A development-specific and elicitor-inducible acyltransferase [hydroxycinnamoyl-CoA: tyramine N-(hydroxycinnamoyl)transferase (THT; EC 2.3.1.110)] that catalyzes the transfer of hydroxycinnamic acids from hydroxycinnamoyl-CoA esters to hydroxyphenethylamines was purified 988-fold to apparent homogeneity from opium poppy (Papaver somniferum L.) cell-suspension cultures. The purification procedure, which resulted in a 6.8% yield, involved hydrophobic interaction and anion-exchange chromatography, followed by affinity chromatography on Reactive Yellow-3-Agarose using the acyl donor (feruloyl-CoA) as eluent. Purified THT had an isoelectric point of 5.2, a native molecular mass of approximately 50 kDa, and consisted of two apparently identical 25-kDa subunits as determined by two-dimensional polyacrylamide gel electrophoresis. The purified enzyme was able to synthesize a variety of amides due to a relatively low specificity for cinnamoyl-CoA derivatives and hydroxyphenethylamines. The best substrates were feruloyl-CoA (VK m −113.4 mkat g−1 M−1) and tyramine (VK m −16.57 mkat g−1 M−1). The THT activity increased during development of opium poppy seedlings, occurred at high levels in roots and stems of mature plants, and was induced in cell-suspension cultures after treatment with a pathogen-derived elicitor. Immunoblot analysis using THT mouse polyclonal antibodies did not always show a correlation between THT polypeptide and enzyme activity levels. For example, despite low THT activity in leaves, an abundant 25-kDa immunoreactive polypeptide was detected. Immunohistochemical localization showed that THT polypeptides occur in cortical and xylem parenchyma, immature xylem vessel elements, root periderm, anthers, ovules, and the inner layer of the seed coat, but are most abundant in phloem sieve-tube members in roots, stems, leaves, and anther filaments. The association behavior of hydrophobically modified ethyl hydroxyethyl cellulose (HM-EHEC) and its interaction with the anionic surfactant sodium dodecyl sulfate (SDS) has been studied in the dilute concentration regime. Steady-state fluorescence probe techniques have been utilized to obtain microstructural information of the system properties and combined with macroscopic bulk information from equilibrium dialysis experiments in order to determine binding isotherms of SDS to HM-EHEC. HM-EHEC was found to self-associate and form polymeric micelles in semi-dilute aqueous solutions. c* for the self-association process was determined to be approximately 0.4%. The microviscosity of the polymeric micelles is much higher, and the micropolarity slightly higher, than that of ordinary SDS micelles. The onset of interaction between HM-EHEC and SDS was evidenced by a simultaneous strong increase in microviscosity and decrease in micropolarity upon successive addition of SDS. There is a minor, noncooperative SDS binding to the HM-EHEC starting from low concentrations of SDS (<5 mM) followed by a highly cooperative binding region at SDS concentrations ≥5 mM. The polymer–surfactant aggregates are rigid and hydrophobic with a maximum in microviscosity in the noncooperative binding region at a very low degree of SDS-adsorption.  相似文献

Calli initiated from mature embryos of Leymus racemosus (Lam. Tzvel. =L. giganteus were transferred onto the AA and DM media to produce friable embryogenic callus,from which embryogenic suspension cultures were established. Protoplasts were isolated from the embryogenic suspension cultures and were cultured either in thin-layer liquid medium or in double-layer (agar/liquid) medium. When visible calli were formed they were transferred onto the NBI agar medium or into the MBL liquid medium for further proliferation. These calli were transferred onto differentiation media of NBII and NR, where green spots were developed. Plants with both shoots and roots can be recovered from these green spots on MS Ⅱ medium containing 0.5 mg/L NAA. The results showed that the Km8p basal medium was favourable to the culture of L. racemosus protoplasts during the early stages of culture. In addition, the composition of the media added to the cultures had a marked influence on the growth of protoplasts, indicating that the nutritional requirements in this plant were different at various stages of protoplast growth and differentiation.  相似文献

自1993年以来,紫杉醇(Taxol)已成为临床上治疗乳腺癌和卵巢癌的重要药物。由于其药源植物—红豆杉属植物(TaxusL.)生长非常缓慢,体内紫杉醇含量很低,且资源有限,所以人们一直在寻找解决紫杉醇药源短缺的方法。用红豆杉属植物组织培养技术生产紫杉醇被认为是一种有潜力的方法之一,受到人们的高度重视。虽然有关这方面的报道较多[1],但该研究仍处在试验阶段,还未见用此方法商业化生产紫杉醇的报道。其重要原因是目前红豆杉属植物离体细胞的紫杉醇产量还比较低,而且不稳。人们仍在继续寻找提高紫杉醇产量的方法…  相似文献

The conformational behavior of different molecular weight fractions of a synthetic C6-oxidized derivative of cellulose were investigated by means of capillary viscometry, CD, and microcalorimetric measurements. Experiments were carried out in the presence of either monovalent or divalent counterions. The experimental data indicated that C6-oxidized cellulose can assume an ordered extended conformation at low ionic strength, induced by the intrachain repulsions of negative charges. This conformation was suggested to be very similar to the fully extended structure of cellulose. In addition to this, upon increasing the ionic strength, a conformational transition of the order-to-disorder type occurred. In fact, the screening of the electrostatic repulsions introduced a number of conformational kinks into the cellulosic backbone, which enabled the polymer to assume a more coiled conformation hence producing less viscous aqueous solutions. © 1998 John Wiley & Sons, Inc. 该文系统地研究了高产纤维素酶生产菌(TrichodermaresiTB9701)与饲料酵母混合共发酵玉米秸秆粉对合成菌体蛋白质和利用纤维素的关系,优选出一条最佳的共发酵工艺途径和条件。研究表明在以氨法处理的玉米秸秆为底物的TB-9701与饲料酵母菌的混合菌共发酵正交实验中,于pH5.0,30℃的条件下200r/min的恒温、恒速摇瓶培养8d,经测定发酵液终产物中粗蛋白(SCP)的含量达到了23.70%,总秸秆纤维的转化率达到70%以上。  相似文献

本文将鱼类抗冻蛋白应用于植物细胞的超低温保存。结果表明,在水稻悬浮细胞的两步法保存中,浓度为0.01mg/ml的抗冻蛋白具有特别的负作用,相对高浓度的抗冻蛋白则能减小细胞存活率的波动性。在玻璃化法保存中,浓度为0.2mg/ml的抗冻蛋白能改善保存效果,更高浓度的抗冻蛋白(>5mg/ml)反而会降低保存效果。环境冰晶量、抗冻蛋白浓度、低温保护剂浓度和细胞膜组成等是影响抗冻蛋白使用效果的几大因素。作者在机理分析中认为,一方面,抗冻蛋白能和冰晶作用,抑制重冰晶,防止去玻璃化;另一方面,抗冻蛋白也能和细胞膜作用,诱发膜表面冰晶形成。  相似文献
