Conidial spores are often used as the infectious agent during insect biocontrol applications of entomopathogenic fungi. Here we show differential virulence of conidia derived from Metarhizium anisopliae strain EAMa 01/58-Su depending upon the solid substrata used for cultivation, where LC50 values differed by up to ~10-fold (5.3×106?4.5×105 conidia/ml) and LT50 values by ~40% (9.8?7.1 d). This fungal strain is also known to secrete proteins that are toxic towards adult Mediterranean fruit flies, Ceratitis capitata, and the Greater wax moth, Galleria mellonella, larvae. In vitro production and intrahemoceol injection using G. mellonella as the host was used to test fractions during purification of the protein toxins, demonstrating that they elicited defence-related responses including melanisation and tissue necrosis. Production of these proteins/peptides along with a number of potential cuticle degrading enzymes was confirmed both in vitro and during the infection process (in vivo). Two-dimensional gel electrophoresis, followed by gel elution and bioassay, was used to identify at least three proteins or peptides (molecular mass=11, 15 and 15 kDa) as mediating the observed insect toxicity. These data demonstrate that in vitro screening for insect toxins can mimic in vivo (i.e. during the infection process) secretion and applies the use of proteomics to invertebrate pathology.  相似文献

The short- to medium-term viability and growth of Metarhizium anisopliae var. acridum conidia were investigated when combined with six insecticides, at three different concentrations. All of the insecticides used in this study were suitable for immediate spraying with M. anisopliae var. acridum conidia except for fenitrothion. Fipronil, teflubenzuron, and fenitrothion formulations significantly reduced conidial viability over time. The 10% teflubenzuron treatment caused loss of viability relatively quickly with 9.9% germination after 28 days. Mycelial growth was affected by all the treatments except fenitrothion.  相似文献

[背景] 表观遗传酶类化学抑制剂对真菌的影响研究主要集中在新次生代谢产物挖掘方面,而对大量已知次生代谢物含量的变化却关注较少。金龟子绿僵菌是一种常用杀虫真菌,能代谢出多种已知生物活性物质,其含量可能会影响到该菌与环境间关系及利用潜力。[目的] 评估组蛋白去乙酰化酶和DNA甲基转移酶的化学抑制剂对金龟子绿僵菌代谢物安全性和可利用性的影响。[方法] 在金龟子绿僵菌培养基中添加表观遗传酶类化学抑制剂,培养一定时间后用高分辨液质联用及标准品对照方法分析次生代谢产物变化。根据差异代谢物的生物活性评估化学抑制剂的影响。[结果] 高分辨液质联用分析结果表明当抑制剂浓度达500μmol/L时,金龟子绿僵菌有16种主要次生代谢产物出现明显量的变化,包括destruxin A、A1、A2、B、B1、B2、E、E2、Ed、didesmethyldestruxin C、dihydrodestruxin A、desmethyldestruxin B、12-hydroxyovalicin、subglutinol C、fungerin和ustilagic Acid C。其中,丁酸钠处理可使15种主要代谢物含量升高。苯甲酰胺可使12种主要代谢物含量升高。伏立诺他虽然仅能使10种主要代谢物含量升高,但部分代谢物的升高幅度明显高于前两者。2种DNA甲基转移酶抑制剂可使金龟子绿僵菌代谢物中绿僵菌素类代谢物含量普遍下降。[结论] 组蛋白去乙酰化酶抑制剂可引起金龟子绿僵菌主要代谢物含量普遍升高,而DNA甲基转移酶抑制剂使金龟子绿僵菌的绿僵菌素含量普遍下降。由于变化的代谢物都具有显著的杀虫、免疫抑制或抗菌抗癌等生物活性,因此上述化学抑制剂可增强或降低金龟子绿僵菌对环境中昆虫毒性,同时也增加或降低其代谢物利用潜力。另外,subglutinol C、fungerin和ustilagic Acid C是首次在金龟子绿僵菌中被发现。  相似文献

This study employed the post-real-time PCR application, high resolution melting (HRM) analysis, in order to differentiate between characterised clinical and reference Cryptosporidium parvum samples obtained from Cork University Hospital (Cork, Ireland) and the Cryptosporidium Reference Unit (Swansea, Wales). A sample set composed of 18 distinct C. parvum gp60-subtypes of the IIa gp60-subtype family (an allele family accounting for over 80% of all cryptosporidiosis cases in Ireland) was employed. HRM analysis-based interrogation of the gp60, MM5 and MS9-Mallon tandem repeat loci was found to completely differentiate between 10 of the 18 studied gp60-subtypes. The remaining eight gp60-subtypes were differentiated into three distinct groupings, with the designations within these groupings resolved to two to three potential gp60-subtypes.The current study aimed to develop a novel, reproducible, real-time PCR based multi-locus genotyping method to distinguish between C. parvum gp60-subtypes. These preliminary results support the further expansion of the multi-locus panel in order to increase the discriminatory capabilities of this novel method.  相似文献

被动游泳运动可诱发小鼠抑郁样行为,游泳环境的改变已成为抑郁样行为严重程度影响因素之一。观察不同水质、水温及持续时间对被动游泳小鼠抑郁样行为的影响,并初步探讨肠道菌群组成与抑郁样行为的关系。通过不同条件下的被动游泳运动建立抑郁样行为小鼠模型。采用糖水偏好实验及强迫游泳实验评价其行为学变化;采用16S rDNA高通量测序技术及实时荧光定量PCR技术对小鼠肠道菌群进行分子生态学分析。被动游泳16周后,各模型组小鼠体质量及糖水偏爱度均较正常对照组降低,而不动时间则有所延长。其中,室温海水游泳15 min小鼠体质量及糖水偏爱度降低程度最大,不动时间最长,与正常对照组比较,均具有显著性差异(P<0.05)。各模型组小鼠肠道菌群Chao指数、Shannon指数及PCoA分析均较正常对照组具有显著性差异(P<0.05),其从门水平到属水平的丰度也发生不同程度改变。其中,室温海水游泳15 min小鼠肠道菌群组成变化程度最大,并发生拟杆菌属、普氏菌属等多个菌属的富集以及乳杆菌属丰度的减少,实时荧光定量PCR实验也得到了较为一致的结果(P<0.05)。以上结果表明,被动游泳运动可导致小鼠抑郁样行为的发生,其肠道菌群组成也发生明显改变;同时,菌群组成的改变会随着抑郁样行为的严重程度而有所变化。  相似文献

Reduced genetic diversity through inbreeding can negatively affect pathogen resistance. This relationship becomes more complicated in social species, such as social insects, since the chance of disease transmission increases with the frequency of interactions among individuals. However, social insects may benefit from social immunity, whereby individual physiological defenses may be bolstered by collective‐level immune responses, such as grooming or sharing of antimicrobial substance through trophallaxis. We set out to determine whether differences in genetic diversity between colonies of the subterranean termite, Reticulitermes flavipes, accounts for colony survival against pathogens. We sampled colonies throughout the United States (Texas, North Carolina, Maryland, and Massachusetts) and determined the level of inbreeding of each colony. To assess whether genetically diverse colonies were better able to survive exposure to diverse pathogens, we challenged groups of termite workers with two strains of a pathogenic fungus, one local strain present in the soil surrounding sampled colonies and another naïve strain, collected outside the range of this species. We found natural variation in the level of inbreeding between colonies, but this variation did not explain differences in susceptibility to either pathogen. Although the naïve strain was found to be more hazardous than the local strain, colony resistance was correlated between two strains, meaning that colonies had either relatively high or low susceptibility to both strains regardless of their inbreeding coefficient. Overall, our findings may reflect differential virulence between the strains, immune priming of the colonies via prior exposure to the local strain, or a coevolved resistance toward this strain. They also suggest that colony survival may rely more upon additional factors, such as different behavioral response thresholds or the influence of a specific genetic background, rather than the overall genetic diversity of the colony.  相似文献

The 37-kDa laminin receptor precursor/67-kDa laminin receptor (LRP/LR, also known as ribosomal protein SA, RPSA) has been reported to be involved in cancer development and prion internalization. Previous studies have shown that the LRP/LR is expressed in a wide variety of tissues. In particular, expression of LRP/LR mRNA may be closely related to the degree of PrPSc propagation. This study presents a detailed investigation of the LRP/LR mRNA expression levels in eleven normal ovine tissues. Using real-time quantitative PCR, the highest LRP/LR expression was found in neocortex (p < 0.05). Slightly lower levels were found in the heart and obex. Intermediate levels were seen in hippocampus, cerebellum, spleen, thalamus, mesenteric lymph node, and the lowest levels were present in liver, kidney, and lung. In general, the LRP/LR mRNA levels were much higher in neuronal tissues than in peripheral tissues. The observation that differences in LRP/LR mRNA expression levels are consistent with the corresponding variation in PrPSc accumulation suggests that the 37-kDa/67-kDa laminin receptor may be involved in the regulation of PrPSc propagation.  相似文献

为建立检测法夫酵母JMU-MVP14中虾青素合成相关基因在不同生长时期表达水平的实时定量PCR方法,构建法夫酵母JMU-MVP14的管家基因β-actin、gpd、18S rRNA的标准质粒,进行实时定量PCR,制作标准曲线及回归方程.β-actin基因标准曲线相关系数(R2)=0.9956,扩增效率(E) =96.93%;gpd基因标准曲线相关系数(R2) =0.9901,扩增效率(E) =93.78%;18S rRNA基因标准曲线相关系数(R2) =0.9981,扩增效率(E)=98.76%.3个基因片段的熔解曲线均呈单峰;扩增曲线呈典型的S型动力学曲线,指数期和平台期明显,为理想的熔解曲线和扩增曲线.用geNorm软件对三个管家基因的稳定性进行分析,三个基因的稳定性排序为β-actin> 18S rRNA> gpd,故β-actin和18S rRNA较适合作为研究法夫酵母JMU-MVP14定量实验的内参基因.  相似文献

Autophagy is a highly conserved process that maintains intracellular homeostasis by degrading proteins or organelles in all eukaryotes. The effect of autophagy on fungal biology and infection of insect pathogens is unknown. Here, we report the function of MrATG8, an ortholog of yeast ATG8, in the entomopathogenic fungus Metarhizium robertsii. MrATG8 can complement an ATG8-defective yeast strain and deletion of MrATG8 impaired autophagy, conidiation and fungal infection biology in M. robertsii. Compared with the wild-type and gene-rescued mutant, Mratg8Δ is not inductive to form the infection-structure appressorium and is impaired in defense response against insect immunity. In addition, accumulation of lipid droplets (LDs) is significantly reduced in the conidia of Mratg8Δ and the pathogenicity of the mutant is drastically impaired. We also found that the cellular level of a LD-specific perilipin-like protein is significantly lowered by deletion of MrATG8 and that the carboxyl terminus beyond the predicted protease cleavage site is dispensable for MrAtg8 function. To corroborate the role of autophagy in fungal physiology, the homologous genes of yeast ATG1, ATG4 and ATG15, designated as MrATG1, MrATG4 and MrATG15, were also deleted in M. robertsii. In contrast to Mratg8Δ, these mutants could form appressoria, however, the LD accumulation and virulence were also considerably impaired in the mutant strains. Our data showed that autophagy is required in M. robertsii for fungal differentiation, lipid biogenesis and insect infection. The results advance our understanding of autophagic process in fungi and provide evidence to connect autophagy with lipid metabolism.  相似文献
