Two matrices have been assessed for their ability to immobilize Lactobacillus casei cells for lactic acid fermentation in whey permeate medium. Agar at 2% concentration was found to be a better gel than polyacrylamide in its effectiveness to entrap the bacterial cells to carry out batch fermentation up to three repeat runs. Of the various physiological parameters studied, temperature and pH were observed to have no significant influence on the fermentation ability of the immobilized organism. A temperature range of 40–50°C and a pH range of 4.5–6.0 rather than specific values, were found to be optimum when fermentation was carried out under stationary conditions. In batch fermentation ~90% conversion of the substrate (lactose) was achieved in 48 h using immobilized cell gel cubes of 4 × 2 × 2 mm size, containing 400 mg dry bacterial cells per flask and 4.5% w/v (initial) whey lactose content as substrate. However, further increase in substrate levels tested (>4.5% w/v) did not improve the process efficiency. Supplementation of Mg2+ (1 mM) and agricultural by-products (mustard oil cake, 6%) in the whey permeate medium further improved the acid production ability of the immobilized cells under study.  相似文献

A strain ofLactobacillus acidophilus (Strain HF) was isolated from human faeces. A chloramphenicol resistant strain (HFCm) and a strain (HFCmSm) restant to both chloramphenicol and streptomycin were developed from the isolated strain (HF). All the three strains showed similarin vitro susceptibility against host defence factors like gastric acid, bile salts and volatile as well as non-volatile fatty acids.In vivo tests were done by feeding these strains to mice. Summary The batch fermentation of whey permeate to lactic acid was improved markedly by the addition of enzymehydrolyzed whey protein. Acid concentrations greater than 90 g/l were achieved at a productivity of 4.3 g/l per h and a 98% substrate use. Cell mass concentration reached 6 g/l. The acid productivity achieved is somewhat higher than that typical for fermentation of whole whey. The process economics, based on in-house hydrolyzate preparation, look promising. Presented in this paper are the experimental results showing the effects of hydrolyzate concentration on acid and cell mass production.  相似文献

本文对31例非特异性阴道炎病人和31例正常人阴道微生物群进行定性、定量分析.结果表明,阴道炎病人的阴道乳杆菌的平均浓度明显低于正常人,而肠杆菌科、葡萄球菌、棒状杆菌、类杆菌属、支原体却明显增多,阴道炎病人未发现特异性病原体,菌群失调可能是其发病因素。乳杆菌为阴道正常优势菌,对改善阴道的微环境,防止条件致病菌引起的内源性感染具有重要的生理作用。  相似文献

提取干酪乳酸杆菌细胞壁成分(LC-CW),研究其抗肿瘤作用及其机理。结果表明:100μgLC-CW,ip,连续4天,可明显抑制小鼠S18腹水瘤移植物的生长,抑瘤率为54%。增强IL-2诱导的LAK杀伤活性,可明显促进小鼠NK杀伤活性,明显促进小鼠T细胞转化,促进ConA和PHA-P诱导的IL-2产生,促进SIL-2R的减少。研究结果表明LC-CW是一种重要的抗肿瘤免疫调节因子。  相似文献

Spontaneous, phenotypically stable mutations at the -galactosidase locus (lacL-lacM) in Lactobacillus helveticus were identified and analyzed. We found that a significant number of mutations were caused by integration of a new IS element, ISL2, into these lac genes. ISL2 is 858 by long, flanked by 16-bp perfect inverted repeats and generates 3-bp target duplications upon insertion. It contains one open reading frame, which shows significant homology (40.1 % identity) to the putative transposase of IS702 from Cyanobacterium calothrix. ISL2 is present in 4–21 copies in the L. helveticus genome, but it is not found in other lactic acid bacteria. Its divergence in copy number and genomic locations in different L. helveticus strains makes it useful as a tool for strain identification by genetic fingerprinting.  相似文献

Abstract A genomic library of Lactobacillus delbrueckii ssp. lactis DSM7290 in the low copy number vector pLG339, was screened for the presence of peptidase genes. Using the chromogenic substrate gly-ala-β-naphthylamide, which is not a substrate for any of the recently cloned peptidases of DSM7290, and the multiple peptidase deficient Escherichia coli strain CM89, allowed the isolation of clones, which contained the equivalent hydrolytic activity. To identify genes encoding the conserved catalytic active site of cysteine proteases, partial nucleotide sequencing with a degenerate oligonucleotide was performed on recombinant plasmids isolated from such clones. This allowed to identify two out of nine clones to carry the Lactobacillus pepC gene. A total of 2026 nucleotides were determined, and sequence analysis revealed a gene with strong homology to the recently cloned Lb. helveticus (73.2%) and Lactococcus lactis (51.03%) pepC genes, and the derived protein showed homology with the active site of a large number of cysteine proteases. The predicted open reading frame consists of 449 codons, coding for a protein of 50 909 Da. The enzyme is functional and extremely overexpressed in E. coli .  相似文献

Abstract Fourier Transform infrared (FTIR) spectroscopy can be used to identify microorganisms. This study describes the influence of culture conditions on FTIR spectra and the discrimination of Lactobacillus species found in breweries. Fifty three Lactobacillus strains were analysed by FTIR spectroscopy and identification at the species level was correct for 94% of the strains, and at the strain level for 91% of the strains.  相似文献

【目的】探究植物乳杆菌培养上清(Lactobacillus plantarum culture supernatant,LPC)对3种血清型沙门氏菌猪霍乱(Salmonella cholerae,SC)、肠炎(Salmonella enteritidis,SE)和鸡白痢(Salmonella pullorum, SP)的生长和致病性的抑制作用效果及机理。【方法】将2%LPC与3种沙门氏菌分别共培养后,采用比浊法及牛津杯抑菌圈试验检测沙门氏菌生长情况及LPC中的主要抑菌物质,使用实时荧光定量PCR (quantitative real-time polymerase chain reaction, qRT-PCR)探究沙门氏菌致病性相关基因表达水平,最后通过结晶紫染色法检测沙门氏菌的生物被膜。【结果】2%LPC能够显著抑制3种沙门氏菌的生长,其作用效果与庆大霉素(gentamicin, GM)相近且对SE的生长抑制效果优于GM,其主要抑菌物质为有机酸;2%LPC对3株沙门氏菌SPI-1编码的主要毒力基因(InvA、InvF、SopE、SopB、SipB、HilA和SipA)、SPI-2毒...  相似文献
