圆背角无齿蚌血细胞培养   总被引:11,自引:1,他引:10  
用新设计的培养基培养了圆背角无齿蚌的血细胞,在倒置相差镜下进行了活体观察及扫描电镜摄影,发现培养的血细胞无颗粒细胞、颗粒细胞、透明细胞和类淋巴细胞,前三者均能伸出长的伪足和突起,与瓶壁紧密贴附,呈体外培养的成纤维细胞型;后者呈圆形,不与瓶壁贴附;四者的比例约为4:2:3:1。在活体内注射或在培养基上加入PHA和ConA,培养2-7d中,每天取部分供加入秋水仙素,用空气干燥法制片,作染色体观察,但未观察到转化细胞和有丝分裂相。研究结果表明,圆背角无齿蚌的颗粒细胞、无细胞和透明细胞在体外贴附玻璃表面的特征与高等动物的巨噬细胞类似,而不贴瓶的圆形细胞与高等动物的淋巴细胞类似,但在体外培养均不能繁殖,它们可能是高度分化的细胞。  相似文献

Hemocytes isolated from the larval hematopoietic organs of the silkworm were classified following staining with acridine orange and propidium iodide. Among the hemocytes isolated from the hematopoietic organs of whole fifth larval and wandering stages, most were prohemocytes (60%–70%) and oenocytoids (30%–40%). Granulocytes comprised only about 0.5%–1% at the wandering stage and were even rarer at other stages; no spherulocytes or plasmatocytes were found. Therefore, hemocyte differentiation inside larval hematopoietic organs is not as extensive as previously thought. Following 10–30 min in vitro culture of hemocytes isolated from larval hematopoietic organs, many young granulocytes and plasmatocytes appeared. Furthermore, during phagocytosis assays, prohemocytes were seen to adopt the morphology of plasmatocytes, containing fragments of phagocytosed cells. Our results underline the similarities between Drosophila and Bombyx hematopoiesis.  相似文献

We have focused attention on functions of Drosophila damaged DNA binding protein 1 (D-DDB1) in Drosophila hematopoiesis and previously reported that its whole body dsRNA over-expression using a GAL4-UAS targeted expression system results in melanotic tumors and complete lethality. Since the lesions appear to arise as a normal and heritable response to abnormal development, forming groups of cells that are recognized by the immune system and encapsulated in melanized cuticle, D-DDB1 appears to be an essential development-associated factor in Drosophila. To probe the possibility that it contributes to hemocyte development, we used a collagen promoter-GAL4 strain to over-express dsRNA of D-DDB1 in Drosophila hemocytes. The D-DDB1 gene silencing caused melanotic tumors and mortality at the end of larval development. Similarly, it interfered with melanization and synthesis of antimicrobial peptides. Transgenic flies with D-DDB1 gene silencing were found to accumulate abnormal large blood cells, reminiscent of human leukemia, suggesting that D-DDB1 has functions in hemocyte development.  相似文献

To identify novel factors involved in Drosophila hematopoiesis, we screened a collection of lethal recessive mutations that also affected normal hemocyte composition in larvae. We present the characterization of the gene yantar (ytr) for which we isolated null and hypomorphic mutations that were associated with severe defects in hemocyte differentiation and proliferation; ytr is predominantly expressed in the hematopoietic tissue during larval development and encodes an evolutionary conserved protein which is predominantly localized in the nucleus. The hematopoietic phenotype in ytr mutants is consistent with a defect or block in differentiation of precursor hemocytes: mutant larvae have enlarged lymph glands (LGs) and have an excess of circulating hemocytes. In addition, many cells exhibit both lamellocyte and crystal cell markers. Ytr function has been preserved in evolution as hematopoietic specific expression of the Drosophila or mouse Ytr proteins rescue the differentiation defects in mutant hemocytes.  相似文献

Hemocytes are crucial cells of the insect immune system because of their involvement in multiple immune responses including coagulation, phagocytosis and encapsulation. There are various types of hemocytes, each having a particular role in immunity, such that variation in their relative abundance affects the outcome of the immune response. This study aims to characterize these various types of hemocytes in larvae of the grapevine pest insect Eupoecilia ambiguella, and to assess variation in their concentration as a function of larval diet and immune challenge. Four types of hemocytes were found in the hemolymph of 5th instar larvae: granulocytes, oenocytoids, plasmatocytes and spherulocytes. We found that the total concentration of hemocytes and the concentration of each hemocyte type varied among diets and in response to the immune challenge. Irrespective of the diet, the concentration of granulocytes increased following a bacterial immune challenge, while the concentration of plasmatocytes and spherulocytes differentially varied between larval diets. The concentration of oenocytoids did not vary among diets before the immune challenge but varied between larval diets in response to the challenge. These results suggest that the resistance of insect larvae to different natural enemies critically depends on the effect of larval diet on the larvae’s investment into the different types of hemocytes.  相似文献

Schistosomiasis is one of the most detrimental neglected tropical diseases. Controlling the spread of this parasitic illness requires effective sanitation, access to chemotherapeutic drugs, and control over populations of the freshwater snails, such as Biomphalaria glabrata, that are essential intermediate hosts for schistosomes. Effectively controlling this disease, while minimising ecological implications of such control, will require an extensive understanding of the immunological interactions between schistosomes and their molluscan intermediate hosts. Here we histologically characterise the clearance of schistosome larvae by snails that exhibit allelic variation at a single genomic region, the Guadeloupe resistance complex. We show that snails with a resistant Guadeloupe resistance complex genotype clear schistosomes within the first 24–48?h, and that this resistance can be transferred to susceptible snails via whole hemolymph but not cell-free plasma. These findings imply that Guadeloupe resistance complex-coded proteins help to coordinate hemocyte-mediated immune responses to schistosome infections in Guadeloupean snails.  相似文献

An active phenoloxidase preparation from the freshwater crayfish Pacifastacus leniusculus exhibited a strong antibacterial effect in vitro on the bacteria Aeromonas hydrophila, Escherichia coli, Streptococcus pneumoniae whereas a weaker but still significant effect against Bacillus cereus, Pseudomonas aeruginosa and Staphylococcus aureus. In most cases reduction of bacterial growth was stronger when dopamine was used as substrate as compared to L-dopa. The effect on bacteria was abolished if no substrate was available for the phenoloxidase or in the presence of the phenoloxidase inhibitor phenylthiourea.  相似文献

Baculoviruses and parasitoids are critically important biological control agents in integrated pest management (IPM). They have been simultaneously and sequentially used to target insect pests. In this study, we examined the impacts of both baculovirus and polydnavirus (PDV) infection on the host cellular immune response. Larvae of the lepidopteran Spodoptera litura were infected by Spodoptera litura multicapsid nucleopolyhedrovirus (SpltMNPV) and then the animals were parasitized by the braconid wasp Microplitis bicoloratus. The fate of the parasitoids in the dually infected hosts was followed and encapsulation of M. bicoloratus first instar larvae was observed. Hemocytes of S. litura larvae underwent apoptosis in naturally parasitized hosts and in non-parasitized larvae after injection of M. bicoloratus ovarian calyx fluid (containing MbPDV) plus venom (CFPV). However, assessments of the percentages of cells undergoing apoptosis under different treatments indicated that SpltMNPV could inhibit MbPDV-induced apoptosis in hemocytes when hosts were first injected with SpltMNPV budded virus (BV) followed by injection with M. bicoloratus CFPV. As the time of injection with SpltMNPV BV increased, the percentages of apoptosis in hemocytes population declined. Furthermore, in vitro, the percentages of apoptosis showed that SpltMNPV BV could inhibit MbPDV-induced granulocytes apoptosis. The occurrence of MbPDV-induced host granulocytes apoptosis was inhibited in the dually infected hosts. As hemocytes apoptosis causes host immunosuppression, the parasitoids are normally protected from the host immune system. However, in larvae infected with both baculovirus and PDV, the parasitoids underwent encapsulation in the host hemocoel.  相似文献
