The hypersensitivity of Nicotiana tabacum cv. Xanthi to tobacco mosaic virus infection leads to the production and accumulation of a great number of phenolics (flavonol glycosides, caffeoylquinic, feruloylquinic and p-coumaroylquinic acids, glucose esters and glucosides of cinnamic and benzoic acids). An increase in temperature inhibits the hypersensitive reaction, resulting in the disappearance of these substances. The differences between the healthy and infected leaves become important when the synthesis of the virus is practically brought to completion and the hypersensitivity taken hold. The phenolic compounds do not appear to be responsible for the necrotic hypersensitivity and their production is one of the secondary effects of the virus infection.  相似文献

Potato tuber phosphofructokinase was purified 19·.6-fold by a combination of ethanol fractionation and DEAE-cellulose column chromatography. The enzyme was very unstable; its pH optimum was 8·0. Km for fructose-6-phosphate, ATP and Mg2+ was 2·1 × 10?4 M, 4·5 × 10?5 M and 4·0 × 10?4 M respectively. ITP, GTP, UTP and CTP can act as phosphate donors, but are less active than ATP. Inhibition of enzyme activity by high levels of ATP was reversed by increasing the concentration of fructose-6-phosphate; the affinity of enzyme for fructose-6-phosphate decreased with increasing concentration of ATP. 5′-AMP, 3′,5′-AMP, 3′-AMP, deoxy AMP, UMP, IMP, CMP, GMP, ADP, CDP, GDP and UDP did not reverse the inhibition of enzyme by ATP. ADP, phosphoenolpyruvate and citrate inhibited phosphofructokinase activity but Pi did not affect it. Phosphofructokinase was not reactivated reversibly by mild change of pH and addition of effectors.  相似文献

Three major peroxidases, designated as A, B2 and B2 from Solanum melongena leaves have been reported. Peroxidases-A, -B2 and -B2 were considered to be true peroxidases on the basis of k1:k4 ratio. The pH optima for the three enzymes were found to be 7·0, 6·0 and 6.0 respectively. These peroxidases differ in their k1:k4 ratio, in the effect of pH on this ratio and in the uric acid/guaiacol and o-dianisidine/guaiacol activity ratio.  相似文献

Roots of Mandragora autumnalis and M. vernalis contain hyoscyamine, hyoscine, cuscohygrine, apoatropine 3α-tigloyloxytropane and 3,6-ditigloyloxytropane. Belladonnine is present in the dried roots but could not be detected in fresh roots. No major differences were found in the alkaloids present in the two species. This is the first time the presence of tiglic acid esters has been reported in Mandragora species and the significance of this in the chemotaxonomy of the genus is indicated.  相似文献

The isoenzyme of potato peroxidase A5 has MW 105 000; C3—94 000; C4 and C5—56 500 C6—48 500. The isoenzymes retain activity on SDS-gels thereby allowing direct measurement of monomeric MW, even in crude extracts. One of the isoperoxidases showed anomalous behaviour on SDS-electrophoresis.  相似文献

Potato microsomes were delipidated by aqueous acetone solutions of increasing concentrations. Lipid extraction did not change the basal ATPase activity of these membranes (measured in the absence of added mineral ions), but affected the Mg2+-dependent ATPase activity. Low acetone concentrations (5–15%) moderately stimulated the Mg2+-ATPase; more concentrated solutions (20–50% acetone) dramatically decreased the activity of this enzyme, but 70 and 90% acetone solutions enhanced it again, as compared to the activity of the 50% acetone-treated fraction. This last stimulation could be explained by the selective extraction of an inhibitor of Mg2+-ATPase by concentrated acetone solutions. After lipid extraction with 50–90% acetone solutions, the initial Mg2+-dependent ATPase activity could not be restored by adding lipids to delipidated microsomes. These results strongly suggest that, in potato microsomes, the Mg2+-dependent ATPase was a lipid-dependent enzyme, but suitable relipidation conditions remain to be found to definitely prove this lipid dependence.  相似文献

A fast procedure was developed for the extraction and assay of quinolinic acid (QA) phosphoribosyl (PR) transferase based on gel filtration through a prepacked disposable PD 10 column and on reversed-phase HPLC. This makes the use of radiolabelled QA unnecessary. The specific activities determined in different organs of tobacco and in cell suspension cultures were determined; they indicate that this enzyme is probably involved in the regulation of nicotine biosynthesis. With a preparation from the roots of Nicotiana tabacum var. Samsun, the Km values for QA and PR-pyrophosphate were 5.1 and 21 μM, respectively.  相似文献

The heritability and genetic basis of nectar traits have been rarely studied in the field, where plants are exposed to environmental factors that could mask underlying genetic effects. Heritabilities and variance components were estimated for nectar and morphological traits of Nicotiana alata , using a partial diallel design. The main experiment was conducted in a Missouri experimental garden using a randomized block design with three plant density treatments, whereas a smaller experiment was conducted near native Brazil habitat to compare the environmental variance in traits between Missouri and Brazil. Significant heritability was detected for nectar volume and energy content, and for corolla tube length. Phenotypic correlations were significant between all traits investigated, whereas significant genetic correlations were only found between nectar volume and energy and between corolla limb width and mouth diameter. There were no significant family-by-density interactions detected in the Missouri field environment. All traits differed significantly between Missouri and Brazil environments, but significant genetic by environment (G × E) interactions between Missouri and Brazil were detected for only one trait. This study shows that nectar traits can be heritable despite considerable environmental variation.  相似文献

Tetranychus evansi Baker and Pritchard and Tetranychus urticae Koch (Acari: Tetranychidae) are important pests of Solanaceae in many countries. Several studies have demonstrated that T. urticae is an acceptable prey to many predatory mites, although the suitability of this prey depends on the host plant. T. evansi, has been shown to be an unfavorable prey to most predatory mites that have been tested against it. The predator Phytoseiulus fragariae Denmark and Schicha (Acari: Phytoseiidae) has been found in association with the two species in Brazil. The objective of this work was to compare biological parameters of P. fragariae on T. evansi and on T. urticae as prey. The study was conducted under laboratory conditions at 10, 15, 20, 25 and 30°C. At all temperatures, survivorship was lower on T. evansi than on T. urticae. No predator reached adulthood at 10°C on the former species; even on the latter species, only about 36% of the predators reached adulthood at 10°C. For both prey, in general, duration of each life stage was shorter, total fecundity was lower and intrinsic rate of population increase (r m ) was higher with increasing temperatures. The slower rate of development of P. fragariae on T. evansi resulted in a slightly higher thermal requirement (103.9 degree-days) on that prey than on T. urticae (97.1 degree-days). The values of net reproduction rate (R 0), intrinsic rate of increase (r m ) and finite rate of increase (λ) were significantly higher on T. urticae, indicating faster population increase of the predator on this prey species. The highest value of r m of the predator was 0.154 and 0.337 female per female per day on T. evansi and on T. urticae, respectively. The results suggested that P. fragariae cannot be considered a good predator of T. evansi.  相似文献
