【目的】探究不同腐殖酸浓度下参与含砷水铁矿转化的微生物类群组成和丰度变化及对砷释放的影响,预测原位高砷含水层中功能微生物群参与有机质—含砷铁矿物转化过程对砷转化释放的作用。【方法】对河套平原高砷地下水和同深度高砷沉积物中的铁还原功能群落进行富集培养,构建室内厌氧微宇宙体系,将富集菌群分别加入到实验室条件下合成的不同浓度腐殖酸(0、1.5、7、14 mg C/L)-含砷水铁矿体系中,通过体系中砷、铁形态及浓度的变化分析,结合高通量测序技术、X-射线衍射(X-ray diffractometer, XRD),探究不同条件下砷的释放固定和群落的演替。【结果】高砷地下水组(G组)和沉积物组(S组)富集得到的铁还原功能群落具有明显差异,G组中以Aeromonadaceae为特殊优势菌群,而S组中以Shewanellaceae为特殊优势菌群。微宇宙实验结果显示,S组的铁还原量相对较高且速率较快;G组与S组中液相砷形态存在明显差异,整个培养期内G组均以As(Ⅴ)为主,而S组中前期以As(Ⅴ)为主,当反应到达20 d时液相As(Ⅲ)高达3.4μmol/L,推测此时具有砷还原功能的群落占优势地位。当反应...  相似文献

Flowering time is one of important agronomic traits determining the crop yield and affected by high temperature. When facing high ambient temperature, plants often initiate early flowering as an adaptive strategy to escape the stress and ensure successful reproduction. However, here we find opposing ways in the short-day crop soybean to respond to different levels of high temperatures, in which flowering accelerates when temperature changes from 25 to 30 °C, but delays when temperature reaches 35 °C under short day. phyA-E1, possibly photoperiodic pathway, is crucial for 35 °C-mediated late flowering, however, does not contribute to promoting flowering at 30 °C. 30 °C-induced up-regulation of FT2a and FT5a leads to early flowering, independent of E1. Therefore, distinct responsive mechanisms are adopted by soybean when facing different levels of high temperatures for successful flowering and reproduction.  相似文献

Global food security is one of the utmost essential challenges in the 21st century in providing enough food for the growing population while coping with the already stressed environment. High temperature (HT) is one of the main factors affecting plant growth, development and reproduction and causes male sterility in plants. In male reproductive tissues, metabolic changes induced by HT involve carbohydrates, lipids, hormones, epigenetics and reactive oxygen species, leading to male sterility and ultimately reducing yield. Understanding the mechanism and genes involved in these pathways during the HT stress response will provide a new path to improve crops by using molecular breeding and biotechnological approaches. Moreover, this review provides insight into male sterility and integrates this with suggested strategies to enhance crop tolerance under HT stress conditions at the reproductive stage.  相似文献

古尔班通古特沙漠短命和旱生植物在沙漠东西走向上形成明显的种群变化梯度,那么沙漠藻类结皮土壤中微生物是否也具有类似地空间分布特征?为研究该科学问题,采用高通量测序技术,探究沙漠东西走向上藻类结皮土壤中细菌和真菌群落结构分布规律。在古尔班通古特沙漠西部、中部和东部区域分别采集藻类结皮土样,进行土壤理化因子分析及细菌和真菌扩增子测序,对比不同区域土壤理化性质和微生物群落空间分异特征,分析微生物多样性与环境因子相关性,最后根据微生物物种种类对其功能进行预测分析。结果表明:藻类结皮土壤氮素、全磷、全钾、pH以及土壤化学计量比碳磷比(C∶P)和氮磷比(N∶P)在沙漠不同区域存在显著性差异,其余指标空间尺度上变化梯度较小。微生物α多样性结果显示3个区域藻类结皮土壤中细菌群落丰富度和多样性存在显著性差异,东部与西部区域产生明显分化;而真菌α多样性指数无显著性差异,群落结构较为均匀,未产生分化。β多样性结果显示细菌群落在西部与东部藻类结皮间存在显著性差异,沙漠中部为沙漠西部和东部的过渡带。门水平细菌优势菌群为蓝藻门、变形菌门和拟杆菌门;真菌为子囊菌门和担子菌门。属水平上,丰度前30个属在沙漠不同区域表现...  相似文献

为探讨甜樱桃对高温胁迫的响应机制,以1年生的甜樱桃嫁接盆栽苗为试材,研究了自然高温处理及胁迫后恢复对其叶片生理和超显微结构的影响。结果表明:经连续3d日最高气温均值达57.7℃胁迫后,甜樱桃叶片过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)活性、超氧阴离子(O2·-)产生速率、非光化学淬灭系数(qN)显著增加,渗透调节物质可溶性蛋白、可溶性糖及脯氨酸含量显著提高,谷胱甘肽还原酶(GR)活性、叶绿素含量、PSII最大光化学效率(Fv/Fm)和电子传递速率(ETR)显著下降,超氧化物歧化酶(SOD)和过氧化物酶(POD)活性及光化学淬灭系数(qP)变化不显著。同时对叶片显超微结构观察发现,胁迫引起叶绿体变性,基质内囊体发生扭曲,部分基粒类囊体片层消失,出现大量巨型淀粉粒及嗜锇颗粒。恢复2d后SOD、POD、CAT、APX活性以及可溶性糖、可溶性蛋白、叶绿素总含量均显著下降,而GR活性和脯氨酸含量显著升高。以上表明,连续3d自然最高气温达57.7℃对甜樱桃叶片抗氧化系统和光合特性等产生了显著影响,叶绿体超显微结构受到明显破坏,对叶片造成了不可逆...  相似文献

规律成簇的间隔短回文重复序列(clustered regularly interspaced short palindromic repeats, CRISPR)及其相关Cas蛋白所构建的CRISPR/Cas系统是古细菌或细菌中特有的一种获得性免疫系统。研究人员将其开发成基因编辑工具之后,凭借其高效、精准和通用性强等优点迅速成为合成生物学领域的热门研究方向,在生命科学、生物工程技术、食品科学及农作物育种等多个领域引发了革命性的影响。目前基于CRISPR/Cas系统单基因编辑与调控技术日益完善,但在多重基因编辑和调控方面仍存在挑战。本文聚焦基于CRISPR/Cas系统的多重基因编辑与调控技术开发及应用,针对单个细胞内实现多位点基因编辑或调控和细胞群体内实现多位点基因编辑或调控技术,依据作用原理对其进行了系统总结和阐述,包括基于CRISPR/Cas系统的双链断裂、单链断裂以及多重基因调控技术等。这些工作丰富了多重基因编辑与调控的工具,为CRISPR/Cas系统在多领域的应用作出了贡献。  相似文献

Strains of the fungal antagonist Gliocladium virens were separated into two distinct groups on the basis of secondary metabolite production in vitro. Strains of the ‘P’ group produced the antibiotics gliovirin and heptelidic acid but not the antibiotic gliotoxin and its companion, dimethylgliotoxin. Strains of the ‘Q’ group produced gliotoxin and dimethylgliotoxin but not gliovirin or heptelidic acid. Strains from both groups produced the antibiotic viridin and phytotoxin viridiol. Gliovirin was very inhibitory to Pythium ultimum but had no activity against Rhizoctonia solani, and strains that produce it were more effective seed treatment biocontrol agents of disease incited by P. ultimum. Conversely, gliotoxin was more active against R. solani than against P. ultimum, and strains that produced it were more effective seed treatments for controlling disease incited by R. solani. These results indicate that the antibiotic profiles of strains should be considered when screening strains for biocontrol efficacy, and that it may be necessary to treat seeds with a combination of strains in order to broaden the disease control spectrum.  相似文献

本研究旨在建立一种基于双荧光素酶报告基因检测体系的法尼醇X受体(farnesoid X receptor, FXR)激动剂细胞筛选体系,以满足对FXR激动剂先导化合物的高通量筛选。通过在报告基因质粒pGL4-luc2P-Hygro中的萤火虫荧光素酶(firefly luciferase,Luc)基因上游克隆并插入来自FXR靶基因的FXR反应元件(FXR response element,FXRE)片段,构建用于筛选FXR激动剂的报告基因质粒,并结合海肾荧光素酶内参质粒,建立能够有效反映药物对FXR激动效应的双荧光素酶报告基因细胞检测体系。通过一系列优化实验,比较了过表达RXR、鼠源和人源FXR、不同的FXRE片段、FXR过表达质粒与报告基因质粒的混合比对筛选体系诱导效率和灵敏度的影响。根据上述结果,最终确定了优化条件,优化后体系Z因子达到0.83。本研究建立了一种用于FXR激动剂筛选的改良的基于双荧光素酶报告基因检测体系的细胞筛选体系,其主要特征在于,使用多段FXR靶基因上的FXRE片段叠加组成一种新型的增强型FXRE元件,而非传统的反向重复序列-1 (inverted repeats...  相似文献

An efficient monitoring and control strategy is the basis for a reliable production process. Conventional optical density (OD) measurements involve superpositions of light absorption and scattering, and the results are only given in arbitrary units. In contrast, photon density wave (PDW) spectroscopy is a dilution-free method that allows independent quantification of both effects with defined units. For the first time, PDW spectroscopy was evaluated as a novel optical process analytical technology tool for real-time monitoring of biomass formation in Escherichia coli high-cell-density fed-batch cultivations. Inline PDW measurements were compared to a commercially available inline turbidity probe and with offline measurements of OD and cell dry weight (CDW). An accurate correlation of the reduced PDW scattering coefficient µs′ with CDW was observed in the range of 5–69 g L−1 (R2 = 0.98). The growth rates calculated based on µs′ were comparable to the rates determined with all reference methods. Furthermore, quantification of the reduced PDW scattering coefficient µs′ as a function of the absorption coefficient µa allowed direct detection of unintended process trends caused by overfeeding and subsequent acetate accumulation. Inline PDW spectroscopy can contribute to more robust bioprocess monitoring and consequently improved process performance.  相似文献

Nucleoside phosphorylases are important biocatalysts for the chemo-enzymatic synthesis of nucleosides and their analogs which are, among others, used for the treatment of viral infections or cancer. S-methyl-5′-thioadenosine phosphorylases (MTAP) are a group of nucleoside phosphorylases and the thermostable MTAP of Aeropyrum pernix (ApMTAP) was described to accept a wide range of modified nucleosides as substrates. Therefore, it is an interesting biocatalyst for the synthesis of nucleoside analogs for industrial and therapeutic applications. To date, thermostable nucleoside phosphorylases were produced in shake flask cultivations using complex media. The drawback of this approach is low volumetric protein yields which hamper the wide-spread application of the thermostable nucleoside phosphorylases in large scale. High cell density (HCD) cultivations allow the production of recombinant proteins with high volumetric yields, as final optical densities >100 can be achieved. Therefore, in this study, we developed a suitable protocol for HCD cultivations of ApMTAP. Initially, optimum expression conditions were determined in 24-well plates using a fed-batch medium. Subsequently, HCD cultivations were performed using E. coli BL21-Gold cells, by employing a glucose-limited fed-batch strategy. Comparing different growth rates in stirred-tank bioreactors, cultivations revealed that growth at maximum growth rates until induction resulted in the highest yields of ApMTAP. On a 500-mL scale, final cell dry weights of 87.1–90.1 g L−1 were observed together with an overproduction of ApMTAP in a 1.9%–3.8% ratio of total protein. Compared to initially applied shake flask cultivations with terrific broth (TB) medium the volumetric yield increased by a factor of 136. After the purification of ApMTAP via heat treatment and affinity chromatography, a purity of more than 90% was determined. Activity testing revealed specific activities in the range of 0.21 ± 0.11 (low growth rate) to 3.99 ± 1.02 U mg−1 (growth at maximum growth rate). Hence, growth at maximum growth rate led to both an increased expression of the target protein and an increased specific enzyme activity. This study paves the way towards the application of thermostable nucleoside phosphorylases in industrial applications due to an improved heterologous expression in Escherichia coli.  相似文献
