Construction and packaging of pseudotype retrovirus containing human N—ras cDNA antisense sequence and its biological effects on human hepatoma cells

N-ras is one of the transforming genes in human hepatic cancer cells.It has been found that N-ras was overexpressed at the mRNA and protein level in hepatoma cells.In order to explore the biological roles of N-ras in human hepatic carcinogenesis and the potential application in control of cancer cell growth,a preudotype retrovirus containing antisense sequence of human N-ras was constructed and packaged.A recombinant retrovirus vector containing antisense or sense sequences of N-ras cDNA was constructed by pZIP-NeoSV(X)1.The pseudotype virus was packaged ang rescued by transfection and infection in PA317 and ψ 2 helper cells.It has been demonstrated that the pseudotype retrovirus containing antisense N-ras sequence did inhibit the growth of human PLC/PRF/5 hepatoma cells accompanied with inhibition of p21 expression,while the retrovirus containing sense sequence had none.The pseudotype virus had no effect on human diploid fibroblasts.     

人黑色素瘤细胞分泌的组织纤溶酶原激活剂(t-PA)的纯化

 本文报道了培养的人黑色素瘤细胞分泌的组织纤溶酶原激活剂(t-PA)的纯化方法。Bowes株人黑色素瘤细胞的分泌产物,经CM-Sephadex C--50层析,赖氨酸-Sepharose 4B,苯甲眯-sepharose 4B亲和层析后,即可得到纯化470倍的蛋白纯品。样品经聚丙烯酰胺凝胶电泳鉴定为均一单带,测得其分子量约为72kD。纯化的t-PA与尿激酶相比较,发现前者有更高亲和纤维蛋白的能力。

一氧化氮与痛觉调制

大量证据表明一氧化氮（ＮＯ）参与外周及中枢水平的痛觉调制：在外周,ＮＯ表现为致痛和镇痛双重作用;在中枢,ＮＯ不但可以促进脊髓水平痛觉过敏的形成和发展,而且参与脑内的致痛作用。     

大肠癌中p53基因突变的研究

应用聚合酶链反应(PCR)──单链构型多态性(SSCP)结合银染法对14例大肠癌p53基因的第4、第5─6和第7外显子进行了点突变的研究,结果共检测出6例点突变,而且发现各外显子的突变频率存在差异。另外,利用购自ATCC的两个探针 (p53cDNA探针和pYNZ22探针)对大肠癌中p53基因的杂合性失去进行了研究,在14例大肠癌中共检出6例杂合性丢失。将点突变检测结果同杂合性丢失结果进行比较分析, 并着重探讨了大肠癌中p53基因失活导致肿瘤的作用方式。 Abstract:The exons 4-7 of p53 gene were examined in 14 colorectal Cancer patients by using PCR-SSCP-silver staining method.The results showed 6 cases of point mutation and the mutation frequencies of exons were different from each other.p53 cDNA and pYNZ22 VNTR were used as probes to examine LOH(Loss of heterozygosity)of 14 colorectal cancers.6 cases with LOH were found.The results of present research suggest that mutation and LOH of p53 gene are critical events in the progress and development of Cancer.There were different kinds of inactivation model of p53 gene in the process of development of cancer and transformation of cells.     

小麦远缘杂种幼胚胚状体的发生,发育及低温对愈伤组织分化能力…

通过普通小麦与滨麦、簇毛麦及山羊草属７个种杂交幼胚培养,获得８个体细胞胚性无性系（８个组合１０个胚）及大量试管苗。愈伤组织及胚性愈伤组织诱导率分别为６５．７９％和２６．３２％。形态学及细胞学鉴定结果,均为真杂种。不同染色体组及同一染色体组不同基因型的幼胚,在组织培养中有明显差异。非部体细胞具有遗传的全能性,但当染色体数目严重偏离双单倍体数目的,其全能性即丧失。胚状体的发育具有与合子胚极相似的典型结

乙烯消减盐渍胁迫对苜蓿种子萌发的抑制作用

在无盐条件下,外源乙烯对苜蓿种子萌发有促进作用,但对最终发芽率无影响。盐渍严重抑制苜蓿种子萌发,加入１～５０μｌ／Ｌ（ｖ／ｖ）外源乙烯或０．１～５．０ｍｍｏｌ／Ｌ１－氨基环丙烷－１－羧酸（ＡＣＣ）或５～１００ｍｇ／Ｌ（ｗ／ｖ）乙烯利（ＥＴＨ）均能极显著地减轻ＮａＣｌ对苜蓿种子萌发的抑制作用。激动素（ＫＴ）也有类似作用,并能促进萌发种子的乙烯产生,它与ＡＣＣ一起使用,则对种子萌发和乙烯产生均显示加成作用。在ＮａＣｌ胁迫下,应用乙烯和乙烯利虽有利萌发,但幼芽鲜重和下胚轴长度明显低于无盐对照。
