Wildflower strips are a management practice increasingly used to provide floral resources to wild bees in agroecosystems. Yet, despite known spatiotemporal variation in wild bee communities, the degree to which different wildflower strip species consistently support wild bee communities is poorly understood. Additionally, whether such consistency is related to the functional roles wildflower species play (e.g., in supporting diverse, rare, or unique suites of bee species) has not been considered. Over three years and on four diversified farms, we evaluated spatiotemporal variation in wild bee communities and bee-flower interactions in wildflower strips to better understand the roles of flower strip species in supporting bees. We documented spatiotemporal variation in the abundance, richness, and composition of local wild bee communities. Certain wildflower species consistently supported the highest richness of wild bees across years. These wildflower species were regularly core members of the bee-flower interaction network, visited by both generalist and specialist bees. By contrast, wildflower species supporting the most unique suites of bees were variable in this role among farms. In order to select plant species for wildflower strips that consistently support a high diversity of wild bee communities within farm landscapes, it is useful to consider several different functional roles that plants may play. Whereas a handful of wildflower species may be visited by the majority of local wild bee species, achieving support for the remaining, and perhaps rarer, bee species will require planting additional flower species, which may appear redundant until the spatiotemporal variation in wild bee communities is more thoroughly considered. This functional approach to selecting wildflower species for bee conservation efforts is important for making practical recommendations to land managers and for guiding best management practices in different regions and with diverse management goals.  相似文献

The use of synthetic antioxidants has been associated with serious concerns for human and environmental health. During ripening stages, tomato fruit is exposed to different abiotic stresses which not only influence its nutritional, mechanical, and functional properties at harvest, but also affect the quality and shelf life of the fruit during storage. This study investigated the pattern of changes in dietary antioxidants during various ripening stages of tomato fruit (cv. Red Rose) and their impact on storage behavior of the fruit during cold storage. Tomato fruits were harvested at mature green, breaker, turning, pink, light-red and red stages of maturity. Then, they were analysed for flesh firmness, soluble solids content, titratable acidity, total sugars, pH, dry matter content, lipophilic (lycopene, β-carotene, and total carotenoids), and hydrophilic (ascorbic acid, phenolic and flavonoids) antioxidants. Additional fruits were harvested at each maturity stage and divided into three equal lots, then were subjected to low-temperature (10 ± 1 °C) storage with 80 ± 5% RH, for 7, 14, and 21 days. Flesh firmness, and the levels of dietary antioxidants were analysed following the subsequent storage periods. The results revealed that the peak of hydrophilic antioxidants such as ascorbic acid, phenolic compounds, and flavonoids was between the ‘pink’ and the ‘light-red’ stages of fruit maturity. Whereas tomatoes harvested at the ‘red’ stage of maturity had the highest levels of lycopene and β-carotene. Both the stage of fruit maturity at harvest and duration of cold storage influenced flesh firmness, organoleptic and functional properties of ‘Red Rose’ tomato fruit. In conclusion, the results of the current investigation have practical implications in formulating foods with improved functional properties at processing industries.  相似文献

The original thyroid scan (TS) was widely used to identify typical imaging patterns, suggesting the widely accepted main following clinical diagnoses: Grave's disease, Toxic adenoma, [hetero]-nodular goiters and thyroiditis. With the diffusion of sensitive TSH assays, considerable advances in the comprehension of the molecular mechanisms of hormonosynthesis, and new quantification possibilities especially using 123I, the TS is a textbook of molecular imaging. The image can be finely quantified with, not only as regards the Uptake (123IUp) and related parameters but also, the quantification of the spatial targeting leading to a Spatial Target Index (STI). Using this new molecular 123I-TS, TSH values, and when required, correlation to Multiparametric Ultrasounds (MPUS), we generated a basic classification system of hyperthyroidism, with well-defined indexed criteria (C11-1 to C17-3), that allows reporting 24 distinct etiologies. Selected criteria involve TS and contrast patterns, precocious 123IUp (p123IUp), maximal TSH-dependent physiological Uptake, lobar concentration, Uptake and concentration ratios, STI, 99mTc-MIBI TS and correlative MPUS. This approach allows to identify 4 subtypes of Graves’ disease, including hyperplastic, nodular and common GD variants entangled with Hashimoto's struma, 4 subtypes of Thyroid Functional Autonomy, including Disseminated Functional Autonomy, that cannot be diagnosed with other conventional procedures. Criteria C14-1 to C17-3 report on hyperthyroidism and iodine overload, factitia, main thyroiditis presentations and rare central or tumoral etiologies of hyperthyroidism. This classification, based on 123I-TS molecular imaging, leads to unprecedented diagnostic finesse and paves the way for a personalized theranostic approach in thyroid pathology. Further development towards artificial intelligence networks is under study.  相似文献

Sphagnum peatlands host a high abundance of protists, especially testate amoebae. Here, we designed a study to investigate the functional diversity of testate amoebae in relation to wetness and forest cover in Baltic bogs. We provided new data on the influence of openness/wetness gradient on testate amoebae communities, showing significant differences in selected testate amoebae (TA) traits. Three key messages emerged from our investigations: 1) we recorded an effect of peatland surface openness on testate amoebae functional traits that led us to accept the hypothesis that TA traits differ according to light intensity and hydrology. Mixotrophic species were recorded in high relative abundance in open plots, whereas they were nearly absent in forested sites; 2) we revealed a hydrological threshold for the occurrence of mixotrophic testate amoebae that might be very important in terms of peatland functioning and carbon sink vs. source context; and 3) mixotrophic species with organic tests were nearly absent in forested sites that were dominated by heterotrophic species with agglutinated or idiosomic tests. An important message from this study is that taxonomy of TA rather indicates the hydrological gradient whereas traits of mixotrophs the openness gradient.  相似文献

N‐Methyl‐D‐aspartate (NMDA) receptors are key components in synaptic communication and are highly relevant in central nervous disorders, where they trigger excessive calcium entry into the neuronal cells causing harmful overproduction of nitric oxide by the neuronal nitric oxide synthase (nNOS) protein. Remarkably, NMDA receptor activation is aided by a second protein, postsynaptic density of 95 kDa (PSD95), forming the ternary protein complex NMDA/PSD95/nNOS. To minimize the potential side effects derived from blocking this ternary complex or either of its protein components, a promising approach points to the disruption of the PSD‐95/nNOS interaction which is mediated by a PDZ/PDZ domain complex. Since the rational development of molecules targeting such protein‐protein interaction relies on energetic and structural information herein, we include a thermodynamic and structural analysis of the PSD95‐PDZ2/nNOS‐PDZ. Two energetically relevant events are structurally linked to a “two‐faced” or two areas of recognition between both domains. First, the assembly of a four‐stranded antiparallel β‐sheet between the β hairpins of nNOS and of PSD95‐PDZ2, mainly enthalpic in nature, contributes 80% to the affinity. Second, binding is entropically reinforced by the hydrophobic interaction between side chains of the same nNOS β‐hairpin with the side chains of α2‐helix at the binding site of PSD95‐PDZ2, contributing the remaining 20% of the total affinity. These results suggest strategies for the future rational design of molecules able to disrupt this complex and constitute the first exhaustive thermodynamic analysis of a PDZ/PDZ interaction.  相似文献

10–23 DNAzyme is an artificially selected catalytic DNA molecule. Its great potential as genetic therapeutics promoted chemical modifications for more efficient DNAzymes. Here, 10–23 DNAzyme was modified on its six deoxyadenosine residues (A5, A9, A11, A12, A15 in the catalytic domain and A0 of the recognition arm next to the cleavage site) with compound 1, an adenosine analogue with 2′-O-[N-(aminoethyl)carbamoyl]methyl group. A positive effect of compound 1 at A15 was observed (HJDS-05, kobs = 0.0111 min−1). Compared to the effect of 2′-H and 2′-OMe at A15, this result provided an approach for more efficient DNAzyme by combining 2′-substituted amino group of adenosine with A15 as the lead structure.  相似文献

目的观察双歧杆菌三联活菌(金双歧)联合乳果糖口服液治疗儿童功能性便秘的临床疗效。探索临床治疗儿童功能性便秘的良好方法。方法选择我院2015年1月至2017年12月收治的110例功能性便秘患儿为研究对象,随机分为治疗组(58例)和对照组(52例)。两组患儿均予以基础治疗,对照组患儿给予乳果糖口服液治疗,治疗组患儿给予金双歧联合乳果糖口服液治疗,比较两组患儿疗效及不良反应。结果治疗组和对照组患儿治疗总有效率分别为91.4%和76.9%,差异有统计学意义(χ~2=4.385 0,P=0.036 2)。治疗组患儿不良反应发生率为3.4%,对照组为11.5%,差异无统计学意义(χ~2=5.153 0,P=0.272 0)。随访1年后,治疗组患儿远期复发率低于对照组。结论双歧杆菌三联活菌联合乳果糖口服液对儿童功能性便的疗效优于单用乳果糖口服液,患儿远期复发少,值得临床推广。  相似文献

目的探讨儿童功能性便秘(FC)患者肠道菌群及血清脑肠肽水平的变化。方法选取2017年1月至2019年4月在我院儿科门诊治疗的80例FC患儿为研究组,选择同期于体检中心检查的无胃肠道疾病的30例健康儿童为健康组。研究组患儿根据病情轻重分为轻中度组(52例)和重度组(28例)。比较各组儿童肠道菌群数量[乳杆菌、双歧杆菌和大肠埃希菌]及血清脑肠肽指标[一氧化氮(NO)、P物质(SP)和血管活性肠肽(VIP)]水平的差异。结果研究组患儿肠道双歧杆菌[(8.16±0.89)lg CFU/g]和乳杆菌数量[(7.92±0.83)lg CFU/g]明显少于健康组[(8.94±0.97)lg CFU/g、(8.63±0.91)lg CFU/g],而大肠埃希菌数量[(8.12±0.81)lg CFU/g]明显多于健康组[(7.30±0.72)lg CFU/g],差异有统计学意义(t=2.37、2.35、2.44,P0.05)。重度组患儿肠道双歧杆菌和乳杆菌数量明显少于轻中度组,而大肠埃希菌数量明显多于轻中度组(t=2.36、2.34、2.29,P0.05)。研究组患儿血清NO和VIP水平[(50.12±8.25)μmol/L、(29.17±5.12)pg/mL]明显高于健康组[(34.46±6.13)μmol/L、(20.43±4.79)pg/mL],SP水平[(5.21±0.82)ng/mL]明显低于健康组[(7.57±1.04)ng/mL],差异有统计学意义(t=2.78、2.54、2.47,P0.05)。重度组患儿血清NO和VIP水平高于轻中度组,SP水平明显低于轻中度组(t=2.31、2.41、2.23,P0.05)。结论 FC患儿存在肠道菌群失调和脑肠肽分泌异常,且随着病情的加重其肠道菌群失调及脑肠肽分泌异常越发明显。  相似文献

ABSTRACT

Hesperidin, a citrus flavonoid, can exert numerous beneficial effects on human health. Interstitial cells of Cajal (ICC) are pacemaker cells in the gastrointestinal (GI) tract. In the present study, we investigated potential effects of hesperidin on pacemaker potential of ICC in murine small intestine and GI motility. A whole-cell patch-clamp configuration was used to record pacemaker potential in ICC, and GI motility was investigated in vivo by recording gastric emptying (GE) and intestinal transit rate (ITR). Hesperidin depolarized pacemaker potentials of ICC in a dose-dependent manner. Pre-treatment with methoctramine or 4-DAMP did not inhibit hesperidin-induced pacemaker potential depolarization. Neither a 5-HT3 receptor antagonist (Y25130) nor a 5-HT7 receptor antagonist (SB269970) reduced the effect of hesperidin on ICC pacemaker potential, whereas the 5-HT4 receptor antagonist RS39604 was found to inhibit this effect. In the presence of GDP–β–S, hesperidin-induced pacemaker potential depolarization was inhibited. Moreover, in the presence of U73122 and calphostin C, hesperidin did not depolarize pacemaker potentials. Furthermore, hesperidin accelerated GE and ITR in vivo. These results imply that hesperidin depolarized ICC pacemaker potential via 5-HT4 receptors, G protein, and PLC/PKC dependent pathways and that it increased GI motility. Therefore, hesperidin may be a promising novel drug to regulate GI motility.  相似文献
