Two new flavonoids from Derris laxiflora Benth

Two new compounds, derriscoumaronochromone (1) and cis-3,4′-dihydroxy- 5,7-dimethoxyflavan (2), as well as trans-4'-O-methylcatechin (3) were isolated from Derris laxiflora, The structures of these compounds were determined by analysis of their spectroscopic data.     

Factors affecting the extraction and the relative proportions of multiple forms of plant α-galactosidases

The extractability of α-galactosidase activity from mature Vicia faba seeds and the conversion of the low molecular weight form II to the larger oligomer I, was examined over a range of salt concentrations. Specific and total activities of the preparations were high when strong salt solutions were used. Extraction of α-galactosidase I, in comparison with II, requires solutions with a high ionic strength (e.g. 0.5 M NaCl). Interpretation of gel filtration patterns are, however, complicated by conversion of II to I which occurs under these conditions. This conversion is also enhanced by routine procedures used for enzyme purification, such as citric acid precipitation.     

Phytochrome controlled acid RNase: An “attached” protein of ribosomes

The light-dependent increment in RNase activity (which is ribosome bound in cell extracts) is distributed as a gradient increasing from base to hook of lupin hypocotyls. No evidence was found of non-specific or of specific activation of pre-formed enzyme molecules following isolation, either before or after (latent activity) destruction of particles. The autodegradation capacity of ribosomes isolated from irradiated cells was almost double that of ribosomes from etiolated tissue. It is proposed that association between the bulk of the light-controlled RNase fraction and lupin ribosomes results from binding of soluble protein. It is not clear whether binding is specific or an artifact of isolation.     

The phylogeny of Lens (Leguminosae): new insight from ITS sequence analysis

 Lens includes L. culinaris subsp. culinaris (the cultivated lentil) and several wild species distributed from the Mediterranean region to western Asia. We compared sequence variation in the ITS region among species of Lens in an effort to end persisting uncertainty regarding the phylogeny of the genus. The parsimony analysis revealed a single minimum-length tree with a topology congruent with patterns derived by previous studies of nuclear and chloroplast DNA RFLPs. The basal and highly divergent status of the L. nigricans clade is depicted, and the progenitor-derivative relationship between L. culinaris subsp. orientalis and L. culinaris subsp. culinaris is reaffirmed. Resolution in the tree was improved by combining the ITS data set with a pre-existing set of chloroplast DNA restriction site data obtained from the same group of samples. Isoaurmillone,an isoflavone from the pods of Milletia auriculata

The pods of Milletia auriculata have yielded a new substance, 5,7-dihydroxy-6-methoxy-4′-O-prenyloxyisoflavone.     

Characterization of gibberellins from light-grown Phaseolus coccineus seedlings by combined GC-MS

Following extensive purification of an extract from 5000 light-grown Phaseolus coccineus seedlings by procedures including countercurrent distribution, Sephadex G10, polyvinylpyrrolidone, charcoal-celite and silicic acid partition column chromatography, TLC preparative GLC, gibberellins A₁, A₄, A₅ and A₂₀ were characterized by combined GC-MS. In addition, an unknown compound isomeric with gibberellin A₁₇ but lacking an hydroxyl group, was also detected.     

2,4-cis-4,5-cis-4,5-Dihyroxypipecolic acid—a naturally occurring imino acid from Calliandra pittieri

2,4-cis-4,5-cis-4,5-Dihydroxypipecolic acid has been isolated from the leaves of Calliandra pittieri. This is the third dihydroxypipecolic acid isomer isolated from Calliandra and the first report of this compound from a natural source.     

Multiple forms of hydroxycinnamate : CoA ligase in etiolated pea seedlings

A survey of a range of plant tissues showed that the hydroxycinnamate CoA ligase in crude extracts of pea shoots had a high relative activity towards sinapic and other methoxycinnamic acids, together with high activity with p-coumaric acid. The pea enzyme has been resolved by chromatography on DEAE-cellulose into two peaks which differ in their substrate specificity. The form which elutes at relatively low salt concentrations has a ratio activity towards p-coumaric and sinapic acids of about 1.8:1 while the form eluting at higher salt concentrations, although showing very high activity with p-coumaric acid, is inactive towards sinapic acid. The pattern of elution of these forms following gel filtration on Ultragel AcA 34 and Sephadex G100 suggests that these two isoenzymes which differ in ionic properties and substrate specificity can exist in two or three molecular weight forms and there is evidence that these forms are under certain circumstances interconvertible.
