In order to understand how inter-subunit association stabilizes oligomeric proteins, a single polypeptide chain variant of heptameric co-chaperonin GroES (tandem GroES) was constructed from Escherichia coli heptameric GroES by linking consecutively the C-terminal of one subunit to the N-terminal of the adjacent subunit with a small linker peptide. The tandem GroES (ESC7) showed properties similar to wild-type GroES in structural aspects and co-chaperonin activity. In unfolding and refolding equilibrium experiments using guanidine hydrochloride (Gdn-HCl) as a denaturant at a low protein concentration (50 microg ml(-1)), ESC7 showed a two-state transition with a greater resistance toward Gdn-HCl denaturation (Cm=1.95 M) compared to wild-type GroES (Cm=1.1 M). ESC7 was found to be about 10 kcal mol(-1) more stable than the wild-type GroES heptamer at 50 microg ml(-1). Kinetic unfolding and refolding experiments of ESC7 revealed that the increased stability was mainly attributed to a slower unfolding rate. Also a transient intermediate was detected in the refolding reaction. Interestingly, at the physiological GroES concentration (>1 mg ml(-1)), the free energy of unfolding for GroES heptamer exceeded that for ESC7. These results showed that at low protein concentrations (<1 mg ml(-1)), the covalent linking of subunits contributes to the stability but also complicates the refolding kinetics. At physiological concentrations of GroES, however, the oligomeric state is energetically preferred and the advantages of covalent linkage are lost. This finding highlights a possible advantage in transitioning from multi-domain proteins to oligomeric proteins with small subunits in order to improve structural and kinetic stabilities.  相似文献

Enzyme activities localized in the luminal compartment of the endoplasmic reticulum are integrated into the cellular metabolism by transmembrane fluxes of their substrates, products and/or cofactors. Most compounds involved are bulky, polar or even charged; hence, they cannot be expected to diffuse through lipid bilayers. Accordingly, transport processes investigated so far have been found protein-mediated. The selective and often rate-limiting transport processes greatly influence the activity, kinetic features and substrate specificity of the corresponding luminal enzymes. Therefore, the phenomenological characterization of endoplasmic reticulum transport contributes largely to the understanding of the metabolic functions of this organelle. Attempts to identify the transporter proteins have only been successful in a few cases, but recent development in molecular biology promises a better progress in this field.  相似文献

本研究旨在探讨细胞外信号调节激酶1/2(extracellular signal-regulated kinase,ERK1/2)信号通路在慢性哮喘模型大鼠支气管平滑肌细胞(bronchial smooth muscle cells,BSMCs)迁移能力改变中的调控作用。应用卵清蛋白致敏和雾化方法制备大鼠慢性哮喘模型,体外培养大鼠BSMCs,采用免疫荧光细胞化学、Western blot和RT-PCR方法检测ERK1/2信号通路的表达,分别用平面迁移实验和跨膜迁移实验来评价BSMCs的活动和趋向迁移能力,并比较用和不用ERK1/2信号通路干预剂的差异。Western blot结果显示慢性哮喘模型大鼠BSMCs中总ERK1/2(9.13±0.87)较对照组(4.68±0.59)明显增加,磷酸化ERK1/2(p-ERK1/2)占总ERK1/2的比值(0.55±0.05)较对照组(0.48±0.04)显著提高(n=10,P<0.01)。慢性哮喘组ERK1和ERK2 mRNA的表达(1.83±0.24和1.07±0.11)较对照组(0.58±0.14和0.51±0.12)明显增高(n=10,P<0.01)。在平面迁移实验中,慢性哮喘大鼠BSMCs的迁移最远距离是对照组的(2.9±0.1)倍,在ERK1/2激动剂表皮生长因子(epidermal growth factor, EGF)刺激下增加到(5.0±0.2)倍,而在30μmol/L PD98059的作用后下降到(1.7±0.2)倍。正常对照大鼠BSMCs平面迁移能力对PD98059的反应较慢性哮喘组弱,仅在100μmol/L PD98059的作用下下降到(0.8±0.1)倍。跨膜迁移实验中,慢性哮喘大鼠BSMCs的跨膜迁移细胞是对照组的(1.9±0.1)倍,在EGF刺激下增加到(3.1±0.2)倍,而在30μmol/L PD98059作用后下降到(1.45±0.2)倍。这些结果表明慢性哮喘模型大鼠BSMCs的迁移能力明显增强,ERK1/2信号通路在该功能变化的调控中可能发挥了重要作用。  相似文献

This study examined whether in vivo exposure to a β2-adrenoceptor agonist, tulobuterol, induces human Period1 (hPer1) mRNA expression in cells from peripheral whole blood. In one experiment, oral tulobuterol was administered to five healthy volunteers at 22:00 h, while in another, a transdermally tulobuterol patch was applied to the same five subjects at 20:00 h. In each experiment, serum tulobuterol concentrations were measured at four time points, and total RNA was isolated from peripheral blood cells for determinations of hPer1 mRNA expression by real-time polymerase chain reaction. Both the tulobuterol tablet and the transdermal patch increased hPer1 mRNA expression, suggesting that analyses of human peripheral blood cells could reliably represent peripheral clock gene mRNA expression in vivo.  相似文献

为建立一种快速鉴别严重急性呼吸综合征冠状病毒2 (severe acute respiratory syndrome coronavirus 2,SARS-CoV-2)的5种主要变异株的Taq Man探针实时荧光定量PCR(real-time quantitative PCR, RT-qPCR)体系,基于SARS-CoV-2野生型及变异株alpha (N501Y、HV69-70del)、beta (E484K、K417N)、gamma (K417T、V1176F)、delta (L452R、T478K)和omicron (H655Y、N679K、P681H)序列设计特异性引物、探针,建立和优化一种鉴别新型冠状病毒(SARS-CoV-2) 5种主要变异株的Taq Man探针RT-qPCR方法,并进行该方法的特异性、敏感性、鉴别能力评价。该方法可准确区分出SARS-CoV-2野生型和突变型,与其他呼吸道病原体(n=21)无交叉,显示高特异性。该方法最低检测限为2×10;拷贝/mL,操作简单、快速、成本廉价,可用于监测SARS-CoV-2毒株的变异,精准指导疫情识别与防控。  相似文献

Interfering with the assembly of Amyloid β (Aβ) peptides from monomer to oligomeric species and fibrils or promoting their clearance from the brain are targets of anti-Aβ-directed therapies in Alzheimer disease. Here we demonstrate that cromolyn sodium (disodium cromoglycate), a Food and Drug Administration-approved drug already in use for the treatment of asthma, efficiently inhibits the aggregation of Aβ monomers into higher-order oligomers and fibrils in vitro without affecting Aβ production. In vivo, the levels of soluble Aβ are decreased by over 50% after only 1 week of daily intraperitoneally administered cromolyn sodium. Additional in vivo microdialysis studies also show that this compound decreases the half-life of soluble Aβ in the brain. These data suggest a clear effect of a peripherally administered, Food and Drug Administration-approved medication on Aβ economy, supporting further investigation of the potential long-term efficacy of cromolyn sodium in Alzheimer disease.  相似文献

目的探讨孟鲁司特联合舒利迭对哮喘急性发作期患者疗效及免疫功能的影响。方法 2012年1月至2014年1月选取在本院收治的120例哮喘急性发作期患者为研究对象,根据随机数字表将患者分为观察组及对照组各60例,对照组给予舒利迭雾化吸入治疗,50 mg/250 mg,2次/d,观察组在对照组基础上口服孟鲁司特,10 mg/次,1次/d,疗程为2周。对比分析两组患者治疗效果及治疗前后肺功能及免疫功能的变化。结果观察组总有效率为96.67%,对照组总有效率为76.67%,差异有统计学意义(P0.05)。观察组患者治疗后第1秒呼吸气体的容积(FEV1)、用力呼吸的肺活量(FVC)及FVC占预计值百分比(FVC%)改善优于对照组(P0.05)。观察组治疗后白细胞介素4(IL-4)、免疫球蛋白(Ig E)水平、IL-4/INF-γ显著低于对照组,而干扰素(IFN-γ)与对照组相比差异无统计学意义。结论孟鲁司特联合舒利迭能有效改善哮喘急性发作期患者肺功能及免疫功能,提高哮喘患者治疗效果。  相似文献
