An in vitro method was developed to regenerate large numbers of phenotypically uniform plants from the basal parts of the leaves of flowering plants of Renanthera imschootiana Rolfe. Differentiation of up to 10 shoot buds free of callus and protocorm-like bodies occurred in 10–12 weeks from the base of a single leaf implanted in Mitra et al. (1976) medium supplemented with 2% sucrose, 2 g l-1 peptone, 44.4 M benzyladenine (BA) and 10.7 M naphthaleneacetic acid (NAA). Subculture of the tissues in medium enriched with 10% coconut water and 35 g l-1 ripe banana pulp resulted in the production of highest average number of 40 shoots in 12 weeks. No difference in the regeneration potential was observed among the three young leaves while mature leaves did not respond. All the leaves of the regenerated shoots were easily recultured to increase shoot multiplication. Shoots readily formed roots on transfer to a medium containing 4.4 M BA, 10.7 M NAA and 1% activated charcoal. All regenerated plants examined were normal diploids with 2n=38. In neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, multiple sclerosis and amyotrophic lateral sclerosis, neuroinflammation can lead to blood‐brain barrier (BBB) breakdown. After intravenous or intra‐arterial injection into mice, endothelial progenitor cells (EPCs) home to the damaged BBB to promote neurovascular repair. Autologous EPCs transfected to express specific therapeutic proteins offer an innovative therapeutic option. Here, we demonstrate that EPC transfection by electroporation with plasmids encoding the reporter protein GFP or an anti‐β‐amyloid antibody fragment (Fab) leads to secretion of each protein. We also demonstrate the secreted anti‐β‐amyloid Fab protein functions in β‐amyloid aggregate solubilization.  相似文献

In this study, we made an attempt to reveal how competition intensity from established plants impacts on palatable and unpalatable grass seedlings recruitment, in a natural mesic grassland of central Argentina. Our objective was to assess the seedling recruitment of a palatable species (Chascolytrum subaristatum) and an unpalatable species (Nassella trichotoma) in microsites differing in competition intensity from established plants. Identity (C. subaristatum and N. trichotoma) and defoliation severity were used as surrogate for competition intensity. In March 2017, we permanently marked established individuals of N. trichotoma and C. subaristatum and placed two circular plots adjacent to each individual. In one plot we added seeds of N. trichotoma and in the other seeds of C. subaristatum. After seeding, established plants were randomly assigned to one of three level of defoliation: without defoliation, low defoliation severity and high defoliation severity. From April to November 2017 (i.e. over a complete annual growing cycle), we measured seedling density, recruitment and growth. Our results supported the hypothesis that seedlings of palatable grasses are more competitive than seedlings of unpalatable grasses. Seedling of the palatable grass C. subaristatum recruited successfully regardless the intensity of competition from established plants, whereas seedlings of the unpalatable grass N. trichotoma recruited better under low competitive pressure from established plants. Our results suggest that the availability of microsites with low competitive pressure from the established vegetation, created by selective grazing of palatable grasses, promotes the recruitment of unpalatable grass seedlings. This mechanism may contribute to the species replacement process commonly observed in heavy grazed grasslands.  相似文献

分子标记可揭示种群遗传和进化信息, 为制定濒危物种保护措施、指导恢复实践提供重要依据。本文主要介绍了分子标记在濒危物种保护过程不同环节中的应用, 包括: (1)正确识别保护单元, 如排除隐存种和杂交种的影响; (2)确定优先保护单元, 包括优先保护区域、优先保护物种、优先保护种群等; (3)指导迁地保护; (4)对保护工作的动态监测和评估。文章最后探讨了分子标记应用于保护的发展方向, 如开展长期的种群遗传组成监测、切实应用于保护管理实践、将基因组学等遗传信息用于全球变化背景下保护策略的制定等, 期望为分子标记技术在生物多样性保护的研究和实践中提供参考。  相似文献

Using the example of Madagascar Fauna and Flora Group (MFG), we look at the factors which contribute to the successful maintenance of an international consortium dedicated to the conservation of Malagasy biodiversity. We discuss the philosophy, mission and set-up of the MFG and how, over its 25-year history, it has enabled the productive collaboration of its diverse international members to achieve the common goal of helping to protect Madagascar's unique biodiversity. We explore the benefits of pooling resources to fund a stable base of personnel and infrastructure to maximise the conservation impact of contributions from organisations that might not otherwise be able to fund viable independent programmes and consider the benefits that accrue to partners in the consortium. We highlight specific examples of plant conservation projects set up as a result of the productive working relationship between MFG and Missouri Botanical Garden to reinforce the argument that like-minded organisations working in successful partnership can far exceed the conservation capacity of individual institutions.  相似文献

The primary goal of captive breeding programmes for endangered species is to prevent extinction, a component of which includes the preservation of genetic diversity and avoidance of inbreeding. This is typically accomplished by minimizing mean kinship in the population, thereby maintaining equal representation of the genetic founders used to initiate the captive population. If errors in the pedigree do exist, such an approach becomes less effective for minimizing inbreeding depression. In this study, both pedigree‐ and DNA‐based methods were used to assess whether inbreeding depression existed in the captive population of the critically endangered Attwater's Prairie‐chicken (Tympanuchus cupido attwateri), a subspecies of prairie grouse that has experienced a significant decline in abundance and concurrent reduction in neutral genetic diversity. When examining the captive population for signs of inbreeding, variation in pedigree‐based inbreeding coefficients (fpedigree) was less than that obtained from DNA‐based methods (fDNA). Mortality of chicks and adults in captivity were also positively correlated with parental relatedness (rDNA) and fDNA, respectively, while no correlation was observed with pedigree‐based measures when controlling for additional variables such as age, breeding facility, gender and captive/release status. Further, individual homozygosity by loci (HL) and parental rDNA values were positively correlated with adult mortality in captivity and the occurrence of a lethal congenital defect in chicks, respectively, suggesting that inbreeding may be a contributing factor increasing the frequency of this condition among Attwater's Prairie‐chickens. This study highlights the importance of using DNA‐based methods to better inform management decisions when pedigrees are incomplete or errors may exist due to uncertainty in pairings.  相似文献

Metastasis is a major cause for cancer-related morbidity and mortality. Metastasis is a multistep process and due to its complexity, the exact cellular and molecular processes that govern metastatic dissemination and growth are still elusive. Live imaging allows visualization of the dynamic and spatial interactions of cells and their microenvironment. Solid tumors commonly metastasize to the lungs. However, the anatomical location of the lungs poses a challenge to intravital imaging. This protocol provides a relatively simple and quick method for ex vivo live imaging of the dynamic interactions between tumor cells and their surrounding stroma within lung metastasis. Using this method, the motility of cancer cells as well as interactions between cancer cells and stromal cells in their microenvironment can be visualized in real time for several hours. By using transgenic fluorescent reporter mice, a fluorescent cell line, injectable fluorescently labeled molecules and/or antibodies, multiple components of the lung microenvironment can be visualized, such as blood vessels and immune cells. To image the different cell types, a spinning disk confocal microscope that allows long-term continuous imaging with rapid, four-color image acquisition has been used. Time-lapse movies compiled from images collected over multiple positions and focal planes show interactions between live metastatic and immune cells for at least 4 hr. This technique can be further used to test chemotherapy or targeted therapy. Moreover, this method could be adapted for the study of other lung-related pathologies that may affect the lung microenvironment.  相似文献

采用不同溶剂对芦苇﹝Phragmites australis ( Cav.) Trin. ex Steud.﹞叶片水提物进行萃取,并以小麦( Triticum aestivum Linn.)和萝卜( Raphanus sativus Linn.)种子为实验材料对不同萃取物的化感效应进行检测;采用薄层层析和柱层析对抑制作用最强的正丁醇萃取物进行进一步分离,并采用GC-MS法对生物活性较高的组分进行组成成分分析;在此基础上,选择相对含量高并具有代表性的潜在化感成分进行生物活性检测,以期筛选出芦苇叶中的潜在化感成分。结果显示:随质量浓度(20、100和500 mg·L-1)提高,芦苇叶水提物的石油醚、二氯甲烷、乙酸乙酯、正丁醇和水萃取物对小麦和萝卜种子萌发的抑制作用均逐渐增强,其中正丁醇萃取物的抑制作用最强。在正丁醇萃取物的11个组分中,Fr.5、Fr.6、Fr.7、Fr.9和Fr.10组分均能显著抑制萝卜或小麦幼苗的生长,经质量浓度500 mg·L-1各组分处理液处理后萝卜或小麦幼苗的株高、根长及单株鲜质量均显著低于对照(P<0.05)。采用GC-MS法从Fr.5、Fr.6、Fr.7、Fr.9和Fr.10组分中分别鉴定出11、15、15、12和22种成分,分别占各组分总相对含量的83.02%、91.31%、87.36%、97.92%和94.34%,主要成分包括糖类、醇类、有机酸类、酮类、酰胺类和酯类。对14种潜在化感成分生物活性的检测结果显示这些成分对小麦幼苗生长有明显的抑制作用,其中,经质量浓度20 mg·L-1油酸酰胺、棕榈酸甲酯、亚油酸、2-苯乙胺、2-甲基烯丙醇和4-羟基-3-甲氧基苦杏仁酸处理后,小麦幼苗的株高、根长及单株鲜质量显著低于对照。综合分析结果显示:芦苇叶水提物具有较强的化感活性,其潜在的化感成分为油酸酰胺、棕榈酸甲酯、亚油酸、2-苯乙胺、2-甲基烯丙醇和4-羟基-3-甲氧基苦杏仁酸。  相似文献
