Developmental plasticity is often correlated with diversity and has been proposed as a facilitator of phenotypic novelty. Yet how a dimorphism arises or how additional morphs are added is not understood, and few systems provide experimental insight into the evolution of polyphenisms. Because plasticity correlates with structural diversity in Pristionchus nematodes, studies in this group can test the role of plasticity in facilitating novelty. Here, we describe three new species, Pristionchus fukushimae sp. nov. , Pristionchus hoplostomus sp. nov. , and the hermaphroditic Pristionchus triformis sp. nov. , which are characterized by a novel polymorphism in their mouthparts. In addition to showing the canonical mouth dimorphism of diplogastrid nematodes, comprising a stenostomatous (‘narrow‐mouthed’) and a eurystomatous (‘wide‐mouthed’) form, the new species exhibit forms with six, 12, or intermediate numbers of cheilostomatal plates. Correlated with this polymorphism is another trait that varies among species: whereas divisions between plates are complete in P. triformis sp. nov. , which is biased towards a novel ‘megastomatous’ form comprising 12 complete plates, the homologous divisions in the other new species are partial and of variable length. In a reconstruction of character evolution, a phylogeny inferred from 26 ribosomal protein genes and a partial small subunit rRNA gene supported the megastomatous form of P. triformis sp. nov. as the derived end of a series of split‐plate forms. Although split‐plate forms were normally only observed in eurystomatous nematodes, a single 12‐plated stenostomatous individual of P. hoplostomus sp. nov. was also observed, suggesting independence of the two types of mouth plasticity. By introducing these new species to the Pristionchus model system, this study provides further insight into the evolution of polymorphisms and their evolutionary intermediates. © 2013 The Linnean Society of London  相似文献

Chagas disease is a highly prevalent zoonosis in Mexico, Central, and South America. Early cardiac involvement is one of the most serious complications of this disease, and conduction disturbances may occur at an early age. We describe a young pregnant woman with Chagas disease and a high degree atrioventricular block, who required implantation of a permanent dual chamber pacemaker. Using an electroanatomic navigation EnSite NavX® system the pacemaker was successfully implanted with minimal fluoroscopic exposure. This case demonstrates the safety and feasibility of using an electroanatomic navigation system to guide permanent pacemaker implantation minimizing x-ray exposure in pregnant patients.  相似文献

Zusammenfassung Zur Bestimmung des cytotischen Membran-Turnovers wurden morphometrische Messungen an über 60 Zellen der Art Amoeba proteus durchgeführt. Danach nehmen diese Amöben 0,14% ihrer Zelloberfläche pro Minute durch permanente Endocytose in das Cytoplasma auf. A. proteus benötigt also insgesamt 12 Std, um die gesamte Zellmembran während der normalen Bewegung einmal zu erneuern. Infolge des geringen Membranturnovers kann der permanenten Endocytose keine aktive Bedeutung für die Erzeugung der Bewegungstriebkraft zugesprochen werden. In Übereinstimmung mit dieser Vermutung ließ sich eine Abhängigkeit zwischen Fortbewegungsgeschwindigkeit und Endocytoseintensität nicht nachweisen.Entsprechende Messungen mit drei verschiedenen Endocytoseinduktoren ergaben für die induzierte Endocytose in Abhängigkeit von der verwendeten Substanz eine wesentlich höhere Ingestionsrate von 0,43–2,25%/min. Derartige Spitzenwerte können allerdings nur innerhalb eng begrenzter Zeiträume von 15–30 min erzielt werden. Vergleicht man dagegen die Membranaufnahme während der permanenten und induzierten Endocytose über längere Zeitintervalle (4–5 Std), so bleibt die induzierte Endocytose mit 0,05–0,12%/min in der Intensität deutlich hinter der permanenten Endocytose (0,14%/min) zurück. Eine Erhöhung der Temperatur auf 30° und eine Erniedrigung auf 15°C bringen beide Endocytoseformen zum Erliegen.Die permanente Endocytose muß bei Amöben neben der Phagocytose als der wichtigste Mechanismus zur kontinuierlichen Aufnahme gelöster und suspendierter Stoffe (bis zur Größenordnung von Bakterien) angesehen werden.
Pinocytosis and locomotion of amoebae
Summary Cytotic membrane turnover of Amoeba proteus was morphometrically studied in more than 60 cells. The results obtained indicate that 0.14% of the total cell membrane area per minute is ingested by permanent endocytosis. Consequently during normal locomotion the total cell membrane area is renewed once within 12 hours.This rate is too low to play any role in the generation of motive force. No correlations were found between the rates of locomotion and permanent endocytosis.Comparative measurements on cells treated with three different substances inducing endocytosis reveal that induced endocytosis leads to an increased rate of membrane ingestion of 0.43–2.25%/min depending on the substance used. These high rates, however, are only maintained during short periods of time (15–30 min). When the rates are calculated on the basis of long periods of time (4–5 hours), it is obvious that induced endocytosis (0.05–0.12%/min) is less effective in long term membrane turnover than permanent endocytosis (0.14%/min). Endocytotic activity is completely abolished by both the increase and decrease in temperature to 30°C and 15°C respectively.In addition to discontinuous phagocytosis permanent endocytosis is an important mechanism for continuous ingestion of fluid including particles up to the size of bacteria.

PACAP exerts neuroprotective effects under various neurotoxic conditions in vitro. In vivo, it reduces brain damage after global and transient focal ischemia. The present study investigated whether PACAP has neuroprotective effects when applied before the onset of permanent ischemia. Rats were given bolus injections of PACAP38 intracerebroventricularly, and then underwent permanent middle cerebral artery occlusion. The results show that 2 μg of PACAP significantly reduced the infarct size measured 12 and 24 h after the onset of ischemia. No further reduction was obtained by a 7-day pretreatment. PACAP also ameliorated certain sensorimotor deficits. Our present study provides further evidence for the neuroprotective effects of PACAP, and implies that it might be a promising preventive therapeutic agent in ameliorating ischemic brain damage.  相似文献

Stomata are consistently patterned so that they are not in contact. This patterning is violated in the too many mouths (tmm) and four lips (flp) mutations of Arabidopsis thaliana (L.) Heynh. which have stomatal clusters in the first-formed leaves. To clarify the function of both genes in stomatal initiation and patterning, the phenotypes of many different organs were quantified. The flp mutation affects dorsiventral and cylindrical organs differentially with respect to the frequency of clustering. The tmm mutation has a more complex region-specific phenotype in that some regions lack stomata entirely, other regions have excess stomata, and the flower stalk exhibits an apex-to-base gradient from excess to no stomata. This suggests that TMM represents an unusual type of gene regulating plant cell development in that it can either influence stomatal initiation in a positive or negative fashion depending on region. Since the frequencies of initiation and clustering can be uncoupled in tmm, these two functions are under separate region-specific control. Analysis of double mutants shows that tmm and flp in some cases show region-specific interactions in both cluster formation and initiation, and that there may be subpopulations of stomata under different genetic control. Abstract. Short-term field experiments are often used to predict and evaluate long-term management effects. Based on a mowing experiment in two calcareous fens near Lake Neuchâtel, Switzerland, we investigated whether short-term treatment effects (i.e. during the first four years) were confirmed by long-term results (13 - 14 yr). Plots were mown in summer or in winter or left unmown. The main long-term trends in overall species composition (based on percentage cover estimates) were already observable in the first four years: mown and unmown plots diverged, whereas summer-cut and winter-cut plots remained similar. At the individual species level, however, short-term and long-term treatment effects differed considerably: many species whose abundance seemed affected by treatments during the first four years showed no response in the long term, and vice versa. These discrepancies were similar when based on cover estimates or on counts of shoots. Species responses did actually depend on the time scale considered. Short-term and long-term treatment effects on species richness were similar (i.e. a decrease in unmown plots), although only long-term effects were significant. Treatment effects on the above-ground biomass varied considerably, and short-term trends (lower biomass in unmown plots) differed from long-term trends (higher biomass in unmown plots). Our sites showed little overall change in species composition during the period investigated, and treatment effects were low compared with other similar experiments. If study sites are less stable or treatment effects more drastic, a short-term evaluation is expected to be even less reliable. Knowledge on species dynamics at a site may help to choose the adequate spatial and temporal scale of investigation, and thus increase the efficiency of management experiments.  相似文献

A three‐dimensional model of the stomatostylet and associated structures has been reconstructed from serial thin sections of Aphelenchus avenae, a representative of Tylenchomorpha, a group including most plant parasitic nematodes. The reconstruction is compared with previous work on bacteriovorous cephalobids and rhabditids to better understand the evolution of the stylet and its associated cells. Two arcade syncytia (“guide ring”) line the stylet shaft, supporting the hypothesis that the stylet shaft and cone (into which the shaft extends and which is not lined by syncytia) are homologous with the gymnostom of cephalobids, the sister taxon of tylenchids. Epidermal syncytia, HypA, HypB, HypC, and HypE, line the cephalic framework, vestibule, and vestibule extension, congruent with the hypothesis that these components are homologous with the cephalobid cheilostom. Relative to outgroups, HypC is expanded in A. avenae, enclosing sensilla that fill most of the cephalic framework. The homolog of syncytium HypD in the cephalobid Acrobeles complexus is not observed in A. avenae. Arcade syncytia are reduced compared with those of cephalobids. Stylet protractor muscles in A. avenae are homologous with the most anterior set of radial muscles of cephalobids. Observations to date test and verify our previous hypotheses of homology of the stomatostylet with respect to the stoma of bacteriovorous outgroups. Reconstruction of the stegostom and pharynx will provide further tests of homology and evolution of feeding structure adaptations for plant parasitism. J. 用不同浓度秋水仙素处理美洲黑杨(Populus deltoides)与小叶杨(Populus simonii)杂种后代以诱导产生多倍体,并采用流式细胞术(FCM)对初选的多倍体材料进行DNA含量分析.结果表明:小于0.1%秋水仙素浓度处理对诱导多倍体无作用;0.3%及其以上浓度处理对诱导产生多倍体效果较好,但随处理浓度增加秋水仙素对杨树的毒害作用增大, 0.5%浓度处理可使顶芽死亡率高达71.43%,且在顶芽死亡的植株中未发现多倍体单株.应用气孔性状对388株扦插苗进行多倍体初选后,采用流式细胞术对初选的5株拟多倍体进行DNA含量分析,鉴定到2株四倍体材料.获得的四倍体材料与二倍体植株在株高和叶绿素含量上差异显著,但在SOD和POD活性上无显著性差异.  相似文献

Questions: 1. Which plant traits and habitat characteristics best explain local above‐ground persistence of vascular plant species and 2. Is there a trade‐off between local above‐ground persistence and the ability for seed dispersal and below‐ground persistence in the soil seed bank? Locations: 845 long‐term permanent plots in terrestrial habitats across the Netherlands. Methods: We analysed the local above‐ground persistence of vascular plants in permanent plots (monitored once a year for ca. 16 year) with respect to functional traits and habitat preferences using survival statistics (Kaplan‐Meier analysis and Cox’ regression). These methods account for censored data and are rarely used in vegetation ecology. Results: Local above‐ground persistence is determined by both functional traits (especially the ability to form long‐lived clonal connections) and habitat preferences (especially nutrient requirements). Above‐ground persistence is negatively related to the ability for dispersal by wind and to the ability to accumulate a long‐term persistent soil seed bank (‘dispersal through time’) and is positively related to the ability for dispersal by water. Conclusions: Most species have a half‐life expectation over 15 years, which may contribute to time lags after changes in habitat quality or ‐configuration (‘extinction debt’). There is evidence for a trade‐off relationship between local above‐ground persistence and below‐ground seed persistence, while the relationship with dispersal in space is vector specific. The rate of species turnover increases with productivity.  相似文献
