激光光漂恢复技术测定了异硫氰基荧光素标记的林蛀卵表面分子在第一次卵裂前的运动。发现固着在玻片上的剥离“细胞膜”的分子运动形式为扩散。扩散系数为(4.6±1.3)×10~(-12)cm~2/s,可动部份为15%。完整卵子上的分子运动形式为流动。细胞膜在不停地流动着。它可能起着协助细胞质运动的作用。细胞膜流动的速度随时间而异,卵裂前不久,在大多数的卵子上,出现两个流动较慢的谷,少数细胞只测到一个谷。这可能与光漂起始时间,光斑与未来分裂沟的距离,和卵子间的差异有关。也讨论了这种速度变化与表面收缩波的关系。 相似文献

“Brush structures” are temporary wooden structures built with unmodified local materials and used as shelters by First Nation Peoples in the forests of the Yukon prior to European contact. This paper reports a preliminary attempt to date these structures using dendrochronology. Investigations were carried out of four njel (“teepee like”) structures and eight män-ku (low 2–3 sided wall structures) at four main sites. The primary material cored was poles (dead spruce trunks), often only 10–20 cm diameter, with narrow, sometimes extremely suppressed ring sequences. These structures are dated between 1865 and 1887, based on the latest (outermost) ring in the sampled material. The limited sampling and use of old wood in these structures (whether fire-kill, standing dead or reused from previous features) makes it difficult to give precise dates for the initial evidence of First Nation activity at these sites: more extensive sampling could provide further insight into the settlement history and construction techniques used. Summary This paper describes the development of pollen grains ofGasteria verrucosa from the late microspore to the mature two-cellular pollen grain. Ultrastructural changes and the distribution of plastids as a result of the first pollen mitosis have been investigated using light and electron microscopy. The microspores as well as the generative and the vegetative cell contain mitochondria and other cytoplasmic organelles during all of the observed developmental stages. In contrast, the generative cell and the vegetative cell show a different plastid content. Plastids are randomly distributed within the microspores before pollen mitosis. During the prophase of the first pollen mitosis the plastids become clustered at the proximal pole of the microspore. The dividing nucleus of the microspore is located at the distal pole of the microspore. Therefore, the plastids are not equally distributed into both the generative and the vegetative cell. The possible reasons for the polarization of plastids within the microspore are briefly discussed. The lack of plastids in the generative cell causes a maternal inheritance of plastids inGasteria verrucosa. 相似文献

The main characteristics of the dominant economic system, including the increasing use of markets and money are described. The global system has expanded trade, including international trade, and production tremendously. While this system has the potential to favour nature conservation, in practice the opposite has occurred. Difficulties raised for conservation of biodiversity by short-term economic crises such as deficits in a country's international payments, the adoption of policies for structural economic adjustment, international capital flows, international loans and foreign aid as well as debt-for-nature swaps are discussed. As explained, it is politically difficult in market economies to support nature conservation at the expense of economic growth and as more economies develop and become market economies this problem spreads. Given global interdependence of nations, an important issue is the distribution of net benefits from biodiversity conservation between developed and less developed countries. Possible distributions of benefits and related issues are discussed. In conclusion, the importance of political lobbying by nature conservation groups in developed market economies is emphasised as a means of ensuring correction of market failures. Unfortunately, no economic system is likely to prove satisfactory in itself in conserving biodiversity so political action by conservationists is always required. 相似文献

Isolated rat pancreatic islets, incubated in the presence of extracellular 32Pi to steady state 32P incorporation into cellular phosphopeptides, were exposed to glucose for 10 min. Glucose (16.7 mM) significantly stimulated the phosphorylation of six phosphoproteins with molecular weights of 15,000, 35,000, 49,000, 64,000, 93,000 and 138,000. Mannoheptulose (16.7 mM) markedly inhibited glucose-stimulated phosphorylation of these six phosphoproteins. The lac region of Escherichia coli, carried on an F′ lacproB episome, was used as a target for the transposition of several transposable elements. Tn9 shows a preferential integration (by a factor of 50) into a region extending from the end of the Z gene through the Y gene. Throughout the remainder of the lacI, Z and Y genes one other short region, located in the middle of the I gene, is favored for integration. Within these favored regions many different integration points are evident. Inspection of the DNA sequence for the I and Y genes, and parts of the Z gene, shows a strong correlation between A + T richness and regions of preferential integration. Tn5 insertions follow a similar pattern, although with less preference; whereas Tn10 insertions (provided by T. J. Foster), also favor the Y gene and the end of Z, but are distributed among fewer integration points. Most of the Tn3 insertions into the episome are accompanied by a nearby or adjacent deletion. 相似文献

Direct evidence was obtained for the presence of hormone-stimulated trypsin-like protease activity in the rat uterus. Ovariectomized rats were either untreated (U), treated with estradiol (E), or estradiol plus progesterone (EP). The uteri were excised and subcellular fractions were prepared. Each fraction was assayed for protease activity using protamine as substrate, the cleavage products being quantitated fluorometrically following reaction with 4-phenylspiro[furan-2(3H),1′-phthalan]-3,3′dione (Fluram). Fractions from U rats yielded negative results, whereas the 12,000g pellets and nuclei from the uteri of E and EP rats exhibited appreciable activities. No significant increase in protease activity was observed in thymus and diaphragm following hormone treatment, indicating organ specificity. The enzyme (or enzymes) from the 12,000g pellet was solubilized and some characteristics were determined. The apparent Km is about 1.0 × 10?6m, the temperature optimum is about 44 °C and maximum velocity is achieved in the alkaline range (pH ~ 8.5). The protease is a plasminogen activator and is inhibited by diisopropyl fluorophosphate, Antipain, and Leupeptin. These properties resemble those of trypsin. 相似文献
