The gram-negative anaerobic oral bacterium Porphyromonas gingivalis initiates periodontal disease through fimbrial attachment to saliva-coated oral surfaces. To study the effects of immunomodulation on enhancement of subunit vaccination, the expression in E. coli and immunogenicity of P. gingivalis fimbrial protein (FimA) linked to the C-terminus of the cholera toxin B subunit (CTB) were investigated. Complementary DNAs encoding the P. gingivalis 381 fimbrillin protein sequence FimA1 (amino acid residues 1-200) and FimA2 (amino acid residues 201-337) were cloned into an E. coli expression vector downstream of a cDNA fragment encoding the immunostimulatory CTB. CTB-FimA1 and CTB-FimA2 fusion proteins synthesized in E. coli BL21 (DE3) cells were purified under denaturing conditions by Ni2+-NTA affinity column chromatography. Renaturation of the CTB-FimA1 and CTB-FimA2 fusion proteins, permitted identification of CTB-FimA pentamers and restored CTB binding activity to GM1-ganglioside to provide a biologically active CTB-FimA fusion protein. Mice orally inoculated with purified CTB-FimA1 or CTB-FimA2 fusion proteins generated measurable FimA1 and FimA2 IgG antibody titers, while no serum fimbrial IgG antibodies were detected when mice were inoculated with FimA1 or FimA2 proteins alone. Immunoblot analysis confirmed that sera from mice immunized with CTB linked to FimA1 or FimA2 contained antibodies specific for P. gingivalis fimbrial proteins. In addition, mice immunized with FimA2 or CTB-FimA2 generated measurable intestinal IgA titers indicating the presence of fimbrial antibody class switching. Further, mice orally immunized with CTB-FimA1 generated higher IgA antibody titers than mice inoculated with FimA1 alone. The experimental data show that the immunostimulatory molecule CTB enhances B cell-mediated immunity against linked P. gingivalis FimA fusion proteins, in comparison to immunization with FimA protein alone. Thus, linkage of CTB to P. gingivalis fimbrial antigens can increase subunit vaccine immunogenicity to provide enhanced protection against periodontal disease.  相似文献

Geotrichum sp. lipase with enhanced activity and operational stability was prepared for use in non-aqueous media. A combined strategy comprising bioimprinting with dual imprint molecules and a co-solvent coupled to pH tuning, KCl salt activation, lecithin coating and immobilization on macroporous resin effectively enhanced the activity and operational stability of Geotrichum sp. lipase. The modified lipase exhibited 18.4-fold enhanced esterification activity towards methyl oleate synthesis, and retained 90% activity following repeated use in 10 cycles. The combined strategy exhibited a significant synergistic effect and was suitable for lipase modification, dramatically enhancing the enzyme activity and operational stability. This approach is applicable to the preparation of other enzyme biocatalysts, since the methods are effective for upgrading crude enzyme to a refined product with high activity and stability for use in non-aqueous media.  相似文献

用51%的Percoll分离液和DEAE-纤维素层析柱,从自然感染的黄鳝(Monopterus albusZuiew)中分离鳝锥虫(Trypanosoma monopteriChen et Hsieh,1964)。对含虫血液样品分别以2.1×103r/min、2.5×103r/min、2.9×103r/min、3.3×103r/min、3.6×103r/min离心5、10、15、20min,观察红细胞的去除和鳝锥虫存留百分数,结果表明:2.9×103r/min离心15min分离效果最佳。离心分离的混悬液含有锥虫、白细胞、血栓细胞和极少的红细胞,悬混液再经DEAE-纤维素过柱,得到纯净的鳝锥虫。过柱后光学显微镜观察,锥虫形态正常,运动活泼,无血细胞。本方法具有耗时短、回收率高的特点,是把梯度分离和离子交换结合在一起的一种快速有效方法,为鳝锥虫的进一步研究奠定了基础。另外,此方法还可以作为快速、准确检测自然状态下黄鳝是否感染锥虫的手段,为鳝锥虫的流行病学研究提供可靠的数据。  相似文献

外来入侵害虫刺槐叶瘿蚊Obolodiplosis robiniae (Haldemann) 原产北美洲东部, 2006 年在中国发现, 危害刺槐叶片, 严重影响刺槐的健康生长, 已经列入我国检疫性害虫名单。在河北省秦皇岛市野外调查, 发现了一种广腹细蜂科天敌--叶瘿蚊广腹细蜂Platygaster robiniae Buhl and Duso, 该寄生蜂属中国新记录种。野外观察表明其对刺槐叶瘿蚊最高寄生率达84.8%, 具有良好的生物防治应用前景。本文对该广腹细蜂的外部形态和初步观察的生物学特性进行了描述。  相似文献

于2012年8月至2013年7月对四明湖水库水体理化特性以及浮游动物的种类、数量和生物量进行逐月采样调查,分析了水库的浮游动物群落特征及其影响因子。调查期间共检测到浮游动物150种,丰度198.0~7 258.0 ind/L,年平均3 795.2 ind/L,生物量0.28~5.64 mg/L,年均值为1.11 mg/L,浮游动物丰度和生物量空间差异不显著,但季节变化明显。对应分析(CA)和后续拟合表明,水温、叶绿素a、总磷、真光层深度、氨氮以及水位是驱动浮游动物结构变化的主要环境因子,各点的影响因子间存在空间差异性。对比24年前历史资料,发现四明湖水库浮游动物数量和生物量均减少,浮游动物出现小型化,推测认为是鱼类捕食的下行效应、蓝藻密度增加及蓝藻产生藻毒素增加影响浮游动物繁殖力所致。水质评价结果表明,四明湖水库目前处于中营养水平,但有向富营养型发展的趋势。  相似文献

Hepatocellular carcinoma (HCC) is regarded as one of the most common malignancies worldwide leading to cancer-related death. Long noncoding RNAs (lncRNAs) are a critical modulator affecting HCC progression. Whereas, the pathogenesis of lncRNA RBM5-AS1 in the development of HCC remains unclear. Quantitative RT-PCR or western blot assays were applied to detect the expression of genes and proteins, respectively. The proliferation and metastasis abilities were assessed using Cell counting kit-8 (CCK-8), EdU and transwell assays. RNA immunoprecipitation (RIP) experiment was employed to validate the molecular interactions. RBM5-AS1 is highly expressed in HCC tissues and cell lines, especially in Hep3B and HepG2 cells. RBM5-AS1 knockdown dramatically restrains cell proliferation, invasion and migration of HCC cells. Importantly, RBM5-AS1 acts as an epigenetic regulator to elevate the H3K27me3 level of miR-132/212 promoter regions via recruiting PRC2 (EZH2, SUZ12, EED), and eventually reducing miR-132/212 expressions. The recovery experiments demonstrated that downregulation of miR-132/212 markedly eliminate the antitumor effects mediated by RBM5-AS1 silencing in HCC cells. The data of this work illustrate that RBM5-AS1 acts as an epigenetic regulator to promote the HCC progression by repressing miR-132/212 expressions, which would provide a new insight for understanding the action mechanism of RBM5-AS1 in HCC development.  相似文献

The methylthiotriazine herbicide, simetryn, is commonly used in Japan, and its concentration in surface water is often high enough to affect natural phytoplankton. To estimate how the plankton community in eutrophic systems respond to short-term exposure of realistic concentrations of simetryn, we collected plankton from a eutrophic lake and exposed them to low (20 μg l−1) and high (100 μg l−1) concentrations of simetryn for 12 days in microcosm tanks (50 l). High concentrations significantly lowered total phytoplankton biomass, particularly green algal density. Consequently, the species composition was severely modified by simetryn application. However, there was no apparent impact of simetryn on microbial food-web components, bacteria, heterotrophic nanoflagellates (HNF), and ciliates. Despite the decreased abundance of algal food, the zooplankton community showed subtle changes with simetryn application. The results indicate that the direct impact of simetryn on planktonic organisms other than phytoplankton, particularly on microbial food-web components, is weak. The indirect impact of simetryn on zooplankton through the change of food quality and quantity was also small. It has been suggested that the persistence of microorganisms, alternative food for zooplankton, probably dilutes the indirect impact of simetryn on zooplankton by compensating for the loss of food phytoplankton. Consequently, the plankton community in eutrophicated systems is resistant to the herbicide at a feasible concentration for a short period of time.  相似文献

Background

The control of soil-transmitted helminth (STH) infections currently relies on the large-scale administration of single-dose oral albendazole or mebendazole. However, these treatment regimens have limited efficacy against hookworm and Trichuris trichiura in terms of cure rates (CR), whereas fecal egg reduction rates (ERR) are generally high for all common STH species. We compared the efficacy of single-dose versus triple-dose treatment against hookworm and other STHs in a community-based randomized controlled trial in the People''s Republic of China.

Methodology/Principal findings

The hookworm CR and fecal ERR were assessed in 314 individuals aged ≥5 years who submitted two stool samples before and 3–4 weeks after administration of single-dose oral albendazole (400 mg) or mebendazole (500 mg) or triple-dose albendazole (3×400 mg over 3 consecutive days) or mebendazole (3×500 mg over 3 consecutive days). Efficacy against T. trichiura, Ascaris lumbricoides, and Taenia spp. was also assessed.Albendazole cured significantly more hookworm infections than mebendazole in both treatment regimens (single dose: respective CRs 69% (95% confidence interval [CI]: 55–81%) and 29% (95% CI: 20–45%); triple dose: respective CRs 92% (95% CI: 81–98%) and 54% (95% CI: 46–71%)). ERRs followed the same pattern (single dose: 97% versus 84%; triple dose: 99.7% versus 96%). Triple-dose regimens outperformed single doses against T. trichiura; three doses of mebendazole – the most efficacious treatment tested – cured 71% (95% CI: 57–82%). Both single and triple doses of either drug were highly efficacious against A. lumbricoides (CR: 93–97%; ERR: all >99.9%). Triple dose regimens cured all Taenia spp. infections, whereas single dose applications cured only half of them.

Conclusions/Significance

Single-dose oral albendazole is more efficacious against hookworm than mebendazole. To achieve high CRs against both hookworm and T. trichiura, triple-dose regimens are warranted.
