森林土壤氮转化的微生物功能研究   总被引:2,自引:1,他引:1  
本文研究了不同林型下土壤(A+6层和A_1层)微生物、土壤酶活性在森林土壤氮转化中的作用。结果表明不同林型下土壤具有不同的固氮作用、反硝化作用、氨化作用和硝化作用速率,即阔叶林>针阔混交林>针叶林。已经证明,固氮作用主要存在于森林土壤的A_1层,反硝化作用主要存在于A_0层。森林土壤存在2种硝化作用过程,即由自养微生物所引起的自养硝化作用过程和异养微生物所引起的异养硝化作用过程。它的存在与林型有关,某些森林土壤中这2种硝化作用过程都存在,如针阔混交林下的A_0层和A_1层。有些林型下土壤,则以异养硝化作用过程为主,如针叶林的A_0层。  相似文献

Bioavailability of dichlorodiphenyltrichloroethanes (DDTs) in surface sediments was evaluated with semipermeable membrane devices (SPMDs) and two different sediment-dwelling benthic mussels, Bellamya aeruginosa (B. aeruginosa) and Corbicula fluminea (C. fluminea). After 28d laboratory exposure, the positive correlations of DDT concentrations between both SPMDs and benthic mussels with sediments documented that the bioavailability of DDTs was mainly affected by surrounding sediments, while the observed differences of DDT concentrations and congener proportions between B. aeruginosa and C. fluminea were due to the specific physiological characteristics of organisms and different physico-chemical properties of contaminants. Comparisons between SPMDs and benthic mussels showed higher values of biota-sediment accumulation factors (BSAF, 0.63-3.61 for B. aeruginosa and 2.19-17.08 for C. fluminea) than device accumulation factors (DAF, 1.00-1.74). This indicated that living organisms bioaccumulated much more DDTs from sediments than SPMDs due to the different exposure and uptake routes. Strong positive associations between DDTs in SPMDs and benthic mussels indicated SPMDs could mimic the bioaccumulation of DDTs, especially in C. fluminea. However, given the distinct differences observed for both concentrations and congener proportions of DDTs in SPMDs and B. aeruginosa, future study should be directed to develop reliable models with various sediment-dwelling organisms before SPMDs are routinely used in field study.  相似文献

Smilacaceae, composed of Smilax and Heterosmilax, are a cosmopolitan family of > 200 species of mostly climbing monocots with alternate leaves characterized by reticulate venation, a pair of petiolar tendrils and usually prickly stems. Although there has been a long history of studying Smilax since Linnaeus named the genus in 1753, the phylogenetic history of this dioecious family remains unclear. Here we present results based on nuclear ribosomal internal transcribed spacer (nrITS) and plastid matK and rpl16 intron DNA sequence data from 125 taxa of Smilacaceae. Our taxon sampling covers all sections of Smilax and Heterosmilax and major distribution zones of the family; species from Ripogonaceae and Philesiaceae are used as outgroups. Our molecular analysis indicates that phylogenetic relationships largely contradict the traditional morphological classification of the family, instead showing a conspicuous geographical pattern among the species clades. The previously recognized genus Heterosmilax was found to be embedded in Smilax. Species in the family are separated into primarily New World and Old World clades, except for a single species lineage, Smilax aspera, that is sister to the remaining species of the family, but with poor statistical support. Ancestral character state reconstructions and examination of distribution patterns among the clades provide important information for future taxonomic revisions and historical biogeography of the group. © 2013 The Linnean Society of London, Botanical Journal of the Linnean Society, 2013, 173 , 535–548.  相似文献

广西拟水龟和艾氏拟水龟的分类和系统发生多年来存在争议。通过测定广西拟水龟、艾氏拟水龟和黄喉拟水龟线粒体ND4基因和核c-mos基因部分序列,合并GenBank中拟水龟属其他物种的ND4基因和c-mos基因部分序列进行分析,从分子水平探讨广西拟水龟和艾氏拟水龟的系统发生。ND4基因数据分析发现NJ、MP和BI树中广西拟水龟与安南拟水龟的聚类分支相互交织聚为一支,二者种内遗传距离均为0.002~0.017,种间遗传距离为0.000~0.035,种间遗传距离明显小于同属内其他种间0.056~0.109的遗传距离,表明广西拟水龟与安南拟水龟可能为同一物种,可能是安南拟水龟的同种异名,或是作为安南拟水龟的一个亚种;NJ树、MP树和BI树均显示,艾氏拟水龟与黄喉拟水龟的位置和关系最为相近,二者间的遗传距离为0.020~0.035,明显小于拟水龟属其他物种间遗传距离,而明显大于同属各物种内遗传距离,艾氏拟水龟与黄喉拟水龟之间系统分类关系是介于种内与种间之间;乌龟、中华花龟、安南拟水龟等物种都是与拟水龟属中其他物种先聚成一支后再与同科的地龟属的地龟形成姐妹支,支持将乌龟属、花龟属和安南龟属并入拟水龟属的分类。c-mos基因数据分析发现,拟水龟属各物种间不存在明显的遗传距离,NJ树和MP树也未能对属中各物种的分子系统发生位置进行有效的界定,但在属及属以上阶元的分子分类系统中c-mos基因可以作为分类依据,并与线粒体基因数据有较好的一致性。  相似文献

[背景] 海洋微生物在活性物质开发方面具有巨大的应用前景,而目前有关南海东海岛微生物的研究鲜少。[方法] 对从东海岛沉积物中分离纯化的海洋细菌,采用形态学观察、生理生化以及16S rRNA基因序列的系统发育分析方法进行鉴定;以大肠杆菌(Escherichia coli)、枯草芽孢杆菌(Bacillus subtilis)和金黄色葡萄球菌(Staphylococcus aureus)作为指示菌,测定其抑菌活性;对具有抑菌活性的菌株扩增聚酮合酶(Polyketide synthase I,PKSI)基因,并与已知的PKSI氨基酸序列比对;选择具有PKSI基因的代表菌株,检测菌株及其发酵抑菌物的稳定性。[结果] 分离纯化到25株海洋细菌,分属于不动杆菌属(Acinetobacter)、交替单胞菌属(Alteromonas)、芽孢杆菌属(Bacillus)、嗜冷杆菌属(Psychrobacter)、假交替单胞菌属(Pseudo-alteromonas)、海洋单胞菌属(Oceanimonas)、葡萄球菌属(Staphylococcus)、微球菌属(Micrococcus)和海杆菌属(Marinobacter)。12株菌株通过基因筛选检测到PKSI编码基因,其中6株菌株具有抑菌活性和PKSI编码基因,并分属于芽孢杆菌属和交替单胞菌属;PKSI氨基酸序列同源性分析推测菌株DHD-15和DHD-a可能产生新的I型聚酮合酶结构。菌株DHD-15和DHD-L生长温度范围为15-40℃,可耐受10% NaCl高盐以及pH 3和pH 11的酸碱条件,但不耐高温;菌株DHD-15产生的抑菌物质可耐受100℃和pH 11的高温碱性条件,在50℃、pH 9条件下制备和室温保藏条件下抑菌活性较高,其稳定性较好。[结论] 南海东海岛沉积物筛选的细菌种具有抑菌活性,具有产聚酮类活性物质的潜力。  相似文献

香蕉枯萎病主要由尖孢镰刀菌 4 号生理小种(Fusarium oxysporum f. sp. cubense,Foc4)引起的一种土传病害,严重威胁香蕉产业的可持续发展。为寻求一种经济有效且环保的防治措施,以韭菜化感物质的衍生物草莓酸(strawberry acid,SA)为材料,通过平板和盆栽实验,研究了SA对Foc4的菌丝生长、香蕉枯萎病病情指数、土壤微生物数量、土壤酶活性的影响。结果表明:(1)随着SA浓度的增加,Foc4的菌落生长直径显著减小,第5天时菌落直径在SA浓度为300、450 μL·L-1时比150 μL·L-1分别减小了49.15%、70.89%; 液体培养条件下SA浓度为600 μL·L-1时Foc4的分生孢子数量显著低于对照处理(相差 470 多倍); pH为5时SA对Foc4的抑制效果显著比pH为7和9时好。(2)随实验处理时间的延长,添加 SA后香蕉幼苗的病情指数显著低于对照。(3)土壤细菌、真菌数量和微生物总量在SA为600 μL·L-1时均为最高; Foc4数量随SA浓度升高而降低,在1 200 μL·L-1时显著降低。(4)各土壤酶在浓度(300~600 μL·L-1)SA处理时活性较高; 1 200 μL·L-1时显著降低,过氧化氢酶和多酚氧化酶较对照分别降低了41.88%、54.82%。(5)相关性分析得出,土壤微生物总量与细菌、真菌数量极显著正相关; 土壤真菌与放线菌显著负相关; 土壤细菌、真菌和放线菌数量均与蔗糖酶、多酚氧化酶显著正相关; 蔗糖酶与脲酶、过氧化氢酶与多酚氧化酶均显著正相关。综上认为,添加SA浓度为600 μL·L-1能较好地抑制Foc4的菌丝生长且能提高其抑制率,病情指数明显降低,有利于改善香蕉的生长环境。该研究结果为有效利用SA防治香蕉枯萎病提供了科学依据。  相似文献

The objectives of this study are to explore the relationships between plant phenology and satellite-sensor-derived measures of greenness, and to advance a new procedure for determining the growing season of land vegetation at the regional scale. Three phenological stations were selected as sample sites to represent different climatic zones and vegetation types in northern China. The mixed data set consists of occurrence dates of all observed phenophases for 50–70 kinds of trees and shrubs from 1983 to 1988. Using these data, we calculated the cumulative frequency of phenophases in every 5-day period (pentad) throughout each year, and also drew the cumulative frequency distribution curve for all station-years, in order to reveal the typical seasonal characteristics of these plant communities. The growing season was set as the time interval between 5% and 95% of the phenological cumulative frequency. Average lengths of the growing season varied between 188 days in the northern, to 259 days in the southern part of the research region. The beginning and end dates of the surface growing season were then applied each year as time thresholds, to determine the corresponding 10-day peak greenness values from normalized difference vegetation index curves for 8-km2 pixels overlying the phenological stations. Our results show that, at the beginning of the growing season, the largest average greenness value occurs in the southern part, then in the northern, and finally the middle part of the research region. In contrast, at the end of the growing season, the largest average greenness value is measured in the northern part, next in the middle and lastly the southern part of the research region. In future studies, these derived NDVI thresholds can be applied to determine the growing season of similar plant communities at other sites, which lack surface phenological data.
