Agelasimine A and agelasimine B, two novel compounds related to adenine, have been isolated from the orange sponge, Agelas mauritiana, and have been tested for a variety of biological activities. Both compounds inhibited proliferation of cultured L1210 leukemia cells at nanomolar concentrations with accumulation in the G1 stage of the cell cycle. However, no prolongation of life was observed in mice bearing P388 leukemia treated with these compounds. In the rat isolated aorta, micromolar concentrations of agelasimines were very effective in inhibiting contractions elicited by potassium chloride but had little or no effect on responses for prostaglandin F2 alpha and had modest effects on the responses to noradrenaline and significant effects on 5-hydroxytryptamine. Agelsamines A and B appeared to be equipotent in causing relaxation in rabbit jejunum and bovine coronary artery, and they also inhibited nucleoside transport into rabbit erythrocytes in micromolar concentrations. 相似文献

Membrane polypeptides (relative mass (Mr) 48,000--55,000) associated with the equilibrative transport of nucleosides were identified in cultured murine leukemia (L1210/C2) cells by site-specific photolabeling with [3H]nitrobenzylthioinosine ([3H]NBMPR). Growth of cells in the presence of tunicamycin resulted in the gradual conversion of 3H-labeled polypeptides to a form that migrated more rapidly (Mr 42,000--47,000) during sodium dodecyl sulfate (SDS)--polyacrylamide gel electrophoresis. When plasma membrane fractions were photolabeled and incubated with O-glycanase or endoglycosidase F, the [3H]NBMPR-labeled polypeptides migrated in SDS-polyacrylamide gels with the same mobility as native NBMPR-binding polypeptides, whereas incubation with either N-glycanase or trifluoromethane sulfonic acid converted [3H]NBMPR-labeled polypeptides to the more rapidly migrating form (Mr 41,000--48,000). These observations are consistent with the presence of N-linked oligosaccharides of the complex type on the NBMPR-binding polypeptides of L1210/C2 cells. Tunicamycin exposures that reduced incorporation of [3H]mannose into plasma membrane fractions by greater than 95% had little, if any, effect on either the affinity (Kd values, 0.1-0.2 nM) or abundance (Bmax values, 200,000--220,000 sites/cell) of NBMPR-binding sites, whereas uridine transport kinetics at 37 degrees C were altered in a complex way. Thus, although N-linked glycosylation is not required for insertion of the NBMPR-binding protein into the plasma membrane or for interaction of NBMPR with the high-affinity binding sites, it is important for function of at least one of the three nucleoside transporters expressed by L1210/C2 cells. 相似文献

It is shown that an inhibited enzyme electrode, using cytochrome oxidase, will respond to H2S, HCN and azide ion. For all three inhibitors the kinetics of the inhibition and recovery processes have been analysed using the theoretical model presented previously (Albery et al., 1990a). Rearrangement of the differential equation describing inhibition and the development of the necessary software has enabled us to obtain values of the concentration of inhibitor in a matter of seconds after exposure of the sensor. Synthesis of heat shock proteins (HSPs) in the leaves of a drought- and heat-resistant (line ZPBL 1304), and a drought- and heat-sensitive (line ZPL 389) line of maize (Zea mays L.) was studied under two environmental stress treatments: (a) soil drying and high temperature and (b) high temperature. In the first treatment 13-day-old plants were exposed to 7-day soil drying followed by high temperature stress (45°C), and in the second treatment 20-day-old plants were exposed to high temperature stress (45°C). Second leaves were labeled with [35S]methionine. During the labeling period line ZPBL 1304 showed no signs of leaf dehydration under soil drying and high temperature stress conditions. In contrast, line ZPL 389 was dehydrated 23%, as determined by relative water content. Incorporation of [35S]methionine into protein was greater in the resistant than in the sensitive line in both treatments. The pattern of synthesis of HSPs in the two lines was similar in treatments 1 and 2. Both lines synthesized a high molecular mass set and a low molecular mass set of HSPs. Proteins from both sets from both lines of maize appeared similar to each other, with respect to the molecular mass. Heated plants of the drought- and heat-resistant line ZPBL 1304 synthesized a band of HSP(s) of approximately 45 kilodaltons which was not found in heated plants of the drought and heat sensitive line ZPL 389. Pepsinogens (PGA) are the inactive precursors of pepsin, the major acid protease found in the stomach. The PGA gene family exhibits polymorphic variation in human populations that can either be demonstrated by electrophoretic analysis of the proteins or by analysis of the respective genes with cDNA probes. Here, we describe the interrelationships between the most common pepsinogen protein phenotypes and the corresponding pepsinogen haplotypes (A, B, and C) containing different combinations of the PGA3, PGA4, and PGA5 genes. We propose that this unusual genetic variation involving haplotypes that contain three, two, and one genes, respectively, is the result of molecular evolution by gene duplication. 相似文献

Mutations in the araC gene of Escherichia coli B/r were isolated which alter both activation of the araBAD operon expression and autoregulation. The mutations were isolated on an araC-containing plasmid by hydroxylamine mutagenesis of plasmid DNA. The mutant phenotype selected was the inability to autoregulate. The DNA sequence of 16 mutants was determined and found to consist of seven different missense mutations located within the distal third of the araC gene. Enzyme activities revealed that each araC mutation had altered both autoregulatory and activator functions of AraC protein. The mutational analysis presented in this paper suggests that both autoregulatory and activator functions are localized to the same determinants of the AraC protein and that the amino acid sequence within the carboxy-terminal region of AraC protein is important for site-specific DNA binding. 相似文献

In vivo electrochemistry was used to investigate the mechanisms contributing to the clearance of locally applied dopamine in the dorsal striatum and nucleus accumbens of urethane-anesthetized rats. Chronoamperometric recordings were continuously made at 5 Hz using Nafion-coated carbon fiber electrodes. When a finite amount of dopamine was pressure-ejected at 5-min intervals from a micropipette adjacent to the electrode, transient and reproducible dopamine signals were detected. Substitution of L-a-methyldopamine, a substrate for the dopamine transporter but not for monoamine oxidase, for dopamine in the micropipette did not substantially alter the time course of the resulting signals. This indicates that metabolism of locally applied dopamine to 3,4-dihydroxyphenylacetic acid is not responsible for the decline in the dopamine signal. Similarly, changing the applied oxidation potential from ±0.45 to ±0.80 V, which allows for detection of 3-methoxytyramine formed from dopamine via catechol-O-methyltransferase, had little effect on signal amplitude or time course. In contrast, lesioning the dopamine terminals with 6-hydroxydopamine, or locally applying the dopamine uptake inhibitors cocaine or nomifensine before pressure ejection of dopamine, significantly increased the amplitude and time course of the dopamine signals in both regions. The effects of cocaine and nomifensine were greater in the nucleus accumbens than in the dorsal striatum. Local application of lidocaine and procaine had no effect on the dopamine signals. Initial attempts at modeling resulted in curves that were in qualitative agreement with our experimental findings. Taken together, these data indicate that (1) uptake of dopamine by the neuronal dopamine transporter, rather than metabolism or diffusion, is the major mechanism for clearing locally applied dopamine from the extracellular milieu of the dorsal striatum and nucleus accumbens, and (2) the nucleus accumbens is more sensitive to the effects of inhibitors of dopamine uptake than is the dorsal striatum. 相似文献

土壤碳(C)、氮(N)、磷(P)是参与植物光合作用和影响生态系统初级生产力的主要元素。甘南高原是黄河流域重要的生态屏障,为了解该区不同林分土壤养分状况的差异,选取该区4种典型林分:云杉林、华北落叶松林、巴山冷杉林以及岷江冷杉糙皮桦混交林为研究对象,研究土壤C、N、P化学计量特征。结果表明:(1)岷江冷杉及糙皮桦混交林土壤C、N含量最高,云杉林土壤N、P含量最低。不同林分间P含量差异显著(P<0.05),不同土层间C、N含量差异均显著(P<0.05)。(2)云杉林土壤C : N值显著高于其他林分,岷江冷杉及糙皮桦混交林土壤N : P及C : P高于其他林分。(3)海拔、土壤pH、容重与土壤含水量是影响土壤养分的重要因素。土壤C含量与N、P含量均显著相关(P<0.05)。总体来说,不同林分土壤化学计量特征具有显著差异,混交林土壤养分状况较纯林好,未来森林管理和植被建设中,可以通过选择合适的树种和提高树种多样性有效改善森林土壤质量。 相似文献
