Six methods for quantification of the endospore concentrations of Pasteuria penetrans from tomato roots are described. Mortar disruption and machine disruption methods gave the highest estimations (endospores per gram of root material) of 83.7 and 79.0 million, respectively. These methods were significantly superior to incubation bioassay (47.7 million), enzymatic disruption (32.1 million), and enzymatic disruption + flotation (25.8 million) methods. A centrifugation bioassay method gave the lowest estimation of 12.7 million.  相似文献

Abstract

Explorative experiments were done to investigate the possibility that tomato plants infected by Botrytis cinerea have a different emission of volatile organic compounds (VOC) than healthy plants. This was done by headspace analysis of volatiles emitted by detached leaves of infected and healthy plants. Principal component analysis (PCA) of GC-FID chromatograms revealed clearly separated clusters between infected and control leaves and identification of an interesting compound. In further analysis by GC-MS, the significantly distinctive component (p≤0.05) was identified as the sesquiterpene α-copaene. In previous work on herbivore damage, α-copaene was not distinctive, which may suggest that α-copaene may be specifically associated to fungal infections in tomato.  相似文献

Summary The competitive ability of the pollen of wild tomato species (Solanum pennellii, Lycopersicum hirsutum and L. minutum) and a cultivar of tomato to affect fertilization at low temperature was investigated. Pollen grains of wild species were mixed with those of the cultivar and pollinated onto the stigma of the cultivar. The number of seedlings resembling the wild species, excluding L. minutum, were significantly higher when the plants were maintained at a low temperature (8°–12° C) than when they were maintained at a normal temperature (22°–28° C). This indicates that the pollen of S. pennellii and L. hirsutum wild species can compete better with pollen of the cultivar at a low temperature, while there was no change in the competitive ability of L. minutum pollen under these conditions.  相似文献

The potential of 24 indigenous isolates of Purpureocillium lilacinum (Paecilomyces lilacinus) (Thom) Samson collected from different agro-climatic zones of India was investigated against the root-knot nematode, Meloidogyne incognita. The studies were conducted in vitro (larvicidal, ovicidal and egg-parasitising capacity) and under naturally infested field conditions with selected strains. Repeated field trials were conducted with talc-based preparations of fungal strains at 10 kg ha?1, which were applied mixed in farm yard manure (FYM) at 1.5 t ha?1. Results (in vitro) showed that all tested isolates were capable to parasitise eggs, inhibit egg hatching and cause juvenile mortality of M. incognita at various levels. Based on the performance under in vitro studies, eight isolates (NDPL-01, ANDPL-02, SHGPL-03, HYBPL-04, AHDPL-05, PTNPL-06, SNGPL-07 and VNSPL-08) were re-tested to confirm the results. HYBDPL-04 was found causing highest mortality (80%), inhibition of egg hatching (90%) as well as parasitisation of M. incognita eggs (75%). Under field trials also, the best protection of root-knot disease of tomato (Lycopersicon esculentum L.), in terms of reduction of galls (61%) and reproductive factor (Pf/Pi (RF) = 0.2) was achieved through application of HYBDPL-04 + FYM compared to control and other tested isolates. It also enhanced marketable yield of tomato up to 43%. It is concluded that the HYBDPL-04 strain of P. lilacinum is highly effective for management of root-knot disease of tomato under naturally infested field conditions. It is the isolate which produced the maximum number of metabolites which were extracted through high pressure liquid chromatography.  相似文献

In the present study, the effect of aqueous fruit extracts of Azadirachta indica on activity of Peroxidase (POX) at different ages of Tomato (Lycopersicon esculentum) leading to induction of systemic resistance against Pseudomonas syringae pv. tomato was evaluated. For this evaluation, four ages, that is, 6, 8, 10 and 12?weeks of plants were selected. A single leaf at the third node from base of each plant was treated either singly or with different combinations of Neem extract and pathogen. Samples were collected at an interval of 24?h for up to five days and after two weeks of the treatment from both treated and untreated nodes. The change in the activity of defence enzyme POX and expression of its isoforms was studied. The results demonstrate that systemic acquired resistance induced by the Neem fruit extract increases as the plant matures but it is not only the limiting factor.  相似文献

Fusarial wilt of tomato (Lycopersicon esculentum Mill.) is a very common and severe disease occurring in most of the vegetable fields in West Bengal, India. Potenciation and formulation of different fungicidal chemicals and phytoextracts were evaluated against the growth of the pathogen wherein carbendazim (bavistin) and leaf extracts of Azadirachta indica (neem) were recorded to be most effective. Combined treatment with 4 ml neem leaf extract and 1 ml captan (0.01%) or with 4 ml garlic bulb extract and 1 ml captan (0.01%) exhibited 100% growth inhibition of the pathogen. Integrated control of the pathogen with phytoextracts, fungicide and biocontrol agents was carried out. Among the treatments, a combination with extracts of neem, captan (0.01%) and metabolites of Trichoderma harzianum was proved to be superior over the other. Field experiment with three fungicides at 0.5% concentration was carried out in randomised block design where application of bavistin showed up to 62.27% reduction of wilt infection in tomato plants. Soil solarisation of tomato field showed 62.50 and 66.69% reduction of infection during the trial years. However, integration of soil solarisation with the applications of T. harzianum, captan (0.01%) and neem resulted in 100% reduction of infection and thus it was recorded as the most effective treatment in reducing the incidence of the disease.  相似文献

Plants at 6, 8, 10 and 12?weeks of age were selected. A single leaf at the third node from the base of each plant was treated either singly or with different combinations of the pathogen and Neem fruit extract. Samples were collected from the non-treated leaves at an interval of 24?h up to five?days for enzyme assay and after two?weeks for disease development from both treated and untreated plants. The results demonstrate that the neem fruit extract could induce additional PPO isoforms both locally as well as systemically. The PPO activity was observed to be elevated in both the treated and non-treated leaves leading to induction of SAR. The induction of SAR enhances with the increase in the age of the plant.  相似文献

Microbiological analysis of seeds and plants of local and foreign selection made it possible to assess quality of the seed material and phytosanitary condition of tomato plants. It's very important to obtain and to cultivate healthy seed material of high quality. Strains of Pseudomonas fluorescens were investigated for biocontrol efficacy against tomato spotted wilt virus (TSWV) in tomato both alone and in mixtures. P. fluorescens strains applied to seed, soil and foliage or as a seedling dip significantly reduced TSWV, with a concomitant increase in growth promotion in both the glasshouse and field. Two native strains (CoP-1 and CoT-1) and one foreign strain (CHAO) reduced TSWV. In P. fluorescens-treated tomato plants, increased activity of polyphenol oxidase, β-1,3-glucanase and chitinase was observed, and induction of chitinase was confirmed by western blot analysis. Induction of new protein (18 kDa) detected by SDS-PAGE in P. fluorescens-treated tomato plants was not found in healthy and P. fluorescens-untreated virus inoculated control plants. Indirect ELISA clearly showed a reduction in viral antigen concentration in P. fluorescens-treated tomato plants corresponding to reduced disease ratings. All the P. fluorescens-treated tomato plants also showed enhanced growth and yield compared to control plants. Hence, plant growth promoting rhizobacteria (PGPR) could play a major role in reducing TSWV and increasing yield in tomato plants.  相似文献
