Summary Developmental changes of thyrotropin-releasing hormone (TRH)-immunoreactive structures in the brain of mallard embryos were studied by means of immunocytochemistry (PAP technique). The primary antibody was generated against synthetic TRH. Immunoreactive neurons were first detected in the hypothalamus of 14-day-old embryos. By day 20, increasing numbers of immunoreactive perikarya were observed in the paraventricular nucleus, anterior preoptic region and supraoptic region. Immunoreactive fiber projections were seen in the median eminence as early as embryonic day 20; they occurred also in some extrahypothalamic regions (lateral septum, accumbens nucleus). The number and staining intensity of the cell bodies increased up to hatching, and continued to increase during the first week after hatching.  相似文献

The objective of this study was to assess the efficacy of a subcutaneous melatonin implant to suppress estrus in queens (felis catus). The hypothesis was that this implant would temporarily and reversibly suppress estrus in queens without producing any clinically detectable side effects. Fourteen adult queens were maintained in cages under artificial illumination (14 h light:10 h dark) for 45 d and then randomly assigned to one of two treatments. At interestrus, queens received a single subcutaneous melatonin implant (18 mg; Melovine [CEVA Sante Animal, Libourne, France]; MEL: n = 9), or a single subcutaneous placebo implant without melatonin (0 mg; PLA; n = 5). At the next estrus, all queens received a second MEL (n = 9) or PLA (n = 5) implant. Blood samples were taken when queens displayed estrous signs and during interestrus to measure estradiol (E2) and progesterone (P4), respectively, by radioimmunoassay. There were no significant differences in duration of the interestrus interval in PLA cats, regardless of whether the implants were placed during interestrus or estrus (6.0 ± 9.7 d vs. 6.0 ± 9.7 d, respectively; least square means [LSM] ± SEM). However, when MEL implants were placed during interestrus, the duration of interestrus was approximately twice as long as that occurring when MEL implants were placed during estrus (113.3 ± 6.1 d vs. 61.1 ± 6.8 d, respectively; P < 0.01). Serum E2 and P4 concentrations were similar in queens with PLA and MEL implants and in queens that received implants in estrus and interestrus. In conclusion, a subcutaneous MEL implant effectively and reversibly suppressed estrus in queens for approximately 2 to 4 mo with no clinically detectable side effects.  相似文献

为了解华东地区家鸭内禽流感病毒的遗传进化情况,对2002~2006年分离自华东地区家鸭的3种主要N1亚型的禽流感病毒:2株H1N1、10株H3N1和14株H5N1,共26株病毒的NA基因进行了遗传进化分析。结果表明,华东地区家鸭中的N1亚型的禽流感病毒正处于不断进化状态中。14株H5N1禽流感病毒均在NA的茎部缺失20个氨基酸(49~68位),而其他N1亚型的禽流感病毒的NA都未见发生此缺失。H3N1病毒可能与H1N1病毒发生了NA基因的重排,但是目前还没有直接证据表明华东地区家鸭中H5N1禽流感参与了基因重排。  相似文献

2012年巫山蓝家寨遗址考古发掘中,出土了春秋时期的马(Equus caballus)、黄牛(Bos taurus)、山羊(Capra hircus)、鸡(Gallus gallus domesticus)、狗(Canis familiaris)、猪(Sus domestica)6种家畜的骨骼。这是三峡地区唯一一个在先秦时期六畜兼备的古文化遗址。遗址中的马、牛,是三峡地区同类家畜中时代最早的发现资料。三峡地区早期历史时期,人类肉食资源以野生动物为主,家畜动物骨骼发现材料较少。蓝家寨遗址出现种类较多的家畜动物种类,为全面分析和认识三峡地区经济形态发展过程、古居民行为模式和区域文化等,提供了重要资料。遗址中在春秋时期出现的家畜马和黄牛,与该时期各种区域文化在三峡地区的碰撞、交流、融合有关。  相似文献

Oviductin is known to be a key player providing a convenient environment for the process of fertilization affecting this by direct interaction with oocytes and sperm. As in vitro embryo production in the context of assisted reproduction for endangered felids is still in the process of optimization, oviductin might be used to improve IVF results. Recombinant His-tagged feline oviductin was expressed by transformed Escherichia coli BL21DE3 cells. The protein was purified by immobilized metal ion affinity chromatography. The effect of the recombinant protein was characterized in three experiments: a hemizona assay for sperm binding analysis, the IVF outcome, and the relative mRNA expression levels in blastocysts after IVF. A significant higher number of bound sperm cells were found after incubation in oviductin. No significant effect on cleavage, morula, and blastocyst rates with or without oviductin incubation during IVF could be observed. However, the relative mRNA abundance of GJA1, a gene, whose expression level is known to be a marker of embryo quality, was significantly increased (P value less than 0.05) in blastocysts after oviductin treatment. In contrast to this, expression of OCT4, HSP70, DNMT1, DNMT3A, BAX, IGF1R, and GAPDH was not significantly affected. We assume that our recombinant oviductin in its current nonglycosylated form is able to enhance sperm binding. Despite of a missing significant effect on IVF outcome, embryo quality in terms of relative GJA1 expression is influenced positively. These promising results demonstrate the value of recombinant oviductin for the IVF in cats.  相似文献

The aim of the study was to find the most stable reference genes from: ACTB, GAPDH, RPL30, CYC, RPL17, RPS7 and YWHAZ in the feline endometrium. Three free software packages, geNorm, NormFinder and BestKeeper were used. In geNorm analysis, the most stable gene was RPS7 (at a primer concentration 1000 nM) or YWHAZ (500 and 250 nM). According to NormFinder and BestKeeper, ACTB (at all examined primer concentrations) followed by RPS7 and CYC were the most stable genes. Based on geNorm results at least two genes from among RPS7, RPL30, ACTB or YWHAZ should be chosen for Real Time-PCR result normalization.  相似文献
