Large conductance Ca2+-activated potassium channels (BK) are targets for research that explores therapeutic means to various diseases, owing to the roles of the channels in mediating multiple physiological processes in various cells and tissues. We investigated the pharmacological effects of curcumin, a compound isolated from the herb Curcuma longa, on BK channels. As recorded by whole-cell patch-clamp, curcumin increased BK (α) and BK (α+β1) currents in transfected HEK293 cells as well as the current density of BK in A7r5 smooth muscle cells in a dose-dependent manner. By incubating with curcumin for 24 hours, the current density of exogenous BK (α) in HEK293 cells and the endogenous BK in A7r5 cells were both enhanced notably, though the steady-state activation of the channels did not shift significantly, except for BK (α+β1). Curcumin up-regulated the BK protein expression without changing its mRNA level in A7r5 cells. The surface expression and the half-life of BK channels were also increased by curcumin in HEK293 cells. These effects of curcumin were abolished by MG-132, a proteasome inhibitor. Curcumin also increased ERK 1/2 phosphorylation, while inhibiting ERK by U0126 attenuated the curcumin-induced up-regulation of BK protein expression. We also observed that the curcumin-induced relaxation in the isolated rat aortic rings was significantly attenuated by paxilline, a BK channel specific blocker. These results show that curcumin enhances the activity of the BK channels by interacting with BK directly as well as enhancing BK protein expression through inhibiting proteasomal degradation and activating ERK signaling pathway. The findings suggest that curcumin is a potential BK channel activator and provide novel insight into its complicated pharmacological effects and the underlying mechanisms.  相似文献

This paper deals with a change in abundance of a population of Fritillaria meleagris L. The results of the investigation were compared with those of a study performed about 40 yr ago. The comparison showed both a change in the vegetation and in the abundance of the fritillary along a topographical gradient. The study reported here was a transplant experiment carried out in order to evaluate the hypothesis that regulation of the water levels of an adjacent lake was the main reason for the change. The transplantations showed that the performance of the fritillary was negatively affected by the high ground water levels and inundations of the site where the fritillary earlier was most abundant.  相似文献

Rapid urbanization in major cities has adversely affected avian biodiversity in both developed and developing countries. Due to over-urbanization and little regard for nature conservation, such cities are usually characterized by poor biodiversity. In contrast to central business district areas, suburbs may support greater levels of biodiversity through an increase in green areas. We examined urban bird species richness, abundance, and composition in Klang Valley, Peninsular Malaysia. We surveyed 141 points for passerines across 80 parks grouped into two different urban zones, namely central business districts and suburbs. Our results revealed that bird richness did not differ significantly (p = 0.994) between central business districts and the suburbs. We found that the abundances of birds were significantly greater in the suburbs than in central business districts. However, species composition was similar between the central business districts and suburbs. We also found that bird richness increased significantly with an increase in size of green areas and park areas, as well as the number of woody trees in both zones. Evidence from this study suggests that creating urban parks and gardens, coupled with tree planting in central business districts, is able to support greater biodiversity, at least for birds in cities similar to those found in the suburbs.  相似文献

Yeast cells (Saccharomyces cerevisiae) and invertase enzyme were immobilized in thiophene capped poly(ethyleneoxide)/polypyrrole (PEO-co-PPy) and 3-methylthienyl methacrylate-co-p-vinyl benzyloxy poly(ethyleneoxide)/polypyrrole (CP-co-PPy) matrices. Immobilization of the enzyme and yeast cells was performed via entrapment in conducting copolymers during electrochemical polymerization of pyrrole through the thiophene moiety of the polymers. Maximum reaction rates, Michaelis–Menten constants, optimum temperature and pH values, operational and storage stabilities of the enzyme and yeast cell electrodes were investigated.  相似文献

We used a non-invasive TOBEC method (Total Body Electric Conductivity) to estimate lean body mass and fat content in mouse-eared batsMyotis myotis (Borkhausen, 1797) hibernating in Poznañ Forts (W Poland) and in a semi-natural cave-mine Miedzianka (SE Poland). In December, fat content averaged 5.5 g in females (body mass = 29.4 g) and 5.3 g in males (body mass = 28.4 g). At the end of hibernation (April), fat content averaged 2.2 g in females (body mass = 25.6 g) and 1.4 g in males (body mass = 23.7 g). Fat content did not differ between the localities either in December or in April, but the pattern of changes of fat content was different. We calculated the rate of energy expenditure in hibernating bats using two methods, based on independent samples (fat content in first-time captured individuals) and based on paired observations (changes of fat content in re-captured individuals), and discussed problems associated with the two approaches. Both methods show that the bats need about 4.9 g of fat (191 kJ) to sustain a 165-day hibernation. However, the rate of fat usage varied considerably between the sites and hibernation phase. Although the average amount of fat remaining in April would be sufficient to support at least six more weeks of hibernation, the level of reserves was close to zero in some individuals.  相似文献

Because estuaries are dynamic on many temporal scales, it is difficult to differentiate long-term shifts from those resulting from erratic pulse events like severe storms. Diatoms are abundant and diverse in estuaries, and may characterize these dynamics across relevant time scales. The climate of south Florida is subtropical, with dry winters and wet summers that may also harbor tropical storms and cyclones. We investigated whether these repeated seasonal drivers of change in estuaries differ from the influence of tropical cyclones on planktonic and benthic diatom assemblages. Diatom assemblages and environmental parameters were measured in the Charlotte Harbor watershed of southwest Florida to identify differences between the wet and dry seasons and changes that occurred following Tropical Storm Debby in 2012. Indicator taxa that were significantly affiliated with each season and post-storm conditions were identified. Diatom assemblages across the watershed were similar in wet and dry seasons, but differences increased following the tropical storm. The reduction in community dispersion following the storm was pronounced in each of the major drainages of the estuary. There were spatially-distinct responses of diatoms to environmental changes driven by the storm. These results suggest that past storm activity could be detected using diatoms preserved in estuarine sediments using both single indicator-species as well as community approaches.  相似文献

Docetaxel resistance remains one of the main problems in clinical treatment of metastatic prostate cancer (PCa). Previous studies identified differently expressed lncRNAs in docetaxel-resistant PCa cell lines, while the potential mechanisms were still unknown. In the present study, we found NEAT1 was expressed at high levels in docetaxel-resistant PCa clinical samples and related cell lines. When knockdown NEAT1, cell proliferation and invasion in docetaxel-resistant PCa cells in vitro and in vivo were downregulated. Our further researches explained that NEAT1 exerts oncogenic function in PCa by competitively ‘sponging’ both miR-34a-5p and miR-204-5p. Inhibition of miR-34a-5p or miR-204-5p expression mimics the docetaxel-resistant activity of NEAT1, whereas ectopic expression of miR-34a-5p or miR-204-5p attenuates the anti-drug function of NEAT1 in PCa cells. Besides, we also found ACSL4 is a target of both miR-34a-5p and miR-204-5p, and ACSL4 was also inhibited by miR-34a-5p and miR-204-5p. Moreover, suppression of miR-34a-5p or/and miR-204-5p greatly restrained the expression of ACSL4 upon NEAT1 overexpression. Joint expression of miR-34a-5p and miR-204a-5p synergistically decreased the expression of ASCL4, indicating miR-34a-5p and miR-204a-5p collaboratively inhibit the expression of ACSL4. Innovatively, we concluded that NEAT1 contributes to the docetaxel resistance by increasing ACSL4 via sponging miR-34a-5p and miR-204-5p in PCa cells.  相似文献

Aiming to identify plants, we propose to evaluate the color texture of the palisade parenchyma, from microscopic images of leaf cross-sections, using a graph based approach. Our texture analysis approach models the image texture as a graph and uses measurements computed from the shortest paths between specific vertices to provide a feasible texture signature. For a more consistent evaluation, we compared our approach to different methods for color texture analysis in a texture classification experiment. The results obtained indicate that our approach is the most suitable for this histological analysis as it surpassed all the other texture approaches using Linear Discriminant Analysis, and obtained the second best accuracy using 1-Nearest Neighbor. These results also corroborate the feasibility of using both histological (as the palisade parenchyma) and computer analysis for identification and delimitation of plant taxa.  相似文献
