为考察温水性鲤科鱼类在不同食物资源条件下标准代谢率(Standard metabolic rate, SMR)与其生长性能的关系, 研究在(25.00.5)℃条件下测定体重相近的30尾锦鲫(Carassius auratus)幼鱼的SMR, 将所有实验鱼置于多单元格水槽进行实验处理(摄食与饥饿)。实验时间为4周(0、7d、14d、21d和28d), 包括两周的生长实验(014d)和两周的饥饿实验(1528d)。摄食期间, 于每天上午9: 00和下午21: 00用商业饲料对每尾鱼饱足投喂并记录投喂量。单尾鱼的体重和体长每隔1周测定一次, 而SMR仅在第0、第14和第28天测定。结果显示:(1)锦鲫幼鱼在摄食期间的体重、体长以及SMR均明显上升, 饥饿期间体重和SMR显著降低(P0.05), 但体长变化不明显; SMR不论摄食期间还是饥饿期间呈现较好的稳定性(二者P0.05)。(2)锦鲫幼鱼在摄食期间开始时的SMR (测定Ⅰ)与摄食率(FR)、摄食转化率(FE)以及特定体重生长率(SGRBM)均无相关性, 摄食期间结束时的SMR (测定Ⅱ)仅与FR呈正相关(P0.05), 但与FE以及SGRBM不相关。(3)摄食期间(饥饿期间)实验鱼的第一周日均体重增(减)量与第二周日均体重增(减)量呈正相关, 其体重的增率与减率无显著差异(P0.05)。(4)实验鱼在摄食期间的特定体重增长率(SGRBM)与饥饿期间的不相关(P0.05), 但摄食期间的特定SMR增长率(SGRSMR)与饥饿期间的呈负相关(P0.05)。研究表明在实验室环境中锦鲫幼鱼的SMR具有稳定性, 该种鱼的日均体重变化量保持相近, 此形态变化特征与SMR不相关, 并且该种鱼在实验起始时的SMR的个体差异未能预测其在实验室食物资源变动下生长性能的适应特征。  相似文献

Summary The distribution of neuropeptide Y (NPY) immunoreactivity has been studied by means of immunocytochemistry and radioimmunoassay in the brain of the goldfish. It was found that NPY had a widespread distribution in the entire brain in particular in the telencephalon, diencephalon, optic tectum and rhombencephalon. In the pituitary gland, positive type-B fibers were observed in the various lobes frequently in direct contact with secretory cells, in particular the gonadotrophs, somatotrophs and MSH (melanocyte-stimulating hormone) secreting cells. When measured by radioimmunoassay, the highest NPY concentrations were found in the pituitary and telencephalon, confirming the results of immunocytochemistry. The displacement curves obtained with serial dilutions of brain extracts were parallel to that of synthetic porcine NPY. Following high performance liquid chromatography, the NPY-like material extracted from goldfish brain co-eluted as a single peak with synthetic porcine NPY. These data demonstrate the presence of an NPY-like substance widely distributed in the goldfish brain. The observation of NPY-immunoreactive fibers in the pituitary gland suggests that, among its other functions, NPY may play a role in the neuroendocrine regulation of pituitary function.  相似文献

本文对鲫鱼后鳃体的形态结构及其功能进行了初步研究。结果表明,鲫鱼后鳃体是一种内分泌腺体。腺体由滤泡组成,滤泡上皮包括二种颗粒细胞(亮细胞和暗细胞)和一种支持细胞。对不同性腺期的鲫鱼后鳃体的观察表明,后鳃体滤泡细胞随性腺发育增加高度,滤泡增生,腺体体积增大。而性腺成熟后期,后鳃体滤泡上皮有崩溃现象。揭示后鳃体可能与性腺发育有关。用免疫组织化学的非标记过氧化物酶技术对鲫鱼后鳃体进行激素降钙素的定性定位研究,观察到滤泡上皮的颗粒细胞发生阳性反应,由此证明颗粒细胞可分泌降钙素。  相似文献

在胚胎发育过程中,用不同比放射性的~(90)锶试液处理受精卵,各处理组都有畸形和死亡现象。畸形的高峰一般出现在囊胚期和晶体形成期,畸形的类型一般为脊椎弯曲、体短、胸腔和卵黄囊扩大,头和眼畸形。随后即出现死亡高峰。例如:游动期的死亡率分别是:21%(对照);36%(5×10~(-11)居里/升);40%(5×1-~(-10)居里/升);45%(5×10~(-9)居里/升)。鲫仔鱼对~(90)锶吸收积累的结果表明:鲫仔鱼体中比放射性大小与试液的比放射性大小成正比,并随试验吋间的延长,生物量的增加而增加,鲫仔鱼的积累系数则不依试液的比放射性大小和生物量的多少为转移。  相似文献

Summary Immunohistochemistry was used to localize regulatory peptides in endocrine cells and nerve fibres in the pancreas of two species of elasmobranchs (starry ray,Raja radiata and spiny dogfish,Squalus acanthias), and in the Brockmann bodies of four teleost species (goldfish,Carassius auratus, brown troutSalmo trutta, rainbow trout,Oncorhynchus mykiss and cod,Gadus morhua). In the elasmobranchs, the classical pancreatic hormones somatostatin, glucagon and insulin were present in endocrine cells of the islets. In addition, endocrine cells were labelled with antisera to enkephalins, FMRF-amide, gastrin/cholecystokinin-(CCK)/caerulein, neurotensin, neuropeptide Y (NPY), and peptide YY (PYY). Nerve fibres were demonstrated with antisera against bombesin, galanin and vasoactive intestinal polypeptide (VIP). These nerve fibres innervated the walls of blood vessels, in the exocrine as well as the endocrine tissue. In the four teleost species immunoreactivity to somatostatin, insulin and glucagon was intense in the Brockmann bodies. Cells were labelled with antisera to enkephalin, neurotensin, FMRFamide, gastrin/CCK/ caerulein, NPY, PYY and VIP. Only a few nerve fibres were found with antisera against dopamine--hydroxylase (DBH, cod), enkephalin (met-enkephalin-Arg-Phe, cod), bombesin (cod), gastrin/CCK/caerulein (cod) and VIP. Galanin-like-immunoreactive fibres were numerous in the Brockmann bodies of all teleosts examined. Immunoreactivity to calcitonin gene-related peptide (CGRP), substance P, tyrosine hydroxylase (TH), and phenyl-N-methyl transferase (PNMT) could not be found in any of the species studied.  相似文献

Summary Nerve terminals forming typical synapses with adrenal chromaffin tissues have been examined in the goldfish, frog (Rana pipiens), hamster and rat. Presumptive secretory inclusions present in the terminals are of two distinct types. Electron-lucent synaptic vesicles 30–50 nm in diameter are densely clustered adjacent to membrane thickenings and presumably discharge their contents into the synaptic clefts. Secretory granules (i.e. large dense-cored vesicles) 60–100 nm in diameter are more abundant in other parts of the terminals. Sites of granule exocytosis have been observed in each of the animals investigated. They are usually encountered within apparently undifferentiated areas of plasmalemma and only rarely occur within synaptic thickenings. Granule exocytosis from within synaptic terminals and chromaffin gland cells is most readily observed in specimens exposed, prior to fixation, to saline solutions containing both tannic acid, and 4-aminopyridine and/or elevated levels of K+. These findings show that the pattern of secretory discharge, involving both synaptic and non-synaptic release, which is widespread in invertebrate central nervous systems, is also characteristic of vertebrate, peripheral cholinergic terminals.  相似文献

During the process of progestogen-induced meiotic maturation in the goldfish oocyte, the oocyte nucleus (germinal vesicle, GV) migrates to the sperm entry site or micropyle at the animal pole. Following GV migration (GVM) to the micropyle, the nuclear membrane undergoes dissolution (GVD) and the cell enters metaphase I in preparation to generate the first polar body. Microtubule destabilizing drugs including colcemid, nocodazole and vinblastine were found to elicit GVM, mimicking the process which occurs just prior to the prophase I-metaphase I transition during steroid induced oocyte meiotic maturation. In addition, these drugs enhanced the induction of GVM by 17 alpha, 20 beta dihydroxy-4-pregnen-3-one, a potent, naturally occurring meiotogenic steroid in this species. By contrast, taxol, a microtubule stabilizing drug, was found to inhibit steroid induced GVM. A new assay for centrifugation induced GVM was applied to the goldfish oocyte in order to assess effects of steroids and drugs on GVM, without the complication of GVD or the restrictions imposed by the slow time course of naturally occurring GVM. The effective centrifugal force (ECF) required to elicit GVM in 50% of the oocytes (ECF50) decreased significantly after short incubations (1-5 hr) of oocytes with either 17 alpha,20 beta dihydroxy-4-pregnen-3-one or microtubule disrupting drugs (i.e., colcemid, nocodazole, or vinblastine). A working hypothesis, modeled after the effects of microtubule disrupting agents on intermediate filament arrays in somatic cells, is proposed in which a small number of microtubules or other polymeric tubulin units are responsible for maintaining a cytoskeletal array.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

本研究以方正银鲫(Carassius auratus gibelio Bloch)普通鲫(Carassius auratus)和滇池高背鲫(Carassius sp.)的各种组织器官为材料,进行酯酶(Esterase)和乳酸脱氢酶(LDH)同工酶电泳图谱的分析比较。结果表明:9种不同组织中酯酶同工酶谱带各不相同,有明显的组织特异性。滇池高背鲫的酯酶谱图有3种表型。方正银鲫和滇池高背鲫同一组织的LDH同工酶酶谱也有明显差异。等电聚焦凝胶电泳(T=7.5%,C=5%)的结果又表明这二种鱼的肝脏、脑、卵的酯酶同工酶酶谱及电泳扫描图亦有差异。这些结果揭示滇池高背鲫与方正银鲫至少在生化水平上已有明显的分化,很可能起源于不同的地区,由不同的祖先,独立演化而形成。滇池高背鲫与云南普通鲫的LDH酶谱较为接近,这说明滇池高背鲫最可能起源于云南本地的普通鲫。  相似文献

Summary Microinjections of dopamine (DA) were made into specific forebrain loci in goldfish (Carassius auratus: 40–85 g) to study the involvement of DA in behavioral thermoregulation. Injections of 25, 50, 100 and 250 ng DA into the anterior aspect of the nucleus preopticus periventricularis (NPP) led to consistent, dose-dependent decreases in selected temperature was observed following injections of 5 or 10 ng DA. Injections of the control solution were without effect.Injections of DA into other forebrain loci, including the posterior half of the NPP, either had no thermoregulatory effect or had minor thermoregulatory effects which, in comparison to injections into the most effective sites, were inconsistent and required larger doses to obtain. The decrease in selected temperature following injections of 100 ng DA into the anterior NPP was blocked by haloperidol, a dopaminergic antagonist, but not by phentolamine, a noradrenergic antagonist. Injections of haloperidol alone resulted in a minor, but statistically significant, increase in selected temperature.The most sensitive DA sites lie caudal to the sites most sensitive to norepinephrine within the anterior NPP. DA acts on the dopaminergic receptors of central thermoregulatory neurons in the anterior NPP of goldfish. In this study, we isolated and characterized nine polymorphic trinucleotide microsatellites (CAG and CCT) from the crucian carp (Carassius auratus). The number of alleles per locus ranged from two to six. Five loci showed a significantly excess homozygosity, and a genetic linkage between CAL0102 and CAL0495 was strongly suggested. Our results confirmed the triploidy of Korean individuals, and the microsatellites were found to be useful for analysing the allelic state of the polyploid crucian carp.  相似文献
