Aedes epactius larvae were utilized to study the infection sequence of the nuclear polyhedrosis virus (NPV) from Aedes sollicitans. From 30 min to 6 hr postinoculation, polyhedra and many free virions were observed in the larval midgut lumen. Penetration of the midgut cells by virions was not observed. The first infected nuclei were observed 12 hr postinoculation. Nucleocapsids initially exhibited electron translucent cores which became electron dense before the nucleocapsids acquired an envelope. Envelope acquisition occurred through a process of de novo membrane morphogenesis. Occlusion of the singly embedded virions began by 18 hr postinoculation with the mature rough-surfaced polyhedra averaging approximately 1 by 2 μm. Unusually long nucleocapsids (approximately two or three times the length of other nucleocapsids) were only observed in late infection period nuclei. There was no evidence that long nucleocapsids represented an early developmental stage for nucleocapsids of standard length. Infection was restricted to midgut nuclei and gastric caecae cells. Infected early instar A. epactius larvae became moribund 36 to 40 hr postinoculation and infected midgut nuclei were observed to undergo lysis. The late stages of NPV infection were observed in larvae of A. annandalei, Wyeomyia smithii, Toxorhynchites brevipalpus, and Eretmapodites quinquevittatus. Virion development and occlusion in these species was basically identical to the sequence observed in A. epactius larvae.  相似文献

The kinetics of infection with S. ratti were quantitated in normal and previously exposed C57B1 /6 mice. In primary infections, larvae penetrated the skin rapidly and were seen in peak numbers 12 h after infection. By 24 h after infection, larval numbers had declined appreciably and there was a slow decrease in numbers thereafter. Larvae were first observed in the lungs at 24 h and maximal recovery occurred at 48 h. It is thought that larval migration through the lungs is rapid. Worms were first seen in the intestines two days after infection. Maximum numbers were seen on the fifth day and worm expulsion was complete by day 10. Two moults took place in the small intestine during days 3 and 4 after infection. Rhabditiform larvae were first noted on the fourth day after infection. Mice exposed to S. ratti four weeks previously had significantly less larvae in the skin 4 and 12 h after infection but by 24 h there was no difference when compared with mice with primary infections. Peak recovery of larvae from the lungs occurred 24 h after infection; significantly less larvae were recovered on days 2 and 3 when compared with normal mice. There was a marked reduction in the adult worm burden in the gut; the number of worms recovered was less than one fifth of that seen in primary infections. Those worms which did mature were less fecund and were expelled from the intestines within 7 days of infection. It is suggested that in previously exposed animals, the migration of larvae from the skin is hastened, many of these larvae are destroyed in the lungs and that expulsion of worms which do mature in the intestines is accelerated.  相似文献

Seakem-9 calcium carrageenan, a reported anti-macrophage agent, was found to confer partial immunity in mice subsequently challenged with 5 × 105Plasmodium berghei NK65A parasitized erythrocytes. Transient parasitemias and significantly extended survival times were evident in carrageenantreated animals. It was suggested that carrageenan may have enhanced nonspecific cellular immunological mechanisms or affected specific immune reactions through the cytotoxicity to suppressor macrophages.  相似文献

A nonsusceptible and a highly susceptible strain of the silkworm, Bombyx mori, to peroral infection with a densonucleosis virus (DNV) were studied by probit analysis. Tests with the susceptible and nonsusceptible parent strains, their reciprocal F1 hybrids, the F2 hybrid, and the backcrossed hybrids to either of the parents demonstrated that the nonsusceptibility of the silkworm to DNV was inherited and controlled by a recessive gene which was not sex-linked.  相似文献

Ascorbic acid (AA) increased the phosphatase activity (pH 6.8) in 10,000 g supernatants from Anacystis nidulans. The enzyme activated by AA was deactivated by dehydroascorbic acid (DHAA). The modulation by AA/DHAA of phosphatase activity in Anacystis appears to be specific; a number of other redox compounds, known to modulate other enzymes, had no effect on the Anacystis phosphatase. A purified phosphatase preparation from Anacystis was also deactivated by DHAA. In contrast, the purified enzyme was not activated by AA, suggesting that a factor mediating the effect of AA was lost during purification. Another factor was found to protect the purified phosphatase against deactivation by DHAA. The enzyme was characterized as a phosphatase with a broad substrate specificity, an apparent molecular weight of 19,000, and a pH optimum of 6.0–7.0. Dialysis of the enzyme preparation against EDTA abolished the phosphatase activity which could be restored by Zn2+ ions and partially restored by Co2+ ions. Crude extracts also contained a latent enzyme, the phosphatase activity of which could be detected in the presence of Co2+ ions only. Zn2+ ions did not activate this enzymatically inactive protein. The Co2+-dependent phosphatase had an apparent mol. wt. of 40,000, a broad substrate specificity, and an alkaline pH-optimum. Infection of Anacystis cultures by cyanophage AS-1 resulted in a decrease in phosphatase activity. 【目的】探究单增李斯特菌溶血素O (listeriolysin O, LLO)中D3区域β8折叠片上第253位氨基酸(谷氨酰胺,Q)和第254位氨基酸(异亮氨酸,I)对单增李斯特菌(Listeria monocytogenes)感染生物学功能的影响。【方法】构建LLOQ253A和LLOI254A突变蛋白的原核表达菌株,以及利用同源重组方法构建hlyQ253AhlyI254A突变株;通过表达纯化突变蛋白,测定溶血活性;比较LLO第253位Q和第254位I均突变成丙氨酸(A)后,对细菌体外生长能力、黏附侵袭、胞内迁移和增殖能力的影响。【结果】相应位点突变后,LLO蛋白均能够正常表达。在pH 6.5条件下,所有突变蛋白和突变株的溶血活性丧失。然而,在pH 5.5条件下,LLOI254AhlyI254A恢复了溶血活性。与野生株相比,突变株的体外生长、黏附能力和胞内增殖能力均无明显差异;突变株的侵袭能力和胞间迁移能力显著低于野生株。【结论】本研究证实第253位Q和第254位I均突变成A后,单增李斯特菌在pH 6.5条件下丧失溶血活性,并降低了感染宿主细胞的能力,但具体机制还有待进一步探索。本研究为深入探究LLO结构对单增李斯特菌生物学功能的影响奠定基础,对单增李斯特菌点突变株的构建具有一定参考意义。  相似文献

A purulent laryngeal air sac infection was diagnosed in a free-ranging adult male mountain gorilla (Gorilla gorilla beringei) in Central Africa. Successful treatment included the administration of antibiotics via blow dart and surgical drainage in the field. This case occurred during an outbreak of respiratory disease of undetermined etiology in this and other gorillas in the population.  相似文献

Recombinant human granulocyte colony-stimulating factor (rhG-CSF) was investigated for its clinical efficacy in the treatment of various types of neutropenia (3 cases with idiopathic neutropenia of suspected drug induction, 5 cases with idiopathic neutropenia of other origin, and 2 cases with cyclic neutropenia). Treatment with glycosylated rhG-CSF produced in the Chinese Hamster Ovary cells at dose levels of 2–5g/kg/day caused rapid increases of neutrophil counts associated with an improvement of the infection. In cyclic neutropenia patients, marked reduction in the duration of the neutropenic period was observed with rhG-CSF administration started before the period. Intercurrent stomatitis, which occurred in 1 patient, was markedly milder as compared to a previous episode which occurred during an untreated neutropenic period.The treatment of rhG-CSF was well tolerated and no adverse events were observed, nor was there any detectable anti-rhG-CSF antibody in any patients studied; hence the clinical use of rhG-CSF is considered to be safe.These results suggest beneficial effects of rhG-CSF on the recovery of neutrophil counts in cyclic and other types of idiopathic neutropenias, as well as for the treatment of neutropenia-associated infection.  相似文献

本文报告了1990年5月对北京市92名性乱者女性和129名健康妇女进行解脲和人型支原体分离培养结果。前者的检出率为71.74%,后者为19.57%,两者同时检出为18.48%,而健康妇女其检出率则分别为43.41%、5.43%和4.65%。说明性乱妇女支原体感染率以显著高于健康妇女。提示北京地区存在支原体性病,这对进一步掌握我国性病病谱提供了新的资料。  相似文献
