Ganoderic acid, from Ganoderma lucidum, at 8 μg/ml inhibited replication of hepatitis B virus (HBV) in HepG2215 cells over 8 days. Production of HBV surface antigen and HBV e antigen were 20 and 44% of controls without ganoderic acid. Male KM mice were significantly protected from liver injury, induced with carbon tetrachloride, by treatment with ganoderic acid at 10 mg and 30 mg/kg·d (by intravenous injection) 7 days. Ganoderic acid at the same dosage also significantly protected the mice from liver injury induced by M. bovis BCG plus lipopolysaccharide (from Escherichia coli 0127:B8).  相似文献

In this paper, a novel reaction catalysed by the soybean lipoxygenase-1 (linoleate:oxygen oxidoreductase, EC 1.13.11.12) (LOX-1) is reported: p-nitrosodimethylaniline (RNO) bleaching. RNO bleaching in the course of the LOX-1 reaction (with linoleate as substrate) was followed by monitoring photometrically at 440 nm the absorbance decrease of the test sample. The appearance of oxodienes, a class of compounds produced during the same reaction, was also studied. This was carried out by monitoring the absorbance increase at 285 nm in the course of the LOX-1 reaction. RNO bleaching and oxodiene formation by soybean LOX-1 were found to occur synchronously. They occurred only under conditions of limited oxygen; moreover, both reactions showed saturation kinetics as expected for enzyme-catalysed reactions, had similar dependence on pH and temperature (pH optima about 9, temperature optima about 45 °C) and were competitively inhibited by n-propylgallate (PG), a free radical scavenger. Finally, the occurrence of both reactions was found to depend on the presence of 13-hydroperoxy-linoleate, the primary product of the aerobic LOX reaction and was found to be not related with the β-carotene bleaching catalysed by the same enzyme. The possible molecular basis of the link between RNO bleaching and oxodiene formation in the course of the anaerobic soybean LOX-1 reaction is discussed.  相似文献

The normal cellular prion protein, PrPc is a highly con served and widely expressed cell surface glycoprotein in all mammals. The expression of PrP is pivotal in the pathogenesis of prion diseases; however, the normal physiological functions of PrPc remain incompletely understood. Based on the studies in cell models, a plethora of functions have been attributed to PrPc. 鼻咽癌(nasopharyngeal carcinoma,NPC)是一种多基因遗传性疾病,好发生于我国华南、东南亚及部分非洲地区。近年来随着分子生物学及其技术的迅速发展,人们对鼻咽癌发生、发展及其生物学行为的研究已进入基因水平。microRNA(miRNA)是一类广泛存在于动植物体内的非编码小RNA,主要参与基因转录后水平调控。随着对miRNA研究的深入,发现肿瘤的细胞分化障碍、增殖失控、细胞永生化与miRNA密切相关。人类肿瘤组织与正常细胞组织间的miRNA表达水平和类型存在明显差异,提示miRNA可能是一类新的参与肿瘤发生的重要分子。本文就鼻咽癌与miRNA相关的研究进展作一综述。  相似文献

The uptake kinetics of phosphate (Pi) by Myriophyllum spicatum was determined from adsorption and absorption under light and dark conditions. Pi uptake was light dependent and showed saturation following the Michaelis-Menten relation (in light: V = 16.91 × [Pi](1.335 + [Pi]), R2 = 0.90, p < 0.001; in the dark: V = 5.13 × [Pi](0.351 + [Pi]), R2 = 0.77, p < 0.001). Around 77% of the loss of Pi in the water column was absorbed into the tissue of M. spicatum, and only 23% was adsorbed on the surface of the plant shoots. Our study shows that M. spicatum shoots have a much higher affinity (in light: 3.9 μmol g−1 dw h−1 μM−1; in the dark: 3.7 μmol g−1 dw h−1 μM−1) and Vmax (maximum uptake rate, shoot light) for Pi uptake than many other aquatic macrophytes (in light: 0.002-0.23 μmol g−1 dw h−1 μM−1; in the dark: 0.002-0.19 μmol g−1 dw h−1 μM−1), which may provide a competitive advantage over other macrophytes across a wide range of Pi concentrations.  相似文献

Although anoxic preconditioning (APC) in the myocardium has been investigated for many years, its physiological mechanism is still not completely understood. Increasing evidence indicates that transiently increased resistance to ischemic damage following APC is dependent on de novo proteins synthesis. However, the key effector pathway(s) associated with APC still remains unclear. The proto-oncogene Pim kinase belongs to a serine/threoine protein kinase family, consists of Pim-1, Pim-2 and Pim-3 and has been implicated in stimulating cell growth and inhibiting cell apoptosis. Therefore we assumed that Pim-3 expression might be aberrantly induced in cardiomyocytes that were subjected to anoxia/reoxygenation (A/R) injury and that Pim-3 might also contribute to cardio-protection after APC. To address this hypothesis, we cloned a Pim-3 expression vector, transfected it into rat cardiomyocytes, and examined Pim-3 expression in rat cardiomyocytes that were subjected to A/R injury. Moreover, we studied the role of three major MAPK pathways, e.g. p38 MAPK, JNK, and ERK1/2, in order to evaluate the molecular mechanism underlying Pim-3 up-regulation and A/R induced cardiomyocyte injury. Our experiments showed that APC induced an up-regulation of Pim-3 and the transfection of Pim-3 gene into the cardiomyocytes attenuated A/R injury. The inhibition of p38 MAPK by SB203580 abolished both the Pim-3 up-regulation and the cardio-protection provided by APC. Overall, these results suggest that APC could act to protect the heart from A/R injury with cooperation from the proto-oncogene Pim-3; in addition, it up-regulates Pim-3 expression through a p38 MAPK signaling pathway.  相似文献

从大戟科植物续随子(Euphorbia lathyris L.)的干燥成熟种子千金子中分离得到6个非萜类化合物.通过光谱分析,分别鉴定为金色酰胺醇酯(1)、蔓荆子黄酮(2)、青蒿亭(3)、胡萝卜苷(4)、对羟基苯甲酸(5)、秦皮乙素(6).其中化合物1~5均为首次从千金子中分离得到.  相似文献
