The sperm ultrastructure of Epipedobates trivittatus and E. hahneli is described. The spermatological characteristics are identical in both species, except for the extension of the acrosomal vesicle. The structure of the acrosomal complex is shared with species of the Bufonoidea neobatrachian lineage, which differs from the condition observed in species included in the ranoid lineage. The phylogenetic significance of the mitochondrial collar, also seen in Eubufonoidea groups, is still unclear. Also the significance of the alternative mitochondrial arrangement seen here is unclear. The perpendicular arrangement of the centrioles is also shared with the Bufonoidea and is considered a synapomorphic condition. An expanded undulating membrane and the absence of a juxtaxonemal fibre appear to be apomorphic characteristics of dendrobatid species, as also observed (homoplastically) for species of Litoria and Cyclorana. However, the notable expansion of the undulating membrane appears to be an autapomorphy of Dendrobatidae frogs. The ultrastructural data presented here do not support the proposed placement of E. trivittatus in a separate genus.  相似文献

Summary Tyrosine hydroxylase (TH) immunocytochemistry was utilized to quantify dopaminergic synapses in the inner plexiform layer of the retina of Bufo marinus. Since dopaminergic cells have bistratified dendritic arborisation in the inner plexiform layer, attention was given to the segregation of synapses between the scleral and the vitreal sublaminae. Light-microscopically, a more elaborate dendritic branching was observed in the scleral than in the vitreal sublamina. In contrast, about 55% of synapses occurred in the vitreal one fifth of the inner plexiform layer, 30% in the scleral fifth, and 15% in the intermediate laminae. Input sources and output targets showed only minor quantitative differences between sublaminae 1 and 5. TH-immunoreactive processes were found in presynaptic (62.8%) and postsynaptic (37.2%) positions. Synapses to the stained dendrites derived from bipolar (40.4%) and amacrine (59.6%) cells, whereas outputs from the TH-positive processes were directed to amacrine cells (56.8%) and to small and medium-sized dendrites (35.4%); at least some of these can be considered as ganglion cell dendrites. TH-positive profiles neither formed synapses with each other nor were presynaptic to bipolar cell terminals. Junctional appositions of the immunoreactive profiles were occasionally seen on non-stained amacrine and ganglion cell dendrites in the scleral sublamina of the inner plexiform layer and on optic axons in the optic fibre layer. Although dopaminergic cells are mainly involved in amacrine-amacrine interactions, inputs from bipolar terminals and outputs to ganglion cell dendrites were also substantial, suggestive of a role also in vertical information processing.  相似文献

Summary The presence and distribution of gastrin-, gastrin-releasing peptide-, neurotensin-and vasoactive intestinal polypeptide-like immunoreactivity in the Harderian gland ofRana esculenta were studied at different times of the annual cycle. Gastrin-releasing peptide, neurotensin and vasoactive intestinal polypeptide-like substances were found either in the glandular cells, or in the nerve fibers surrounding the glandular acini. Gastrin-like immunoreactivity was confined to the glandular cells. The immunoreactivity varied during the annual cycle, with the greatest concentration being noted during the recovery phase of glandular secretory activity.  相似文献

Zusammenfassung Mit der prolongierten Osmierung nach Friend wird an den Außengliedern der pinealen Sinneszellen von Rana temporaria eine körnige Schwärzung erzielt. Diese Partikel haben einen Durchmesser von etwa 50 Å. In Analogie zu den Befunden an den retinalen Stäbchen (Eakin und Brandenburger) wird das geschwärzte Material als das Reaktionsprodukt eines Photopigments gedeutet. Weiterhin kann man mit dieser Methode in den pinealen Stützzellen 300–2500 Å große Granula darstellen, die im Golgi-Apparat entstehen. Die für Stützzellen charakteristischen Granula ermöglichen eine sichere Erkennung kleiner Stützzellanschnitte. Obwohl der Chemismus der Friendschen Reaktion nicht bekannt ist, bewährt sich diese Fixierungsmethode bei der elektiven Darstellung bestimmter Zellstrukturen. So war es möglich, den Degenerationsablauf an den Sinneszellaußengliedern und das Verhalten von Zellfortsätzen im perivaskulären Raum präziser als bisher zu untersuchen. Mit der Acetylcholinesterase-Reaktion können in der Anurenepiphyse nur Ganglienzellen und deren Neuriten elektiv hervorgehoben werden; basaler Rezeptorenfuß und die plexiformen Zonen sind dagegen negativ.
The anuran pineal organ after prolonged osmium tetroxide fixation and in acetylcholinesterase preparations
Summary 50 Å particles are selectively demonstrated in the cytoplasmic lamellae of the outer segments of the pineal sensory cells inRana temporaria after prolonged osmium tetroxide fixation according to Friend. In analogy to the interpretations of Eakin and Brandenburger concerning the rods of the lateral eye, it is assumed that a photopigment is present in the outer segment of the anuran pineal receptors. Furthermore, 300–2500 Å granules are demonstrated in the supportive cells; these originate in the Golgi zone. The characteristic granules of supportive cells are an important feature in the demonstration of small supportive cell segments. The chemical basis of the Friend reaction is not completely understood. However, the reaction proves to be useful in the demonstration of certain cell organelles. A more precise picture of degenerating outer segments of pineal sensory cells and of the distribution of terminal processes of different pineal cell types in the perivascular spaces can be obtained by the prolonged osmium tetroxide fixation. The nerve cells of the anuran pineal organ and their axons are demonstrated selectively by the acetylcholinesterase technique. The basal feet-like processes of the receptor cells and the plexiform zones of the pineal organ are negative in acetylcholinesterase preparations.

The distribution of actin, myosin, and tubulin has been investigated in coelomic oocytes, unfertilized and fertilized eggs of Discoglossus pictus utilizing: (1) immunofluorescence; (2) electron microscopy; (3) incubation with heavy meromyosin (HMM), and (4) SDS-polyacrylamide gel electrophoresis (PAGE). In coelomic oocytes, the germinative area (GA) has long, irregular microvilli containing microfilaments. In the rest of the oocyte, the microvilli are shallow. During the transit of the oocyte in the oviduct, a dimple forms by the invagination of the GA. A palisade of microfialment bundles is present in the finger-shaped microvilli of the dimple and extends for about 10 μm in the cytoplasm. In the rest of the egg, microvilli are absent and only random filaments appear in the cortex. Following HMM incubation, the dimple microfilaments are decorated with arrowheads pointing toward the bulk of the cytoplasm. SDS-PAGE of egg extracts shows bands co-migrating with actin (43K), pyruvate kinase (57K), and phosphorylase (94K). As result fertilization, the pattern of microfilament bundles in the dimple disappers in parallel with the dimple invergination itself. Generally, the entire oocyte cortex is positive to immunofluorescent staining with anti-actin, antimyosin, and antitubulin antibodies. However, the pattern of distribution and intensity of immunofluorescent staining changes for each antiserum, during different stages. It is concluded that a contractile system is present in Discoglossus eggs, and it is particularly developed in the dimple. The dimple is probably a major compartment for the storage of unpolymerized tubulin.  相似文献

Using histological techniques and computer‐aided three‐dimensional reconstructions of histological serial sections, we studied the development of the olfactory and vomeronasal organs in the discoglossid frog Discoglossus pictus. The olfactory epithelium in larval D. pictus represents one continuous unit of tissue not divided into two separate portions. However, a small pouch of olfactory epithelium (the “ventromedial diverticulum”) is embedded into the roof of the buccal cavity, anteromedial to the internal naris. The lateral appendix is present in D. pictus through the entire larval period and disappears during the onset of metamorphosis. The disappearance of the lateral appendix at this time suggests that it is a typical larval organ related to aquatic life. The vomeronasal organ develops during hindlimb development, which is comparatively late for anurans. The development of the vomeronasal organ in D. pictus follows the same general developmental pattern recognized for neobatrachians. As with most anurans, the vomeronasal glands appear later than the vomeronasal organ. After metamorphosis, the olfactory organ of adult D. pictus is composed of a series of three interconnected chambers: the cavum principale, cavum medium, and cavum inferius. We suggest that the ventromedial diverticulum at the anterior border of the internal naris of larval D. pictus might be homologous with the ventral olfactory epithelium of bufonids and with the similar diverticulum of Alytes. Laying eggs out of water was crucial to the transition to land and has evolved repeatedly in multiple animal phyla. However, testing hypotheses about this transition has been difficult because extant species only breed in one environment. The pantless treefrog, Dendropsophus ebraccatus, makes such tests possible because they lay both aquatic and arboreal eggs. Here, we test the oviposition site choices of D. ebraccatus under conflicting risks of arboreal egg desiccation and aquatic egg predation, thereby estimating the relative importance of each selective agent on reproduction. We also measured discrimination between habitats with and without predators and development of naturally laid aquatic and arboreal eggs. Aquatic embryos in nature developed faster than arboreal embryos, implying no cost to aquatic egg laying. In choice tests, D. ebraccatus avoided habitats with fish, showing that they can detect aquatic egg predators. Most importantly, D. ebraccatus laid most eggs in the water when faced with only desiccation risk, but switched to laying eggs arboreally when desiccation risk and aquatic predators were both present. This provides the first experimental evidence to our knowledge that aquatic predation risk influences non-aquatic oviposition and strongly supports the hypothesis that it was a driver of the evolution of terrestrial reproduction.  相似文献

The fork-tongued frogs, members of the amphibian Order Anura, belong to the family Dicroglossidae and are one of the most diverse groups of Anuran frogs; however, their taxonomy and phylogeny remain controversial. In the present study, sixteen dicroglossine mitochondrial genomes representing nine dicroglossine genera and 23 other neobatrachian taxa, were used to reconstruct the phylogenetic relationships of the family Dicroglossidae using different partitioned maximum likelihood and partitioned Bayesian inference methods at both the nucleotide and amino acid levels. The sampled fork-tongued frogs form a strongly supported monophyletic group that is the sister taxon to another well-supported clade that includes representatives of the families Ranidae, Rhacophoridae, and Mantellidae. The monophyly of the subfamily Occidozyginae and Dicroglossinae was revealed with strong supports, and two major clades were supported within Dicroglossinae. The sister-group relationship between the genera Limnonectes and the tribe Paini was supported. In addition, a sister-group relationships between Fejervarya and Euphlyctis + Hoplobatrachus, between Quasipaa and Yerana, and between Feirana and Nanorana are well supported. Estimates of divergence times revealed the divergence of Dicroglossidae during the Late Upper Cretaceous to the Early Eocene, and diversification of the major dicroglossine genera from the Early Eocene to the Middle Miocene.  相似文献
