栀子黄色素的醇提制备及其稳定性研究   总被引:1,自引:0,他引:1  
通过单因素实验,考察栀子黄色素乙醇提取工艺。乙醇提取最优工艺条件为:乙醇浓度30%、液固比8:1、提取温度60℃、提取次数2次、每次1h。在此条件下,栀子黄色素、栀子苷和总三萜酸的提取率分别为1.32、110.76和94.91mg/g。  相似文献

通过静态吸附解析实验,确定了利用大孔吸附树脂初步纯化唐古特白刺红色素的条件,并根据所得条件做了扩大实验。结果表明.X-5树脂的吸附能力最好,吸附率为76.85%,pH在1—3范围内对吸附效果无影响;较高的乙醇浓度和pH值有利于色素的解析,解析液以80%酸化乙醇(含1%HCI)效果最佳,解析时间为40min;扩大实验得到紫红色色素粉末,得率为1.24%,水溶性好,不含糖类、蛋白质等水溶性杂质;树脂通过回收再生可重复利用。本实验为色素产品的开发及白刺果实的综合利用提供了理论依据。  相似文献

The patterns of pigment distribution in the shell of the gastropod Littorina obtusata were studied. It was determined that the background coloration of the periwinkle shell resulted from the coloration interaction of the external and internal parts of the ostracum. The coloration of the latter can be monochromatic or twolayer. The external and/or internal zones of the ostracum can be colored purple, white, orange, yellow and greenish. Only 11 of 25 possible color combinations were recorded. Four additional versions of shell background coloration resulted from partial depigmentation of external or internal parts of ostracum. The mottling pattern corresponded to white or depigmented lenticular sites in the external part of the ostracum. The bands on the shell may be subdivided into two groups according to their means of formation. The bands that appeared due to redistribution of pigments forming background coloration belonged to the first group, the bands arising as a layer of additional (white, yellow, orange) pigment belonged to the second group. Various band versions could occur simultaneously in the same individual.  相似文献

本研究以蛹虫草菌丝体为供试材料,采用高效液相色谱对提取的蛹虫草菌丝体水溶性黄色素的组分进行了分离纯化与测定分析,并以其主要组分YP-4为检测指标,采用温度、光照、pH、金属离子、常见食品添加剂等条件对蛹虫草菌丝体水溶性黄色素稳定性的影响进行了较为系统的研究。结果表明,蛹虫草菌丝体水溶性黄色素由5种结构相似的类胡萝卜素组分组成。其中,主要组分YP-4占到WSYPs总量的57.43%,YP-4的MS data [M-H]为521.2657,紫外特征吸收峰为:209nm、239nm、421nm、445nm、474nm,在445nm处有最大吸收峰。以YP-4为检测指标,对提取蛹虫草菌丝体黄色素的稳定性测试结果表明,该类黄色素具有良好的加工稳定性,但在加工过程中应尽量缩减紫外光照射时间,注意酸性添加剂和Fe 3+的用量。这一研究结果将为蛹虫草菌丝体水溶性黄色素乃至其他优质天然黄色素的开发利用提供参考依据。  相似文献

本实验对前期研究得到的具有高产红色素能力的产紫青霉突变菌株Penicillium purpurogenum Li-3-9的发酵条件进行优化的同时作出了对红色素的安全性评价。在单因素试验的基础上,采用Box-Behnken试验设计优化了蔗糖浓度、酵母膏浓度和装液量对红色素色价的影响;并且研究了P. purpurogenum Li-3-9产生的红色素对斑马鱼胚胎的毒性效应。结果表明,最佳发酵条件为:蔗糖40g/L、酵母膏4g/L、装液量为45mL、接种量4%、培养温度32℃、初始pH值6.9、培养时间168h、摇床转速150r/min,红色素色价最高达到了11.4U/mL。通过计算斑马鱼胚胎致死率、心率、畸形率及孵化率可知,该色素无致死毒性,并且对斑马鱼胚胎发育具有一定的促进作用。  相似文献

Skin pigment pattern formation is a paradigmatic example of pattern formation. In zebrafish, the adult body stripes are generated by coordinated rearrangement of three distinct pigment cell‐types, black melanocytes, shiny iridophores and yellow xanthophores. A stem cell origin of melanocytes and iridophores has been proposed although the potency of those stem cells has remained unclear. Xanthophores, however, seemed to originate predominantly from proliferation of embryonic xanthophores. Now, data from Singh et al. shows that all three cell‐types derive from shared stem cells, and that these cells generate peripheral neural cell‐types too. Furthermore, clonal compositions are best explained by a progressive fate restriction model generating the individual cell‐types. The numbers of adult pigment stem cells associated with the dorsal root ganglia remain low, but progenitor numbers increase significantly during larval development up to metamorphosis, likely via production of partially restricted progenitors on the spinal nerves.  相似文献

The Retinal Pigment Epithelium (RPE) forms the primary site of pathology in several blinding retinopathies. RPE cultures are being continuously refined so that dynamic disease processes in this important monolayer can be faithfully studied outside the eye over longer periods. The RPE substrate, which mimics the supportive Bruch’s membrane (BrM), plays a key role in determining how well in-vitro cultures recapitulate native RPE cells. Here, we evaluate how two different types of BrM substrates; (1) a commercially-available polyester transwell membrane, and (2) a novel electrospun scaffold developed in our laboratory, could support the generation of realistic RPE tissues in culture. Our findings reveal that both substrates were capable of supporting long-lasting RPE monolayers with structural and functional specialisations of in-situ RPE cells. These cultures were used to study autofluorescence and barrier formation, as well as activities such as outer-segment internalisation/trafficking and directional secretion of key proteins; the impairment of which underlies retinal disease. Hence, both substrates fulfilled important criteria for generating authentic in-vitro cultures and act as powerful tools to study RPE pathophysiology. However, RPE grown on electrospun scaffolds may be better suited to studying complex RPE-BrM interactions such as the formation of drusen-like deposits associated with early retinal disease.  相似文献

Organogenesis is regulated by a complex network of intrinsic cues, diffusible signals and cell/cell or cell/matrix interactions that drive the cells of a prospective organ to differentiate and collectively organize in three dimensions. Generating organs in vitro from embryonic stem (ES) cells may provide a simplified system to decipher how these processes are orchestrated in time and space within particular and between neighboring tissues. Recently, this field of stem cell research has also gained considerable interest for its potential applications in regenerative medicine. Among human pathologies for which stem cell-based therapy is foreseen as a promising therapeutic strategy are many retinal degenerative diseases, like retinitis pigmentosa and age-related macular degeneration. Over the last decade, progress has been made in producing ES-derived retinal cells in vitro, but engineering entire synthetic retinas was considered beyond reach. Recently however, major breakthroughs have been achieved with pioneer works describing the extraordinary self-organization of murine and human ES cells into a three dimensional structure highly resembling a retina. ES-derived retinal cells indeed assemble to form a cohesive neuroepithelial sheet that is endowed with the intrinsic capacity to recapitulate, outside an embryonic environment, the main steps of retinal morphogenesis as observed in vivo. This represents a tremendous advance that should help resolving fundamental questions related to retinogenesis. Here, we will discuss these studies, and the potential applications of such stem cell-based systems for regenerative medicine.  相似文献

In order to provide theoretical evidences for bamboo cultivation under the background of climate change, the method of open top chambers (OTCs) was employed to approach the effects of elevated ozone on photosynthetic pigments content, lipid peroxidation and anti oxidation enzymes in the leaves of Phyllostachys edulis. The result indicated that with the increasing of ozone concentration, the content of the chlorophyll, carotenoid and SOD activity decreased, while the content of souble protein, relative electrical conductivity, contents of MDA, O2- and the activity of POD increased.Correlation analyses showed that there was significant negative correlation between O2- content and SOD activity as well as photosynthetic pigment,whereas significant positive correlation between O2- and relative electeical conductivity, MDA, POD as well as soluble protein. It was summarried that, on one hand, long time exposure to high ozone concentration exposure, accelerated the aging of leaves, intensified the damage to membrane lipid, and destroyed the structure of cell membrane and the function of anti oxidation system as well. It affected the regular growth of P.edulis. This research was conducted to provide theoretical evidence for bamboo cultivation during this time of possible climate change.  相似文献

蓝藻结皮在干旱和半干旱区广泛分布,它们在环境状态的维持和改良过程中发挥着重要的作用。在荒漠藻固沙技术的应用过程中,温度不仅影响藻类在培养池中的培养,还影响接种后藻类的生长。因此,摸清优势固沙藻类的温度生长特征及对不同温度的生长适应性,对于接种蓝藻固沙技术的应用具有积极的意义。实验分3部分:研究了室温培养条件下3种荒漠优势蓝藻-具鞘微鞘藻(Microcoleus vaginatus)、爪哇伪枝藻(Scytonema javanicum)、纤细席藻(Phormidium tenue)的生长曲线,在不同温度(2、5、10、15、25、35℃)、开放式载体培养状态下3种蓝藻的生长状况及形态观察,以及爪哇伪枝藻在不同温度(10、15、20、25、30℃)培养条件下的光合活性、光合色素含量和伪枝藻素含量的变化。实验结果表明:(1)在液体培养基中,纤细席藻生长速率最快,高于具鞘微鞘藻和爪哇伪枝藻;(2)开放式载体培养条件下,藻株的生长速率低于液体培养,因此荒漠优势藻类的培养优先选择液体培养,具鞘微鞘藻和爪哇伪枝藻不易被细菌污染,纤细席藻容易受到细菌污染,在培养该藻株时要考虑添加抗生素类药物抑制细菌过度繁殖;(3)爪哇伪枝藻短期培养(18d)宜选择相对较高的培养温度(25-30℃),而长期培养(30d)宜选择相对较低的培养温度(15-20℃)。    相似文献
