Angiogenesis is essential for transplantation of mesenchymal stem cells (MSCs). Vascular endothelial growth factor (VEGF) is one of the most potent angiogenic factors identified to date. Elevated VEGF levels in MSCs correlate with the potential of MSCs transplantation. As an indirect angiogenic agent, transforming growth factor-β1 (TGF-β1) plays a pivotal role in the regulation of vasculogenesis and angiogenesis. However, the effect of TGF-β1 on VEGF synthesis in MSCs is still unknown. Besides, the intracellular signaling mechanism by which TGF-β1 stimulates this process remains poorly understood. In this article, we demonstrated that exposure of MSCs to TGF-β1 stimulated the synthesis of VEGF. Meanwhile, TGF-β1 stimulated the phosphorylation of Akt and extracellular signal-regulated kinase 1/2 (ERK1/2). Moreover, Ly 294002, a specific inhibitor of phosphatidylinositol-3-kinase (PI3K)/Akt significantly attenuated the VEGF synthesis stimulated by TGF-β1. Additionally, U0126, a specific inhibitor of ERK1/2, also significantly attenuated the TGF-β1-stimulated VEGF synthesis. These results indicated that TGF-β1 enhanced VEGF synthesis in MSCs, and the Akt and ERK1/2 activation were involved in this process.  相似文献

As the frequent emergence of the resistant bacteria, the development of new agents with a new action mode attracts a great deal of interest. It is now widely accepted that antimicrobial peptides (AMPs) are promising alternatives to conventional antibiotics. In this study, antimicrobial peptide polybia-MPI and its analogs were synthesized and their antibacterial activity was studied. Our results revealed that polybia-MPI has potent antibacterial activity against both Gram-positive and Gram-negative bacteria. Its ability to make PI permeate into bacteria and lead to the leakage of calcein from model membrane LUVs, suggests a killing mechanism involving membrane perturbation. SEM and TEM microscopy experiments verified that the morphology of bacteria was changed greatly under the treatment of polybia-MPI. Compared with the conventional chemotherapy, polybia-MPI targets the cell membrane rather than entering into the cell to exert its antibacterial activity. Furthermore, molecular dynamics (MD) simulations were employed to investigate the mechanism of membrane perturbation. The results indicated that the α-helical conformation in the membrane is required for the exhibition of antibacterial activity and the membrane disturbance by polybia-MPI is a cooperative process. In conclusion, with the increasing resistance to conventional antibiotics, there is no doubt that polybia-MPI could offer a new strategy to defend the resistant bacteria.  相似文献

Sinowilsonia henryi Hemsi., the only representative of the monotypic genus Sinowilsonia Hemsi. (Hamamelidaceae), is a threatened plant endemic to China with high phylogenetical, ecological and economical values. In the present study, inter-simple sequence repeat (ISSR) markers were employed to investigate the genetic diversity and differentiation of 214 individuals sampled from 14 populations. Fifteen selected primers yielded a total of 178 bright and discernible bands. The genetic diversity was low at the population level (h = 0.1025; I = 0.1506; PPL = 26.7%), but quite high at the species level (h = 0.2449; I = 0.3690; PPL = 72.5%). In line with the limited gene flow (Nm = 0.3537), the hierarchical analysis of molecular variance (AMOVA) revealed pronounced genetic differentiation among populations (ΦST = 0.6639). Furthermore, the Mantel test revealed a significant correlation between genetic and geographic distances among populations (r = 0.688, P = 0.001), indicating the role of geographic isolation in shaping its present population genetic structure. The present patterns of genetic diversity of S. henryi were assumed to result largely from its evolutionary history and geographic factors. Based on these findings, conservation strategies were proposed to preserve this threatened plant.  相似文献

栖息地退化会使环境中蜥蜴的可利用食物资源发生改变。采用剖胃法对内蒙古自治区鄂尔多斯市准格尔旗和达拉特旗地区采集的草原沙蜥样本进行了食性分析。对固定沙丘、半固定沙丘和流动沙丘三类样地中77个实胃样本的652只(套)食物进行鉴定和分析的结果表明,这些食物主要为节肢动物,分为35类,隶属3纲11目32科;也在少部分个体(4.69%)的胃容物中发现少量(1.95%)植物碎片。上述三类栖息地中,草原沙蜥对蚁科、蚜科、瓢虫科、叶蝉科和茧蜂科昆虫均有较大的捕食比例;但相比固定沙区栖息地,草原沙蜥在半固定及流动沙丘中增减的食物种类均比较多,导致草原沙蜥在流动沙区与其他两类样地之间有较大的食物相异性。从固定沙丘、半固定沙丘到流动沙丘,随栖息地荒漠化程度的增加,草原沙蜥捕食食物类群的丰富度逐步减少,辛普森优势度指数相应地下降,但营养生态位宽度、香农威纳多样性指数和皮洛均匀性指数却在流动沙丘栖息地上明显增大。这表明鄂尔多斯地区的草原沙蜥是以捕食昆虫等节肢动物为主、兼食极少量植物的杂食性动物。栖息地荒漠化所致的环境变化显著地影响草原沙蜥的食物组成,使其觅食的食物种类逐步减少。  相似文献

以陕南地区的3种主栽柑橘离体叶片为材料,分别以5、0、-5和-10℃进行低温处理,分析与耐寒性相关的生理响应,并采用隶属函数法分析3种柑橘的抗寒能力以及通过主成分分析评价各个测定指标的贡献率。结果表明:随着处理温度的降低,3种柑橘离体叶片均依次出现叶片卷曲、叶片细胞内的水分外渗,细胞内结冰导致叶片僵硬和最终的叶片内外结冰情况,叶片形态变化结果显示3种柑橘抗寒能力强弱依次为日南1号兴津泛亚特早。随处理温度的降低,3种柑橘离体叶片的相对电导率(REC)和丙二醛(MDA)均从0℃处理时呈显著升高趋势,-10℃处理时达到最高值,同一低温处理条件下REC和MDA值的大小顺序均为泛亚特早兴津日南1号。O_2~-·、H_2O_2和可溶性糖含量总体保持升高的趋势,而脯氨酸含量呈先显著升高后降低的趋势。抗氧化酶SOD、CAT和APX的活性及抗氧化物质ASC含量均呈现先升高后下降的趋势。REC和MDA综合指标的累积贡献率高于85%,具有较强的代表性,可作为柑橘抗寒性分析的重要指标,平均隶属度分析结果表明3种柑橘抗寒能力依次为日南1号兴津泛亚特早。  相似文献

野生朱鹮的种群数量和分布现状   总被引:2,自引:0,他引:2  
2012年9-10月,我们对野生朱鹮(Nipponia nippon)的分布区和潜在分布区进行了调查,共发现其游荡期夜宿地23个,其中20个有朱鹮夜宿,分布在洋县(16个)、宁陕县(3个)和城固县(1个)。对这些夜宿地进行了的同步调查,共统计到野生朱鹮1 090只,其中97.2%分布在洋县境内。最大的夜宿集群数量为184只,集群数量超过40只的夜宿地共11个,累计停歇的朱鹮占总数的91.7%,表明野生朱鹮在游荡期有趋于集结较大群体夜宿的习性。朱鹮野生种群中当年出生的幼鸟占19.0%,据此估算,截至秋季朱鹮幼鸟的存活率约为67.2%。加强对保护区以外,尤其是野生朱鹮新扩散地区的的保护管理,将促进野生朱鹮种群的扩散,进一步增加这一濒危物种抵御风险的能力。  相似文献

A method for the determination of the proportions of major fiber-forming collagens (types I, III and V) in soft connective tissue was elaborated. The method is based on the release of insoluble collagen by CNBr with subsequent separation of the arising peptides. For routine application the peptides are separated by capillary electrophoresis (50 mM phosphate pH 2.5, 15 kV, 50°C, 70/60 cm×70 μm I.D. capillary with UV detection at 200 nm). Quantitation of collagen type I can be done either on the basis of spiking the sample with a peptide mixture obtained from a known amount of collagen type I, or by spiking the sample with an equimolar mixture of the two peptides [α1(I)CB2 and α1(I)CB4] (constituting a fused peak) along with α1(III)CB2 and α1(V)CB1. Compared to the previously published methods the procedure is faster and does not require isolation of marker peptides by tedious chromatographic procedures in a preceding preparatory step. Good results are obtained within a wide range of run buffer concentrations and applied voltages; conversely, intensive cleaning of the capillary after every three runs is recommended with a new capillary after 20–30 runs.  相似文献

本研究从陕西留坝紫柏山采集的七叶树Aesculus chinensis根和茎中分离出一株具有高产七叶皂苷C的内生真菌,将其编号为EA-LJS80。利用ITS序列分析鉴定其种属并对其形态进行观察;采用紫外-可见分光光度计进行全波长扫描,确定其最大紫外吸收峰;采用HPLC测定七叶皂苷C的产量;采用滤纸片扩散法研究其发酵液提取物的抑菌活性;采用CCK-8法测定提纯后七叶皂苷对肺腺癌A549细胞的增殖抑制率,探究其生物活性。结果显示,该菌株EA-LJS80为曲霉菌属Aspergillus真菌,最大紫外吸收波长为230nm,测得提纯后七叶皂苷C的产量为9.23mg/mL。生物活性实验表明:菌株EA-LJS80的发酵液提取物对大肠杆菌Escherichia coli等4种致病菌有明显的抑菌效果;经CCK-8法测定,提纯后的七叶皂苷对肺腺癌A549细胞的增殖有明显的抑制作用。综上所述,菌株EA-LJS80具有产七叶皂苷活性,尤其是七叶皂苷C的产量较高,且其产生的七叶皂苷具有较高的生物活性,这对于改进七叶皂苷生产模式,提高生产效率具有重要意义,同时对于七叶皂苷C的进一步研究有一定的促进作用。  相似文献

The effect of lanthanum on the metabolism of ascorbate (AsA) and glutathione (GSH) in the leaves of maize seedlings under cadmium stress was investigated. The findings showed that Cd remarkably increased electrolyte leakage (EL), the activities of ascorbate peroxidase, dehydroascorbate reductase, monodehydroascorbate reductase (MDHAR), glutathione reductase, L-galactono-1,4-lactone dehydrogenase, and γ-glutamylcysteine synthetase, and the content of reduced AsA, reduced GSH, total AsA, total GSH, malondialdehyde (MDA), and Cd, compared with control. However, Cd significantly decreased the dry biomass of roots and shoots. Treatment with La + Cd evidently increased the activities of above enzymes except MDHAR, the content of reduced AsA, reduced GSH, total AsA and total GSH, and the dry biomass of roots and shoots, compared with Cd stress alone. Meanwhile, treatment with La + Cd remarkably decreased EL and the content of Cd and MDA compared with Cd stress alone. Our results suggested that La could be used as a regulator to improve the Cd tolerance of maize for its role in the alleviation of Cd-induced oxidative damage by regulating the metabolism of AsA and GSH.  相似文献
