in the value of KA1A2 obtained by this method from that measured by the method of Parson ((1969) Biochim. Biophys. Acta 189, 384–396), namely: (4.5±1.4) · 103s?1 and (6.9±1.2) · 103 s?1, respectively.The proposed method has also been used for the estimation of the KA1A2 value in chromatophores of Rhodospirillum rubrum deprived of constitutive electron donors which are capable of reducing P+ at a rate exceeding this for the transfer of electron from A1 to A2. The method of Parson cannot be used in this case. The value of KA1A2 has been found to be (2.7±0.8) · 103 s?1.The activation energies for the A1 to A2 electron transfer have also been determined. They are 12.4 kcal/mol and 9.9 kcal/mol for E. shaposhnikovii and R. rubrum, respectively.  相似文献

Gammarus lacustris and Hyalella azteca (hereafter G. lacustris and H. azteca, respectively) are important components of secondary production in wetlands and shallow lakes of the upper Midwest, USA. Within the past 50 years, amphipod densities have decreased while occurrences of fish and intensity of agricultural land use have increased markedly across this landscape. We investigated influences of fish, sedimentation, and submerged aquatic vegetation (SAV) on densities of G. lacustris and H. azteca in semipermanent and permanent wetlands and shallow lakes (n = 283) throughout seven eco-physiographic regions of Iowa, Minnesota, and North Dakota during 2004–2005. G. lacustris and H. azteca densities were positively correlated with densities of SAV (P < 0.001 and P < 0.001, respectively). Both species were negatively correlated with densities of large fish (non-Cyprinidae; P = 0.01 and P = 0.013, respectively) and with high densities of fathead minnows (Pimephales promelas; P < 0.001 and P = 0.033, respectively). H. azteca densities also were negatively correlated with densities of small fish (e.g., other minnows [Cyprinidae] and sticklebacks [Gasterosteidae]; P = 0.048) and common carp (Cyprinus spp.; P = 0.022). G. lacustris densities were negatively correlated with high levels of suspended solids (an index for sedimentation; P < 0.001). H. azteca densities were positively correlated with the width of upland-vegetation buffers (P = 0.004). Our results indicate that sedimentation and fish reduce amphipod densities and may contribute to the current low densities of amphipods in the upper Midwest. Thus, removing/excluding fish, and providing a thick buffer of upland vegetation around wetlands may help restore amphipod densities and wetland and water quality within this landscape.  相似文献

Five species of epibenthic pycnogonids collected on the giant lion´s-paw scallop Nodipecten subnodosus are recorded. A new species of Eurycyde, Eurycyde bamberi, is described. Of the 19 species known in this genus; the new species is closest to Eurycyde hispida Kroyer, 1844 but differs from it in the absence of plumose spines and the shapes of the lateral process, first coxa, and ocular tubercle. The new species represents the third member of Eurycyde from the eastern Pacific in addition to Eurycyde spinosa Hilton, 1916 and Eurycyde clitellaria Stock, 1955. Besides Eurycyde bamberi, the following species were collected: Nymphopsis duodorsospinosa Hilton, 1942c; Callipallene californiensis (Hall, 1913); Nymphon lituus Child, 1979; and Pycnogonum rickettsi Schmitt, 1934. Pycnogonum rickettsi is recorded for first time from Mexican waters, as is Nymphon lituus from the western coast of Baja California Peninsula. Each of these four species are re-described and re-illustrated in order to fill in existing gaps in the literature of the region.  相似文献

The rate of light-induced electron transport by isolated spinachthylakoids under phosphorylating conditions, as affected bydifferent concentrations of Br, Cl, NO3,HCO3, SO42– and CO32– has been investigated.The results show that both low and high concentrations of HCO32–stimulated the oxygen evolution capacity under phosphorylatingconditions, whereas only low concentrations of CO32–,SO42– and Cl stimulated the oxygen evolution capacity.However, irrespective of concentration, both Br and NO3reduced this capacity. The rate of photosynthetic electron transportwas generally stimulated by addition of ADP, even in cases whereelectron transport was inhibited by addition of bromide andnitrate. The different concentrations of these anions also causedreduction of the power generated by proton pumping and usedfor phosphorylation.
