The minichromosome maintenance (MCM) complex, consisting of six subunits, Mcm2-7, is loaded onto replication origins through loading factors (origin recognition complex [ORC], Cdc6, and Cdt1) and forms an MCM double hexamer that licenses the initiation of DNA replication. Previous studies with Xenopus egg extracts showed that loading factors, especially Cdc6, dissociate from chromatin on MCM loading, but the molecular mechanism and physiological significance remain largely unknown. Using a cell-free system for MCM loading onto plasmid DNA in Xenopus egg extracts, we found that MCM loaded onto DNA prevents DNA binding of the loading factors ORC, Cdc6, and Cdt1. We further report that a peptide of the C-terminal region of MCM3 (MCM3-C), previously implicated in the initial association with ORC/Cdc6 in budding yeast, prevents ORC/Cdc6/Cdt1 binding to DNA in the absence of MCM loading. ATP-γ-S suppresses inhibitory activities of both the MCM loaded onto DNA and the MCM3-C peptide. Other soluble factors in the extract, but neither MCM nor Cdt1, are required for the activity. Conservation of the amino acid sequences of MCM3-C and its activity in vertebrates implies a novel negative autoregulatory mechanism that interferes with MCM loading in the vicinity of licensed origins to ensure proper origin licensing.  相似文献

Sclerotinia sclerotiorum is an important plant pathogen with worldwide distribution that causes severe economic losses of various agricultural crops such as soybean. This fungus is normally controlled with synthetic chemical fungicides that pose risks to the environment, and can be harmful to human health, and they can also induce resistance in pests. The aim of this study was to investigate the potential of Trichoderma asperelloides as a biocontrol agent towards white mold disease on soybeans crops. The antagonism of two strains of T. asperelloides (T25 and T42) isolated from soil samples was determined in-vitro by dual-culture confrontation testing on nine S. sclerotiorum strains obtained from sclerotia collected on diseased soybean plants. The mycelial growth and inhibition of carpogenic and ascospore germination by T. asperelloides extracts, as well as the efficacy of these on white mold control in soybeans were evaluated. Both strains of T. asperelloides exhibited high potential of antagonism. Methanolic and ethyl acetate extracts of the two T. asperelloides strains showed excellent growth inhibition (60–100%) on all of the pathogens tested. The ethyl acetate extracts of both T. asperelloides strains exhibited the highest efficacy against carpogenic germination, decreasing by 20–30% the number of ascospores per apothecium. Strains of T. asperelloides tested were more efficient in controlling white mold than two commercial products made from Trichoderma harzianum. The new strains of T. asperelloides have potential for successful biological control of white mold disease of soybean crops in the field.  相似文献

Nuclear receptors (NRs) represent attractive targets for the treatment of metabolic syndrome-related diseases. In addition, natural products are an interesting pool of potential ligands since they have been refined under evolutionary pressure to interact with proteins or other biological targets.This review aims to briefly summarize current basic knowledge regarding the liver X (LXR) and farnesoid X receptors (FXR) that form permissive heterodimers with retinoid X receptors (RXR). Natural product-based ligands for these receptors are summarized and the potential of LXR, FXR and RXR as targets in precision medicine is discussed.  相似文献

Centrosomes are powerful and exclusive parthenogenetic agents in the Xenopus egg. We have previously shown that heterologous centrosomes from various vertebrate species were able to promote egg cleavage in Xenopus and that human centrosome activity was associated with an insoluble proteinacious structure that is not significantly simpler than the native centrosome. In this work, we have investigated the parthenogenetic capacity of more evolutionary distant centrosomes. We show that centrosomes devoid of centrioles, such as SPBs isolated from Saccharomyces cerevisiae, do not form asters of microtubules in cytoplasmic extracts from Xenopus eggs, and are inactive in the parthenogenetic test. We further show that Drosophila centrosomes which possess a typical centriole architecture, and are quite active to nucleate microtubules in Xenopus cytoplasmic extracts, are unable to trigger egg cleavage. This was observed both with centrosomes isolated from Drosophila syncytial embryos and nucleus-centrosome complexes from the Drosophila Kc23 cell line. We demonstrate that this inability could not be restored after pre-incubation of Drosophila centrosomes in the egg cytoplasm before injection. We conclude that the parthenogenetic activity of a centrosome is not directly linked to its capacity to nucleate microtubules from the egg tubulin, and that the evolutionary conserved nine-fold symmetrical structure of the centriole cannot be considered as sufficient for triggering procentriole assembly.  相似文献

The structural basis of mitotic condensation of chromosomes is one of the problems of cell biology yet to be elucidated. A variety of approaches have been used to study this problem and a large number of hypotheses have been proposed to explain the different levels of compaction of chromatin. Xenopus egg extracts, now widely used to study various aspects of cell biology, provide a valuable tool to study mitotic condensation of chromosomes. No detailed study has however yet been reported on the submicroscopic organization of condensed chromosomes in vitro in egg extracts. We present here the results of our electron microscopic studies on the organization of condensed chromosomes in vitro, using demembranated sperm nuclei and mitotic (CSF-arrested) extracts of Xenopus laevis eggs, clarified by high speed centrifugation. Upon introduction of sperm nuclei in egg extracts, the nuclei swell and the chromatin undergoes a rapid decondensation; at this stage the chromatin is formed of 10 nm fibrils. After longer incubation, the chromatin condenses, and by 2 h chromosomal structures can be visualized by staining with DAPI or Hoechst 33258. Our results on the organization of chromosomes in different stages of condensation are discussed in relation to the different hypotheses proposed to explain the process of mitotic condensation of chromosomes. Finally, this study demonstrates the feasibility of high-resolution analysis of the process of chromosome condensation.  相似文献

1 Thirty extracts of wood and bark of hardwood trees from Eastern North America were examined for insect growth‐reducing activity in a bioassay with European corn borer, Ostrinia nubilalis, and an antifeedant bioassay with the rice weevil, Sitophilus oryzae. 2 Nine of the bark extracts and four of the wood extracts showed significant growth reducing effects at 0.5% in meridic diets, whereas only two bark extracts and one wood extract showed significant antifeedant effect at the same concentration. 3 Slower growing tree species were more biologically active than fast growing ones. Isolation of the bioactive compounds in one of the active species, Prunus serotina, showed that naringenin, its derivative methoxynaringenin, and eriodictyol were responsible for the antifeedant effects.  相似文献

The allelopathic effects of Juniperus rigida litter aqueous extract (LE) on wheat and Pinus tabuliformis were studied, as well as the physiological responses to the extract. High concentration LE (0.10 g Dw/ml) significantly inhibited the seed germination and seedling growth in receptor plants. The chlorophyll content and root activity in the wheat seedlings were reduced significantly across all treatments; however, those were more prominently reduced at high concentration (0.10 g Dw/ml) but received little stimulation at low concentration (0.025 g Dw/ml) in P. tabuliformis. The content of malonaldehyde (MDA) increased with increasing concentrations of LE, except at 0.025 g Dw/ml. Activities of antioxidant enzymes (POD, CAT and SOD) in receptor plants were all significantly inhibited at high concentrations but stimulated at low concentrations. These results demonstrate that the aqueous extract from J. rigida litter has allelopathic potential. Various phenolic compounds were identified in litter aqueous extract and litter ethanol extract by HPLC. The phenolic compound content in the aqueous extract was significantly lower than that in the ethanol extract. Chlorogenic acid and podophyllotoxin were the predominant phenolic compounds in both types of litter extracts. These findings suggest that the seed germination and seedling growth of P. tabuliformis and wheat would be inhibited when planted near large amounts J. rigida litter.  相似文献

【目的】探讨入侵杂草薇甘菊Mikania micrantha对棕榈害虫红脉穗螟Tirathaba rufivena的产卵忌避活性。【方法】采用室内生物测定法,研究薇甘菊不同溶剂提取物对红脉穗螟的产卵忌避作用和杀卵活性。【结果】产卵忌避试验结果表明,薇甘菊各提取物中,以正己烷和三氯甲烷提取物对红脉穗螟的产卵忌避效果最好,两者的选择性忌避率分别为43.64%和44.20%,非选择性忌避率分别为51.60%和59.20%。对卵孵化率的影响显示,各溶剂提取物均对红脉穗螟具有一定的杀卵活性,其中三氯甲烷提取物对卵孵化率的影响最大,校正孵化率最低,仅为53.39%,而且三氯甲烷提取物引起的1龄幼虫的死亡率最高,可达42.64%。在薇甘菊三氯甲烷提取物的不同溶剂萃取物中,正丁醇萃取物对红脉穗螟的产卵忌避和杀卵活性均显著高于其他溶剂萃取物。【结论】薇甘菊提取物具有一定的产卵忌避和杀卵活性,具有用于红脉穗螟的生态防控的潜力。  相似文献

马缨丹提取物对黄曲条跳甲成虫的生物活性   总被引:4,自引:1,他引:3  
测定了马缨丹(Lantana camaraL.)地上部位(茎叶花)的不同溶剂提取物及萃取物对黄曲条跳甲Phyllotreta striolata(Fabricius)成虫的取食忌避、拒食、胃毒和触杀作用。结果表明:处理后48 h,以马缨丹乙醇回流提取物效果最好,其对黄曲条跳甲成虫的取食忌避率达94.94%;非选择性拒食率为85.48%;除正丁醇萃取物对黄曲条跳甲成虫的胃毒作用达到56.67%外,其它处理对黄曲条跳甲成虫的胃毒作用不明显;石油醚萃取物、石油醚冷浸物、乙醇回流物对黄曲条跳甲成虫的触杀作用效果最好,分别为56.67,53.33和53.33%。据实验结果可知马缨丹不同溶剂提取物及萃取物对黄曲条跳甲成虫的生物活性主要表现在对成虫较强的忌避和拒食作用。  相似文献
