The frequency of sandfly–host contacts can be measured by host antibody levels against sandfly salivary proteins. Recombinant salivary proteins are suggested to represent a valid replacement for salivary gland homogenate (SGH); however, it is necessary to prove that such antigens are recognized by antibodies against various populations of the same species. Phlebotomus perniciosus (Diptera: Psychodidae) is the main vector of Leishmania infantum (Trypanosomatida: Trypanosomatidae) in southwest Europe and is widespread from Portugal to Italy. In this study, sera were sampled from naturally exposed dogs from distant regions, including Campania (southern Italy), Umbria (central Italy) and the metropolitan Lisbon region (Portugal), where P. perniciosus is the unique or principal vector species. Sera were screened for anti‐P. perniciosus antibodies using SGH and 43‐kDa yellow‐related recombinant protein (rSP03B). A robust correlation between antibodies recognizing SGH and rSP03B was detected in all regions, suggesting substantial antigenic cross‐reactivity among different P. perniciosus populations. No significant differences in this relationship were detected between regions. Moreover, rSP03B and the native yellow‐related protein were shown to share similar antigenic epitopes, as canine immunoglobulin G (IgG) binding to the native protein was inhibited by pre‐incubation with the recombinant form. These findings suggest that rSP03B should be regarded as a universal marker of sandfly exposure throughout the geographical distribution of P. perniciosus. 相似文献
Potential long-term health effects from tobacco products can be estimated by measuring changes in biochemical indicators of disease mechanisms like inflammation. This study assesses the potential relationships between biomarkers of potential harm (BOPH) and biomarkers of cigarette smoke exposure (BOE) based on data from the NHANES (2007–2012, n?=?17,293 respondents). Statistically significant relationships were observed between white blood cells (WBC) and high-density lipoprotein (HDL) and BOE; between WBC and high-sensitivity C-reactive protein and smoking status; and between WBC and HDL and smoking intensity. This analysis suggests that WBC and HDL are useful BOPH in studies assessing the health risks of cigarette smoking. 相似文献
Context: Genotoxicity assays are widely employed in human biomonitoring studies to assess genetic damage inflicted by genotoxic agents.
Objective: Evaluation of micronuclei (MN) as a screening marker of occupational ionizing radiation (IR) exposure.
Materials and methods: Using micronucleus test, peripheral blood lymphocytes (PBL) of 402 control and exposed subjects were screened for genetic damage.
Results: The mean frequencies of micronucleus test parameters were significantly higher in exposed persons. Increase of micronucleus yield with duration of exposure (DOE) by 0.303MN/year was revealed.
Discussion and conclusion: The obtained data encourage us to consider MN as valuable markers for preventive medical screening of occupationally exposed groups. 相似文献
We aimed to examine the effects of night work on salivary melatonin concentration during and subsequent to night work and the mediating role of light. We included 254 day workers and 87 night workers who were followed during 322 work days and 301 days off work. Each day was defined as the 24 hour period starting from the beginning of a night shift or from waking in the mornings with day work and days off. Light levels were recorded and synchronized with diary information (start and end of sleep and work). On average, participants provided four saliva samples per day, and these were analyzed for melatonin concentration by liquid chromatography tandem mass spectrometry (LC-MS/MS). Differences between day and night workers on work days and days off were assessed with multilevel regression models with melatonin concentration as the primary outcome. All models were stratified or adjusted by time of day. For light exposure, we estimated the total, direct and indirect effects of night work on melatonin concentrations obtaining 95% confidence intervals through bootstrapping. On work days, night workers showed 15% lower salivary melatonin concentrations compared with day workers (?15.0%; 95% CI: ?31.4%; 5.2%). During the night, light exposure mediated a melatonin suppression of approximately 6% (?5.9%, 95% CI: ?10.2%; ?1.5%). No mediating effect of light was seen during the day time. On days off, we observed no difference in melatonin concentrations between day and night workers. The circadian time structure (CTS) and its disruption by rotating and nightshift schedules relative to work performance, accident risk, and health/wellbeing have long been areas of occupational medicine research. Yet, there has been little exploration of the relevance of the CTS to setting short-term, time-weighted, and ceiling threshold limit values (TLVs); conducting employee biological monitoring (BM); and establishing normative reference biological exposure indices (BEIs). Numerous publications during the past six decades document the CTS substantially affects the disposition – absorption, distribution, metabolism, and elimination – and effects of medications. Additionally, laboratory animal and human studies verify the tolerance to chemical, biological (contagious), and physical agents can differ extensively according to the circadian time of exposure. Because of slow and usually incomplete CTS adjustment by rotating and permanent nightshift workers, occupational chemical and other contaminant encounters occur during a different circadian stage than for dayshift workers. Thus, the intended protection of some TLVs when working the nightshift compared to dayshift might be insufficient, especially in high-risk settings. The CTS is germane to employee BM in that large-amplitude predictable-in-time 24h variation can occur in the concentration of urine, blood, and saliva of monitored chemical contaminants and their metabolites plus biomarkers indicative of adverse xenobiotic exposure. The concept of biological time-qualified (for rhythms) reference values, currently of interest to clinical laboratory pathology practice, is seemingly applicable to industrial medicine as circadian time and workshift-specific BEIs to improve surveillance of night workers, in particular. Furthermore, BM as serial assessments performed frequently both during and off work, exemplified by employee self-measurement of lung function using a small portable peak expiratory flow meter, can easily identify intolerance before induction of pathology. 相似文献
The aim of this study was to assess the associations between respiratory symptoms and pulmonary defects of Para rubber sawmill workers in the South of Thailand. A cross-sectional study was conducted among 687 workers in Para rubber wood sawmills and a subset of sawmill workers from four factories participated in spirometric measurements and personal dust samplings. Multiple logistic regression analysis was used to explore risk factors pulmonary impairment. The exposed workers had higher respirable wood dust exposure (0.902 mg/m3) compared with the unexposed groups (0.185 mg/m3). The exposed group had significantly higher prevalence than the unexposed group for chest tightness (odds ratio (OR) = 2.79) and shortness of breath (OR = 2.27). The ventilatory function values (FEV1 and FVC) were lower for the exposed group compared with the unexposed group (2.41 vs 2.55 L/s and 2.91 vs 3.01 L/s, respectively). The results suggest that sawmill factories using Para rubber tree wood should implement appropriate exposure control measures to reduce wood dust exposure to protect their employees. 相似文献
To investigate the contamination level, distribution, possible source, and human exposure risk of polycyclic aromatic hydrocarbons (PAHs) in the urban traffic environment, 15 PAHs were measured in 34 road dust samples (particle size < 25 μm) collected from three grades of roads and park paths in Xinxiang, China. ΣPAHs concentrations ranged from 311 to 21200 ng g?1, with a mean of 5890 ng g?1 and decreased in the following order: main roads (7650 ng g?1) > collector streets (7410 ng g?1) > bypasses (2970 ng g?1) > park paths (1570 ng g?1), indicating that significant positive correlation existed between PAH contamination and traffic density. PAHs in all samples were dominantly composed of 4-ring PAHs, accounting for 44.8% of the total. Pyrene, fluoranthene, and chrysene were the predominant individual components and accounted for 14.7% (1.2–19.2%), 12.9% (3.3–20.3%), and 11.0% (2.5–18.6%) of ΣPAHs, respectively. The specific isomer ratios indicated that traffic emission was the dominant source of PAHs in road dust. The incremental lifetime cancer risk values showed that cancer risk from exposure to road dust–borne PAHs was acceptable for local residents in Xinxiang. 相似文献
