Connexin43 (Cx43) gap-junction channels are highly abundant in intestinal smooth muscle but their functional impact has not been studied so far. Here, we have aimed to elucidate the functional role of Cx43 in the tunica muscularis of the mouse intestine in vivo. Transgenic mice with conditional deletion of Cx43 in smooth muscle cells (SMC) were generated. Histological investigations by immunofluorescence analyses and organ-bath recordings to assess the contractility of intestinal tissue strips were carried out. Measurements of gastrointestinal transit and of the visceromotor response by utilizing a standardized colorectal distension model to quantify alterations of visceral sensory function were also performed in SMC-specific Cx43 null mice and control littermates. Histologically, we found thickening of the tunica muscularis and a 13-fold increase of neutrophil infiltration of the gastrointestinal wall of SMC-specific Cx43 null mice. These animals also exhibited a decrease of 29% in gastrointestinal transit time. In contrast, the visceromotor response to a standardized colorectal distension was elevated, as was the contractility in SMC-specific Cx43 null mice, compared with controls. Thus, SMC-specific ablation of Cx43 in mice leads to morphological and functional alterations of the intestinal tunica muscularis, to gastrointestinal motor dysfunction and to altered visceral sensory function. We consider a function-valued trait z(t) whose pre-selection distribution is Gaussian, and a fitness function W that models optimizing selection, subject to certain natural assumptions. We show that the post-selection distribution of z(t) is also Gaussian, compute the selection differential, and derive an equation that expresses the selection gradient in terms of the parameters of W and of the pre-selection distribution.   • 1 The mammal fauna of the British Isles has seen major perturbations since the end of the last ice age, some natural and some anthropogenic. Today, 61 species of terrestrial mammals breed in the British Isles, but only 39 of them are native species, the rest have been introduced. Furthermore, 19 native species have disappeared from the fauna including all the large predators, lynx Lynx lynx, wolf Canis lupus and brown bear Ursus arctos.
  • 2 Inevitably, these changes in species composition have been accompanied by major changes in community function including changes in patterns of energy flow through the mammalian community. For example, a high percentage of all the energy now flows through the introduced rabbit Oryctolagus cuniculus population and red deer Cervus elaphus, which, in the absence of natural predators, are living at extremely high densities.
  • 3 Could the reintroduction of species help to reverse such changes in community structure and function? The successful return to the wild of species such as the Arabian oryx Oryx leucoryx and beaver Castor fiber has shown that reintroductions are certainly possible. However, the impact on community function of returned species is more difficult to evaluate.
  • 4 This question is addressed in relation to the consequences for deer populations of any possible reintroduction of the wolf to Scotland. Based on what we know of wolves elsewhere, predictions are made about the likely demography and patterns of killing behaviour of introduced wolves. These values are then used to parameterize a Leslie matrix simulation of the impact of wolves on contemporary populations of red deer in Scotland. The simulations suggest very strongly that wolves are very unlikely to have any significant impact on the high‐density populations of deer now living in the Scottish landscape.

We develop a nonparametric imputation technique to test for the treatment effects in a nonparametric two-factor mixed model with incomplete data. Within each block, an arbitrary covariance structure of the repeated measurements is assumed without the explicit parametrization of the joint multivariate distribution. The number of repeated measurements is uniformly bounded whereas the number of blocks tends to infinity. The essential idea of the nonparametric imputation is to replace the unknown indicator functions of pairwise comparisons by the corresponding empirical distribution functions. The proposed nonparametric imputation method holds valid under the missing completely at random (MCAR) mechanism. We apply the nonparametric imputation on Brunner and Dette's method for the nonparametric two-factor mixed model and this extension results in a weighted partial rank transform statistic. Asymptotic relative efficiency of the nonparametric imputation method with the complete data versus the incomplete data is derived to quantify the efficiency loss due to the missing data. Monte Carlo simulation studies are conducted to demonstrate the validity and power of the proposed method in comparison with other existing methods. A migraine severity score data set is analyzed to demonstrate the application of the proposed method in the analysis of missing data.  相似文献

In this paper I propose a reinforcement learning model for a predator preying upon two types of prey, the unpalatable (noxious) models, and the palatable mimics. The latter type of prey resembles the models in appearance so as to derive some protection from the predator who must avoid the unpalatable models. Essentially the predator is treated as a learning automaton adopting a simple reinforcement learning strategy in order to increase its consumption of palatable prey and reduce the consumption of unpalatable ones. The populations of both mimics and models are assumed to grow logistically.  相似文献

Some pan-histone-deacetylase (HDAC) inhibitors have recently been reported to exert their anti-leukemia effect by inhibiting the activity of class IIB HDAC6, which is the deacetylase of Hsp90 and α-tubulin, thereby leading to hyperacetylation of Hsp90, disruption of its chaperone function and apoptosis. In this study, we compared the effect of a class I HDAC inhibitor FK228 with the pan-HDAC inhibitor suberoylanilide hydroxamic acid (SAHA) on the Hsp90 chaperone function of K562 cells. We demonstrated that, although having a weaker inhibitory effect on HDAC6, FK228 mediated a similar disruption of Hsp90 chaperone function compared to SAHA. Unlike SAHA, FK228 did not mediate hyperacetylation of Hsp90, instead the acetylation of Hsp70 was increased and Bcr-Abl was increasingly associated with Hsp70 rather than Hsp90, forming an unstable complex that promotes Bcr-Abl degradation. These results indicated that FK228 may disrupt the function of Hsp90 indirectly through acetylation of Hsp70 and inhibition of its function.  相似文献

During winter, increased thermoregulatory demands coincide with limited food availability necessitating physiological tradeoffs among expensive physiological processes resulting in seasonal breeding among small mammals. In the laboratory, short winter-like day lengths induce regression of the reproductive tract, but also enhance many aspects of immune function. It remains unspecified the extent to which bolstered immune responses in short days represent enhanced immune function per se compared to long days or represents energetic disinhibition mediated by the regression of the reproductive tract. Cohabitation of male Siberian hamsters with intact female conspecifics can block short-day reproductive regression. We sought to determine whether female cohabitation could also block the enhanced immune function associated with short days. Adult male Siberian hamsters were housed in long or short day lengths in one of three housing conditions: (1) single-housed, (2) housed with a same sex littermate, or (3) housed with an ovariectomized female. Delayed-type hypersensitivity (DTH) responses were assessed after 8 weeks of photoperiod treatment. Housing with an ovariectomized female was not sufficient to block short-day reproductive regression, but prevented short-day enhancement of DTH responses. Housing with a male littermate did not alter reproductive or immune responses in either photoperiod. These data suggest that short day enhancement of immune function is independent of photoperiod-mediated changes in the reproductive system.  相似文献

1. Range size, population size and body size, the key macroecological variables, vary temporally both within and across species in response to anthropogenic and natural environmental change. However, resulting temporal trends in the relationships between these variables (i.e. macroecological patterns) have received little attention. 2. Positive relationships between the local abundance and regional occupancy of species (abundance-occupancy relationships) are among the most pervasive of all macroecological patterns. In the absence of formal predictions of how abundance-occupancy relationships may vary temporally, we outline several scenarios of how changes in abundance within species might affect interspecific patterns. 3. We use data on the distribution and abundance of 73 farmland and 55 woodland bird species in Britain over a 32-year period encompassing substantial habitat modification to assess the likelihood of these scenarios. 4. In both farmland and woodland habitats, the interspecific abundance-occupancy relationship changed markedly over the period 1968-99, with a significant decline in the strength of the relationship. 5. Consideration of intraspecific dynamics shows that this has been due to a decoupling of abundance and occupancy particularly in rare and declining species. Insights into the intraspecific processes responsible for the interspecific trend are obtained by analysis of temporal trends in the distribution of individuals between sites, which show patterns consistent with habitat quality declines. 6. This study shows that a profitable approach to ascertaining the nature of human impacts is to link intra- and interspecific processes. In the case of British farmland and woodland birds, changes to the environment lead to species-specific responses in large-scale distributions. These species-specific changes are the driver of the observed changes in the form and strength of the interspecific relationship.  相似文献

用电镜和细胞化学技术对毛竹[Phyllostachys edulis(Carr.)H.De Lehaie]节部“韧皮部结”发育过程中Ca^2+-ATP酶进行了超微细胞化学定位研究.结果显示:在“韧皮部结”形成期,仅细胞质膜和细胞核上具有很高的Ca^2+-ATP酶活性;随着“韧皮部结”的发育,发育期细胞质膜上的Ca^2+-ATP酶活性较形成期有所降低,而细胞核上仍保持较高的Ca^2+-ATP酶活性,胞间连丝、运输小泡膜上都具有Ca^2+-ATP酶活性;发育后期,液泡膜及内质网上也开始出现Ca^2+-ATP酶沉积物;成熟期的“韧皮部结”细胞质膜上的Ca^2+-ATP酶活性较发育期有所升高,并且在“韧皮部结”成熟的过程中,细胞核、内质网、胞间连丝、质体膜和细胞质降解物上始终都有较高的Ca^2+-ATP酶活性.实验结果表明“韧皮部结”细胞具有活跃的生理代谢以及频繁的共质体运输和信息交流.  相似文献
