Decoloration of a Carpet Dye Effluent Using Trametes Versicolor   总被引:1,自引:0,他引:1  
Although a non-sterile, undiluted carpet dye effluent (containing two anthraquinone dyes) did not support growth of Trametes versicolor, the pre-grown fungus removed 95% of its color in shake-flasks after 10 h of incubation. After decoloration, the COD of the cell-free supernatant increased and the toxicity was unchanged as determined by the Microtox assay using Vibrio fischeri. Decoloration rates decreased when either glucose alone or Mn2+ and glucose were added. T. versicolor, immobilized on jute twine in a rotating biological contacting reactor, also decolorized four successive batches of the effluent. There was no decoloration in any of the uninoculated, non-sterile controls.  相似文献

Malachite green (MG) is a triphenylmethane dye used as a fungicide but also possesses a high toxicity to mammalian cells. The toxicity of MG to Fomes sclerodermeus and Phanerochaete chrysosporium was assessed. P. chrysosporium was highly sensitive to the dye and it was unable to grow on solid media containing 64 microM of MG, lower concentrations caused a delay in growth. The radial growth of F. sclerodermeus was not affected at this concentration and up to 128 microM. In liquid media both fungi were more sensitive. F. sclerodermeus not only was able to grow in the presence of high concentrations of MG, but also it was able to decolorize and detoxify the dye. MG treated with supernatants containing high laccase activity in the presence or absence of 1-hydroxybenzotriazole (1-HBT) gave a colorless product (DMG) that was not toxic to P. chrysosporium and other white rot fungi tested. On the basis of the data of maximal absorbance, it is probable that the mechanism involved in the modification of the dye was different if 1-HBT was added to the reaction.  相似文献

Stalk-eyed flies are exemplars of sexual selection leading to the evolution of exaggerated male ornaments (eyespan). In Sphyracephala beccarri, there is no evidence for female mate choice for exaggerated male eyespan and only minor sex differences in eyespan. We used S. beccarri to test whether heightened condition dependence only evolves when male eyespan becomes sexually exaggerated. Male eyespan showed heightened condition dependence under food stress compared with a control trait (wing length). However, female eyespan displayed a similar pattern and there was no sex difference in the degree of increased eyespan sensitivity. The finding that eyespan is a sensitive indicator of food stress, even in an unexaggerated state, suggests that this may have acted as a pre-adaptation to the role of eyespan in sexual signalling in other Diopsid species. These results are consistent with handicap theory and Fisher's view of how sexual selection is initiated.  相似文献

Oligomycin has long been known as an inhibitor of mitochondrial ATP synthase, putatively binding the Fo subunits 9 and 6 that contribute to proton channel function of the complex. As its name implies, OSCP is the oligomycin sensitivity-conferring protein necessary for the intact enzyme complex to display sensitivity to oligomycin. Recent advances concerning the structure and mechanism of mitochondrial ATP synthase have led to OSCP now being considered a component of the peripheral stator stalk rather than a central stalk component. How OSCP confers oligomycin sensitivity on the enzyme is unknown, but probably reflects important protein–protein interactions made within the assembled complex and transmitted down the stator stalk, thereby influencing proton channel function. We review here our studies directed toward establishing the stoichiometry, assembly, and function of OSCP in the context of knowledge of the organization of the stator stalk and the proton channel.  相似文献

Biological decolorization of the synthetic dye RBBR in contaminated soil   总被引:1,自引:0,他引:1  
Soil contaminated with the synthetic dye Remazol Brilliant Blue R (RBBR) was treated independently with the wheat straw-grown white rot fungus Irpex lacteus, a bacterial consortium isolated from a dye-polluted soil and a coculture comprising both I. lacteus and the bacterial consortium. Both I. lacteus and the coculture removed RBBR (decrease in absorbance at 578 nm) gradually during a 49-day incubation time to 76 and 78%, respectively. The bacterial consortium alone, however, decolorized RBBR starting after 14 days with a final RBBR removal of 89%. Using controls with heat-killed cultures almost no decolorization occurred. The decolorization by the coculture did not show an increased RBBR removal as compared to the individual cultures. This might be explained by the observation that I. lacteus inhibited growth of the bacterial consortium.  相似文献

The toxicity of thirteen isomers of mono-, di-, tri- and pentachlorophenols was tested in potato-dextrose agar cultures of the white rot fungi Panus tigrinus and Coriolus versicolor. 2,4,6-Trichlorophenol (2,4,6-TCP) was chosen for further study of its toxicity and transformation in liquid cultures of these fungi. Two schemes of 2,4,6-TCP addition were tested to minimize its toxic effect to fungal cultures: stepwise addition from the moment of inoculation and single addition after five days of growth. In both cases the ligninolytic enzyme systems of both fungi were found to be responsible for 2,4,6-TCP transformation. 2,6-Dichloro-1,4-hydroquinol and 2,6-dichloro-1,4-benzoquinone were found as products of primary oxidation of 2,4,6-TCP by intact fungal cultures and purified ligninolytic enzymes, Mn-peroxidases and laccases of both fungi. However, primary attack of 2,4,6-TCP in P. tigrinus culture was conducted mainly by Mn-peroxidase, while in C. versicolor it was catalyzed predominantly by laccase, suggesting a different mode of regulation of these enzymes in the two fungi.  相似文献

The elaborate morphologies of sexually selected ornaments are ubiquitous across the animal kingdom. In studies investigating these traits, ornament size is frequently the focus, and empirical evidence supports its positive correlation with fitness. Yet shape plays an important role and, surprisingly, is often overlooked. Shape frequently changes with size, influenced by biomechanical, developmental, or performance constraints. Therefore, shape can provide additional insights into the morphological differences between individuals and the potential limits on sexual trait exaggeration. Here, we used landmark‐based geometric morphometric methods on a sexually dimorphic species of stalk‐eyed fly (Teleopsis dalmanni) to examine patterns of sexual shape dimorphism. Our analyses reveal a significant difference in head shape between the sexes, with males exhibiting smaller eye bulbs, thinner stalks, and smaller heads than females. Additionally, as eyestalk length increases within each sex, a similar pattern of shape change was observed as that observed between sexes. This pattern of shape change may be the result of constraints acting against further ornament exaggeration, and we suggest that this may significantly impact the whole‐organism performance in stalk‐eyed flies. © 2012 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, 106 , 104–113.  相似文献

Abstract Sugarcane borer, Diatraea saccharalis (F.), is a major target of transgenic maize expressing Bacillus thuringiensis (Bt) proteins in South America and the US mid‐south region. Resistance development in target pest populations is a major threat to the sustainable use of Bt crops. In our field trials in 2009, a significant number of live borers and plant injury from D. saccharalis were observed in an experimental SmartStax? maize line. The objective of this study was to assess the relative susceptibility of two field populations of D. saccharalis collected from non‐Bt and Bt maize plants containing SmartStax? traits to five individual Cry proteins. The five Bt proteins included two proteins (Cry1A.105 and Cry2Ab2) that were expressed in SmartStax? maize plants and three other common Bt proteins (Cry1Aa, Cry1Ab and Cry1Ac) that were not produced in SmartStax?. Larval mortality and growth inhibition on Bt diet of the fourth generation after field collections were evaluated 7 days after release of neonates on the diet surface. The laboratory bioassays showed that 50% lethal concentration (LC50) values for Cry1A.105 and Cry2Ab2 for the population originated from Bt plants were 3.55‐ and 1.34‐fold greater, respectively, than those of the population collected from non‐Bt plants. In contrast, relative to the population from non‐Bt plants, the LC50 of the population sampled from Bt plants were 3.85‐, 2.5‐ and 1.64‐fold more sensitive to Cry1Aa, Cry1Ab and Cry1Ac, respectively. The results did not provide clear evidence to conclude that the observed field survival of D. saccharalis on Bt plants was associated with increased levels of resistance.  相似文献

以白腐菌为出发菌株,利用紫外线(UV)进行诱变,筛选高产漆酶突变菌株。通过测定致死率绘制出发菌株的致死曲线,采用PDA-RBBR平板变色法进行初筛,ABTS检测酶活对突变株进行摇瓶复筛。结果表明:利用15 w紫外灯在照射距离为30 cm,照射时间为120 s,致死率为72.1%的条件下进行诱变处理,获得一株高产菌株,其酶活提高79.54%,经过5代传代培养,未见酶活下降,具有较好的遗传稳定性,进一步研究了初始pH值,接种量和培养基装液量等对诱变菌株产酶的影响,结果表明在最佳的培养条件pH值6.0,15%的装液量于28℃下,酶活达214.9 U/L。  相似文献
