提高榨菜离体培养植株再生频率 总被引:11,自引:0,他引:11
采用榨菜“浙桐1号”品种为材料,以MS为基本培养基,通过对不同植物生长调节剂的组合和不同外植体等主要因素的筛选,大幅度提高了榨菜离体培养植株再生频率。结果表明,2mg/L6.BA 0.2mg/L2,4-D的组合较为适宜,其不定芽再生频率可达50%,且外植体以下胚轴为好:而CPPU和2,4-D的适宜组合为1.5mg/L 0.2mg/L,其不定芽再生频率高达66.67%,最适外植体为带柄子叶。同时,研究结果显示,添加0.25~1mg/L的GA,对榨菜已分化的不定芽的伸长有抑制作用;子叶柄和下胚轴外植体的分化具有极性现象。 相似文献

The effect of different hermetic and non-hermetic closure-types (aluminum foil, cotton bung, cotton plug, polypropylene cap and Steristopper) on potato (Solanum tuberosum L.) plantlets growth and chlorophyll contents was studied in three genotypes belonging to different maturity groups. Plantlets grown in culture tubes closed with aluminum foils and polypropylene caps had higher fresh mass and shoot length, but lower chlorophyll contents, higher senescence index and various morphological abnormalities. Non-hermetic closures like cotton plugs and Steristoppers were found optimum for plant growth without any morphological abnormalities. Besides, these plantlets exhibited low senescence index and had higher chlorophyll contents that favour acclimation to ex vitro conditions. 相似文献

本文研究白鹤芋属观赏品种绿巨人白掌的茎顶、叶片、叶柄和幼花序的组织培养和快速繁殖技术。茎顶培养以0.2mg/LNAA 3.0mg/L6-BA培养效果较好;叶片诱导适宜的培养基为0.5mg/LNAA 5.0mg/L6-BA,分化培养基为0.2mg/LNAA 3.0mg/L6-BA;叶柄以0.5mg/L2,4-D 3.0mg/L6-BA诱导效果最好,分化适宜培养基为0.5mg/LNAA 3.0mg/L6-BA;幼花序胚状体的诱导则以0.5mg/L2,4-D 5.0mg/L6-BA效果最好,成苗培养基为0.5mg/LNAA 3.0mg/L6-BA;255mg/L的KH2P04比较/比较适合于绿巨人白掌丛芽的增殖;生根培养基以1/2MS 0.50mg/LNAA较适宜。 相似文献

制备了脑组织腺苷A2A受体基因缺失的小鼠模型并对该模型进行评价。在采用2次6.2 Gy X线间隔照射对敲除A2A受体基因的雌性C57BL/6小鼠进行清髓处理后, 将野生型雄性C57BL/6小鼠骨髓细胞移植到其体内, 使其脑组织的A2A受体仍保持缺失型。然后对移植效果进行鉴定, 并对模型的生理指标进行观察和评价。结果发现: 骨髓移植6周后, 受体小鼠的白细胞性染色体基因由雌性变为雄性; 骨髓中A2A受体阳性细胞率为94.85%, 而脑内A2A受体mRNA与A2A受体基因敲除小鼠比较, 无显著差异。模型小鼠除心率略低于野生型小鼠外, 在呼吸频率、脑含水量以及脑内谷氨酸含量等生理指标上均与野生型小鼠和A2A基因敲除小鼠无显著差异。 相似文献

Advances in our understanding of skeletal stem cells and their role in bone development and repair, offer the potential to open new frontiers in bone regeneration. Tissue engineering seeks to harness the regenerative capacity innate to bone for the replacement of tissue lost or damaged through a broad range of conditions associated with an increasingly aged population. The strategy entails ex vivo expansion of multipotential populations followed by delivery to the site of damage on dynamically durable-biodegradable three-dimensional structures which provide the requisite extracellular microenvironment for stem cell driven tissue development. This review will examine bone stem cell biology, and current advances in skeletal tissue engineering through the enhancement and marrying of biologically informed and clinically relevant strategies. 相似文献

Objective: To directly ascertain the physiological roles in adipocytes of hormone‐sensitive lipase (HSL; E.C. 3.1.1.3), a multifunctional hydrolase that can mediate triacylglycerol cleavage in adipocytes. Research Methods and Procedures: We performed constitutive gene targeting of the mouse HSL gene (Lipe), subsequently studied the adipose tissue phenotype clinically and histologically, and measured lipolysis in isolated adipocytes. Results: Homozygous HSL?/? mice have no detectable HSL peptide or cholesteryl esterase activity in adipose tissue, and heterozygous mice have intermediate levels with respect to wild‐type and deficient littermates. HSL‐deficient mice have normal body weight but reduced abdominal fat mass compared with normal littermates. Histologically, both white and brown adipose tissues in HSL?/? mice show marked heterogeneity in cell size, with markedly enlarged adipocytes juxtaposed to cells of normal morphology. In isolated HSL?/? adipocytes, lipolysis is not significantly increased by β3‐adrenergic stimulation, but under basal conditions in the absence of added catecholamines, the lipolytic rate of isolated HSL?/? adipocytes is at least as high as that of cells from normal controls. Cold tolerance during a 48‐hour period at 4 °C was similar in HSL?/? mice and controls. Overnight fasting was well‐tolerated clinically by HSL?/? mice, but after fasting, liver triglyceride content was significantly lower in HSL?/? mice compared with wild‐type controls. Conclusions: In isolated fat cells, the lipolytic rate after β‐adrenergic stimulation is mainly dependent on HSL. However, the observation of a normal rate of lipolysis in unstimulated HSL?/? adipocytes suggests that HSL‐independent lipolytic pathway(s) exist in fat. Physiologically, HSL deficiency in mice has a modest effect under normal fed conditions and is compatible with normal maintenance of core body temperature during cold stress. Interspecific Variation in RGR and the Underlying Traits among 24 Grass Species Grown in Full Daylight 总被引:4,自引:0,他引:4
Abstract: A growth analysis was conducted with 24 central European grass species in full daylight to test whether traits underlying interspecific variation in relative growth rate (RGR) are the same in full daylight as they are at lower light, and whether this depends on the ecological characteristics of the studied species, i.e., their requirements with respect to nutrient and light availability.
In contrast to studies with herbaceous species at lower light, net assimilation rate (NAR) contributed more than leaf area ratio (LAR) or specific leaf area (SLA) to interspecific variation in RGR. This was associated with a larger interspecific variation in NAR than found in experiments with lower light. Without the two most shade-tolerant species, however, the contribution of LAR and its components to interspecific variation in RGR was similar or even higher than that of NAR.
Leaf dry matter content correlated negatively with RGR and was the only component of LAR contributing in a similar manner to variation in LAR and RGR. There was a positive correlation between NAR and biomass allocation to roots, which may be a result of nutrient-limited growth. RGR correlated negatively with biomass allocation to leaves. Leaf thickness did not correlate with RGR, as the positive effect of thin leaves was counterbalanced by their lower NAR.
Low inherent RGR was associated with species from nutrient-poor or shady habitats. Different components constrained growth for these two groups of species, those from nutrient-poor habitats having high leaf dry matter content, while those from shady habitats had thin leaves with low NAR. 相似文献

目的 探讨中华眼镜蛇伤致局部组织损伤的最佳治疗方法。方法 用中华眼镜蛇毒作家兔局部组织损伤模型,分别采用抗蛇素血清局部注射、糜蛋白酶局部注射、蛇伤药酒外敷、坏死组织早期切除、局部烧灼法、局部组织切开冲洗,共6种处理方法进行局部治疗,观察其疗效。结果 6种治疗方法从优到劣依次是:抗蛇毒血清局部注射、糜蛋白酶局部注射、蛇伤药酒外敷、坏死组织早期切除、局部烧灼法、局部组织切开冲洗。结论 中华眼镜蛇伤致局部组织损伤的治疗方法应首选抗蛇毒血清局部注射和糜蛋白酶局部注射,其次选用蛇伤药酒外敷。 相似文献
