Involvement in DNA repair of the ruvA gene of Escherichia coli

Summary The ruv operon of Escherichia coli consists of two genes, orfl1 and ruv, which encode 22 and 37 kilodalton proteins, respectively, and are regulated by the SOS system. Although the distal gene, ruv, is known to be involved in DNA repair, the function of orf1 has not been studied. To examine whether orf1 is also involved in DNA repair, we constructed a strain with a deletion of the entire ruv operon. The strain was sensitive to UV even after introduction of low copy number plasmids carrying either orf1 or ruv, but UV resistance was restored by introduction of a plasmid carrying both orfl and ruv. These results suggest that orf1 as well as ruv is involved in DNA repair. Therefore, orf1 and ruv should be renamed ruvA and ruvB, respectively.     

Mutagenicity of ptaquiloside, the carcinogen in bracken, and its related illudane-type sesquiterpenes II. Chromosomal aberration tests with cultured mammalian cells

The chromosomal aberration test using a Chinese hamster lung cell line (CHL) was carried out on ptaquiloside and its related compounds, hypoloside B, hypoloside C, illudin M and illudin S. Ptaquiloside induced chromosomal aberrations at doses as low as 4.5 μg/ml (0.0113 mM). The clastogenic effect was ph-dependent. The same activity was observed at a 90-fold higher dose at pH 5.3 in the culture medium compared with the activity at pH 74. or pH 8.0. Both hypoloside B and hypoloside C were also clastogenic at almost the same dose levels as that of ptaquiloside. Illudin M and illudin S were also potet clastogens and induced aberrations at much lower doses than ptaquiloside. These results suggest that the clastogenic effect is involved in the mechanism of carcinogenic potency of ptaquiloside in animals.     

虫生真菌双生座壳孢的研究

双生座壳孢(Aschersonia duplex Berk.)系日本卷毛蚧的病原真菌。此菌在人工培养基和自然基物上生长好,并能产生大量分生孢子。对 C 源中的蔗糖、葡萄糖、甘露醇、山梨糖;N 源中的蛋白胨,天冬素、L-酪氨酸、KNO_3利用好。生长适温22—26℃。病原流行季节相对湿度在80％以上。田间接种,对寄主致病性强,有应用潜力。     

应用明胶颗粒凝集试验检测人免疫缺陷病病毒(HIV-1)抗体

明胶颗粒凝集试验是测定HIV-1抗体的新方法。本研究将明胶颗粒凝集试验与ELISA法、蛋白印迹法和间接免疫荧光试验做了比较,观察本方法的敏感性和特异性。共检测了195份来自法国和非洲象牙海岸的血清,凡是蛋白印迹法阳性的血清,明胶颗粒凝试验都是阳性。这表明本方法是特异和敏感的,方法简便,不需特殊仪器,省时,可用于HIV-1抗体的筛选,但多数蛋白印迹法可疑的血清,明胶颗粒试验均阴性。因此,对蛋白印迹法测出的可疑者应该用数种方法进行追踪检测。     

污水和污水土地处理系统中各种水质对华西蟾蜍蝌蚪红细胞微核率的影响

本文使用蝌蚪红细胞微核率作为指示器研究明通河污水和用污水土地处理系统处理后水质的致突变性。蝌蚪在各种水样品中暴露7天。采心脏血制片。在对照组中,微核率分别为4.40‰和4.68‰。1/4明通河污水组诱发蝌蚪的微核率是17.01‰。同对照组相比有明显的差异。     

Allergy against chironomids (non biting midges) in adult atopic patients

Summary The aim of this investigation was to evaluate the prevalence of atopic sensitization to chironomids (CHI) in patients with asthma and/or rhinitis (A/R), and to study concomitant sensitization to CHI and other allergens. Skin prick tests were performed with 3 different CHI extracts as well as with common inhalant allergens in 600 consecutive patients, 495 of which had A/R. Allergen specific IgE antibodies in the sera against CHI, shell fish and cockroaches were analyzed with Magic Lite.59 (12%) of the patients with A/R had a positive skin test with CHI. Positive skin tests with house dust mites and a storage mite were more common in CHI allergic patients than in other atopic patients. Nasal or conjunctival provocation tests, performed on 23 of the patients with positive skin test with CHI, were clearly positive in 7 cases (30%), questionable in 8 (35%) and negative in 8 cases (35%).Magic Lite, performed on sera from 50 of the patients with positive skin test against CHI, was positive with CHI in 39 cases (78%), with crayfish in 33 (66%), shrimp 20 (40%), cockroach 21 (40%) and with crab in 3 cases (6%).It is concluded that sensitization against CHI is common in patients with A/R. The clinical relevance of the positive test results is, however, unknown. Concomitant sensitization with CHI, crustaceans and cockroach is common.     

Storage mites sensitivity and allergic respiratory disease

Summary In order to evaluate the frequency of skin sensitivity to Storage Mites and the role of such sensitization in respiratory allergic disease in workers with occupational exposure to stored items we studied 217 dock workers, 93 farmers and 104 white collars.From the results of skin prick tests the sensitization to sole Storage Mites appears significantly higher among people working in docks or farms, compared with a control group. This confirms the role of the working environment in inducing sensitization to Storage Mites. Rhinitis and asthma however affect nearly always (27/29 cases) people with an associated sensitization to House Dust Mites. Further studies are needed to define the allergenic importance of Storage Mites in working environments.     

Genetics of leaf blight resistance in wheat

Summary Studies on the genetics of leaf blight caused byAlternaria triticina using generation mean analysis revealed that additive components played a major role, but that dominance components also contributed significantly in controlling the variability for leaf blight resistance in wheat crosses. Furthermore, the additive x additive type of epistasis was predominant in the first three crosses, whereas in the fourth cross additive x dominance (j) and dominance x dominance (1) components of epistasis were most significant. Because of this it may be desirable to follow a simple recurrent selection scheme for higher tolerance, to isolate resistant plants from the segregating populations derived from crosses of parents of diverse origin following the pedigree method of breeding. CPAN-1887 was very tolerant to leaf blight in the present study and should be utilized in hybridization programs to develop leaf-blight-resistant varieties.     

Evidence of excited state absorption in PS II membrane fragments

Utilizing a two-beam technique in the frequency domain, the pumped absorption of PS II membrane fragments from spinach and of acetonic chlorophyll-a solutions was measured at room temperature. In a very narrow wavelength region (0.2 nm around the pump pulse wavelength) the relative test beam transmission exhibited either a decrease or an increase, respectively, dependent on the intensity of a strong pump beam. In contrast, the transmission changes of chl-a solutions were not affected by the wavelength mistuning between pump and test beam. The data obtained for PS II membrane fragments were interpreted in terms of excited state absorption of pigment-protein clusters within the light-harvesting complex of PS II. The interpretation of the small absorption band as a homogeneously broadened line led to a transversal relaxation time for chlorophyll in vivo of about 1 ps.Abbreviations PS I photosystem I of green plants - PS II photosystem II of green plants - P700 primary donor of PS I - P680 primary donor of PS II     

树干径流过程的动态响应模型

本文用实验测试法测量了干流过程。结果表明,干流过程曲线与R-L-C电路瞬态响应过程(包括输入、输出曲线)极为相似。据此,可用R-L-C电路瞬态响应过程曲线拟合干流过程,求出解析解。在计算机上调试参数,讨论其水文学意义,定性分析参数与影响干流因子的关系,取得了满意的结果。
