Cynaropicrin is a guaianolide sesquiterpene lactone with a 5-7-5 tricyclic skeleton, four exo-olefins, and two hydroxyl groups. Recently, it was found that the compound is a potent in vitro and in vivo inhibitor of the protozoan parasite Trypanosoma brucei, which causes human African trypanosomiasis (HAT; sleeping sickness). In this Letter, chemical derivatization of cynaropicrin and the structure–activity-relationship (SAR) study against T. brucei is described.  相似文献

长期施肥对紫色土农田土壤动物群落的影响   总被引:6,自引:0,他引:6  
土壤动物在陆地生态系统物质循环和能量流动中起着重要作用,直接或间接的参与土壤有机质的分解与矿化;长期施肥对土壤理化性质产生影响的同时,改变了土壤动物群落组成.为查明紫色土长期施肥对土壤动物群落的影响及其响应关系,于2008年的5、7、9和11月分别对紫色土农田无肥对照(CK)、单施氮肥(N)、常规化肥氮磷钾(NPK)、有机肥(OM)、有机肥与化肥氮磷钾混施(OMNPK)、秸秆还田(RSD)和秸秆还田与化肥氮磷钾混施(RSDNPK)等7种长期施肥定位试验地的土壤动物群落进行调查,采用改良的干漏斗和湿漏斗两种方法,共获得土壤动物9454只,隶属7门17纲24目.分析表明,OM和RSDNPK两种施肥方式下土壤动物群落的多样性显著高于CK、N和NPK等3种施肥方式,说明有机物料的长期投入有利于提高土壤动物群落丰富度和多样性.方差分析表明施肥方式对土壤动物主要类群密度的影响差异性极显著(F=42.412,P=0.0001),对土壤动物群落类群影响存在不均衡性.施肥方式主要影响农田土壤动物类群的种群个体数量、线虫动物门个体数量、大蚓类个体数量、甲螨亚目个体数量、密度-类群指数DG及土壤动物群落类群数等六个指标,初步认为这些主要类群因素能够预测长期施肥引起的土壤肥力变化,可能对指示土壤质量的变化具有一定潜力.  相似文献

A direct electrochemical DNA biosensor based on zero current potentiometry was fabricated by immobilization of ssDNA onto gold nanoparticles (AuNPs) coated pencil graphite electrode (PGE). One ssDNA/AuNPs/PGE was connected in series between clips of working and counter electrodes of a potentiostat, and then immersed into the solution together with a reference electrode, establishing a novel DNA biosensor for specific DNA detection. The variation of zero current potential difference (ΔE(zcp)) before and after hybridization of the self-assembled probe DNA with the target DNA was used as a signal to characterize and quantify the target DNA sequence. The whole DNA biosensor fabrication process was characterized by cyclic voltammetry and electrochemical impedance spectroscopy with the use of ferricyanide as an electrochemical redox indicator. Under the optimized conditions, ΔE(zcp) was linear with the concentrations of the complementary target DNA in the range from 10nM to 1μM, with a detection limit of 6.9nM. The DNA biosensor showed a good reproducibility and selectivity. Prepared DNA biosensor is facile and sensitive, and it eliminates the need of using exogenous reagents to monitor the oligonucleotides hybridization.  相似文献

【背景】盐胁迫环境严重影响大豆幼苗生长,内生菌可提高作物的抗逆性。【目的】探究接种内生枯草芽孢杆菌127和解蛋白芽孢杆菌133对盐胁迫下大豆幼苗体内超氧化物歧化酶(superoxide dismutase,SOD)和过氧化物酶(peroxidase,POD)活性的影响。【方法】以“徐豆20”为实验材料,采用盆栽实验法,设置对照组、盐胁迫组和盐胁迫接菌组,在人工气候培养条件下,用不同NaCl浓度(50、100、150、200、250和300 mmol/L)处理大豆幼苗,并接种不同OD600值(OD0.33、OD0.50和OD0.75)的菌悬液。【结果】培养14 d,接种枯草芽孢杆菌127的菌悬液OD0.33和OD0.75分别在盐浓度300 mmol/L和100 mmol/L时,SOD活性均为1.04 U/g-FW;接种解蛋白芽孢杆菌133的菌悬液OD0.50在盐浓度300 mmol/L胁迫下POD活性最高为7 820 U/(g·min),对大豆幼苗修复效果较显著。培养28 d,接种枯草芽孢杆菌127的菌悬液OD0.50,在150 mmol/L时SOD活性最高(0.88 U/g-FW);接...  相似文献

Chamaecrista mimosoides is an annual herb legume widely distributed in tropical and subtropical Asia and Africa. It may have primitive and independently-evolved root nodule types but its rhizobia have not been systematically studied. Therefore, in order to learn the diversity and species affinity of its rhizobia, root nodules were sampled from C. mimosoides plants growing in seven geographical sites along the coast line of Shandong Peninsula, China. A total of 422 rhizobial isolates were obtained from nodules, and they were classified into 28 recA haplotypes. By using multilocus sequence analysis of the concatenated housekeeping genes dnaK, glnII, gyrB, recA and rpoB, the representative strains for these haplotypes were designated as eight defined and five candidate novel genospecies in the genus Bradyrhizobium. Bradyrhizobium elkanii and Bradyrhizobium ferriligni were predominant and universally distributed. The symbiotic genes nodC and nifH of the representative strains showed very similar topology in their phylogenetic trees indicating their co-evolution history. All the representative strains formed effective root nodules in nodulation tests. The correlation between genospecies and soil characteristics analyzed by CANOCO software indicated that available potassium (AK), organic carbon (OC) and available nitrogen (AN) in the soil samples were the main factors affecting the distribution of the symbionts involved in this current study. The study is the first systematic survey of Chamaecrista mimosoides-nodulating rhizobia, and it showed that Chamaecrista spp. were nodulated by bradyrhizobia in natural environments. In addition, the host spectrum of the corresponding rhizobial species was extended, and the study provided novel information on the biodiversity and biogeography of rhizobia.  相似文献

溶磷性大豆根瘤内生菌的筛选、抗性及系统发育和促生   总被引:4,自引:0,他引:4  
对采自河南省不同地区的大豆根瘤进行内生菌分离纯化、溶磷性筛选试验。根据能否产生溶磷圈及溶磷圈直径(D)、菌落直径(d)和D/d值大小确定菌株溶磷能力,采用钼锑抗比色法测定培养液中有效磷含量;平板筛选法对筛选菌株进行耐盐性、耐酸碱、重金属等抗性测定,并对筛选菌株进行理化特性、16S r DNA、rec A序列和系统发育分析。结果表明,从分离纯化的324株内生菌中筛选出36株具有溶磷特性,其中20株有较强溶磷性。菌株DD291发酵液中可溶性磷含量最高(452 mg/L),发酵液p H与对照相比均有不同程度下降,最大降幅达2.92。大部分溶磷性内生菌具有较强耐盐碱性,对Pb2+、Cr6+和Cu2+有较高耐受性,对Ni2+和Hg2+抗性较弱。结合细胞形态、生理生化、16S r DNA、rec A序列和系统发育分析结果,菌株确定为Bacillus cereus,Enterobacter cancerogenus,E.cloacae和Pseudomonas putida。部分溶磷菌株对大豆的生长有促进作用,显示出潜在的应用前景。  相似文献

目的探讨普罗布考防治动脉粥样硬化(AS)的机制。方法选用雄性大鼠,复制大鼠AS模型,随机分为动脉粥样硬化模型组、普罗布考组和正常对照组。大鼠造模成功后给予普罗布考治疗,6周后处死大鼠,采用流式细胞术检测平滑肌细胞凋亡率及凋亡相关基因p53和Fas蛋白的表达。结果模型组大鼠血管平滑肌细胞凋亡率明显高于对照组(P〈0.05),p53和Fas蛋白的表达增强(P%0.05),主动脉壁可肉眼观测典型斑块。普罗布考组大鼠平滑肌细胞的凋亡率明显低于模型组(P〈0.05),p53和Fas蛋白表达下调(P〈0.05),主动脉斑块面积较模型组减小明显。结论普罗布考通过调节p53和Fas蛋白表达来调节AS大鼠平滑肌细胞的凋亡。  相似文献

【目的】采用优良抗病性内生菌资源来控制棉花枯萎病是一种有效的措施。本研究从大豆根瘤中筛选棉花枯萎病拮抗性内生细菌,探索其对棉花枯萎病菌丝的抑制作用和代表菌株特性,为发掘和应用防病、抗逆优良菌株提供理论基础。【方法】采用对峙法和代谢液培养法对大豆根瘤内生细菌进行棉花枯萎病菌抑菌性筛选,显微观察法研究筛选菌株引起病原菌菌丝变化,通过菌株培养特征、理化特性和16S r DNA序列同源性分析确定菌株系统发育地位,比色法测定DD174耐盐碱性,盆栽试验验证防病效果。【结果】经复筛和代谢液试验有5株拮抗性较强菌株,被作用病原菌菌丝畸形、细胞壁消失、自溶,菌丝基部加粗、分支增多,呈树根状;菌丝被菌苔包埋而溶解、断裂,菌丝末端球形膨大。对棉花枯萎病菌的抑制作用主要通过菌体产生胞外代谢物发挥作用。菌株DD174、DD176和DD179最相似菌株分别为Bacillus oceanisediminis H2T(GQ292772)和B.thuringiensis ATCC 10792T(AF290545),菌株DD165和DD166最相似菌株均为Stenotrophomonas maltophilia LMG 958T(X95923)。DD174能耐受6%盐浓度,p H 10生长良好,具有一定耐盐碱能力。DD174处理组防治效果达76.32%,其他防效均在62%以上,可作为棉花枯萎病的生防菌株资源。【结论】大豆根瘤内存在棉花枯萎病内生拮抗细菌,其中有些菌株具有一定耐盐碱能力,对棉花枯萎病病原菌及病害有一定抑菌和防病作用。  相似文献

Semi-natural grassland communities are of great interest in conservation because of their high species richness. These communities are being threatened by both land abandonment and nitrogen eutrophication, and their continued existence will depend upon correct management. However, there is a distinct lack of studies of the ecological mechanisms that regulate species diversity and productivity in Mediterranean grasslands. We have conducted a 3-year field experiment in a species-poor grassland in central Italy to investigate the effects of nitrogen fertilization coupled with removal of plant litter and artificial cutting on species diversity and community productivity. Vegetation cutting reduced living biomass but increased species diversity. In fact, cutting had positive effects on the cover of almost all of the annual and biennial species, while it had a negative effect on the dominant perennial grasses Brachypodium rupestre and Dactylis glomerata. Litter removal had similar effects to cutting, although it was far less effective in increasing species diversity. In contrast, nitrogen enrichment strongly increased the living biomass while maintaining very low species diversity. Our results have indicated that semi-natural Mediterranean grasslands need specific management regimes for maintenance and restoration of species diversity. In the management of these grasslands, attention should be paid to the potential threat from nitrogen enrichment, especially when coupled with land abandonment.  相似文献

利用封顶式生长室模拟未来变化的气候条件,研究了亚高山林线优势物种岷江冷杉(Abies faxoniana)和4种草本植物形态与竞争指标对CO2浓度和温度升高的响应.结果表明:处理2个生长季后,高CO2浓度条件下,岷江冷杉冠体积增加42%,比叶面积、比冠体积和比根长分别增加17%、65%和19%;温度升高使岷江冷杉冠形更纵向生长,冠体积增加22%,根冠比和比根长均比对照增加17%;二者同时升高使岷江冷杉冠体积增加79%,比叶面积、比冠体积和比根长分别增加17%、197%和18%.CO2浓度升高处理下糙野青茅(Deyeuxia scabrescen)的株高、基茎和每株叶片数增加,但比叶面积降低;甘肃苔草(Carexkansuensis)、东方草莓(Fragaria orientali)和紫花碎米荠(Cardamine tangutorum)的各项指标变化与青茅相反.温度升高下青茅、苔草、草莓株高、基茎和根冠比下降.二者同时升高条件下4种草本植物的基茎和每株叶片数增加,但比叶面积和根冠比降低.这表明,在CO2浓度和温度升高处理下,岷江冷杉形成有利于生长的冠层结构且单位质量的竞争力增加,而4种草本植物的形态结构和竞争力均受到不同程度的负面影响.  相似文献
