Arctic shrubs have a strong potential for climate and environmental reconstructions in the chronically understudied regions of the high northern latitudes. The climate dynamics of these regions are important to understand because of large-scale feedbacks to the global climate system. However, little is known about other factors influencing shrub ring growth, possibly obscuring their climate signal. For example, as of yet we are not able to differentiate between herbivory or climatically induced growth depressions. Here, we use one of the most common Arctic shrubs, Alnus viridis as a test case to address this question. We sampled Alnus in Kobbefjord, Greenland, measured shrub-ring width and cell wall thickness and built site chronologies of each parameter. We analysed climate-growth relationships, tested their stability over time and employed a pointer-year analysis to detect growth depressions. We employed bootstrapped transfer function stability tests (BTFS) to assess the suitability of our shrub chronologies for climate reconstruction. Correlations with climate data showed strong significantly positive and stable correlations between summer temperature and ring-width with the exception of the recent decade. A climate reconstruction model failed stability tests, when the complete period of record was used for calibration and verification. Wood anatomy analysis uncovered the occurrence of unusual cell structure (very thin cell walls) in the exceptionally narrow ring of 2004, a recorded insect outbreak year in other parts of Greenland. When excluding the affected ring and a recovery period, the reconstruction model passed all tests, suggesting that the unusual 2004 ring was not climate driven, but rather the result of an insect attack. When combining anatomical analysis with traditional ring-width measurements, we move a step further in potentially distinguishing small rings caused by insect attacks from small rings formed in climatically challenging years. While this study does not provide unambiguous evidence, it does provide potential useful methodological combinations to enable more robust climate reconstructions in areas where climatic records are extremely sparse.  相似文献

To determine relative host preference rates and to establish whether a pair of animals in close proximity (between–host interaction) modified Culicoides attack and abundance profiles, compared to those tethered in isolation (host independence), Culicoides midges were pooted hourly from two sets of experimental animals: (1) a heifer cow and Fjord horse tethered close together (5 m apart), and (2) a heifer and Fjord tethered in isolation (45 m apart). Over 12 days, 570 3‐min observations yielded 23 090 midges, representing 24 species. Approximately 95% belonged to the Culicoides obsoletus (Meigen) complex (two species), Culicoides dewulfi Goetghebuer, Culicoides chiopterus (Meigen), Culicoides punctatus (Meigen), Culicoides pulicaris (L.), and Culicoides achrayi Kettle & Lawson (Diptera: Ceratopogonidae) combined. There was no evidence for between‐host interaction. Mean Culicoides species‐specific attack rates did not differ between animal species, except that C. chiopterus was 7× more abundant on the legs of the heifer compared to the horse, and C. dewulfi twice as abundant on the upper half of the horse compared to the heifer. By contrast, mean species–specific biting rates of the C. obsoletus complex, C. chiopterus, C. dewulfi, C. punctatus, and C. pulicaris midges were 5×, 100×, 1.7×, 2×, and 2.5× lower in the horses compared to the heifers, respectively. It is not clear why high Culicoides attack rates observed in the horses do not convert into high biting rates as seen in cattle; this should be a subject for future research. In light of its apparent predilection for equids, the ability of C. dewulfi to replicate African horse sickness virus (AHSV) extrinsically should be investigated in the laboratory.  相似文献

Cerebral ischemia/reperfusion is known to induce the generation of reactive oxygen species and inflammatory responses. Numerous studies have demonstrated that naringin (NGIN) has anti-oxidant and anti-inflammatory properties. We previously reported that Citrus kawachiensis contains a large quantity of NGIN in its peel. In the present study, we orally (p.o.) administered dried peel powder of C. kawachiensis to mice of a transient global ischemia model and found in the hippocampus region that it 1) suppressed neuronal cell death, 2) reversed the reduction in the level of phosphorylated calcium-calmodulin-dependent protein kinase II, 3) had the tendency to reverse the reduction in the level of glutathione, and 4) blocked excessive activation of microglia and astrocytes. These results suggested that the dried peel powder of C. kawachiensis had a neuroprotective effect against ischemic brain via anti-oxidative and anti-inflammatory effects. We also showed that these effects of the dried peel powder were more powerful than those obtained with a comparable amount of NGIN alone.  相似文献

Aggression between male conspecifics is a complex social behavior that is likely modulated by multiple gene variants. In this study, the BXD recombinant inbred mouse strains (RIS) were used to map quantitative trait loci (QTLs) underlying behaviors associated with intermale aggression. Four hundred and fifty‐seven males from 55 strains (including the parentals) were observed at an age of 13 ± 1 week in a resident‐intruder test following 10 days of isolation. Attack latency was measured directly within a 10‐minute time period and the test was repeated 24 hours later. The variables we analyzed were the proportion of attacking males in a given strain as well as the attack latency (on days 1 and 2, and both days combined). On day 1, 29% of males attacked, and this increased to 37% on day 2. Large strain differences were obtained for all measures of aggression, indicating substantial heritability (intraclass correlations 0.10‐0.18). We identified a significant QTL on chromosome (Chr) 1 and suggestive QTLs on mouse Chrs 1 and 12 for both attack and latency variables. The significant Chr 1 locus maps to a gene‐sparse region between 82 and 88.5 Mb with the C57BL/6J allele increasing aggression and explaining about 18% of the variance. The most likely candidate gene modulating this trait is Htr2b which encodes the serotonin 2B receptor and has been implicated in aggressive and impulsive behavior in mice, humans and other species.  相似文献

Predation risk has a profound influence on the behavior of marine mammals, affecting grouping patterns and habitat use. Dolphins frequently bear evidence of shark bites, which can provide an indirect measure of predation pressure. Using photo‐identification data, we investigated the prevalence of shark bites on three sympatric species of inshore dolphin, the Australian snubfin (Orcaella heinsohni), Australian humpback (Sousa sahulensis), and Indo‐Pacific bottlenose dolphin (Tursiops aduncus), among four study sites in northwestern Australia. Bite prevalence varied markedly between species, with 72% of snubfin, 46% of humpback, and 18% of bottlenose dolphins exhibiting evidence of shark bites. Binomial logistic regression confirmed a high likelihood of bite presence on snubfin dolphins, and at one particular site for snubfin and bottlenose dolphins. The prevalence of tiger shark (Galeocerdo cuvier) bites on snubfin dolphins was high, and bites attributed to other carcharhinid sharks were observed on all species. While acknowledging methodological differences with other studies, the prevalence of shark bites on snubfin dolphins is among the highest reported for any dolphins, suggesting predation risk represents an important but varying influence thereon. This study provides a baseline for future investigations into the affect of predation risk on the behavioral ecology of these sympatric species.  相似文献

The effects of light level, developmental pathway, and previous growth history on the foraging attack distances of juvenile Atlantic salmon Salmo salar were examined in circular rearing tanks. Former manipulation of growth rates had no significant influence on distances moved to intercept food items despite the fact that it caused substantial differences in post-treatment growth. Attack distances of fish that were entering a state of overwinter dormancy (lower modal group; LMG) were shorter than those of actively feeding (and growing) fish (upper modal group; UMG). These differences were explained generally by differences in body size between the two groups, suggesting that actual effort per attack was unassociated with growth requirements. Significant differences between growing and non-growing fish in attack distances could contribute to the variation in growth rates through their effect on feeding rates, but were unlikely to have affected energetic costs. This may be due to the fact that attack distances were consistently short throughout the study period as is also evident from the pattern of change between night and daytime. Whereas in the first experiment (daylight v . twilight) fish moved further to reach food during the day, in the second (daylight v . overcast night) nocturnal attack distances matched (LMG fish) or exceeded (UMG fish) diurnal attack distances. Thus diurnal attack distances were probably minimized in the second experiment. These results are interpreted within a framework of overwintering strategies.  相似文献

Background

Flap necrosis due to insufficient blood supply is a common postoperative complication in random pattern flaps. Stem cell therapies have emerged as promising biologics for tissue ischemia. A novel fat derived product, stromal vascular fraction gel (SVF-gel), can be prepared with lipoaspirate through simple mechanical processing, removing only the lipid content. SVF-gel enriches adipose-derived stem cells and potentially beneficial for flap necrosis.

Methods

Nude mice ischemic flaps were treated with human SVF-gel, stromal vascular fraction (SVF) cell suspension or saline (n = 10). They were injected to the flap recipient beds, and necrosis and vascularization was assessed on postoperative day 14. We harvested the necrosis-free distal to evaluated skin healthiness and neovasculogenesis by Masson's trichrome stain and immunofluorescence, etc. Pro-angiogenic factors were assessed with tissue qRT-PCR. Finally, we traced the grafted human tissue with immunofluorescence.

Results

SVF-gel-treated flaps have the smallest necrotic zones (22.05% ± 0.0438) compared with the saline controls (53.78% ± 0.1412) or SVF-treated ones (35.54% ± 0.0850, p = 0.039). Numerous functional musculocutaneous perforators were developed around SVF-gel grafts. The SVF-gel-treated skin had the best fat restoration (231.3 ± 48.1 μm) among three groups (F = 10.83, p = 0.0102) while saline-treated flap distal appeared fibrotic. SVF-gel-treated flaps also had ~43% more CD31 + capillaries (p = 0.0152) with ~3 folds more gene expression of angiogenic cytokines of VEGF and bFGF (p = 0.0310 and 0.0303, respectively) than saline-treated controls. Furthermore, we found hSVF-gel cells (hGolgi+) had directly engrafted as vessel component (α-smooth muscle actin, α-SMA+) to the flap.

Conclusion

Adipose cellular matrix enhanced flap neovascularization partly by direct incorporation, improved flap survival and fat restoration. The composition-selective fat grafting with SVF-gel demonstrated efficacy comparable with stem cell therapy and is especially valuable for clinical translation.  相似文献

Trees alter their use and allocation of nutrients in response to drought, and changes in soil nutrient cycling and trace gas flux (N2O and CH4) are observed when experimental drought is imposed on forests. In extreme droughts, trees are increasingly susceptible to attack by pests and pathogens, which can lead to major changes in nutrient flux to the soil. Extreme droughts often lead to more common and more intense forest fires, causing dramatic changes in the nutrient storage and loss from forest ecosystems. Changes in the future manifestation of drought will affect carbon uptake and storage in forests, leading to feedbacks to the Earth's climate system. We must improve the recognition of drought in nature, our ability to manage our forests in the face of drought, and the parameterization of drought in earth system models for improved predictions of carbon uptake and storage in the world's forests.  相似文献

Thioredoxin-1 maintains the cellular redox status and decreases the infarct size in ischemia/reperfusion injury. However, whether the increase of thioredoxin-1 expression or its lack of activity modifies the protection conferred by ischemic postconditioning has not been yet elucidated. The aim was to evaluate if the thioredoxin-1 overexpression enhances the posctconditioning protective effect, and whether the lack of the activity abolishes the reduction of the infarct size. Wild type mice hearts, transgenic mice hearts overexpressing thioredoxin-1, and a dominant negative mutant (C32S/C35S) of thioredoxin-1 were used. The hearts were subjected to 30 min of ischemia and 120 min of reperfusion (Langendorff) (I/R group) or to postconditioning protocol (PostC group). The infarct size in the Wt-PostC group decreased in comparison to the Wt-I/R group (54.6 ± 2.4 vs. 39.2 ± 2.1%, p < 0.05), but this protection was abolished in DN-Trx1-PostC group (49.7 ± 1.1%). The ischemia/reperfusion and postconditioning in mice overexpressing thioredoxin-1 reduced infarct size at the same magnitude (35.9 ± 2.1 and 38.4 ± 1.3%, p < 0.05 vs. Wt-I/R). In Wt-PostC, Trx1-I/R and Trx1- PostC, Akt and GSK3β phosphorylation increased compared to Wt-I/R, without changes in DN-Trx1 groups. In conclusion, given that the cardioprotection conferred by thioredoxin-1 overexpression and postconditioning, is accomplished through the activation of the Akt/GSK3β survival pathway, no synergic effect was evidenced. Thioredoxin-1 plays a key role in the postconditioning, given that when this protein is inactive the cardioprotective mechanism was abolished. Thus, diverse comorbidities or situations modifying the thioredoxin activity, could explain the absence of this strong mechanism of protection in different clinical situations.  相似文献

 The amino acid sequences of the human terminal complement components show extensive structural similarity to each other. In this study the C8β and C9 cDNAs of Japanese flounder, Paralichthys olivaceus, were cloned and analyzed. The derived deduced amino acid sequences of the two terminal components were homologous to those of humans, in that the sequences of both species contained LDL receptor, EGF precursor, and two thrombospondin domains. Japanese flounder C9 was found to have a second thrombospondin region in the C-terminus, similar to that reported for rainbow trout and pufferfish. Moreover, these two complement component cDNAs of Japanese flounder had partial similarity to human perforin. These findings show that Japanese flounder C8β and C9 have similar structures, which supports the hypothesis that the terminal complement genes originated from the same ancestral gene.
