The photochemical apparatus organization in Synechococcus 6301 (Cyanophyceae) was investigated under various experimental conditions. Wild type (WT) Synechococcus produced phycobilisomes (PBSs) containing normal levels of phycocyanin (Phc) and allophycocyanin (Aphc). The ratio of reaction centers(RC) RCII/RCI of 0.4 was the same in WT and the mutant strain AN112, whereas RCH/PBS was 1.9:1 in WT and 1:1 in AN112. Excitation of WT cells with broad-band 620 nm light, which is absorbed primarily by Phc and Aphc and to a much lesser extent by chlorophyll (Chl), sensitized the RC of photosystem (PS) II at about 15 times the rate it sensitized RCI. This implies that PBSs are associated exclusively with PSII complexes and that PBS excitation is not transferred to PSI. The AN112 mutant of Synechococcus produced smaller PBSs consisting of the Aphc-containing core and of only six Phc-containing hexamers, respectively. It lacked about 65% of the Phccontaining rod substractures. Under our experimental conditions, the effective absorption cross section of the mutant PBS was only about half that of the WT. In agreement, the rate of RCII excitation by 620 nm light was also about half of that measured in the WT. Thus, the rate of light absorption by PSII depends directly on PBS size and composition. The low rate of RCI excitation with 620 nm light was the same in WT and AN112 cells, apparently independent of the PBS effective absorption cross section. We propose a strict structural-functional association between PBS and PSII complex. In greening leaf segments amino-oxyacetate inhibited both chlorophyll and carotenoid formation by ca 60 % at 0.5 mM inhibitor concentration. In greening tissue serine: glyoxylate aminotransferase was the only enzyme of the glycollate pathway whose activity was markedly decreased after inhibitor treatment. The inhibition of pigment formation in barley and maize could be alleviated by glyoxylate, pyruvate and acetaldehyde; in the latter case there is probably a preferential reaction with inhibitor which displaces it from combination with enzymic pyridoxal 5′-phosphate.  相似文献

Summary Previous experiments indicate that the apical membrane of the frog retinal pigment epithelium contains electrogenic NaK pumps. In the pressent experiments net potassium and rubidium transport across the epithelium was measured as a function of extracellular potassium (rubidium) concentration, [K] o ([Rb] o ). The net rate of retina-to-choroid42K(86Rb) transport increased monotonically as [K] o ([Rb] o ), increased from approximately 0.2 to 5mm on both sides of the tissue or on the apical (neural retinal) side of the tissue. No further increase was observed when [K] o ([Rb] o ) was elevated to 10mm. Net sodium transport was also stimulated by elevating [K] o . The net K transport was completely inhibited by 10–4 m ouabain in the solution bathing the apical membrane. Ouabain inhibited the unidirectional K flux in the direction of net flux but had not effect on the back-flux in the choroid-to-retina direction. The magnitude of the ouabain-inhibitable42K(86Rb) flux increased with [K] o ([Rb] o ). These results show that the apical membrane NaK pumps play an important role in the net active transport of potassium (rubidium) across the epithelium. The [K] o changes that modulate potassium transport coincide with the light-induced [K] o changes that occur in the extracellular space separating the photoreceptors and the apical membrane of the pigment epithelium.  相似文献

Membrane specializations of the peripheral retina of the housefly (Musca domestica) are revealed in thin sections and freeze fracture/etch replicas. Septate junctions are abundant in corner areas of the pseudocone enclosure bonding: between homologous corneal pigment cells (CPC); between homologous large pigment cells (LPC); between CPC-LPC; between Semper cells (SC); between SC-CPC. Spot desmosomes are present between Semper cells. It is likely that septate junctions function as strengthening adhesions in this area. A new membrane specialization similar to a continuous junction was observed between retinular cells of the same or adjacent ommatidium. This junction has indistinct septa in the 115 A intermembrane cleft and is intermittent in character. When this junction is absent, the apposed cells gape apart. In freeze fracture studies, this junction is characterized by bridges composed of fused membrane particles and randomly arranged particles on the P face, and noncorresponding grooves on the E face. The ridges are elongate and roughly parallel and sometimes they form enclosures. Mitochondria line up along these junctions, often within 90 A of the unit membrane. This membrane specialization has characteristics of tight and continuous junctions. In line with previous findings, we suggest that this junction assists in retinular cell orientation, possibly in enforcing the ommatidial twist and in maintaining localized ionic concentration gradients between retinular cells.  相似文献

Predicted increase in ultraviolet-B (UV-B: 280–320 mn) radiation may have adverse impacts on growth and yield of rice ( Oryza sativa L.), as has been found in studies hitherto. However, most of the studies were conducted in growth chambers or greenhouses where the plants are generally more sensitive to UV-B than in the field, presumably because of the distorted balance between UV-B and ultraviolet-A as well as PAR. This study was conducted to address the effects of enhanced UV-B on growth and yield of rice under a realistic spectral balance in the field. Three cultivars, "Koshihikari",'IR 45'and'IR 74'were pot-grown and irradiated with enhanced UV-B for most of the growing season in the field at Tsukuba, Japan (36°01'N, 140°07'E). The UV-B enhancement simulated ca 38% depletion of stratospheric ozone at Tsukuba. The results showed no UV-B effects on plant height, numbers of tillers and panicles, dry weight of the plant parts or the grain yield for any of the 3 cultivars. Natural abundance of 13C in the flag leaves was not altered by the UV-B enhancement either. While UV-absorbing compounds showed no response to the UV-B enhancement, chlorophyll contents decreased with enhanced UV-B. However, the decrease of chlorophyll was limited to an early growth stage with no effect later. We thus found no extraordinary impact of the nearly doubled UV-B radiation on rice in the field, and it would appear that a reliable prediction of the effects of UV-B will require experiments carried out over a number of years under various climatic and solar UV-B regimes.  相似文献

According to a recent hypothesis the melanin granules in the retinal pigment epithelium of mammals originate from photosensory membrane degradation. To test this hypothesis the retinal pigment epithelium of cattle was kept in tissue culture and exposed to gold-labelled rod outer segments. Gold granules were later detected inside phagosomes, melanosomes and mature melanin granules. Tyrosinase, the key enzyme in melanogenesis, was additionally localized inside phagosomes. These results indicate that in cultured retinal pigment epithelium the matrix of the melanosome can originate from phagosomes. therefore, the melanosome is a specialized lysosome.  相似文献

1.  We examined microspectrophotometrically the retinas of 3 species of stomatopods in the superfamily Gonodactyloidea, all of which live in environments that are reduced both in the intensity and spectral range of natural illumination. Species examined were Odontodactylus brevirostris, O. scyllarus, and Hemisquilla ensigera.
2.  All 3 species had the typical gonodactyloid diversity of visual pigments, with 8 different photopigments residing in the 4 tiered rows of the midband and 2 additional types in the untiered classes of photoreceptors in the midband and peripheral retina. The spectral range covered by the max values of the visual pigments of each species was similar to that of other gonodactyloid and lysiosquilloid species.
3.  Apparent retinal adaptations in species of Odontodactylus for vision in dimly lit, spectrally narrow photic environments were seen primarily as specializations of the intrarhabdomal filters. These were of reduced diversity, and had reduced absorption at long wavelengths compared to the filters of other gonodactyloid stomatopods. Retinas of H. ensigera lacked both proximal classes of intrarhabdomal filter, and had the smallest total range of visual pigment max yet observed in mantis shrimps. These modifications decrease the spectral range and number of types of narrow-band spectral classes of phooreceptors, while increasing their sensitivity.

研究了在光胁迫下南方红豆杉叶片中叶绿体色素含量和紫杉醇含量的动态变化。结果表明,与对照相比,叶绿素a、叶绿素b和总叶绿素的含量以及叶绿素a/叶绿素b比值在光胁迫下均减少,随胁迫时间的延长,叶绿素a、叶绿素b和总叶绿素的含量逐渐上升,叶绿素a与叶绿素b的比值先上升后下降;类胡萝卜素含量在前两周与对照相比降低,2周后高于对照;类胡萝卜素与叶绿素的比值高于对照,随胁迫时间的延长,其趋势先降低后上升。光胁迫处理后,南方红豆杉叶片中紫杉醇含量与对照相比,在第1周时略有下降,而后随胁迫时间的延长,紫杉醇含量在第2周开始迅速大量积累,而在处理3周时达到较高值,已增加到1.5倍,这对提高人工种植的南方红豆杉中紫杉醇含量具有非常重要的意义。  相似文献
