Neuropeptide Y modulates the action of vasodilator agents in guinea-pig epicardial coronary arteries

Recently, we have demonstrated that guinea-pig epicardial coronary arteries are supplied by numerous nerve fibres containing neuropeptide Y (NPY) immunoreactivity. However, examination of vasomotor responses revealed that NPY did not elicit a contractile response in these arteries. In contrast, acetylcholine (ACh), calcitonin gene-related peptide (CGRP), substance P and vasoactive intestinal polypeptide (VIP) all relaxed precontracted arteries. In the present study, we have used histochemical, immunohistochemical and in vitro pharmacological techniques, in order to further investigate the possible role of NPY in guinea-pig epicardial coronary arteries. A double-immunofluorescence staining technique revealed that CGRP and substance P were co-localized in nerve fibres distinct from those displaying NPY immunoreactivity. Furthermore, using a method combining immunofluorescence and histochemical techniques, we observed that putative cholinergic nerve fibres (identified by their acetylcholinesterase content) and NPY-immunoreactive nerve fibres are two different nerve populations. An in vitro pharmacological method demonstrated that NPY markedly inhibited the relaxant responses mediated by ACh, VIP, substance P and isoprenaline but had no effect on CGRP. These results suggest that NPY-containing nerves associated with guinea-pig epicardial coronary arteries may be predominantly involved in modulating the action of vasodilator agents. 相似文献

A new biosensor for specific determination of sucrose was developed using an oxidoreductase of Zymomonas mobilis and invertase. Cells of Z. mobilis were permeabilized with toluene in order to utilize the enzymes of glucose-fructose oxidoreductase and gluconolactonase inside the intact cells. Permeabilized cells and invertase were coimmobilized in a gelatin membrane, and a whole cell enzyme electrode was constructed by fixing the membrane on a pH electrode. The production of hydrogen ion was detected using the biosensor-connected microcomputer, and the concentration of sucrose was determined by using both the initial rate and the steady-state methods. Optimum conditions for biosensor response were pH 6.2 and temperature 35 degrees C. The effect of interfering compounds on the electrode response was investigated, and the interference by various sugars was eliminated by determining sucrose concentration using the steady-state method. The biosensor developed is simple and reproducible, and the calibration curve for sucrose is linear up to 70 g/L. 相似文献

4-chloro-methoxyindole is a naturally occurring compound in Vicia faba which can easily react with nitrite to form a N-nitroso compound. In this in vitro study, the potential genotoxic effects of nitrosated 4-chloro-6-methoxyindole and its structural analogue 4-chloroindole were evaluated for the first time by using both Salmonella and Chinese hamster V79 cells. Additionally, the inhibition of gap junctional intercellular communication in V79 cells by these compounds was determined; this is a validated parameter for tumor-promoting activity. Most assays were also performed with nitrosated indole-3-acetonitrile, a naturally occurring compound in brassicas. Both nitrosated chloroindoles were highly mutagenic to Salmonella typhimurium TA100 without the need of exogenous metabolic activation and were potent inducers of Sister Chromatid Exchanges. Nitrosated indole-3-acetonitrile generated the same effects, although at much higher concentrations. Equivocal results were obtained for the nitrosated chloroindoles in a forward mutation assay using the hypoxanthine guaninephosphoribosyltransferase locus. All nitrosated indole compounds significantly inhibited gap junctional intercellular communication. These results indicate that nitrosated chloroindoles and nitrosated indole-3-acetonitrile should be considered as mutagens and agents with potential tumor-promoting capacity.Abbreviations BrdU

Permanent preparations of air dried synovial fluids were prepared by staining calcium compounds with alizarin red S stain; each slide was coverslipped with Permount. Variables studied were: (a) concentration of the solution of alizarin red S, (b) pH of staining solution, (c) time of incubation in staining solution and aqueous and ethanolic content of staining solution. The staining effect of each solution was tested on calcium pyrophosphate dihydrate, calcium oxalate, apatite and monosodium urate (MSU). Of all the solutions, best results were obtained with 0.25% alizarin red S in 50% ethanol at pH 7.0 for 30 min. With this solution, the calcium-containing compounds were well stained. MSU did not stain and still preserved negative birefringence on polarizaton. Fixation of smears with ethanol served a double purpose: It fixed the slides without dissolving or removing MSU or the calcium compounds, yet it did dissolve five corticosteroids commonly used for intra-articular injection which may interfere with interpretation of compensated polarized light microscopy of synovial fluids. 相似文献

In view of the development of al-carnitine deficiency, the metabolism ofl-carnitine and structure-related trimethylammonium compounds was studied inSalmonella typhimurium LT2 by means of thin-layer chromatography (TLC).l-Carnitine, crotonobetaine and acetyl-l-carnitine stimulated the anaerobic growth in a complex medium significantly. The stimulation depended on the formation of -butyrobetaine. The reduction ofl-carnitine proceeded in two steps: (1) Dehydration of thel-carnitine to crotonobetaine, (2) hydrogenation of crotonobetaine to -butyrobetaine. The reduction of crotonobetaine was responsible for the growth stimulation. Terminal electron acceptors of the anaerobic respiration such as nitrate and trimethylamine N-oxide, but not fumarate, suppressed the catabolism ofl-carnitine completely. Glucose fermentation, too, inhibited the reduction ofl-carnitine but optimal growth with a high carnitine catabolism was achieved byd-ribose. The esters of carnitine with medium- and long-chain fatty acids inhibited the growth considerably because of their detergent properties.Abbreviations TLC

Summary The course of the CO2 evolution rates of soil samples has been followed continuously in the absence and in the presence of various organic compounds. After an incubation period of 300 hours at 13 and 20°C the CO2 evolution from pasture soil (containing 1.76% soil organic carbon) amounted to 0.13 and 0.44g CO2–C.g soil–1.h–1, respectively. For arable soil (containing 1.20% soil organic carbon) the rates amounted to 0.04 and 0.09 g CO2–C.g soil–1.h–1, respectively.At 20°C larger amounts of the organic substrates added to the soil supplied with 20 g NH4NO3–N.g soil–1 were lost as CO2 than at 13°C, indicating a higher efficiency of the growth of microorganisms at lower temperatures. In the absence of NH4NO3 the respiration rates were initially higher than in its presence, suggesting that a part of the soil microflora is inhibited by low concentrations of NH4NO3. The amounts of carbon lost were low for phenolcarboxylic acids with OH groups in the ortho position. The replacement of one of these groups by a methoxyl group resulted in a larger amount of the C lost as CO2. The replacement of the COOH group by a C=C–COOH group had a decreasing effect on the decomposition of the phenolic acids tested. The decomposition of vanillic acid,p-hydroxybenzoic acid, and of the benzoic acids with OH groups in the meta position was as complete as that of glucose, amino acids or casein. The decomposition of bacterial cells to CO2 was considerably less than that of glucose.No evidence could be obtained that the low percentage of substrate converted to CO2 at the time of maximal respiration rate was due to the decreasing diffusion rate of substrate to the microbial colonies in the soil during the consumption of substrate. 相似文献

Tracheal sap was extracted from sections of stems (0.5 to 1.5 cm in diameter and 7.5 to 15.0 cm in length) of orange trees (Citrus sinensis (L.) Osbeck cv. Washington Navel) by using a combination of the vacuum and liquid displacement methods. The volume of sap obtained and its concentration of nitrogenous compounds were dependent on the volume of displacing liquid used for the extraction. Four ml of water-saturated 1-butanol extracted essentially all of the xylem fluid present in the stem sections without apparent production of artifacts. The time of sampling affected the nitrogen concentration of the tracheal sap, but not the content of xylem nitrogen per volume of stem material. The orientation of the stems in the tree and the diameter of the stems had an effect on their contents of xylem nitrogen, with southeastern orientation and thinner stems showing higher concentrations. We could not detect the presence of ammonium, nitrites or proteins in the tracheal sap of orange trees. Most of the nitrogen was present as amino acids and about 2% of the total in the form of nitrates. The qualitative composition of amino acids, as determined by TLC, was the same both in winter and spring tracheal sap. The main components of the sap were proline and arginine in winter, and these amino acids together with asparagine and aspartic acid in spring. 相似文献

We have shown that tellurite and tellurate require the interaction with reduced glutathione (GSH) to hemolyze human erythrocytes.
The study of the nature of this interaction is the main object of this paper. The degree of hemolysis was determined by the
method of Angelone. The addition of extracellular 1 mM GSH or cysteine increased the rate of hemolysis. Concanavalin A (0.3
mg/mL) and/or 4 mg/mL adenosine did not affect the hemolysis by 0.1 mM tellurite. One tenth to 1 mM 4-acetamido-4′-isothiocyanostilbene-2,2′-disulfonate
(SITS) inhibited this hemolysis by 60–100%. Millimolar GSH released this inhibition. Incubation of 0.1 mM tellurite with 1
mM GSH for 90 min at 37°C, produced a hemolytic agent when prepared and tested under nitrogen, but one that was not active
when prepared in air. The hemolysis byp-hydroxymercuribenzoate orp-hydroxymercuriphenylsulfonate did not involve GSH. Scanning electron micrographs showed a sphero-echinocyte transformation,
in the pre-hemolytic stage, with all the agents tested. The rate of penetration of tellurite plays a role in determining the
rate of hemolysis, as shown by the effect of SITS. The release by GSH of the inhibition by SITS poses questions concerning
the site of action and cell membrane penetration of the hemolytic agent. Telluride or some intermediate in the interaction
of GSH with tellurite is the actual hemolytic agent. 相似文献
