The effect of a temperature close to the freezing point (chilling) on the nitrate reductase system of leaf discs of Cucumis sativus L. cv. Kleine Groene Scherpe was determined in the absence and presence of light. The capacity of leaf discs in the light (250 μE m−2 s−1 ) at 20°C to increase in vivo and in vitro nitrate reductase activity, was unaffected by chilling pretreatment in the dark, but 4 h of chilling pretreatment in the light (250 μE m−2 s−1 ) decreased the capacity to less than 50% of the unchilled control. The chilling inhibition of the capacity to increase nitrate reductase activity was of a photooxidative nature since it only occurred in the presence of light and oxygen. Plants grown at a low light intensity (65 μE m−2 s−1 ) lost 95% of their capacity to increase nitrate reductase activity, while plants grown at 195 μE m−2 s−1 retained 80% of their nitrate reducing capacity after 6 h chilling pretreatment in the 250 μE m−2 s−1 light. Previously induced nitrate reductase activity was also affected by light during chilling. A lag phase of 7 h preceded a fast phase of decrease in activity. Both in vivo and in vitro activity decreased to 15% of the control value after 18 h of chilling in the light. It is concluded that the induction mechanism of nitrate reductase is primarily affected by photooxidation during chilling. The decrease in nitrate reductase activity is attributed to a decrease in the amount of activity enzyme. 相似文献

The aim of the present study was to isolate and identify Aspergillus species associated with saffron plants in the city of Birjand (South Khorasan Province, Iran) as well as to assess their aflatoxin B1 production. Sampling was performed during 2013–2014 crop season. Aspergillus species were isolated and purified using general and specific culture media. Growth rates and macroscopic and microscopic characteristics of the isolates were determined using yeast extract, Czapek yeast extract, malt extract and creatine sucrose agar media at 25 and 37 °C. DNA was extracted by the modified CTAB method and beta-tubulin, calmodulin and internal transcribed spacer genes were amplified and sequenced. Phylogenetic position of the isolates was determined against other Aspergillus species. Thin layer chromatography was used to investigate the production of aflatoxin B1 by Aspergillus isolates. Based on the morphological characteristics, shape and colour of the colonies, and sequencing results, the isolates belonged to Aspergillus terreus, A. flavus, A. flavipes and A. niger species. Only A. flavus isolates were aflatoxin B1 producers. We concluded that the soil of the studied saffron fields contained several species of Aspergillus, with A. flavus significantly affecting crop production through contamination of the crop by aflatoxin. 相似文献

There are conflicting opinions on the relative importance of photosynthetic limitations under salinity. Quantitative limitation analysis of photosynthesis provides insight into the contributions of different photosynthetic limitations, but it has only been applied under saturating light conditions. Using experimental data and modelling approaches, we examined the influence of light intensity on photosynthetic limitations and quantified the osmotic and ionic effects of salinity on stomatal (LS), mesophyll (LM), biochemical (LB) and light (LL) limitations in cucumber (Cucumis sativus L.) under different light intensities. Non‐linear dependencies of LS, LM and LL to light intensity were found. Osmotic effects on LS and LM increased with the salt concentration in the nutrient solution (Ss) and the magnitude of LM depended on light intensity. LS increased with the Na+ concentration in the leaf water (Sl) and its magnitude depended on Ss. Biochemical capacity declined linearly with Sl but, surprisingly, the relationship between LB and Sl was influenced by Ss. Our results suggest that (1) improvement of stomatal regulation under ionic stress would be the most effective way to alleviate salinity stress in cucumber and (2) osmotic stress may alleviate the ionic effects on LB but aggravate the ionic effects on LS. 相似文献

Photosynthetic activities and the redox states of photosystem I (PSI) and photosystem II (PSII) in intact leaves of cucumber plants (Cucumis sativus L.), as well as the sucrose and starch contents were examined under conditions of ongoing soil water deficit imposed by the cessation of watering. As the soil drought progressed, the maximum rate of photosynthetic CO2 fixation was shown to decrease. These changes in the maximum photosynthetic rate occurred synchronously with changes in the maximum quantum yield of photosynthesis. A was accumulated only at photon flux densities of about 100 mol/(m2 s). The conditions of soil water deficit promoted sucrose accumulation in the source leaf, which was paralleled by a substantial decrease in the amount of starch in the same leaf. The highest content of sucrose in the leaf after a 7-day drought was correlated with the largest decrease in photosynthetic activity. The shoot apex development during the life cycle of Crocus sativus L. was characterized by light microscopy and two-dimensional gel electrophoresis. Using silver staining of polyacrylamide gels, numerous quantitative and qualitative changes in the populations of polypeptides were observed during transition from vegetative to prefloral and from prefloral to floral stages. Using 80 g protein, we were able to detect 352 polypeptidic spots. In comparison with the vegetative apex, 89 new polypeptides were identified in the prefloral meristem and 29 polypeptides were missing. In the reproductive meristem, 94 new spots were identified and 44 spots were missing. Thus, substantial quantitative and qualitative changes in the populations of polypeptides occurred during the prefloral stage, a point of no return in plant development, i.e., and before floral primordia initiation. 相似文献

Summary. Neurolathyrisim is a motor neuron disease characterized by spastic paraparesis in the hind legs, and is caused by grass pea, Lathyrus sativus, which contains the excitotoxic amino acid, 3-N-oxalyl-L-2,3-diaminopropanoic acid (L--ODAP), an -amino-3-hydroxy-5-methyl-4-isoxazole propionic acid (AMPA)-type glutamatergic receptor agonist. In an attempt to make a useful model of this disease, the CNS distribution and toxicity of L--ODAP was studied in rat neonates after parenteral administration. L--ODAP was detected in the spinal cord as well as in the pons/medulla oblongata, though only small amounts in the latter. Repeated injection of L--ODAP resulted in rats with paraparesis of the legs, though at a low incidence rate of 0.032. These paralyzed rats displayed the severe atrophy of the ventral root of the lumbar cord as well as degenerations of motor neuron. The rats were useful models for the study of motor neuron degeneration in the spinal cord. 相似文献

We studied the surface topography of protoplasts from callus of Daucus sativus(Hofm.) Roehl. and from mesophyll cells of Nicotiana tabacumL. We also followed the distribution of actin elements of the cytomatrix in the subcortical cytoplasm layer. Protoplasts were prepared for scanning electron microscopy by a modified method without drying in the critical point apparatus. After postfixation with OsO4, carrot and tobacco protoplasts had a similar topography of the surface: it was rough and had few pores. When carrot protoplasts were not postfixed with OsO4, their surface looked different: it was folded and had 1.5-m pores, which sometimes were bordered with globules 0.3 m in diameter, or it consisted of conical cells varying in depth and size of their bases. We believe that, when protoplasts were not fixed with OsO4, they lost lipid-containing structures from their surface, and what remained was the protein carcass of the plasmalemma and the underlying layer of the cytoplasm. The inner surface of opened carrot protoplasts had elaborate topography, apparently produced by the elements of the cytomatrix, that is, a relatively thick layer of the cortical cytoplasm, where, using phalloidin–colloidal gold and transmission electron microscopy, we could observe a dense network of actin filaments. 相似文献

Sinapis alba, Raphanus sativus and Brassica napus. The chromosomes of S. alba carried four 5S and six 25S gene sites, and those of R. sativus four sites of each gene, respectively. These two species have one chromosome pair with both rDNA genes; the two are closely
located on a short arm of S. alba, while in R. sativus one is distal on the short arm (5S) and the other more proximal on the long arm (25S). In B. napus we have confirmed 12sites of 25S rDNA. The detection of 5S rDNA genes revealed 14 signals on 12 chromosomes. Of these, six
chromosomes had signals for both rDNA genes. The FISH with 5S rDNA probes detected two sites closely adjacent in four chromosomes
of B napus. These results are discussed in relation to a probable homoeologous chromosome pair in B. oleracea.
