Summary Information is limited on soil contamination of leaves from field-grown row crops, especially with respect to aluminum (Al) analyses. The objective of this study was to determine the influence of washing leaf samples with either deionized water or detergent solution on elemental analyses for several agronomic crop plants. The crop plants sampled were corn (Zea mays L.), soybean (Glycine max L. Merr.), grain sorghum (Sorghum bicolor L. Moench), and wheat (Triticum aestivum L.). The crops were grown on a range of soil types, soil pH values, and tillage practices. Samples of upper leaves and lower leaves were collected separately. The samples were either not washed, washed with deionized water, or washed with detergent solution. After drying, grinding, and digesting, the samples were analyzed for Al, nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), iron (Fe), manganese (Mn), zinc (Zn), and copper (Cu). For all crop plants and conditions studied, there was no effect on measured N, P, K, Ca, Mg, Mn, Zn, or Cu concentrations, but measured Al and Fe concentrations were influenced by washing. In general, washing had a greater effect on Al analyses than on Fe analyses. Soybean samples were most affected by washing, while wheat samples seemed to be least affected. The results reflected greater contamination of lower leaves than upper leaves. Decontamination procedures appear necessary prior to Al and Fe analyses of field-grown crop plants.  相似文献

Summary The effects of P and Mn on growth response and uptake of Fe, Mn and P by grain sorghum were investigated using nutrient culture. High P and Mn concentrations in solution (greater than 40 and 1 mg/l for P and Mn, respectively) markedly reduced plant height and shoot and root dry weight of 4-week-old sorghum plants. High Mn concentrations in solution increased the concentrations of Mn and P in shoot tissue and uptake of Mn, but depressed the uptake of P. High levels of P enhanced Mn uptake by sorghum and accentuated Mn toxicity at low Mn levels. The tissue Fe and total uptake of Fe were both reduced markedly by the high levels of P and Mn concentrations in solution. The increases of P, Mn and Fe concentrations in root tissue with a concomitant decrease of Fe in shoots suggested that the translocation of Fe from roots to shoots was hindered under high P and Mn conditions. Since coating occurred on root surfaces and intensified with increasing Mn concentrations in the substrate, part of the reduction of Fe in shoots could be attributed to the formation of high valent manganese oxides on the root surfaces which may retain Fe and reduce its absorption by sorghum.Contribution from the Department of Agronomy and Range Sci., University of California, Davis, CA.  相似文献

Summary Amino acid composition of proteins from anthers of milo and Indian origin male steriles were determined. Comparison of amino acid between A and B lines showed lower contents of histidine, threonine, glutamic acid, glycine, leucine and phenylalanine and higher contents of alanine, serine, proline and tyrosine in line A compared to line B. Alanine content in anthers of A lines was more than two fold higher than that in the anthers from B lines. Marked differences in amino acid composition of anthers of A and B lines are suggestive of their involvement in male sterility. Cytoplasmic male steriles of Indian origin M35-1A and M31-2A showed greater similarity but differed from milo, VZM2A and B.  相似文献

Carboxylesterase activities toward pesticide esters in crops and weeds   总被引:1,自引:0,他引:1  
Proteins were extracted from maize, rice, sorghum, soybean, flax and lucerne; the weeds Abutilon theophrasti, Echinochloa crus-galli, Phalaris canariensis, Setaria faberii, Setaria viridis, Sorghum halepense and the model plant Arabidopsis thaliana and assayed for carboxylesterase activity toward a range of xenobiotics. These included the pro-herbicidal esters clodinafop-propargyl, fenoxaprop-ethyl, fenthioprop-ethyl, methyl-2,4-dichlorophenoxyacetic acid (2,4-d-methyl), bromoxynil-octanoate, the herbicide-safener cloquintocet-mexyl and the pyrethroid insecticide permethrin. Highest activities were recorded with alpha-naphthyl acetate and methylumbelliferyl acetate. Esters of p-nitrophenol were also readily hydrolysed, with turnover declining as the chain length of the acyl component increased. Activities determined with model substrates were much higher than those observed with pesticide esters and were of limited value in predicting the relative rates of hydrolysis of the crop protection agents. Substrate preferences with the herbicides were typically 2,4-d-methyl>clodinafop-propargyl>fenthioprop-ethyl, fenoxaprop-ethyl and bromoxynil-octanoate. Isoelectric focussing in conjunction with staining for esterase activity using alpha-naphthyl acetate as substrate confirmed the presence of multiple carboxylesterase isoenzymes in each plant, with major qualitative differences observed between species. The presence of serine hydrolases among the resolved isoenzymes was confirmed through their selective inhibition by the organophosphate insecticide paraoxon. Our studies identify potentially exploitable differences between crops and weeds in their ability to bioactivate herbicides by enzymic hydrolysis and also highlight the usefulness of Arabidopsis as a plant model to study xenobiotic biotransformation.  相似文献

Summary The use of plant genetic engineering to augment plant breeding programs is significantly strengthened if novel trait(s) can be introduced directly into elite germplasm. Implementing this technology to sorghum breeding programs has been hampered by the lack of an efficient and transferable protocol that is suitable with elite genotypes. This study was conducted to identify parameters that maximize in vitro culture performance in sorghum targeting a specific elite genotype, C2-97, which possesses enhanced agronomic characteristics. Three different tissue culture media formulations, MS, N6, and M11 were evaluated. M11 medium contains approximately 16% and 85% more total nitrogen and sevenfold and threefold higher levels of potassium phosphate than MS and N6 formulations, respectively. Culture performance of C2-97 across the three media formulations was compared to sorghum genotypes that were previously reported to be amenable to genetic engineering, namely Tx430, P898012. Bwheatland, and C401. Maximum embryogenesis induction was observed on M11 medium for all genotypes tested, with greater than 70% embryogenic calluses occurring on immature embryos derived from the C2-97 genotype cultured on M11 medium.  相似文献

Most studies of tiller development have not related the physiological and morphological features of each culm to its subsequent fertility. This introduces problems when trying to account for the effects of tillering on yield in crop models. The objective of this study was to detect the most likely early determinants of tiller fertility in sorghum by identifying hierarchies for emergence, fertility and grain number of tillers over a wide range of assimilate availabilities. Emergence, phenology, leaf area development and dry weight partitioning were quantified weekly for individual tillers and main culms of tillering and uniculm plants grown at one of four densities, from two to 16 plants m(-2). For a given plant in any given density, the same tiller hierarchy applied for emergence of tillers, fertility of the emerged tillers and their subsequent grain number. These results were observed over a range of tiller fertility rates (from 7 to 91%), fertile tiller number per plant at maturity (from 0.2 to 4.7), and tiller contribution to grain yield (from 5 to 78 %). Tiller emergence was most probably related to assimilate supply and light quality. Development, fertility and contribution to yield of a specific tiller were highly dependent on growing conditions at the time of tiller emergence, particularly via early leaf area development of the tiller, which affected its subsequent leaf area accumulation. Assimilate availability in the main culm at the time of tiller emergence was the most likely early determinant of subsequent tiller fertility in this study.  相似文献

The prediction of tillering is poor or absent in existing sorghum crop models even though fertile tillers contribute significantly to grain yield. The objective of this study was to identify general quantitative relationships underpinning tiller dynamics of sorghum for a broad range of assimilate availabilities. Emergence, phenology, leaf area development and fertility of individual main culms and tillers were quantified weekly in plants grown at one of four plant densities ranging from two to 16 plants m(-2). On any given day, a tiller was considered potentially fertile (a posteriori) if its number of leaves continued to increase thereafter. The dynamics of potentially fertile tiller number per plant varied greatly with plant density, but could generally be described by three determinants, stable across plant densities: tiller emergence rate aligned with leaf ligule appearance rate; cessation of tiller emergence occurred at a stable leaf area index; and rate of decrease in potentially fertile tillers was linearly related to the ratio of realized to potential leaf area growth. Realized leaf area growth is the measured increase in leaf area, whereas potential leaf area growth is the estimated increase in leaf area if all potentially fertile tillers were to continue to develop. Procedures to predict this ratio, by estimating realized leaf area per plant from intercepted radiation and potential leaf area per plant from the number and type of developing axes, are presented. While it is suitable for modelling tiller dynamics in grain sorghum, this general framework needs to be validated by testing it in different environments and for other cultivars.  相似文献

Summary Cefotaxime ( 50 and 100 mg/1 ), a cephalosporin antibiotic and the amino acids asparagine and proline (200 mg/l) enhanced the production of embryogenic callus, increased the frequency of plant regeneration, and delayed the loss of regeneration potential in immature embryo-derived callus cultures ofSorghum bicolor (L.) Moench. Although these compounds did not promote callus induction or growth of callus, they influenced plant regeneration considerably in 10 low responding genotypes of grain and high anthocyanin containing sweet sorghums.  相似文献
