福建金线莲的化学成分研究Ⅲ

从福建金线莲(Anoectochilus roxburghii)乙醇提取物的己烷萃取部分分得3个化合物和-甾体混合物,三个化合物结构分别鉴定为Sorghumol(1),木栓酮(2)和棕榈酸(3),甾体混合物经EI-MS、ESI-MS／MS技术确定含有24-异丙烯基胆甾醇、开唇兰甾醇、β-谷甾醇、豆甾醇和菜油甾醇。所有化合物均为该植物首次报道,其中化合物1为首次从兰科植物中分得,并首次对其光谱数据进行了详细的分析,化合物2为从该属植物中首次分得。     

Bioconversion of Hydrocortisone by Cyanobacterium Fischerella ambigua PTCC 1635

Summary The bioconversion of hydrocortisone by a locally isolated strain of cyanobacterium Fischerella ambigua PTCC 1635 was investigated. Fischerella ambigua had not been previously examined for this potential. The fermentation led to production of 11β,17α, 20β, 21-tetrahydroxypregn-4-en-3-one and 11β-hydroxyandrost-4-en-3,17-dione. The metabolites were isolated and purified by chromatographic methods and identified using instrumental analyses.     

饲料中n-3 HUFA含量对花尾胡椒鲷亲鱼的生殖性能及血浆性类固醇激素水平季节变化的影响

以n-3 HUFA含量为0．16％、1．27％、2．36％和3．47％的4种人工配合饲料(D1～D4)及天然饲料(冰鲜杂鱼,D5)饲养花尾胡椒鲷(Plectorhynchus cinctus)雌亲鱼一周年,通过比较各饲料组亲鱼的产卵量、卵和仔鱼质量以及各月份的血浆性类固醇激素水平,研究n-3 HUFA对生殖性能及性类固醇激素水平季节性变化的影响。平均每kg雌鱼的产卵量、卵受精率、仔鱼存活率、开口仔鱼体长等,D2和D3组与D5组相近,但D1和D4组显著低于D5组;饲料中n-3 HUFA含量对血浆17β-雌二醇(E2)和睾酮(T)水平的季节性变化规律无明显影响,但亲鱼性腺发育与成熟时期的E2和T水平．D1和D4组较D5组显著降低。n-3 HUFA含量对亲鱼离体卵泡E2和T的分泌也有一定影响：D4组卵泡E2的基础分泌很少;绒毛膜促性腺激素(HCG,100IU／mL)可刺激D2-D5组卵泡分泌E2和T,但D1组卵泡对HCG无反应。结果提示,花尾胡椒鲷亲鱼饲料中n-3 HUFA的适宜含量为1．27％-2．36％,不足或过高对亲鱼的生殖性能均有不利影响;通过影响性类固醇激素的产生可能是饲料中n-3 HUFA含量影响鱼类生殖性能的机制之一。     

茶条木化学成分的研究

茶条木（Delavaya yunnanensis Franch)是无患子科独属独种植物,也是亚洲特有植物,是一种很好的油料植物,有润发、治疗疥癣等作用。本文采用多 方法首次从茶条木乙醇提取物中分离得到了十余种化学成分,并运用现代谐波学技术（MS,NMR)鉴定了其中九种化合物的结构,它们分别是：Nonaconsane,Nentriacontane,Triacontano,Dotriacontanol,Tatratriacontaol,β－Sitosterol,2-O-methylinositol,(-)-Epicatechin和Quercetin-3-O-β-D-glucoside.     

Studies on Bacillus stearothermophilus. Part 1. Transformation of progesterone to a new metabolite 9,10-seco-4-pregnene-3,9,20-trione

When Bacillus stearothermophilus, a thermophilic bacterium isolated from the Kuwaiti desert, was incubated with exogenous progesterone for 24 h, three monohydroxylated metabolites were produced. 20α-Hydroxyprogesterone was the major metabolite produced in 60.8 relative percentage yield. The other two monohydroxylated metabolites were identified as 6β-hydroxyprogesterone and the rare 6α-hydroxyprogesterone in 21.0 and 13.6 relative percentage yields, respectively. A new metabolite 9,10-seco-4-pregnene-3,9,20-trione was isolated in 3.7 relative percentage yield. All metabolites were purified by preparative TLC and HPLC followed by their identification using ¹H, ¹³C NMR and other spectroscopic data.     

New method for determination of fecal sterols in urine using non-chlorinated solvents

A new method has been developed to determine a number of sterols in urine using non-chlorinated solvents, namely methyl tert.-butyl ether and methanol (the MTB method). The method involves liquid–liquid extraction, saponification, reextraction, silylation and final identification and quantification by GC–FID. The sterols determined were coprostanol, epicoprostanol, cholesterol and dihydrocholesterol. 5α-Cholestane was used as internal standard. The limit of detection for the sterols in this experiment was 2 μg l⁻¹ urine. Recovery of coprostanol over the range 5–100 μg l⁻¹ urine by this method was between 80 and 92%.     

Melanogenesis‐Inhibitory and Cytotoxic Activities of Chemical Constituents from the Leaves of Sauropus androgynus L. Merr. (Euphorbiaceae)

A new steroid, 20‐hydroxyisofucosterol (stigmasta‐5,24(28)‐diene‐3β,20β‐diol) ( 7 ), along with six known compounds 1 – 6 were isolated from the MeOH extract of the leaves of Sauropus androgynus L. Merr . (Euphorbiaceae). The structure of new steroid was determined by HR‐APCI‐MS and various NMR techniques in combination with literature data. Subsequently, their anti‐inflammatory, cytotoxic activities against five human cell lines, as well as inhibitory activities against the α‐MSH induced melanogenesis on the B16 cell line were evaluated. As the results, steroid compounds, 6 and 7 exhibited moderate cytotoxic to HL60, AZ521, SKBR3, and A549 tumor cell lines (IC₅₀ 26.9 – 45.1 μm ) with high tumor selectivity for A549 relative to WI38 cell lines (SI 2.6 and 3.0, resp.). And, flavonoid compounds, 4 and 5 exhibited superior inhibitory activities against melanogenesis (67.0 – 94.7% melanin content), even with no or low toxicity to the cells (90.1 – 99.6% cell viability) at the concentrations from 10 to 100 μm . Furthermore, Western blot analysis suggested that compound 5 could inhibit melanogenesis by suppressing the protein expressions of MITF, TRP‐1, TRP‐2, and tyrosinase.

Cold-induced phase separation for the simple and reliable extraction of sex hormones for subsequent LC-MS/MS analysis

Sex hormones, including androgens, estrogens, and progestogens, are important biomarkers for various diseases. Quantification of sex hormones is typically conducted by LC-MS/MS. At present, most methods require liquid-liquid extraction or solid phase extraction for sample preparation. However, these pretreatments are prone to compromise LC-MS/MS throughput. To improve on the current standard practices, we investigated cold-induced phase separation for sex hormone extraction. After protein precipitation with acetonitrile and adjusting the solution constitution with water, samples were stored at ?30°C for 10 min to generate two distinct phases: an acetonitrile-rich layer on top of a water-rich layer. During this process, the hydrophobic sex hormones spontaneously separate into the upper layer. This simple and reliable cold-induced phase separation-based LC-MS/MS methodology was used here to simultaneously detect estrone, estradiol, estriol, testosterone, androstenedione, dehydroepiandrosterone, progesterone, and 17-hydroxyprogesterone in serum. Validation of this method indicated satisfactory performance, including acceptable linearity, accuracy, precision, and tractability. Compared with the mainstream liquid-liquid extraction-based method, this new method exhibits significant progress in throughput, which shortens the time cost of sample preparation from 90 to 40 min. We propose that this method can be an excellent alternative for sex hormone analysis in routine clinical laboratories.     

The regulation of maternal behaviour in marmosets and tamarins

This article reviews a long-term investigation into the roles of (1) experience obtained with infants during development and postnatally, and (2) levels of reproductive-steroid hormones during late pregnancy, in the regulation of maternal caregiving motivation and behaviour in the red-bellied tamarin (Saguinus labiatus) and the common marmoset (Callithrix jacchus) (family Callitrichidae, Order Primates). Both observational studies, e.g. post hoc analysis of the relationship between developmental caregiving experience and infant-rearing success, and manipulative studies, e.g. direct analysis of the effects of sex-steroid administration on maternal motivation, were performed. In red-bellied tamarins, a breeding female's infant-rearing success was related to its caregiving experience obtained during development and postpartum/postnatally. In multiparous females lacking developmental caregiving experience, postpartum maternal caregiving behaviour and infant-rearing success were related to prepartum urinary oestradiol levels. In nulliparous common marmosets, caregiving of infant-siblings increased as a consequence of physical interaction with parent-infant dyads, late pregnancy, and exposure to late pregnancy-like levels of progesterone and oestradiol. The evidence for a dual mechanism of endocrine and experiential regulation of caregiving in these monkeys is discussed in terms its evolution and function.
