Hydrogen production by dark fermentation may suffer of inhibition or instability due to pH deviations from optimality. The co-fermentation of promptly degradable feedstock with alkali-rich materials, such as livestock wastes, may represent a feasible and easy to implement approach to avoid external adjustments of pH.Experiments were designed to investigate the effect of the mixing ratio of fruit-vegetable waste with swine manure with the aim of maximizing biohydrogen production while obtaining process stability through the endogenous alkalinity of manure.Fruit-vegetable/swine manure ratio of 35/65 and HRT of 2 d resulted to give the highest production rate of 3.27 ± 0.51 LH2 L−1 d−1, with a corresponding hydrogen yield of 126 ± 22 mLH2 g−1VS-added and H2 content in the biogas of 42 ± 5%. At these operating conditions the process exhibited also one of the highest measured stability, with daily productions deviating for less than 14% from the average. 相似文献

The isolation and analysis of glycoproteins by coupling lectin affinity chromatography with MS has emerged as a powerful strategy to study the glycoproteome of mammalian cells. However, this approach has not been used extensively for the analysis of plant glycoproteins. As with all eukaryotes, N-glycosylation is a common post-translational modification for plant proteins traveling through the secretory pathway. Many such proteins are destined for the cell wall, or apoplast, where they play important roles in processes such as modifying cell wall structure, sugar metabolism, signaling, and defense against pathogens. Here, we describe a strategy to enrich for and identify secreted plant proteins based on affinity chromatography using the lectin Concanavalin A and two-dimensional liquid chromatography, together with matrix-assisted laser desorption/ionization MS analysis. The value of this approach is illustrated through the characterization of glycoproteins that are expressed in ripe tomato (Solanum lycopersicum) fruit, a developmental stage that is fundamentally linked with significant changes in cell wall structure and composition. This glycoprotein trap strategy allowed the isolation of a sub-proteome with an extremely high proportion of proteins that are predicted to be resident in the cell wall or secretory pathway, and the identification of new putative cell wall proteins. 相似文献

Adverse effects of gibberellin applications on pear trees after frost such as small fruit size, abnormal fruit shape and poor return bloom are often attributed both to the sole use of GA3 and its overdose. It is unclear whether protection against spring frosts before flower opening is more efficient when GA is applied directly after frost, i.e. before flower opening, or at full bloom or both. In April 2003, early spring frosts at Klein-Altendorf near Bonn, Germany damaged ca. 88% flowers of the early flowering cv. Alexander Lucas, 64% of cv. Conference and ca. 25% of flowers of the later flowering cv. Comice pears. Hence, the objective of the present work was to investigate the optimum timing of the application of low doses of the combined GA3 and GA4 + 7 to improve parthenocarpic fruit set in pears, while maximising fruit quality and size for trees affected by a severe spring frost before full bloom. Return bloom was also considered and quantified. Frost-affected pear trees were treated with gibberellin GA3+GA4 + 7, either immediately after the frost, at the white bud stage, or at full bloom or both to improve parthenocarpic set. Early flowering cv. Alexander Lucas pear was most affected by the early spring frost, but lost only 25% of fruitlets at June drop, irrespective of GA treatment. June drop was, however, severe in the two other cultivars least affected by frost, i.e. by 33% in cv. Conference and 55% in cv. Comice. Both initial and final fruit set were significantly increased by a combined application of GA3+GA4 + 7 at full bloom, without affecting return bloom, but June drop was also enhanced by GA application. The largest positive effect of GA application on fruit yield, an additional 2 kg of fruit per tree equivalent to €1200/ha, was apparent with the cv. Alexander Lucas, i.e. the cultivar most affected by frost. There was no loss in fruit quality viz fruit size after any of the GA applications with any of the pear cultivars examined and no increase in abnormally-shaped, elongated fruit. 相似文献

The localized application of the synthetic cytokinin CPPU ((2-chloro-4-pyridyl)-N-phenyl urea) to ovaries at flower opening was as effective as free pollination in setting parthenocarpic fruit in the triploid watermelon cultivar ‘Reina de Corazones’, and increased yield per unit land area by at least 50%, simply due to the lack of requirement for diploid pollen producing plants within the orchard. The application of the synthetic auxin 2,4-D (2,4-dichlorophenoxy acetic acid) as a full coverage spray, was also effective in setting fruit; total yield was however 10% smaller than in the CPPU-treated plots, but the cost of application was much less expensive. These applications had no adverse effect on fruit quality, and their effectiveness in commercial watermelon production was evaluated over 4 years. Plant hormones play an important regulatory role in growth and development of fleshy fruits and often these are synthesized in a sequence that is tissue- and/or fruit-specific. Previous studies using Hass avocado and its small-fruit phenotype as a model demonstrated a relationship between the cytokinin/abscisic acid ration and final fruit size. More detailed studies revealed that differential activity of molybdenum-cofactor-containing enzymes was in part responsible for the observed increase in abscisic acid in small Hass fruit and that the same suite of enzymes likely impacted levels of indole-3-acetic acid. The present study confirms that the inability of the seed to produce/accumulate indole-3-acetic acid is a major contributing factor and characteristic of fruit that display a small-fruit phenotype, that reduced indole-3-acetic acid levels occur concomitant with increased abscisic acid, and that cytokinin metabolism appears unchanged irrespective of fruit size. Results are discussed in relation to the role of molybdenum-cofactor-requiring enzymes in indole-3-acetic acid and abscisic acid metabolism. 相似文献

We describe here a novel plant-specific gene, Lefsm1 (fruit SANT/MYB-like 1) harboring a single SANT/MYB domain. The expression of Lefsm1 is specific to the very early stages of tomato (Lycopersicon esculentum) fruit development. Ectopic expression of Lefsm1 results in severe developmental alterations manifested in retarded growth, and reduced apical dominance during tomato and Arabidopsis seedling development. A promoter sequence residing 1.0 kb upstream to the translation initiation codon confers the organ-specific expression of the gene. Lefsm1 belongs to a novel small gene family consisting of five to six members in tomato, Arabidopsis and rice. The SANT/MYB domain of LeFSM1 and its orthologs in Arabidopsis and rice differs from that of all other plant or animal MYB proteins and from the SANT domains found in part of the chromatin remodeling proteins. Together, our results indicate that Lefsm1 is a founding member of a small family of proteins containing a novel MYB/SANT domain which is likely to participate in the regulation of a plant-specific developmental program. 相似文献

Microarray analysis of Vitis vinifera cv. Shiraz developing berries has revealed the expression patterns of several categories of genes. Microarray slides were constructed from 4,608 PCR-amplified cDNA clones derived from a ripening grape berry cDNA library. The mRNA expression levels of the genes represented by these cDNAs were measured in flowers, week 2 post-flowering whole berries, week 5, week 8, week 10 (véraison, green berries), week 12 and week 13 berry skin. In addition, a comparison of RNA expression in pigmented and unpigmented berry skin at véraison (week 10) was undertaken. Image and statistical analysis revealed four sets of genes with distinctive and similar expression profiles over the course of berry development. The first set was composed of genes which had maximum RNA expression in flowers, followed by a steady decrease in expression. The most prominent group within this set were genes which have a role in photosynthesis. The second set of cDNAs was dominated by genes involved in flavonoid biosynthesis and had a peak of expression week 2 post-flowering. The data indicate co-ordinate regulation of flavonoid biosynthetic genes which code for the enzymes 4-coumarate-CoA ligase, chalcone synthase, chalcone isomerase, flavonone hydroxylase, anthocyanidin reductase and cytochrome b5. The third set of cDNAs exhibited maximum expression week 5 post-flowering, midway between flowering and véraison, a period of rapid berry growth. This set of cDNAs is dominated by genes which code for structural cell wall proteins. The fourth set of genes was dramatically up-regulated at véraison and remained up-regulated until 13 weeks post-flowering. Peach fruit are handled, processed, and marketed according to their stone adhesion and fruit softening type. Uncertainty exists over whether these simply inherited traits are controlled by two linked loci, Freestone (F) and Melting flesh (M) or one multi-allelic locus, and whether M is controlled by the cell wall degrading enzyme, endopolygalacturonase. From morphological and molecular analysis of two related segregating populations of peach, we conclude that a single locus containing at least one gene for endopolygalacturonase, controls both F and M with at least three effective alleles. A simple diagnostic PCR test is now available for the three major phenotypes of freestone melting flesh (FMF), clingstone melting flesh (CMF), and clingstone non-melting flesh (CNMF). 相似文献

对广州地区引进的19个龙眼(Dimocarps longan)品种的果实品质进行了评价与分类研究。结果表明:在表观性状方面,除品种间肉质、果形差异较大外,其它性状差异不大,部分性状之间具有一定的关联性;小果或薄皮的品种一般可溶性固形物含量较高,薄皮、厚肉或横扁的品种一般可食率较高。运用分层聚类的方法,依据各品质指标将品种划分为3类; 运用基于主成分分析的因子分析法,将8个品质指标用3个主成分来表示(累计贡献率达88.27%);确定简化后的品质评价指标为单果质量、果形指数、可食率和肉厚率,并依据因子得分和简化指标对19个品种进行聚类,两种聚类结果基本一致。 相似文献
