我们以前的工作指出,生长抑素可以预防链佐霉素对大鼠胰岛B细胞的损伤。本工作测定给药前后大鼠血清和组织中Zn、Cu含量,以进一步观察链佐霉素破坏大鼠胰岛B细胞后生长抑素预防效应的可能机制。结果为:链佐霉素(STZ)(35mg/kg,ip)注射后24h的大鼠,血清Zn浓度下降,Cu浓度升高,Zn/Cu比值倒置;用生长抑素作预防性注射后,血清Zn浓度与正常对照水平相同,Cu浓度虽稍有升高,但Zn/Cu比值正常。保护组的血清Zn与损伤组比较,差异极为显著。两组的Zn/Cu比值同样也具有显著差异。在给药后72h,血清Zn/Cu比值在各组之间仍呈上述关系,但差异减小。在大鼠胰腺,注射链佐霉素后胰腺组织Zn浓度未见改变,但生长抑素作预防性注射可使组织锌含量增加。上述结果提示,在生长抑素对胰岛B细胞的保护机制中可能有Zn和Cu的参与。预防性注射生长抑素所引起的高Zn状态可能有利于机体细胞含Zn酶类的活性,增强已损伤细胞的修复。 相似文献

The purpose of this study was threefold: 1. to determine the long-term effects of interactions between lactational zinc deficiency
and gender on bone mineral composition in repleted rat offspring, 2. to determine the nutritional efficacy of the second of
two commercially designed, modified Luecke diets (ML2) during the gestational and lactational stress, and 3. determine the
ultratrace element contents of Ralston Rodent Laboratory Chow #5001. The ML2 basal diet, based on dextrose, sprayed egg white,
and corn oil contained 0.420 μg Zn/g, was supplemented with Zn (as zinc acetate) at 0 (diet 0ML2) or 30 (diet 30ML2) μg/g,
and was mixed and pelleted commercially. all rat dams were fed the 30ML2 diet ad libitum during gestation. Beginning at parturition,
the dams were fed either the 1. 0ML2, 2. 30ML2 (food restricted), or 3. 30ML2 (ad libitum) diets. All pups were fed the 30ML2
diet ad libitum from 23 to 40 d of age. From d 40 to 150, all pups were fed Ralston Rodent Laboratory Chow. The 30ML2 diet
was found to be nutritionally efficacious; litter size and pup growth were normal and pup mortality was only 1.2%. Pups (ZD)
with access to the 0ML2 diet until 23 d of age and nursed by dams fed the 0ML2 diet, when compared to pups (PF) fed restricted
amounts of the 30ML2 diet, exhibited increased mortality and decreased concentrations of tibial zinc but no change in growth.
Inadequate zinc nutriture during infancy, despite postlactational zinc repletion, induced imbalances in adult bone mineral
metabolism. Thus, at 150 d of age, the ZD pups exhibited increased levels of bone P and Mg and decreased concentrations of

Serum copper and zinc levels were determined in 20 healthy women and in 100 women with gynecological tumors. Malignant and
benign tumor cases were separated according to their postoperative, histopathological examinations. The stages of malignant
and benign tumors were also established histologically. Seventy benign and 30 malignant genital tumors (carcinoma of cervix
in situ, cervix, ovary endometrium, and vulva) of the patients were differentiated histopathologically.
The serum Cu/Zn ratios of patients were increased significantly from the control group (0.32±0.35) to the benign group (1.22±0.63)
and from the benign group to the malignant group (2.24±1.03). Nine of 30 malignant cases were determined as false negative
(30%) and 15 of 70 benign cases were determined as false positive (14.2%) according to the serum Cu/Zn ratios of patients.
Serum copper levels of 30 malignant and 10 benign tumor cases showed linear correlation with serum ceruloplasmin values. 相似文献

Copper absorption was measured at two levels of dietary zinc in six healthy young men who were confined to a metabolic unit
for a 75 d study of zinc utilization. A diet of conventional foods was fed, providing either 16.5 or 5.5 mg zinc and 1.3 mg
copper daily. Copper absorption was determined by feeding65Cu, a stable isotope of copper, once during the 16.5 mg Zn diet and near the beginning and end of the 5.5 mg Zn diet. Apparent
copper absorption averaged 48.1% when the 16.5 mg Zn diet was fed. This was significantly higher than the averages of 37.2
and 38.5% when the 5.5 mg Zn diet was fed. Absorption also differed significantly among subjects. Fecal copper did not differ
between diets or among subjects. All subjects were in positive copper balance at both levels of dietary zinc. These results
suggest that a dietary zinc intake slightly above the Recommended Dietary Allowance of 15 mg/d does not increase fecal copper
loss and does not interfere with copper absorption. 相似文献

Although various proteins and some electrolytes have been measured in human saliva, little systematic data about the major
and minor elemental components of this body fluid have been obtained. In order to obtain such data, concentrations of C, Na,
P, Cl, K, Ca, Sc, Cr, Fe, Co, Zn, Se, Br, Rb, Sb, I, and Cs in human parotid saliva were measured by instrumental nuclear
methods. The data obtained confirmed the relative lack of Zn in saliva of patients with hypogeusia (decreased taste acuity)
and suggested that concentrations of Na, Cl, Br, and Ca followed the order: normals > hypogeusia > hyposmia (decreased smell
acuity). To compare concentrations of elements in saliva with those in blood and urine, absolute concentrations were normalized
to that of Na through the use of a concept called an enrichment factor. On this basis, parotid saliva is relatively depleted
in Se, Zn, and Fe and enriched for most other elements relative to blood plasma indicating that the fluid is not simply a
transudate of blood plasma. Using this same technique, saliva composition was found more similar to urine than blood plasma,
being relatively depleted in Se, Cs, and Co, being enriched in I, Br, and Cr and having about the same relative concentrations
of P, Cl, Zn, Fe, Ca, K, and Rb. As the total body concentrations of many of the enriched elements in saliva are extremely
small, their enrichment in saliva suggests special roles for these elements in the oral cavity. Because of its accessibility,
ease of collection, and interaction with some body constituents, saliva represents a useful, albeit neglected, tool in the
diagnosis of some physiological and pathological changes in body function and in understanding important aspects of trace

Mycorrhizal incidence was studied at two forested locations in south-central Virgina. At each location, one site with soil
naturally enriched in copper, lead and zinc was designated as mineralized, and an adjacent site, with significantly lower
levels of these metals, was used as a control. A total of 223 soil samples were collected during the summer of 1984 and assayed
for active mycorrhizal tips. A reduced active mycorrhizal root tip count was found in those samples collected from the mineralized

The level of vesicular-arbuscular mycorrhizal (VAM) infection in the roots of Leucaena grown in a sand-soil mixture in the
greenhouse increased rapidly with time and reached a peak value of 84% at 30 days from planting. The pattern of immobile nutrient
uptake and accumulation closely paralleled that of the development of infection, particularly during the first 10–30 days
after planting. Significant changes in dry matter yield were also observed only after a significant portion of the root length
was colonized byGlomus aggregatum. The development of VAM infection was not accompanied by growth depression at any of the sampling periods. However, VAM roots
had very high levels of Cu which was not translocated to shoots. It is hypothesized that such a diversion of Cu by the endophyte
from the host could cause growth depression under conditions where the soil volume is supplied with sub-optimal levels of
Cu.

A recombinant 19-kDa human fibroblast collagenase catalytic fragment modeled on a naturally occurring proteolytic product was purified from E. coli inclusion bodies. Following renaturation in the presence of zinc and calcium, the fragment demonstrated catalytic activity with the same primary sequence specificity against small synthetic substrates as the full-length collagenase. Unlike the parent enzyme, it rapidly cleaved casein and gelatin but not native type I collagen. Intrinsic fluorescence of the three tryptophan residues was used to monitor the conformational state of the enzyme, which underwent a 24-nm red shift in emission upon denaturation accompanied by quenching of the fluorescence and loss of catalytic activity. Low concentrations of denaturant unfolded the fragment while the full-length enzyme displayed a shallow extended denaturation curve. Calcium remarkably stabilized the 19-kDa fragment, zinc less so, while together they were synergistically stabilizing. Among divalent cations, calcium was the most effective stabilizer, EC50 approximately 60 microM, and similar amounts were required for substrate hydrolysis. Catalytic activity was more sensitive to denaturation than was tryptophan fluorescence. Least sensitive was the polypeptide backbone secondary structure assessed by CD. These observations suggest that the folding of the 19-kDa collagenase fragment is a multistep process stabilized by calcium. 相似文献

Phytosiderophores (PS) are released in graminaceous species (Gramineae) under iron (Fe) and zinc (Zn) deficiency stress and are of great ecological significance for acquisition of Fe and presumably also of Zn. The potential for release of PS is much higher than reported up to now. Rapid microbial degradation during PS collection from nutrient solution-grown plants is the main cause of this underestimation. Due to spatial separation of PS release and microbial activity in the rhizosphere a much slower degradation of PS can be assumed in soil-grown plants. Concentrations of PS up to molar levels have been calculated under non-sterile conditions in the rhizosphere of Fe-deficient barley plants.Besides Fe, PS mobilize also Zn, Mn and Cu. Despite this unspecific mobilization, PS mobilize appreciable amounts of Fe in calcareous soils and are of significance for chlorosis resistance of graminaceous species. In most species the rate of PS release is high enough to satisfy the Fe demand for optimal growth on calcareous soils.In contrast to the chelates ZnPS and MnPS, FePS are preferentially taken up in comparison with other soluble Fe compounds. In addition, the specific uptake system for FePS (translocator) is regulated exclusively by the Fe nutritional status. The role of the trace minerals, copper (Cu) and zinc (Zn) are important in maintaining blood pressure. Copper has been found to inhibit the activity of angiotensin's converting enzyme. An interrelationship has been found to exist between Cu and Zn. Data in renal (RH) and spontaneous hypertensive rates (SHR) regarding Cu and Zn is lacking. The purpose of this report was to measure Cu and Zn levels in two types of experimental animal models of hypertension compared to normotensive (NT) rats. Blood samples were drawn to measure serum levels of Cu and Zn in three types of animals, RH, SHR, and NT. Serum Cu values were found to be lower, whereas Zn levels were elevated in the SHR animals. Serum levels of Cu and Zn in the RH animals were similar to those found in the NT animals. Further study of the interaction of those trace minerals is documented, and extends over knowledge of the role of minerals in blood pressure control. 相似文献
