We have made an extensive allozyme survey of 21 enzyme and protein loci in populations of the alpine grasshopper Podisma pedestris. This species occurs in two races, differing by a chromosomal fusion which separates the ancestral XO/XX race from a derived neo-XY race. These races also differ in DNA content, and hybrids between them have reduced viability. Electrophoresis reveals that the amount of genetic differentiation between these races is no greater than the variation among populations within each race. Both larger-scale surveys and a detailed survey of an area where the races hybridize, show that the chromosomal change is not correlated with gene frequency changes at any of the 21 loci studied. These findings are consistent with recently developed theory concerning the strength of the barrier to gene flow posed by a hybrid zone with characteristics such as those measured experimentally in Podisma. It is argued that hybrid zones in other species which involve allozymic differences do so because of stronger selection against hybrids rather than through mating isolation.  相似文献

The structure and function of the centrosomes from Chinese hamster ovary (CHO) cells were investigated by electron microscopy of negatively stained wholemount preparations of cell lysates. Cells were trypsinized from culture dishes, lysed with Triton X-100, sedimented onto ionized, carbon-coated grids, and negatively stained with phosphotungstate. The centrosomes from both interphase and dividing cells consisted of pairs of centrioles, a fibrous pericentriolar material, and a group of virus-like particles which were characteristic of the CHO cells and which served as markers for the pericentriolar material. Interphase centrosomes anchored up to two dozen microtubules when cells were lysed under conditions which preserved native microtubules. When Colcemid-blocked mitotic cells, initially devoid of microtubules, were allowed to recover for 10 min, microtubules formed at the pericentriolar material, but not at the centrioles. When lysates of Colcemid-blocked cells were incubated in vitro with micotubule protein purified from porcine brain tissue, up to 250 microtubules assembled at the centrosomes, similar to the number of microtubules that would normally form at the centrosome during cell division. A few microtubules could also be assembled in vitro onto the ends of isolated centrioles from which the pericentriolar material had been removed, forming characteristic axoneme- like bundles. In addition, microtubules; were assembled onto fragments of densely staining, fibrous material which was tentatively identified as periocentriolar material by its association of CHO can initiate and anchor microtubules both in vivo and in vitro.  相似文献

Two strains of a virus, designated cymbidium ringspot virus (CyRSV), were isolated from cymbidium orchids and from Trifolium repens respectively in Britain. Experimentally infected cymbidiums developed slight chlorotic ring-mottle; T. repens developed flecks and mottling in the leaves, and slight stunting. Of 101 plant species tested, the cymbidium strain infected sixty-one (thirteen systemically) in twenty-three of thirty-five families; the clover strain infected sixty-four species (eighteen systemically) in twenty-two families. Both strains were propagated in Nicotiana clevelandii and assayed in Chenopodium quinoa. CyRSV was readily transmitted by inoculation of sap, and by foliage contact between plants, but not by the aphids Myzus persicae or Acyrtho-siphon pisum, nor through seed of T. incarnatum, Phaseolus vulgaris or N. clevelandii. Highly infective virus was released into soil from roots of infected N. clevelandii, and acquired by bait seedlings planted in such soil. Similar transmission occurred when purified virus was applied to the surface of sterilized soil containing bait plants; there was no evidence for any living soil vector. The virus was eliminated from 96 % of small cuttings taken from infected N. clevelandii plants grown at 35–37 °C for 9 wk. CyRSV was still infective in sap of N. clevelandii after dilution to 10?5-io–6 (only 2 × 10_1 in cymbidium sap), or after 10min at 85–90 °C. It survived at least 10 months at c. 20 °C and more than 12 yr at 2 °C. Lyophilized sap was highly infective after over 13 yr at laboratory temperatures under high vacuum. Purified preparations made by clarification with n-butanol, followed by differential centrifugation and exclusion chromatography on controlled-pore glass beads, contained isometric particles c. 30 nm diam., with s°20W= 137 S, and had a buoyant density in caesium chloride of 1–36 g/ml. The A 260/A 280 ratio was 1–55, and A max(26o)/A min(242) was 1–17. The virus contained c. 15 % of single-stranded RNA of mol. wt 1–7 × 106; the nucleotide base ratios were: G27'8; A24/9; C2I-3; U26-I. There was one capsid polypeptide of mol. wt 43600. The virus was a good immunogen and a strongly reacting antigen in vitro; in Immunoelectrophoresis, each strain migrated as a single antigenic component towards the cathode. The cymbidium and clover strains were serologically closely related, although spurs were produced in immunodiffusion. No serological relationship was found to forty-three other isometric viruses, including eighteen tombusvirus isolates; CyRSV nevertheless shares many properties with tombusviruses, and we assign it provisionally to this group. Understanding nitrous oxide (N2O) and methane (CH4) fluxes from agricultural soils in semi‐arid climates is necessary to fully assess greenhouse gas emissions from bioenergy cropping systems, and to improve our knowledge of global terrestrial gaseous exchange. Canola is grown globally as a feedstock for biodiesel production, however, resulting soil greenhouse gas fluxes are rarely reported for semi‐arid climates. We measured soil N2O and CH4 fluxes from a rain‐fed canola crop in a semi‐arid region of south‐western Australia for 1 year on a subdaily basis. The site included N fertilized (75 kg N ha?1 yr?1) and nonfertilized plots. Daily N2O fluxes were low (?1.5 to 4.7 g N2O‐N ha?1 day?1) and culminated in an annual loss of 128 g N2O‐N ha?1 (standard error, 12 g N2O‐N ha?1) from N fertilized soil and 80 g N2O‐N ha?1 (standard error, 11 g N2O‐N ha?1) from nonfertilized soil. Daily CH4 fluxes were also low (?10.3 to 11.9 g CH4‐C ha?1 day?1), and did not differ with treatments, with an average annual net emission of 6.7 g CH4–C ha?1 (standard error, 20 g CH4–C ha?1). Greatest daily N2O fluxes occurred when the soil was fallow, and following a series of summer rainfall events. Summer rainfall increased soil water contents and available N, and occurred when soil temperatures were >25 °C, and when there was no active plant growth to compete with soil microorganisms for mineralized N; conditions known to promote N2O production. The proportion of N fertilizer emitted as N2O, after correction for emissions from the no N fertilizer treatment, was 0.06%; 17 times lower than IPCC default value for the application of synthetic N fertilizers to land (1.0%). Soil greenhouse gas fluxes from bioenergy crop production in semi‐arid regions are likely to have less influence on the net global warming potential of biofuel production than in temperate climates.  相似文献

Abstract: Rapid growth in off-highway vehicle (OHV) use in North America leads to concerns about potential impacts on wildlife populations. We studied the relationship between distance to active OHV trail and songbird nesting success and abundance in northeastern California, USA, from 2002 to 2004. We found evidence of greater nest desertion and abandonment and reduced predation on shrub nests <100 m from OHV trails than at nests >100 m from OHV trails. Two of 18 species studied were less abundant at sites on trails than at sites 250 m from trails, and no species were more abundant on trails. Management of OHV trail development should consider possible negative impacts on nesting success and abundance of breeding birds.  相似文献

Models of vegetation function are widely used to predict the effects of climate change on carbon, water and nutrient cycles of terrestrial ecosystems, and their feedbacks to climate. Stomatal conductance, the process that governs plant water use and carbon uptake, is fundamental to such models. In this paper, we reconcile two long‐standing theories of stomatal conductance. The empirical approach, which is most commonly used in vegetation models, is phenomenological, based on experimental observations of stomatal behaviour in response to environmental conditions. The optimal approach is based on the theoretical argument that stomata should act to minimize the amount of water used per unit carbon gained. We reconcile these two approaches by showing that the theory of optimal stomatal conductance can be used to derive a model of stomatal conductance that is closely analogous to the empirical models. Consequently, we obtain a unified stomatal model which has a similar form to existing empirical models, but which now provides a theoretical interpretation for model parameter values. The key model parameter, g1, is predicted to increase with growth temperature and with the marginal water cost of carbon gain. The new model is fitted to a range of datasets ranging from tropical to boreal trees. The parameter g1 is shown to vary with growth temperature, as predicted, and also with plant functional type. The model is shown to correctly capture responses of stomatal conductance to changing atmospheric CO2, and thus can be used to test for stomatal acclimation to elevated CO2. The reconciliation of the optimal and empirical approaches to modelling stomatal conductance is important for global change biology because it provides a simple theoretical framework for analyzing, and simulating, the coupling between carbon and water cycles under environmental change.  相似文献

Background

Porcine reproductive and respiratory syndrome virus (PRRSV) is the etiologic agent of PRRS, causing widespread chronic infections which are largely uncontrolled by currently available vaccines or other antiviral measures. Cultured monkey kidney (MARC-145) cells provide an important tool for the study of PRRSV replication. For the present study, flow cytometric and fluorescence antibody (FA) analyses of PRRSV infection of cultured MARC-145 cells were carried out in experiments designed to clarify viral dynamics and the mechanism of viral spread. The roles of viral permissiveness and the cytoskeleton in PRRSV infection and transmission were examined in conjunction with antiviral and cytotoxic drugs.

Results

Flow cytometric and FA analyses of PRRSV antigen expression revealed distinct primary and secondary phases of MARC-145 cell infection. PRRSV antigen was randomly expressed in a few percent of cells during the primary phase of infection (up to about 20–22 h p.i.), but the logarithmic infection phase (days 2–3 p.i.), was characterized by secondary spread to clusters of infected cells. The formation of secondary clusters of PRRSV-infected cells preceded the development of CPE in MARC-145 cells, and both primary and secondary PRRSV infection were inhibited by colchicine and cytochalasin D, demonstrating a critical role of the cytoskeleton in viral permissiveness as well as cell-to-cell transmission from a subpopulation of cells permissive for free virus to secondary targets. Cellular expression of actin also appeared to correlate with PRRSV resistance, suggesting a second role of the actin cytoskeleton as a potential barrier to cell-to-cell transmission. PRRSV infection and cell-to-cell transmission were efficiently suppressed by interferon-γ (IFN-γ), as well as the more-potent experimental antiviral agent AK-2.

Conclusion

The results demonstrate two distinct mechanisms of PRRSV infection: primary infection of a relatively small subpopulation of innately PRRSV-permissive cells, and secondary cell-to-cell transmission to contiguous cells which appear non-permissive to free virus. The results also indicate that an intact cytoskeleton is critical for PRRSV infection, and that viral permissiveness is a highly efficient drug target to control PRRSV infection. The data from this experimental system have important implications for the mechanisms of PRRSV persistence and pathology, as well as for a better understanding of arterivirus regulation.  相似文献
