Aims: To investigate the effect of glycine betaine (GB) on the survival of Listeria monocytogenes on leaf surfaces under low relative humidity (RH). Methods and Results: The addition of GB (≥25 mmol l?1) improved the survival of L. monocytogenes under low RH on parsley leaves, thus suggesting that GB can improve the tolerance of L. monocytogenes to desiccation. Ten times less GB was needed to improve L. monocytogenes survival under low RH on nonbiological surfaces compared with parsley leaves, suggesting that, on the leaf surface, L. monocytogenes may have to compete for the available GB with autochthonous bacteria and/or the plant itself. Wild type and mutants carrying deletions in the three GB uptake systems, BetL, Gbu and OpuC, behaved similarly with and without added GB on parsley leaves (P > 0·05). In addition, preaccumulation of GB, triggered by osmotic stress prior to inoculation, failed to improve survival under low RH compared with osmotic stress without GB accumulation. Conclusions: Exogenous GB had a protective effect on L. monocytogenes cells from desiccation during survival on parsley leaves. This effect was independent of intracellular GB accumulation by the known uptake systems. Significance and Impact of the Study: Presence of GB could improve the survival of L. monocytogenes to desiccation on leaf surfaces and nonbiological surfaces.  相似文献

栗山天牛Massicus raddei (Blessig)是危害我国东北辽东栎Quercus liaotungensis和蒙古栎Q.mongolicus天然林的重大害虫,生活周期长,3a发生1代,而且生活史整齐,在东北林区的辽宁、吉林和内蒙发育历期基本一致,因而绝大部分的成虫羽化时期出现在同一年份.为了有效防治栗山天牛,利用栗山天牛成虫具有趋光性这一特点,研究发明了诱集捕杀成虫的专用黑光灯,诱杀了大量的栗山天牛成虫,吉林省2008年共诱杀栗山天牛成虫达24.16t.根据每天晚上所诱到的栗山天牛的成虫数量(平均lkg栗山天牛成虫数量为430头,按此折合成重量kg统计),分析栗山天牛在成虫发生期其羽化数量与温湿度因子的关系.结果表明,栗山天牛成虫的羽化高峰期集中在7月下旬至8月上旬;成虫的羽化与林间气温呈显著的正相关关系,与相对湿度则呈显著负相关关系.研究对栗山天牛成虫的发生量与温湿度的关系进行了拟合.研究结果为我国东北林区栗山天牛成虫羽化年份预测预报成虫出现和发生期以及在成虫发生期开展防治提供了依据.  相似文献

Aeration is a promising alternative to the use of pesticides for the control of storage insects by cooling bulk grain, but its effectiveness against mite pests is neither fully understood nor optimised. For this reason, the productivity of three species of storage mites, Acarus siro, Lepidoglyphus destructor and Tyrophagus longior, was studied in a laboratory-based experiment at four combinations of temperature and humidity (10°C and 70% RH, 10°C and 80% RH, 20°C and 70% RH, 20°C and 80% RH) with and without an airflow (at 10 m3/h/tonne, equalling 2.5 l/s/tonne, in tubes containing 15 g of grain). This is the first time that a study has examined the three principal components of aeration separately from each other. The effect of these factors was different for each species. For A. siro, temperature was the most important factor, while airflow and humidity were of similar but lesser importance. For T. longior, temperature was more important than humidity, while the reverse was true for L. destructor. For these two species, airflow was the least important factor. The airflow decreased the productivity of L. destructor and T. longior but increased the productivity of A. siro. This increase in productivity confirms that, in practice, prevention of mite infestations, in particular A. siro, will require storage of grain at low temperature, relative humidity and moisture content. A model has been derived for the enrichment of heavy isotopes of water in leaves, including progressive enrichment along the leaf. In the model, lighter water is preferentially transpired leaving heavier water to diffuse back into the xylem and be carried further along the leaf. For this pattern to be pronounced, the ratio of advection to diffusion (Péclet number) has to be large in the longitudinal direction, and small in the radial direction. The progressive enrichment along the xylem is less than that occurring at the sites of evaporation in the mesophyll, depending on the isolation afforded by the radial Péclet number. There is an upper bound on enrichment, and effects of ground tissue associated with major veins are included. When transpiration rate is spatially nonuniform, averaging of enrichment occurs more naturally with transpiration weighting than with area-based weighting. This gives zero average enrichment of transpired water, the modified Craig-Gordon equation for average enrichment at the sites of evaporation and the Farquhar and Lloyd (In Stable Isotopes and Plant Carbon-Water Relations, pp. 47-70. Academic Press, New York, USA, 1993) prediction for mesophyll water. Earlier results on the isotopic composition of evolved oxygen and of retro-diffused carbon dioxide are preserved if these processes vary in parallel with transpiration rate. Parallel variation should be indicated approximately by uniform carbon isotope discrimination across the leaf.  相似文献

Internodal elongation in floating rice (Oryza sativa) is known to be enhanced by treatment with ethylene or gibberellic acid (GA3) at high relative humidity (RH). However, ethylene-induced internodal elongation is inhibited at low RH, while GA3-induced internodal elongation is hardly affected by humidity. We examined the effects of ethylene and GA3 on the rate of transpiration in stem segments incubated at 30% or 100% RH. Ethylene promoted the transpiration of stem segments at 30% RH, but not at 100% RH, while GA3 had little effect on transpiration at either 30% or 100% RH. We propose that the absence of ethylene-induced internodal elongation at low RH is due, at least in part, to ethylene-induced transpiration.  相似文献

A field experiment was established to examine the effects of temperature and moisture modifications on the nematode fauna of a semiarid grassland. Several combinations of drying, wetting, warming and cooling were applied to plots and compared with untreated control plots. The experiment was performed from July to October 1996. A significant shift was observed in the structure of the nematode fauna between late summer and early autumn. This shift was manifested in the disappearance of four rare genera; Ecumenicus, Eucephalobus, Paraphelenchus and Pungentus. A significant decrease was found in the density of Acrobeles, Aphelenchoides, Ditylenchus and Prismatolaimus; in addition there was a significant increase in the density of Cephalobus, Helicotylenchus, Paratylenchus and Tylenchorhynchus. Community structural change was represented by an initial decrease in nematode generic richness of 30–50%, and in a statistically significant decrease of nematode diversity in the control and all treated plots. Thereafter, emergence of a new community was demonstrated. Data show that temperature manipulation was the main factor to influence nematode diversity, Maturity Index, and Plant Parasite Index. However, nematode population density was influenced predominantly by the soil moisture content. Coenological analysis of soil nematode fauna appears to be a useful tool for the biological monitoring of the effects of global change on semiarid grasslands.  相似文献

Collaborative research was conducted at the USDA-ARS Subtropical Agricultural Research Center in southern Texas to assess the microbial control potential of Beauveria bassiana and Paecilomyces fumosoroseus against Bemisia whiteflies. Laboratory assays demonstrated the capacity of both pathogens to infect Bemisia argentifolii nymphs on excised hibiscus leaves incubated at relative humidities as low as 25% at 23 ± 2°C (ca. 35% infection by B. bassiana and P. fumosoroseus resulted from applications of 0.6–1.4 × 103 conidia/mm2 of leaf surface). In small-scale field trials using portable air-assist sprayers, applications at a high rate of 5 × 1013 conidia in 180 liters water/ha produced conidial densities of ca. 1–2.5 × 103 conidia/mm2 on the lower surfaces of cucurbit leaves. Multiple applications of one isolate of P. fumosoroseus and four isolates of B. bassiana made at this rate at 4- to 5-day intervals provided >90% control of large (third- and fourth-instar) nymphs on cucumbers and cantaloupe melons. The same rate applied at 7-day intervals also provided >90% control in zucchini squash, and a one-fourth rate (1.25 × 1013 conidia/ha) applied at 4- to 5-day intervals reduced numbers of large nymphs by >85% in cantaloupe melons. In contrast to the high efficacy of the fungal applications against nymphs, effects against adult whiteflies were minimal. The results indicated that both B. bassiana and P. fumosoroseus have strong potential for microbial control of nymphal whiteflies infesting cucurbit crops.  相似文献

Spray drying is a way to generate protein solids (powders), which is also true for lyophilization. Sugars are used to protect proteins from conformational changes and chemical degradations arising from drying processes and storage conditions such as the humidity. The influence of trehalose and humidity on the conformation and hydration of spray-dried recombinant human granolucyte colony stimulating factor (rhG-CSF) and recombinant consensus interferon-alpha (rConIFN) was investigated using Fourier transform IR spectroscopy. The spectral analysis of spray-dried powders in the amide I region demonstrated that trehalose stabilized the alpha-helical conformation of both rhG-CSF and rConIFN proteins. Exposure of the pure protein powders to 33% relative humidity (RH) resulted in the formation of beta sheets and loss of turns but no change in alpha-helical structure. Trehalose reduced the magnitude of the changes in beta sheets and turns. Exposure of the pure protein powders to 75% RH resulted in the loss of alpha-helical conformation with a corresponding increase in beta structures (beta sheets and turns). Trehalose did not protect proteins from the loss of alpha-helical structures, but it reduced the formation of antiparallel beta sheets. Hydrogen-deuterium exchange (H-D exchange) was used to further characterize these hydration-induced conformational changes. At 33% RH the percent exchange of the protein decreased with increasing trehalose content, indicating a greater protection of the protein from H-D exchange by a higher concentration of trehalose. Such protection correlates with decreased conformational changes of the protein by trehalose at this humidity. At 75% RH the degree of H-D exchange of the protein was insensitive to the powder composition in all powders. Surprisingly, the H-D exchange of trehalose was low at about 20-25%, which was nearly independent of the protein/trehalose ratio and humidity, indicating that the exchangeable protons on trehalose molecules are highly protected in protein-containing powders. The observed protein hydration is related to the effect of trehalose on the conformational changes of the protein under humidity.  相似文献

We present here a study of European Neogene primate occurrences in the context of changing humidity. We studied the differences of primate localities versus non-primate localities by using the mammal communities and the ecomorphological data of the taxa present in the communities. The distribution of primates is influenced by humidity changes during the whole Neogene, and the results suggest that the primates track the changes in humidity through time. The exception to this is the Superfamily Cercopithecoidea which shows a wider range of choices in habitats. All primate localities seem to differ from non-primate localities in that the mammal community structure is more closed habitat oriented, while in non-primate localities the community structure changes towards open-habitat oriented in the late Neogene. The differences in primate and non-primate localities are stronger during the times of deep environmental change, when primates are found in their preferred habitats and non-primate localities have faunas better able to adapt to changing conditions.  相似文献

The effect of different irrigation and air humidity conditioning treatments on the morphological and physiological responses of Rosmarinus officinalis in nursery conditions was investigated in order to evaluate the degree of hardening resulting from these conditions. Rosmarinus officinalis seedlings were pot-grown during 4 months in two greenhouses (nursery period), in which two irrigation treatments were used (control and deficit). In one of these greenhouses, air humidity was controlled using a dehumidifying system (low humidity), in the other greenhouse the air conditions were not artificially modified (control humidity). After the nursery period, the plants of all treatments were transplanted and well watered (100% water holding capacity for 1 month, transplanting period). After this period, they received no water (establishment period). At the end of the nursery period it was seen that deficit irrigation had altered the morphology of the R. officinalis plants by reducing plant height, stem diameter, leaf area, total dry weight, and root length, while humidity influenced the parameters related with plant water relations. Low air humidity and deficit irrigation-induced tissue dehydration and lower stomatal conductance values (gs). The plants subjected to deficit irrigation developed leaf osmotic adjustment, which was maintained during the transplanting period. At that time, the plants that had been exposed to deficit irrigation and low humidity showed efficient stomatal regulation (lower gs values). After transplanting and during the establishment period, these plants showed a better water status (higher psil and gs values). Their post-planting survival rate improved as a result of acclimation processes.  相似文献
