One type of soil collected from Maoer-shan in Heilongjiang Province,China was selected to induce hyphal growth of Tricholoma matsutake by a soil screening experiment.It was confirmed that hyphal growth of all the tested T.matsutake isolates was significantly stimulated in soil by supplemented with 0.5%~2.0% olive oil.The aggregation of hyphae and soil resembled natural Shiro.The biomass of hyphae in the soil increases with increasing olive oil concentrations.Moreover,seedlings of Pinus densiflora grew well in the soil containing 0.5%~1.0% olive oil and were also successfully infected by T.matsutake isolate A in the soil containing 1.0% olive oil.This study established a culture system of artificial Shiro formation and also provided a premise for formulation of culture substratum for fruit body formation of T.matsutake.  相似文献

Considerable attention has been paid on the allelopathic potential of aquatic macrophytes against blue-green algae. This study compared inhibitory activity of volatile compounds that were contained in two submerged macrophytes, Ceratophyllum demersum and Vallisneria spiralis, on Microcystin aeruginosa. When the concentration of essential oils was 100 mg/L, the antialgal activities for fresh macrophytes were similar between C. demersum and V. spiralis but stronger than those for dry macrophytes. Essential oils from C. demersum in dry macrophytes showed stronger antialgal activity than those from V. spiralis. There exists a positive correlation between the concentration of the essential oils and the inhibitory activity. The essential oils were composed of fatty compounds, terpenoids, phenolic compounds, phthalates, and some unknown compounds. More than 40% of the essential oils from fresh macrophytes were composed of phthalates, whereas approximately 70% of those from dry macrophytes were composed of lipid compounds and terpenoids. However, approximately 20% of the essential oils from fresh macrophytes were composed of unknown compounds, which were unstable and undetectable in dry macrophytes. Therefore, their chemical structures and bioactivities need further study.  相似文献

The hydrolysis of olive oil catalyzed by Chromobacterium viscosum lipase (EC 3.1.1.3) in a water/isooctane two-phase system was carried out both under ultrasound and conventional stirring. The maximum activity of lipase in the ultrasonicated system was 1.75 times higher than that in the stirred system. The lipase activity was dependent on ultrasonic power and volume ratio of isooctane to water. The optimum reaction temperature in both systems was around 25°C. The stability of lipase at 25°C in the ultrasonicated system decreased more rapidly than that in the stirred system. In the presence of exogenous oleic acid, however the half-life of lipase in the ultrasonicated system was improved to a value, which was respectively half and twice of that in stirred systems with and without oleic acid. The maximum reaction rate (Vmax) was increased by ultrasonication whereas the Michaelis constant (Km) remained unaltered.  相似文献

AIMS: The mechanisms of Salmonella contamination in an oilmeal plant were investigated and the basic data were collected in order to achieve control of Salmonella in oilmeal. METHODS AND RESULTS: Salmonella was detected in all contamination vectors and environmental factors investigated, namely: operators, processing floor, dust in the air and rodents. In particular, high concentrations of Salmonella were detected on the processing floor of the manufacturing area, which has high oil content. Steam was the most effective disinfection method used for the processing floor, as the effects of heat sterilization and disinfection may work in tandem. In addition, restricting the movement of operators of the production chain remarkably reduced Salmonella contamination, even in areas of otherwise high contamination. CONCLUSIONS: Within the oilmeal plant, high Salmonella contamination rates for the processing floor represent the greatest risk of contamination of oilmeal via operators, dust in the air and rodents. Therefore, control of the processing floor is the most important means for reducing the oilmeal contamination rate. SIGNIFICANCE AND IMPACT OF THE STUDY: Specific Salmonella control methods for oilmeal plants have been established.  相似文献

AIMS: Naphthenic acids (NAs) are naturally occurring, linear and cyclic carboxylic surfactants associated with the acidic fraction of petroleum. NAs account for most of the acute aquatic toxicity of oil sands process-affected water (OSPW). The toxicity of OSPW can be reduced by microbial degradation. The aim of this research was to determine the extent of NA degradation by sediment microbial communities exposed to varying amounts of OSPW. METHODS AND RESULTS: Eleven wetlands, both natural and process-affected, and one tailings settling pond in Northern Alberta were studied. The natural wetlands and process-affected sites fell into two distinct groups based on their water chemistry. The extent of degradation of a 14C-labelled monocyclic NA surrogate [14C-cyclohexane carboxylic acid (CCA)] was relatively uniform in all sediments (approximately 30%) after 14 days. In contrast, degradation of a bicyclic NA surrogate [14C-decahydronaphthoic acid (DHNA)] was significantly lower in non process-affected sediments. Enrichment cultures, obtained from an active tailings settling pond, using commercially available NAs as the sole carbon source, resulted in the isolation of a co-culture containing Pseudomonas putida and Pseudomonas fluorescens. Quantitative GC-MS analysis showed that the co-culture removed >95% of the commercial NAs, and partially degraded the process NAs from OSPW with a resulting NA profile similar to that from 'aged wetlands'. CONCLUSIONS: Exposure to NAs induced and/or selected micro-organisms capable of more effectively degrading bicyclic NAs. Native Pseudomonas spp. extensively degraded fresh, commercial NA. The recalcitrant NAs resembled those found in process-affected wetlands. SIGNIFICANCE AND IMPACT OF THE STUDY: These results suggest that it may be possible to manipulate the existing environmental conditions to select for a microbial community exhibiting higher rates of NA degradation. This will have significant impact on the design of artificial wetlands for water treatment.  相似文献

为更好地挖掘八角(Illicium verum)挥发油合成相关基因,该文对挥发油性状差异显著的优良无性系桂角69号及普通品种砧01号叶片进行了转录组测序及组装注释,并对差异表达基因进行了GO分类和KEGG通路分析。结果表明:(1)转录本经组装后获得84 182条序列,使用NR、NT、Swiss-Prot、KEGG、KOG、GO和Pfam数据库进行序列比对,共注释了59 161条序列,筛选出30 572个差异表达基因。与砧01号相比,桂角69号叶片中上调基因有15 025个,下调基因有15 547个。(2)GO分类结果显示共有20 287个差异基因被注释。KEGG分析结果表明,有21 600个差异基因被注释到133条KEGG通路上,其中挥发油合成相关的单萜生物合成通路、萜类骨架生物合成通路、苯丙素合成通路中的芳樟醇合酶、月桂烯合酶、香叶基香叶基焦磷酸合酶、肉桂酰辅酶A还原酶、咖啡酸3-O-甲基转移酶、肉桂醇脱氢酶等关键酶基因呈差异表达。(3)转录因子分析发现差异表达基因分布于31个转录因子家族,其中MYB家族序列数量最多。该文利用转录组测序技术分析八角优良无性系与普通品种叶片的差异基因及...  相似文献

Aim: To compare the survival of four bacterial strains (Escherichia coli, Proteus mirabilis, Staphylococcus aureus, Pseudomonas aeruginosa) in pharmaceutical oils, including jojoba oil/tea tree oil, carbol oil, jojoba oil and sesame oil. Methods and Results: Oils were spiked with the test bacteria in a concentration of 104 CFU ml?1. Bacteria were extracted from oils with phosphate‐buffered saline containing 0·5% Tween 20. Aliquots of the pooled water layers were analysed by solid‐phase cytometry and plate counting. Plate counts dropped to zero for all test strains exposed for 24 h to three of the four oils. In contrast, significant numbers of viable cells were still detected by SPC, except in the jojoba oil/tea tree oil mixture and partly in sesame oil. Conclusions: Exposure of bacteria for 24 h to the two oils containing an antimicrobial led to a loss of their culturability but not necessarily of their viability. The antibacterial activity of the jojoba oil/tea tree oil mixture supersedes that of carbol oil. Significance and Impact of the Study: These in vitro data suggest that the jojoba oil/tea tree oil mixture more than carbol oil inhibits bacterial proliferation when used for intermittent self‐catherization.  相似文献
