AIMS: A better understanding of the role of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT) in the protection of Aspergillus niger spores against thermal stress. METHODS AND RESULTS: Conidiospores from A. niger 26 were subjected to wide range of temperatures (30, 50, 60 and 80 degrees C). The stress response was investigated by the determination of spore germination and mycelial growth of survivors under submerged cultivation. Exposure to any temperature above the optimal value induced an increase in SOD and CAT activities. PAGE demonstrated enhanced level of Cu/ZnSOD under stress conditions. We compared the influence of heat shock and superoxide-generating agent paraquat on growth and antioxidant enzyme defence and found different response to the both type of stresses. CONCLUSIONS: Heat stress elicits the enhanced synthesis of enzymes whose functions are to scavenge reactive oxygen species. These results suggested an association between thermal and oxidative stress. SIGNIFICANCE AND IMPACT OF THE STUDY: Evidence is provided for the possibility that oxidative stress plays a major role in the effect of heat in low eucaryotes such as A. niger. This knowledge may be of importance in controlling both fermentation and pathogenicity.  相似文献

Summary Spore production of Coniothyrium minitans was optimized by using response surface methodology (RSM), which is a powerful mathematical approach widely applied in the optimization of fermentation process. In the first step of optimization, with Plackett–Burman design, soluble starch, urea and KH2PO4 were found to be the important factors affecting C. minitans spore production significantly. In the second step, a 23 full factorial central composite design and RSM were applied to determine the optimal concentration of each significant variable. A second-order polynomial was determined by the multiple regression analysis of the experimental data. The optimum values for the critical components for the maximum were obtained as follows: soluble starch 0.643 (36.43 g. l−1), urea −0.544 (3.91 g l−1) and KH2PO4 0.049 (1.02 g l−1) with a predicted value of maximum spore production of 9.94 × 109 spores/g IDM. Under the optimal conditions, the practical spore production was 1.04 × 1010 spores/g IDM. The determination coefficient (R2) was 0.923, which ensure an adequate credibility of the model.  相似文献

Common polypody (Polypodium vulgare L.) belongs to desiccation-tolerant ferns. The structure of storage parenchyma of their rhizome was examined by transmission electron microscopy after dehydration and subsequent rewetting. Analysis revealed that treatment with supplemental abscisic acid resulted in protection of cells against ultrastructural damage compared to untreated ones. Dehydration rate appears to modify the ability of rhizome parenchyma to stand water stress.  相似文献

Five altitudinal pteridophyte zones are established along the northeastern altitudinal slopes of Mt. Banahaw de Lucban, Quezon, Luzon Is., Philippines using cluster- and ordination analyses, namely: Zone 1, Cyathea contaminans – Dicranopteris–Nephrolepis–Diplazium patches at 700–800 m a.s.l.; Zone 2, Sphaerostephanos hirsutus var. hirsutus – Selaginella delicatula patches at 750–900 m a.s.l.; Zone 3, Cyathea philippinensis – Selaginella patches at 900–1200 m a.s.l.; Zone 4, Cyathea philippinensis – Cyathea callosa–Asplenium cymbifolium–Selaginella cumingiana patches at 1200–1550 m a.s.l. and; Zone 5 which is further divided into Sub-zone 5A, Cyathea callosa – Cyathea loheri-Hymenophyllaceae patches at 1550–1800 m a.s.l. and Sub-zone 5B, Cyathea loheriCephalomanes apiifolia patches at 1800–1875 m a.s.l. These pteridophyte zones coincide with the woody species zones of Mt. Banahaw de Lucban but differ significantly with the altitudinal fern zones of Mt. Makiling. Stepwise multiple regression analysis reveals that altitude exhibits a linear relationship with pteridophyte species distribution. Altitude and soil pH influence 65% of the variation in principal component 1 [PC1 = 0.0839 + 0.0010(altitude) − 0.2072(soil pH); r = 0.8058] and 27% of the variation in principal component 2 [PC2 = 2.0453 − 0.0005(altitude) − 0.2560(soil pH); r = 0.5206]. Thirty-three (33) species are preferential to specific microenvironments along the altitudinal gradient, making them effective altitudinal zone markers and biodiversity conservation indicators for the forest ecosystem of Mt. Background and Aims

Human-mediated environmental change is increasing selection pressure for the capacity in plants to colonize new areas. Habitat fragmentation combined with climate change, in general, forces species to colonize areas over longer distances. Mating systems and genetic load are important determinants of the establishment and long-term survival of new populations. Here, the mating system of Asplenium scolopendrium, a diploid homosporous fern species, is examined in relation to colonization processes.

Methods

A common environment experiment was conducted with 13 pairs of sporophytes, each from a different site. Together they constitute at least nine distinct genotypes, representing an estimated approx. 95 % of the non-private intraspecific genetic variation in Europe. Sporophyte production was recorded for gametophytes derived from each parent sporophyte. Gametophytes were grown in vitro in three different ways: (I) in isolation, (II) with a gametophyte from a different sporophyte within the same site or (III) with a partner from a different site.

Key Results

Sporophyte production was highest in among-site crosses (III), intermediate in within-site crosses (II) and was lowest in isolated gametophytes (I), strongly indicating inbreeding depression. However, intragametophytic selfing was observed in most of the genotypes tested (eight out of nine).

Conclusions

The results imply a mixed mating system in A. scolopendrium, with outcrossing when possible and occasional selfing when needed. Occasional intragametophytic selfing facilitates the successful colonization of new sites from a single spore. The resulting sporophyte, which will be completely homozygous, will shed large amounts of spores over time. Each year this creates a bed of gametophytes in the vicinity of the parent. Any unrelated spore which arrives is then selectively favoured to reproduce and contribute its genes to the new population. Thus, while selfing facilitates initial colonization success, inbreeding depression promotes genetically diverse populations through outcrossing. The results provide further evidence against the overly simple dichotomous distinction of fern species as either selfing or outcrossing.  相似文献

针对生物威胁的现场处置工作,建立气溶胶芽胞表面滞留抗力的智能预测模型,以准确预测环境表面芽胞污染状况,为大规模的现场洗消任务提供重要依据,有利于实现及时反应、恰当反应和准确防护的目标。以枯草杆菌芽胞为试验菌,在气溶胶实验室进行芽胞的环境因素暴露及活力测定,以模拟环境中芽胞抗力变化规律数据为依据,采用Matlab6.1软件包中的神经网络工具箱进行抗力预测模型研究。根据研究目的、模拟环境条件和数据训练的平滑曲线等特征,设定了5个输入神经元,8个隐层节点和1个输出神经元。‘tansig’、‘purelin’为传递函数,trainlm为训练函数,网络迭代100次。模型回顾预测效率达到100%,前瞻预测效率达到91%。以实验室数据为依据,利用Matlab平台中的BP神经网络建立的芽胞气溶胶表面滞留抗力预测模型能利用环境因素信息有效预测芽胞抗力。  相似文献

以L-丙氨酸缓冲液为发芽剂,结合芬顿反应原理,观察发芽-氧化损伤效应对芽胞的杀灭效果,以期为新型炭疽疫源地净化方法的深入研究奠定基础。以腊样芽胞为试验菌,采用透射电镜、激光扫描共聚焦显微镜、活菌计数等方法观察芽胞发芽过程的超微结构、核酸含量变化,以及在芬顿反应的联合作用下发芽体的活性变化。在20~30 min的发芽过程中,芽胞核心密度降低,核心与皮质、皮质与外壁之间界限模糊,芽胞外壁和芽胞衣有破裂,通透性增加,进一步有皮质消失、细胞核与细胞质融合、细胞膜基本形成的现象;发芽体荧光强度不断增加,显示菌体中核酸的活性和含量不断增加;发芽体对化学因子的抗力明显下降,H2O2浓度为0.20 mol/L的Fenton反应系统作用60 min时,发芽体灭活可达到3.016个对数级。诱导发芽和反应的联合处理程序可显著提高芽胞的灭活水平。  相似文献

Resting spore formation and Si:N drawdown ratios were investigated under iron (Fe)- and nitrogen (N)-limited conditions using a unialgal culture of Thalassiosira nordenskioeldii and natural phytoplankton assemblages during the spring bloom in the Oyashio region. In the unialgal culture of T. nordenskioeldii, 20% and 100% of the cells formed resting spores under Fe- and N-limited conditions, respectively. The Si:N drawdown ratios were 2- and 14-fold higher in Fe- and N-limited conditions, respectively, compared to Fe- and N-sufficient conditions. At the start of the natural phytoplankton incubation, 18 among 47 identified diatom species were known resting spore-forming species. Approximately 15 common diatom species formed resting spores under Fe- and N-limited conditions. During the natural phytoplankton incubation, the percentage of the resting spores increased with time under both Fe- and N-limited conditions, reaching 25% and 40% of total diatom abundance, respectively. The Si:N drawdown ratios significantly increased with an increase in the contribution of resting spores in both the unialgal culture and natural phytoplankton incubations. These results suggest that if the bloom dominated by neritic, resting spore-forming diatom species decline by either Fe- or N-depletion, Si may be utilized preferentially to N in the upper mixed layer due to the formation of heavily silicified resting spores.  相似文献

The concentrations of airborne fungal spores were measured during 2001–2002 in two sites in Poland—one in the city and the other in the countryside. The sites differed in habitat characteristics, such as urbanisation level, vegetation and microclimate. The aim of the study was to determine if, and in which way, land use type would affect spore occurrence. The volumetric method was used, and ten easily identifiable spore types were sampled and anyalysed: Alternaria, Botrytis, Cladosporium, Epiccocum, Ganoderma, Pithomyces, Polythrincium, Stemphylium, Torula and Drechslera. The season of spore occurrence was determined using the 90% method. The fungal spores studied were very frequent in the air (in most instances at a frequency higher than 50%). The most common spores were those of Cladosporium, with a frequency range of 83.1–90.5%. In both years the proportion of Cladosporium spores was statistically significantly higher in the city. In both 2001 and 2002 the total seasonal sum of all the spores was higher in the countryside than in the city as was the Seasonal Fungal Index (SFI) values and average concentrations of Botrytis, Ganoderma and Torula. These latter three genera are usually represented as pathogens of plants. The mean spore concentrations of most taxa were significantly higher in the rural environment. Correlation coefficients between daily concentrations at both sites for most of the taxa studied were significant, but with lower correlation values between variables. Such results indicate that the values from the sites are weakly interdependent. The study confirms that land use type may very likely have an impact on the course of spore occurrence, the mean daily concentrations of spores as well as SFI values.  相似文献
