The continuously arising of SARS-CoV-2 variants has been posting a great threat to public health safety globally, from B.1.17 (Alpha), B.1.351 (Beta), P.1 (Gamma), B.1.617.2 (Delta) to B.1.1.529 (Omicron). The emerging or reemerging of the SARS-CoV-2 variants of concern is calling for the constant monitoring of their epidemics, pathogenicity and immune escape. In this study, we aimed to characterize replication and pathogenicity of the Alpha and Delta variant strains isolated from patients infected in Laos. The amino acid mutations within the spike fragment of the isolates were determined via sequencing. The more efficient replication of the Alpha and Delta isolates was documented than the prototyped SARS-CoV-2 in Calu-3 and Caco-2 cells, while such features were not observed in Huh-7, Vero E6 and HPA-3 cells. We utilized both animal models of human ACE2 (hACE2) transgenic mice and hamsters to evaluate the pathogenesis of the isolates. The Alpha and Delta can replicate well in multiple organs and cause moderate to severe lung pathology in these animals. In conclusion, the spike protein of the isolated Alpha and Delta variant strains was characterized, and the replication and pathogenicity of the strains in the cells and animal models were also evaluated.  相似文献

Adult somatic cell cloned calves were produced by somatic cell nuclear transfer prepared by fusion of cultured ear fibroblast from a Holstein cow into enucleated oocytes of Luxi Yellow cow. In order to determinate the source of mitochondrial DNA of cloned calves, we designed the breed-specific PCR primers by aligning the known D-loop sequences of Bos taurus and analyzed the displacement loop sequences of five live cloned calves by breed-specific primers PCR. The results demonstrated that mtDNA originated from Holstein breed and that from Luxi breed co-exist in all five live calves.  相似文献

Compound trans-4-(p-bromophenyl)-4-(dimethylamino)-1-(2-thiophen-2-yl-ethyl)-cyclohexanol (C8813), structurally unrelated to morphine, is a novel analgesic. The present study examined the antinociception, opioid receptor selectivity and in vitro activity of C8813. The antinociceptive activity was evaluated using mouse hot plate and acetic acid writhing tests. In mouse hot plate test, the antinociceptive ED(50) of C8813 was 11.5 microg/kg, being 591 times and 3.4 times more potent than morphine and fentanyl respectively. In mouse writhing test, the antinociceptive ED(50) of C8813 was 16.9 microg/kg, being 55 times and 2.3 times more active than morphine and fentanyl respectively. In the opioid receptor binding assay, C8813 showed high affinity for mu-opioid receptor (K(i) = 1.37 nM) and delta-opioid receptor (K(i) = 3.24 nM) but almost no affinity for kappa-opioid receptor (at 1 microM). In the bioassay, the inhibitory effect of C8813 in the guinea-pig ileum (GPI) was 16.5 times more potent than in the mouse vas deferens (MVD). The inhibitory effects of C8813 in the GPI and MVD could be antagonized by mu-opioid receptor antagonist naloxone and delta-opioid receptor antagonist ICI174,864 respectively. However, the inhibitory effect of C8813 in the rabbit vas deferens was very weak. These results indicated that C8813 was a potent analgesic and a high affinity agonist for the mu- and delta-opioid receptors.  相似文献

Angiotensin-converting enzyme 2 (ACE2 or ACEH) is a novel angiotensin-converting enzyme-related carboxypeptidase that cleaves a single amino acid from angiotensin I, des-Arg bradykinin, and many other bioactive peptides. Using des-Arg bradykinin as a template, we designed a series of intramolecularly quenched fluorogenic peptide substrates for ACE2. The general structure of the substrates was F-X-Q, in which F was the fluorescent group, Abz, Q was the quenching group (either Phe(NO(2)) or Tyr(NO(2))), and X was the intervening peptide. These substrates were selectively cleaved by recombinant human ACE2, as shown by MS and HPLC. Quenching efficiency increased as the peptide sequence was shortened from 8 to 3 aa, and also when Tyr(NO(2)) was used as a quenching group instead of Phe(NO(2)). Two of the optimized substrates, TBC5180 and TBC5182, produced a signal:noise ratio of better than 20 when hydrolyzed by ACE2. Kinetic measurements with ACE2 were as follows: TBC5180, K(m)=58 microM and k(cat)/K(m)=1.3x10(5)M(-1)s(-1); TBC5182, K(m)=23 microM and k(cat)/K(m)=3.5 x 10(4)M(-1)s(-1). Thus, based on hydrolysis rate, TBC5180 was a better substrate than TBC5182. However, TBC5180 was also hydrolyzed by ACE, whereas TBC5182 was not cleaved, suggesting that TBC5182 was a selective for ACE2. We conclude that these two peptides can be used as fluorescent substrates for high-throughput screening for selective inhibitors of ACE2 enzyme.  相似文献

Fermentative hydrogen production, as a process for clean energy recovery from organic wastewater, is limited by its low hydrogen yield due to incomplete conversion of substrates, with most of the fermentation products being volatile fatty acids (VFAs). Thus, further recovery of the energy from VFAs is expected. In this work, microbial fuel cell (MFC) was applied to recover energy in the form of electricity from mixed VFAs of acetate, propionate, and butyrate. Response surface methodology was adopted to investigate the relative contribution and possible interactions of the three components of VFAs. A stable electricity generation was demonstrated in MFCs after the enrichment of electrochemically active bacteria. Analysis showed that power density was more sensitive to the composition of mixed VFAs than coulombic efficiency. The electricity generation could mainly be attributed to the portion of acetate and propionate. However, the two components showed an antagonistic effect when propionate exceeded 19%, causing a decrease in coulombic efficiency. Butyrate was found to exert a negative impact on both power density and coulombic efficiency. Denaturing gradient gel electrophoresis profiles revealed the enrichment of electrochemically active bacteria from the inoculum sludge. Proteobacteria (Beta-, Delta-) and Bacteroidetes were predominant in all VFA-fed MFCs. Shifts in bacterial community structures were observed when different compositions of VFA mixtures were used as the electron donor. The overall electron recovery efficiency may be increased from 15.7% to 27.4% if fermentative hydrogen production and MFC processes are integrated.  相似文献

c-mos基因在动物卵母细胞减数分裂调控中起作用,但其作用机制目前仍不清楚。本实验通过RT-PCR、免疫荧光激光共聚焦检测方法检测了猪卵母细胞在体外成熟培养过程中c-mos基因在转录水平、翻译水平上的表达以及蛋白的分布,并应用注射小干扰RNA(siRNA)方法对其进行了RNA干扰(RNAi)研究。结果显示,猪卵母细胞在体外成熟培养过程中c-mos基因mRNA量逐渐增高,电激活后6h接近完全降解;MOS(c-mos基因蛋白产物)在GV卵母细胞生发泡中有一定量的表达,生发泡破裂(GVBD)前表达量增加且开始向卵母细胞胞质弥散,成熟培养44h未成熟卵母细胞中的MOS表达量要高于成熟卵母细胞,激活后6h核区MOS明显减少,但仍然有少量MOS分布于胞质中;成熟培养前干扰c-mos基因,所用三个siRNA都能成功敲低mRNA量,分别是同时期对照组mRNA量的0.08±0.03,0.11±0.06和0.20±0.06倍,干扰后虽然没有完全剔除MOS,但MOS量比同期卵母细胞有明显下降,仍可以引发成熟卵母细胞染色体解凝集。研究结果揭示了猪卵母细胞体外成熟及发育进程中c-mos基因在转录和翻译水平上的动态表达规律,建立了猪卵母细胞c-mos基因RNAi体系,为MOS在猪卵母细胞发育过程中的功能研究建立了重要的基础。  相似文献

对隆肛蛙属Feirana中隆肛蛙F.quadrana和太行隆肛蛙F.taihangnicus的15个种群565只标本的28项形态性状进行了测量,并运用典型判别分析法对其进行分析,分析结果表明:1)太行隆肛蛙和隆肛蛙的形态差异明显,形态度量信息支持太行隆肛蛙与隆肛蛙是不同的物种;2)原隆肛蛙河南伏牛山种群和山西中条山种群均为太行隆肛蛙的地理种群;3)隆肛蛙的种群间形态差异明显,其中四川安县种群、陕西周至种群和湖北利川种群与模式产地重庆巫山种群的差异可能达到了亚种或亚种以上分化水平.  相似文献

Noncanonical Wnt signals control morphogenetic movements during vertebrate gastrulation. Casein kinase I epsilon (CKIvarepsilon) is a Wnt-regulated kinase that regulates Wnt/beta-catenin signaling and has a beta-catenin-independent role(s) in morphogenesis that is poorly understood. Here we report the identification of a CKIvarepsilon binding partner, SIPA1L1/E6TP1, a GAP (GTPase activating protein) of the Rap small GTPase family. We show that CKIvarepsilon phosphorylates SIPA1L1 to reduce its stability and thereby increase Rap1 activation. Wnt-8, which activates CKIvarepsilon, enhances the CKIvarepsilon-dependent phosphorylation and degradation of SIPA1L1. In early Xenopus or zebrafish development, inactivation of the Rap1 pathway results in abnormal gastrulation and a shortened anterior-posterior axis. Although CKIvarepsilon also transduces Wnt/beta-catenin signaling, inhibition of Rap1 does not alter beta-catenin-regulated gene expression. Our data demonstrate a role for CKIvarepsilon in noncanonical Wnt signaling and indicate that Wnt regulates morphogenesis in part through CKIvarepsilon-mediated control of Rap1 signaling.  相似文献

The electricity production of Shewanella-inoculated microbial fuel cells (MFCs) under magnetic field (MF) exposure was investigated in different reactor systems. The persistency of the MF effect and the influences of MF intensity and direction on MFC performance were also studied. Application of a 100-mT static MF to the MFCs improved electricity production considerably, with an increase in the maximum voltage by 20-27% in both single- and two-chamber MFCs, while a more conspicuous improvement in the electricity generation was observed in a three-electrode cell. The MF effects were found to be immediate and reversible, and adverse effects seemed to occur when the MF was suddenly removed. The medium components analysis demonstrated that the application of MF led to an enhanced bioelectrochemical activity of Shewanella, and no significant promotion in mediator secretion was found. The improvement in the electricity production of MFCs under MF was mainly attributed to the enhanced bioelectrochemical activity, possibly through the oxidative stress mechanism. An accelerated cell growth under MF might also contribute to the enhanced substrate degradation and power generation.  相似文献
