We examined the changes of hemocytes in the diamondback moth, Plutella xylostella (Lepidoptera: Plutellidae), due to parasitism by the endoparasitoid Diadegma semiclausum (Hymenoptera: Ichneumonidae). Necrosis of prohemocytes in different stages was observed while cell death was absent in the mature hemocytes in the parasitized larvae, which was related to the declined total hemocyte count per microliter (THC). THC in the host hemolymph declined sharply by 12 h post-parasitization and then remained at a low level. When hemocytes of the parasitized larvae were cultured in vitro, encapsulation ability was suppressed coincidently with the inhibited spreading ability; however, such effects were transient. Simultaneously, activation of the prophenoloxidae from the hemocytes was inhibited. Unlike the results of previous studies, the decrease in hemocytes, which was due to the necrosis of the prohemocytes instead of the mature hemocytes in our study, was not responsible for the impaired encapsulation. Our studies suggest that parasitism by D. semiclausum have some effects on hematopoietic regulation and on hemocyte immune reaction of P. xylostella larvae.  相似文献

  • 1 An integrated pest management (IPM) system incorporating the introduction and field release of Diadegma semiclausum (Hellén), a parasitoid of diamondback moth (DBM) Plutella xylostella (L.), comprising the worst insect pest of the cabbage family, has been developed in Kenya to replace the pesticides‐only approach.
  • 2 Mathematical modelling using differential equations has been used in theoretical studies of host–parasitoid systems. Although, this method helps in gaining an understanding of the system's dynamics, it is generally less accurate when used for prediction. The artificial neural network (ANN) approach was therefore chosen to aid prediction.
  • 3 The ANN methodology was applied to predict the population density of the DBM and D. semiclausum, its larval parasitoid. Two data sets, each from different release areas in the Kenya highlands, and both collected during a 3‐year period after the release of the parasitoid, were used in the present study. Two ANN models were developed using these data.
  • 4 The ANN approach gave satisfactory results for DBM and for D. semiclausum. Sensitivity analysis suggested that pest populations may be naturally controlled by rainfall.
  • 5 The ANN provides a powerful tool for predicting host–parasitoid population densities and made few assumptions on the field data. The approach allowed the use of data collected at any appropriate scale of the system, bypassing the assumptions and uncertainties that could have occurred when parameters are imported from other systems. The methodology can be explored with respect to the development of tools for monitoring and forecasting the population densities of a pest and its natural enemies. In addition, the model can be used to evaluate the relative effectiveness of the natural enemies and to investigate augmentative biological control strategies.

半闭弯尾姬蜂Diadegma semiclausum是小菜蛾Plutella xylostella的优势内寄生蜂, 拥有毒液、多分DNA病毒(PDV)等寄生因子,能有效调控寄主幼虫的营养生理和免疫系统, 但其毒液在这过程中的功能不明。本文利用SDS-PAGE方法分析了半闭弯尾姬蜂毒液的蛋白组分,利用寄主幼虫血细胞体外原代培养的方法,研究了小菜蛾幼虫血细胞噬菌能力在半闭弯尾姬蜂寄生后的变化情况。结果表明:半闭弯尾姬蜂毒液蛋白分子量主要集中在35~220 kDa之间,少数小于15 kDa,但分子量处于35~70 kDa之间的蛋白含量较高,与其他寄生蜂毒液蛋白相似。半闭弯尾姬蜂毒液单独对寄主小菜蛾幼虫功能血细胞(浆血细胞和颗粒血细胞)的延展能力和吞噬功能不产生破坏作用。但半闭弯尾姬蜂寄生后短时间内,寄主功能血细胞的延展受到抑制,然而功能血细胞仍然能识别外源异物, 却无法进一步吞噬外源物; 寄生后24 h,功能血细胞的延展力恢复,颗粒血细胞的吞噬作用可顺利完成。本研究证明了半闭弯尾姬蜂寄生能暂时性地抑制颗粒血细胞的延展性从而影响其噬菌过程。  相似文献

Knowledge of the biological attributes of parasitoids plays a fundamental role in improved management of their pest hosts. Diadegma mollipla is a major indigenous parasitoid of the Diamondback Moth (DBM), Plutella xylostella (Linnaeus), in Eastern Africa. Two laboratory experiments were conducted to determine the DBM larval stage preferences of D. mollipla and their influence on life history traits and the effect of different diets (20% honey, 20% sugar solution, water or nothing) on adult parasitoid longevity. In the first study, 60 DBM larvae (15 each of larval stages L1–L4) were exposed to either one adult parasitoid female or four competing females in choice experiments. When DBM larvae were exposed to one parasitoid, in terms of parasitism, L1 and L2 were found to be the most preferred stages, followed by L3 and then L4. Sex ratio (females/males) increased with host larval stage from 1.3 to 3.1. Pre-imaginal development time was longer in L1 than in the other three stages. When exposed to four parasitoids, no significant differences in parasitism rates and progeny production were obtained between DBM larval stages. However, sex ratio increased from 0.8 in L1 to 4.1 in L4. Developmental time was longest in L4, intermediate in L1 and L3, and shortest in L2. In the diet experiment, honey and sugar were found to significantly increase longevity for D. mollipla adults of both sexes. Overall, combined adult male and female longevity was 24.5, 9.9, 2.2 and 1.9 days when fed on honey, sugar, water or nothing, respectively.  相似文献

Host discrimination was investigated in two related species of ichneumonid wasps: Diadegma eucerophaga Horst. and D. fenestralis (Holmgr.) attacking a common host, the diamond-back moth Plutella xylostella (L.). In the laboratory, I studied: (1) the relative amounts of time spent between patches containing unparasitized or parasitized hosts, (2) the results of parasitoid encounters with parasitized and unparasitized hosts, (3) dissections to reveal whether or not the wasps preferred to oviposit in unparasitized hosts. D. eucerophaga showed discrimination by (1) avoiding patches containing parasitized hosts and by (2) preference to oviposit in unparasitized larvae. No discrimination was detected for D. fenestralis at any of the three levels.In the field, a population consisting primarity of D. eucerophaga was presented to plants containin either unparasitized or previously-parasitized larvae. Although superparasitism occurred, the wasps preferred to oviposit in the unparasitized larvae.
Résumé La sélection a été examinée sur P. xylostella attaqué par deux espèces voisines Diadegma eucerophaga et D. fenestralis. La sélection a été examinée examinée à 3 niveaux au laboratoire: (1) par la comparaison des temps passés sur feuilles portant des chenilles parasitées ou non; (2) par l'analyse des résultats des rencontres avec chenilles parasitées ou non; (3) par des dissections pour examiner dans quel type la ponte a lieu de préférence. Les résultats prouvent une sélection chez D. eucerophaga qui évite les feuilles portant des chenilles parasitées, et pond de préférence dans les chenilles non parasitées. Les chenilles parasitées sont autant attaquées que les saines (insertion de la tarière), ce qui suggère peut-être un signal interne et l'aptitude à ne pas pondre après sa détection. Aucune preuve de sélection à ces trois niveaux n'a pu être apportée pour D. fenestralis. Une explication est suggérée.La sélection par une population naturelle de D. eucerophaga a aussi été étudiée dans la nature en lui présentant des chenilles parasitées ou non. Bien qu'il y ait eu un superparasitisme, les ichneumonides ont préféré pondre dans les chenilles non parasitées.

Laboratory and greenhouse studies were conducted to investigate the suitability of 2 hymenopterous parasites,Diadegma eucerophaga Horstmann andApanteles plutellae Kurdjumov for introduction to control diamondback moth (DBM),Plutella xylostella (L.), a destructive pest of crucifers in tropical to subtropical Southeast Asia. Parasitism byD. eucerophaga was high at temperature range of 15°C to 25°C and that ofA. plutellae, at 20°C to 35°C. Both parasites were active in searching for host and oviposited only during photophase. No parasitism was observed during darkness. WhereasA. plutellae could parasitize all instars of DBM larvae,D. eucerophaga parasitized only the first 3 instars and failed to parasitize the 4th. Parasitism byD. eucerophaga was greater when DBM larvae were feeding on common cabbage (Brassica oleracea var.capitata L.), than on cauliflower (B. oleracea var.italica L.), broccoli (B. oleracea var.botrytis L.) or Chinese cabbage [Brassica campestris L. ssp.pekinensis (Lour) Olsson].A. plutellae parasitism was greater when DBM larvac were feeding on Chinese cabbage than on common cabbage, cauliflower or broccoli. Storage of pupae at 0°C and 4°C to 6°C for up to 2 weeks reduced emergence ofD. eucerophaga adults more than that ofA. plutellae. A non-selective insecticide, deltamethrin, was toxic to adults of both parasites but selective ones such asBacillus thuringiensis, teflubenzuron, and pirimicarb were not. Pupae were more tolerant than adults to insecticides. The insecticide-resistant Luchu strain and susceptible laboratory strain of DBM suffered an equal level of parasitism by both parasites.   相似文献

Abstract 1. The use of flowering vegetation has been widely advocated as a strategy for providing parasitoids and predators with nectar and pollen. However, their herbivorous hosts and prey may exploit floral food sources as well. 2. Previous laboratory studies have shown that not all flower species are equally suitable in providing accessible nectar. Relatively little is known about actual nectar exploitation under field conditions. 3. The present study investigates nectar exploitation by the pest, Plutella xylostella, and its parasitoid, Diadegma semiclausum, under field conditions and examines whether floral nectar exploitation in the field can be predicted based on controlled laboratory studies. 4. Insects were collected from fields bordered by flowering margins containing Fagopyrum esculentum, Lobularia maritima, Anethum graveolens, Centaurea jacea or the grass Lolium perenne (control). Whole insect bodies were individually assayed by HPLC to establish their sugar profile as a measure of the level of energy reserves and the degree of food source use. 5. The average overall sugar content of P. xylostella and D. semiclausum collected in fields bordered by flowering margins was significantly higher than those of individuals collected from grass‐bordered control plots. To the authors’ knowledge, this represents the first demonstration that nectar‐providing plants enhance the energetic state of herbivores under field conditions. 6. In contrast to earlier laboratory studies, the present study did not find elevated sugar contents in P. xylostella and D. semiclausum individuals collected from fields bordered by buckwheat (F. esculentum). 7. The present study shows widespread sugar feeding by both the herbivore and its parasitoid. It also shows that laboratory studies establishing nectar exploitation under controlled conditions can not always be extrapolated to actual exploitation under field conditions. This emphasises the importance of studying field‐collected insects with regard to food source use and nutritional status.  相似文献

The major constraint to the production of good quality cabbage in Papua New Guinea (PNG) is posed by a complex of insect pests which includes the Diamondback Moth Plutella xylostella (L.) as the most prominent species. In order to evaluate the prospects of a classical control approach economically important major pests and their natural enemies were surveyed in three cabbage growing areas with distinct climatic regimes at low-, mid- and high-elevation. In the PNG highlands (Goroka and Tambul area, Western Highlands Province) the release and successful establishment of the P. xylostella -specific parasitoid Diadegma semiclausum (Hellen) reduced crop losses remarkably since 1995. Favourable circumstances, such as continuous brassica cultivation and low status of associated lepidopteran pests, particularly at Tambul (2400 m above sea level) with an almost temperate climate, were supportive for this approach. In the arid PNG lowlands (Laloki area, Central Province) the introduced P.xylostella -parasitoid Cotesia plutellae (Kurdjumov) exerted 80% parasitism in an experimental site, although a lasting establishment was not achieved in that location. Among several indigenous P. xylostella parasitoids Brachymeria phya (Walker) was a ubiquitous species but did not reduce the pest status of its host. Natural enemies of associated lepidoptera such as Crocidolomia pavonana (Zeller), Spodoptera litura (F.), Helicoverpa armigera (Hübner) and Hellula undalis (F.) were economically insignificant. Thus, insecticidal control, preferably selective, was still a requirement. In comparison to Bacillus thuringiensis -products (Delfin ® and Thuricide ® ) and the synthetic insect growth regulator Chlorfluazuron (Atabron ® ), best results were achieved in field trials with the commercial neem formulation NeemAzal ® ( Azadirachta indica A. Juss.) and an aqueous neem seed kernel extract from seeds of PNG grown trees. Besides its high efficacy against lepidopterous pests, neem also controlled the false mustard aphid Lipaphis erysimi (Kalt.), being of economic importance in the arid PNG lowlands. The prospects of integrating C. plutellae into a reduced insecticide spraying program as well as alternative cultural control measures are discussed.  相似文献

在22℃,相对度60%~70%的实验室条件下,研究了半闭弯尾姬蜂Diadegma semiclausum的羽化、交配与产卵行为。结果表明:半闭弯尾姬蜂蜂蛹从开始羽化到羽化结束历经4d,羽化高峰期出现在羽化的第3d,羽化数量为25头,占全部羽化数量的58.82%;第3d中羽化高峰出现在8:00~13:00,羽化数量为16.5头,占全天羽化数量的66.00%。半闭弯尾姬蜂交配过程大致分为3个阶段:准备阶段、交配阶段、结束阶段;交配后开始产卵,产卵过程大致分为寄主寻找和确定、穿刺和产卵、产卵结束和梳理。  相似文献
