The hypopharyngeal gland (HG) of the honeybee (Apis mellifera L.) produces royal jelly (RJ) that is essential to feed and raise broods and queens. A strain of bees (high royal jelly producing bee, RJb) has been selected for its high RJ production, but the mechanisms of its higher yield are not understood. In this study, we compared HG acini size, RJ production, and protein differential expressions between the RJb and nonselected honeybee (Italian bee, ITb) using proteomics in combination with an electron microscopy, Western blot, and quantitative real-time PCR (qRT-PCR). Generally, the HG of both bees showed age-dependent changes in acini sizes and protein expression as worker behaviors changed from brood nursing to nectar ripening, foraging, and storage activities. The electron microscopic analysis revealed that the HG acini diameter of the RJb strain was large and produced 5 times more RJ than the ITb, demonstrating a positive correlation between the yield and HG acini size. In addition, the proteomic analysis showed that RJb significantly upregulated a large group of proteins involved in carbohydrate metabolism and energy production, those involved in protein biosynthesis, development, amino acid metabolism, nucleotide and fatty acid, transporter, protein folding, cytoskeleton, and antioxidation, which coincides with the fact that the HGs of the RJb strain produce more RJ than the ITb strain that is owing to selection pressure. We also observed age-dependent major royal jelly proteins (MRJPs) changing both in form and expressional intensity concurrent with task-switching. In addition to MRJPs, the RJb overexpressed proteins such as enolase and transitional endoplasmic reticulum ATPase, protein biosynthesis, and development proteins compared to the ITb strain to support its large HG growth and RJ secretion. Because of selection pressure, RJb pursued a different strategy of increased RJ production by involving additional proteins compared to its original counterpart ITb. To our knowledge, this morphological and proteomic comparison study on the HG of the two strains of worker honeybees associated with their age-dependent division of labor is the first of its kind. The study provided not only the quantity and quality differences in the HG from the RJb and the ITb, but also addressed the cellular and behavioral biology development question of how the RJb strain can produce RJ more efficiently than its wild type strain (ITb).  相似文献

Barley, Hordeum vulgare L., is well adapted to subarctic Alaska growing conditions, but little is known about its response to grasshopper defoliation. A field experiment was conducted to study dry matter and grain yield in response to a combination of grasshopper defoliation and weeds in 2002 and 2003 near Delta Junction, AK (63 degrees 55' N, 145 degrees 20' W). Barley plants at third to fourth leaf stage were exposed to a combination of two levels of weeds (present or absent) and four densities of grasshoppers (equivalent to 0, 25, 50, and 75 grasshoppers per m2) of third to fourth instars of Melanoplus sanguinipes (F). Dry matter accumulation by the barley plants was determined at three times during the growing seasons: approximately 10 d after introduction of the grasshoppers, shortly after anthesis, and at maturity. Dry matter accumulation and grain yield were much lower in 2003 than in 2002, probably due to very low levels of soil moisture early in the growing season of 2003. Head clipping accounted for a greater portion of yield loss in 2003 than in 2002. The percentage of reduction in harvestable yield due to grasshoppers remained fairly constant between years (1.9 and 1.4 g per grasshopper per m2 in 2002 and 2003, respectively) despite a large difference in overall yield. Examination of the yield components suggest that yields were reduced by the early season drought in 2003 primarily through fewer seeds per head, whereas grasshoppers in both years reduced average seed weight, but not numbers of seeds.  相似文献

Taste quality and palatability are two of the most important properties measured in the evaluation of taste stimuli. Human panels can report both aspects, but are of limited experimental flexibility and throughput capacity. Relatively efficient animal models for taste evaluation have been developed, but each of them is designed to measure either taste quality or palatability as independent experimental endpoints. We present here a new apparatus and method for high throughput quantification of both taste quality and palatability using rats in an operant taste discrimination paradigm. Cohorts of four rats were trained in a modified operant chamber to sample taste stimuli by licking solutions from a 96-well plate that moved in a randomized pattern beneath the chamber floor. As a rat’s tongue entered the well it disrupted a laser beam projecting across the top of the 96-well plate, consequently producing two retractable levers that operated a pellet dispenser. The taste of sucrose was associated with food reinforcement by presses on a sucrose-designated lever, whereas the taste of water and other basic tastes were associated with the alternative lever. Each disruption of the laser was counted as a lick. Using this procedure, rats were trained to discriminate 100 mM sucrose from water, quinine, citric acid, and NaCl with 90-100% accuracy. Palatability was determined by the number of licks per trial and, due to intermediate rates of licking for water, was quantifiable along the entire spectrum of appetitiveness to aversiveness. All 96 samples were evaluated within 90 minute test sessions with no evidence of desensitization or fatigue. The technology is capable of generating multiple concentration–response functions within a single session, is suitable for in vivo primary screening of tastant libraries, and potentially can be used to evaluate stimuli for any taste system.  相似文献

Light availability has a profound effect on plant growth and development. One of the ways to study the effects of light intensity on plant growth and development without the confounding problem of photosynthate availability is sucrose injection/supplementation. A greenhouse experiment was conducted to evaluate the effects of light levels (0% and 75% shade) and sucrose injection (distilled water or 150 g sucrose l(-1)) on three weed species: redroot pigweed (Amaranthus retroflexus L., C4), lambsquarters (Chenopodium album L., C3) and velvetleaf (Abutilon theophrasti Medic., C3). The average total sucrose uptake was 7.6 and 5.9 g per plant for 0% and 75% shading, respectively, representing 47% of the average total weed dry weight. Plants injected with sucrose had greater dry weights and shoot-to-root ratios under both light levels. In spite of sucrose supplementation the reduction in dry matter due to shading was greater for roots and reproductive structures than vegetative shoot tissues, indicating light level regulation of morphological changes resulting in changed C allocation that are independent of photosynthate availability. Dry weights of plants injected with sucrose under 75% shading were not different from distilled water-injected unshaded plants. However, both sucrose-injected and control plants, regardless of their photosynthetic pathways, underwent similar changes in allocation of dry matter and morphology due to shading, suggesting that these effects are strictly due to light intensity and not related to photosynthate availability.  相似文献

Pollinators, honey bees in particular, are continuously exposed to various mixtures of pesticides, which contribute to their population decline. Both amitraz and thiacloprid have been proven less toxic to honey bees and are frequently applied in- and out-hive, respectively. We examined the sub-lethal effects of amitraz, thiacloprid and their sequential exposure on learning, memory and sugar responsiveness in Apis mellifera using the Proboscis extension response (PER). Sub-lethal doses of amitraz (0.1, 0.2 and 0.4 µg/bee) and thiacloprid (0.05, 0.1 and 0.2 µg/bee) were tested. Sub-lethal effects were observed only at the highest doses of each pesticide treatment; amitraz (0.4 µg/bee) and thiacloprid (0.2 µg/bee) but not in lower doses. In sequential treatment of amitraz and thiacloprid, reduced acquisition and memory retention were significant across all tested doses. The same profile was also obtained on sugar responsiveness of foragers. Our results suggest that the sequential exposure would pose higher risk to honey bee compared to single pesticide exposure by reducing the bees’ appetitive olfactory learning, memory and sugar acuity more than individual pesticide exposures.  相似文献

Despite their similar genetic makeup, honeybee (A. mellifera) queens and workers show alternative morphologies driven by nutritional difference during the larval stage. Although much research have been done to investigate the causes of honeybee caste polymorphism, information at subcellular protein levels is limited. We analyzed queen- and worker-destined larvae mitochondrial proteome at three early developmental stages using combinations of differential centrifugation, two-dimensional electrophoresis, mass spectrometry, bioinformatics, and quantitative real time PCR. In total, 67, 69, and 97 protein spots were reproducibly identified as mitochondrial proteins at 72, 96, and 120 h, respectively. There were significant qualitative and quantitative protein expression differences between the two castes at three developmental stages. In general, the queen-destined larvae up-regulated large proportions of proteins at all of the developmental stages and, in particular, 95% at 72 h. An overwhelming majority of the queen larvae up-regulated proteins were physiometabolic-enriched proteins (metabolism of carbohydrate and energy, amino acid, and fatty acid) and involved in protein folding, and this was further verified by functional enrichment and biological interaction network analyses as a direct link with metabolic rates and cellular responses to hormones. Although wide-ranging mitochondrial proteomes participate to shape the metabolic, physiologic, and anatomic differences between the two castes at 72 h, physiometabolic-enriched proteins were found as the major modulators of the profound marking of this caste differentiation. Owing to nutritional difference, prospective queen larvae showed enhanced growth, and this was manifested through the overexpression of metabolic enzymes. Differently from similar studies targeting the causes of honeybee caste polymorphism, this subcellular level study provides an in-depth insight into mitochondrial proteins-mediated caste polymorphism and greatly improves protein coverage involved during honeybee caste determination. Hence, it is a major step forward in the analysis of the fundamental causes of honeybee caste pathway decision and greatly contributes to the knowledge of honeybee biology. In particular, the consistency between the 22 proteins and mRNA expressions provides us important target genes for the reverse genetic analysis of caste pathway modulation through RNA interference.  相似文献

Local communities play an increasingly important role in the management and conservation of forests at local and global scales. Conventional analyses of community forest management tend to view the outcomes of these efforts, as with common pool resources (CPRs) more generally, as contingent on the ability of local institutions to control collective levels of extractive use and enforce group rules. This paper provides a case study of a community forest in southern Michigan, in the Midwestern United States, that challenges these assumptions about community-based forest management. The factors driving change in this forest are not tied to excessive extraction or disturbance by human agents but rather the proliferation of shade-tolerant invasive species. The community institutions and values that made it possible for the forest to grow and mature now threaten its very existence. By discouraging any form of active management, the forest has become susceptible to the growing pressures of human-induced environmental change such as the introduction of exotic plant species. Biodiversity conservation in such contexts consequently relies not only on restraining local forest utilization practices or the preservation of land from development, but on active management interventions by local forest users. Understanding the impact of community management on CPRs in human-dominated ecosystems will require broadening the scope of analysis to account for the importance of active management and the potentially deleterious effects of preservationist approaches on native biota.

Drosophila''s dorsal closure provides an excellent model system with which to analyze biomechanical processes during morphogenesis. During native closure, the amnioserosa, flanked by two lateral epidermal sheets, forms an eye-shaped opening with canthi at each corner. The dynamics of amnioserosa cells and actomyosin purse strings in the leading edges of epidermal cells promote closure, whereas the bulk of the lateral epidermis opposes closure. Canthi maintain purse string curvature (necessary for their dorsalward forces), and zipping at the canthi shortens leading edges, ensuring a continuous epithelium at closure completion. We investigated the requirement for intact canthi during closure with laser dissection approaches. Dissection of one or both canthi resulted in tissue recoil and flattening of each purse string. After recoil and a temporary pause, closure resumed at approximately native rates until slowing near the completion of closure. Thus the amnioserosa alone can drive closure after dissection of one or both canthi, requiring neither substantial purse string curvature nor zipping during the bulk of closure. How the embryo coordinates multiple, large forces (each of which is orders of magnitude greater than the net force) during native closure and is also resilient to multiple perturbations are key extant questions.  相似文献
