Herein, the impact of the halloysite nanotubes to suppress the side effects of Asparaginase (ANase) cellular proliferation was investigated. Methods: A total of 100 adult male mice was employed. These mice were divided into four equal groups; Group 1 (control), Group 2 (ESC group) of a single dose of 0.15 ml Ehrlich cells (2 × 106) intraperitoneal infusion(IP), Group 3 (ESC + ANase group) received six doses equal treatments of Intratumoral (IT) 0.07 ml Aspragnase (7 mg/kg) over two weeks. For two weeks, Group 4 (ESC + ASNase + HNTs) received an IT administration of 0.07 ml Asparaginase stocked on Halloysite nanotubes (HNTs) (30 mg/kg) three times per week. A blood specimen was collected, and the liver was removed to be investigated histologically. Results: TEM measurements for the Halloysite nanoclay showed their tubular cylindrical shape with a mean diameter of 50 nm and an average length of 1 μm, whereas The X-ray diffraction pattern of the Halloysite nanoclay showed their characteristic peaks. ESC increases the serum levels of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, and bilirubin than control and other groups, even as albumin and total protein were decreasing. After using Halloysite Nanotube, the rates of these variables were enhanced up to 75%. The hepatocytes histological studies showed protection against Ehrlich Solid carcinoma-induced degenerative, necrotic, and inflammatory changes up to 70%. In conclusion, halloysite nanotubes have demonstrated effective removal of Ehrlich solid carcinoma in mice using an ASNase delivery system. It promoted the ASNase to inhibit the adverse effect of ANase's on the liver and remove the tumour cells.  相似文献

目的:分析先天性肝纤维化的临床及病理特点.方法:回顾性分析2008年1月-2012年8月在解放军第302医院住院治疗、资料完整的先天性肝纤维化患者的临床及病理资料.结果:先天性肝纤维化47例,男女比例25:22,平均发病年龄17.85±12.49岁,临床以门脉高压症表现为主,肝功能无明显减退,主要并发症为腹水及上消化道出血,45例肝组织病理主要表现为肝细胞板排列基本正常,汇管区增宽,炎细胞浸润不明显,较致密的纤维间隔穿插、包绕大致正常的肝实质,但不形成典型的假小叶,可伴有小胆管增生、扩张和畸形,其中35.6%伴有calori病.结论:先天性肝纤维化多以青少年时期起病,无明显性别差异,门脉高压与肝功能损害不一致,可伴有calori病,确诊需经肝组织病理学检查.  相似文献

目的:探讨肝硬化患者肝脏右叶、左叶体积变化,检测肝硬化患者门静脉血流情况,分析二者之间的关系,以及门静脉血流与肝功能之间关系。方法:本研究纳入54例肝硬化患者和40例正常人,采用超声多普勒方法分析这些受试者的肝脏体积和门静脉主干及左右分支的内径、血流速、流量数据,并通过静脉血检测白蛋白、胆红素、胆碱酯酶水平等评估患者肝功能水平。结果:肝硬化组平均年龄46.3岁,男性32例,其中childA级患者16例,childB级患者27例,childC级患者11例;正常对照组平均年龄41.8岁,男性24例。肝硬化组患者右左肝叶之比明显低于正常对照组(p〈0.05),门静脉内径和血流量明显高于正常对照组(p〈0.05).随着child分级升高,门静脉血流量也明显升高。肝硬化组门静脉右支血流量明显低于左支血流量(p〈0.05);此外肝硬化患者门静脉右支和左支血流量之比明显低于正常人群门静脉右左支之比(p〈0.05);而且肝硬化患者门静脉右左支血流量之比与右左肝叶具有明显的相关性与右左肝叶之比具有明显的相关性(r=0.64,p〈0.05)。结论:评估肝硬化病人门静脉血流情况,对于判断肝脏病理变化程度,评价治疗效果,以及选择治疗方案方面都具有重要的临床价值  相似文献

A CCD-based EPID using new crystal-assembly X-ray (CAX) converters is investigated for radiotherapy dosimetry. The proposed EPID design consists in replacing the common phosphor X-ray converters of current CCD-based EPIDs with high-stopping-power CAX converters. A Test Imaging Device (TID), consisting of a 30-mm-thick CAX converter made of Bismuth Germanate (BGO), coupled to a highly sensitive CCD camera, was used to evaluate the accessible imaging and dosimetric performance of the proposed design. The system response to dose and its dependence on photon beam energy were investigated. The effects of ghosting, dose rate, field size and phantom thickness were evaluated as well. The same measurements were also performed with our clinically used aSi-EPID so that comparisons of performance could be directly inferred. The TID displayed no detectable ghosting or sensitivity to dose rate. Its response to MU exposure was found to be linear within about ±1%. The level of glare induced in the TID and the aSi-EPID were equivalent. The TID resolution was higher than that of the aSi-EPID on the axis, but was found to decrease with off-axis distance. Finally, the image quality, assessed on the basis of signal-to-noise ratio in low dose radiographs of the larynx of a patient, was higher for the TID. The imaging performance accessible with the TID proved to be satisfying and its dosimetric capability was found to be superior to that of the current aSi-EPID.  相似文献

目的:探讨门静脉高压症脾切断流术后门静脉系统血栓形成的相关原因。方法:回顾性分析2010年4月-2011年12月我科450例因肝硬化门静脉高压症行脾切断流术患者的临床资料,应用超声多普勒检测手术前后门静脉血流速度、门静脉直径及脾静脉、肠系膜上静脉、门静脉血栓情况,用Logistic回归分析术前肝功能Child-Pugh分级、门静脉直径、门静脉血流速度、脾脏的质量及术后血小板数量与门静脉系统血栓形成的关系。结果:术前门静脉系统有血栓患者75例,占16.7%。术后门静脉血栓再形成率52.9%。Logistic单因素分析提示门静脉系统血栓形成与门静脉内径、门静脉血流速度、脾脏质量、血清总胆红素、术后血小板数量有关。多因素分析发现门静脉系统血栓形成与门静脉内径、门静脉血流速度、脾脏质量有关,而与血清总胆红素、术后血小板数量无关。结论:肝硬化门静脉高压症脾切除术后门静脉系统血栓形成与门静脉内径、门静脉血流速度、脾脏质量有关。  相似文献

Decellularization and recellularization of parenchymal organs may enable the generation of functional organs in vitro, and several protocols for rodent liver decellularization have already been published. We aimed to improve the decellularization process by construction of a proprietary perfusion device enabling selective perfusion via the portal vein and/or the hepatic artery. Furthermore, we sought to perform perfusion under oscillating surrounding pressure conditions to improve the homogeneity of decellularization. The homogeneity of perfusion decellularization has been an underestimated factor to date. During decellularization, areas within the organ that are poorly perfused may still contain cells, whereas the extracellular matrix (ECM) in well-perfused areas may already be affected by alkaline detergents. Oscillating pressure changes can mimic the intraabdominal pressure changes that occur during respiration to optimize microperfusion inside the liver. In the study presented here, decellularized rat liver matrices were analyzed by histological staining, DNA content analysis and corrosion casting. Perfusion via the hepatic artery showed more homogenous results than portal venous perfusion did. The application of oscillating pressure conditions improved the effectiveness of perfusion decellularization. Livers perfused via the hepatic artery and under oscillating pressure conditions showed the best results. The presented techniques for liver harvesting, cannulation and perfusion using our proprietary device enable sophisticated perfusion set-ups to improve decellularization and recellularization experiments in rat livers.  相似文献

目的:研究经肝动脉化疗栓塞术(transcatheter arterial chemoembolization,TACE)联合胸腺肽α1治疗原发性肝癌并发门静脉癌栓的疗效及对免疫功能的影响。方法:选择2018年9月~2020年2月我院的55例原发性肝癌并发门静脉癌栓患者,根据治疗方法分为两组。两组均开展TACE术,观察组联用胸腺肽α1。比较两组的白蛋白(Albumin,ALB)、谷氨酸转氨酶(glutamate aminotransferase,ALT)、总胆红素(total bilirubin,TBIL)、天冬氨酸转氨酶(aspartate aminotransferase,AST)、血清白介素(interleukin,IL)-4、IL-2和IL-6水平。结果:观察组的有效率明显高于对照组(35.71%vs 18.52%,P<0.05);治疗后,两组的ALB、CD4+/CD8+、CD3+、CD4+水平明显升高(P<0.05),ALT、TBIL、AST、血清IL-4、IL-2、IL-6水平、CD8+明显降低(P<0.05),且观察组上述指标明显优于对照组(P<0.05)。结论:TACE术联合胸腺肽α1对于原发性肝癌并发门静脉癌栓有显著的疗效,能改善其肝功能和免疫功能,降低血清IL-4、IL-2和IL-6水平。  相似文献

Hepatic blood vessels consist of the hepatic artery and three types of venous channels (the portal veins, the sinusoids, and the hepatic veins). This study was undertaken to analyze, by immunohistochemistry, connexin expression throughout the vascular development of the fetal mouse liver with special attention being given to portal vein development. In the adult liver, connexin37 and connexin40 were expressed in the endothelium of the portal vein and hepatic artery, but not in those of the hepatic vein and sinusoids. Connexin43 was expressed in mesothelial cells and smooth muscle cells of the portal veins. The preferential expression of connexin37 and connexin40 in portal veins was seen throughout liver development, including its primordium formation stage (10.5-day or 11.5-day stage), although connexin37 expression was transiently seen in free nonparenchymal cells in fetal stages. The differentiation of each blood vessel in the hepatic vascular system may occur in early developmental stages, soon after hepatic primordium formation. Cholesterol 7alpha-hydroxylase (CYP7A1) is regulated by bile acids through the farnesoid X receptor (FXR) mechanism in a negative feedback fashion. However, the fact that CYP7A1 is down-regulated by intraduodenal administration of bile acid, but not by intravenous administration may not be explained only by this mechanism. The aim of this study was to establish a new rat model with reconstructed or simulated enterohepatic circulation to examine if intravenous or portal administration of bile acid can regulate CYP7A1. Under biliary drainage, taurocholate (0 or 6 micromol/h/100g body weight) was administered continuously for 48h into the duodenum (ID-0/ID-6), femoral vein (IV-0/IV-6), or portal vein (IP-0/IP-6) to create a condition in which biliary bile acids were continuously lost, and a similar dose of taurocholate was supplied to the liver simultaneously. CYP7A1 activity and mRNA expression of the ID-0 group were significantly increased compared with the no treatment (NT) group. CYP7A1 activity and mRNA expression of the ID-6 group were suppressed significantly to 41 and 46% of those of the ID-0 group, respectively. In the IV-6 and IP-6 groups, however, enzyme activity and mRNA expression were decreased slightly, but the suppression was not statistically significant. The results suggested that portal as well as intravenous administration of bile acids cannot suppress bile acid synthesis as effectively as intraduodenal administration. It was concluded that an unidentified regulatory factor other than the nuclear receptors may be involved in bile acid synthesis in vivo.  相似文献

Summary The intra-glandular vascular arrangement in the adrenal has been studied using non-radioactive microspheres injected by three different routes: in-vivo injection into the left ventricle under pentobarbital anesthesia, postmortem orthograde, and postmortem retrograde injection. The doses of microspheres were 105 (average size 24.7 m), 106 (15.8 m) and 107 (9.9 m). The entrapment rate of microspheres by the medulla as compared with the whole gland was measured in the serially sectioned tissue (section thickness 60 m).The entrapment rates of 25-m microspheres differed between the orthograde and retrograde injections, while the entrapment rates for 15-m microspheres were essentially similar irrespective of the route of injection.Our results support the conclusion from previous microangiographic studies that the adrenal cortex and medulla are supplied by different arteries but have a common venous outflow, and that direct communication between cortical and medullary sinusoids is not likely. The medullary blood flow per gram tissue weight is estimated to be larger than cortical blood flow.  相似文献
